Structural and Electrical Properties of Nickel Hydroxide Electrode Prepared by Hydrothermal Synthesis on Nickel Foam (니켈 폼(Ni foam)에 수열 합성법으로 제조한 수산화니켈(Ni(OH)2) 전극의 구조적 및 전기적 특성)

  • In this study, the nickel hydroxide (Ni(OH)2) electrode for supercapacitor was prepared via hydrothermal method. Based on the nickel (Ni) foam, the electrode does not require any additional binder material or post-processing. Nickel nitrate (Ni(NO3)2) and hexamethylenetetramine (C6H12N4) were used for synthesis, and the synthesis condition was 12 hours at 80 ℃. X-ray diffraction (XRD) and field-emission scanning electron microscopy (FE-SEM) were used to analyze the structural characteristics of the electrode, and it shown that the nickel hydroxide was successfully prepared after only the one-step hydrothermal synthesis. The electrochemical properties were analyzed through the half-cell test. The prepared electrode shown a pair of oxidation/reduction peaks, indicating that the driving method included the redox reaction on the electrode surface. After the charge/discharge test, the specific capacitance was calculated as the value of 438 F/g at 3 A/g.

  • Some ionic liquids are suitable for catalysts and solvents which are applicable to $CO_2$ fixation reaction converting $CO_2$ to carbonate. Using the ionic liquids, the synthesis process will become greener and simpler because of easy catalyst recycling and unnecessary use of volatile and harmful organic solvents. In this work, the synthesis of propylene carbonate from propylene oxide using carbon dioxide and ionic liquids were measured at high pressures up to ~140 bar and at temperatures between $60^{\circ}C$ and $80^{\circ}C$. As a results, we found the optimum condition and obtained the maximum yield under that condition.

X-Ray Scattering Studies on Molecular Structures of Star and Dendritic Polymers

  • We studied the molecular shapes and structural characteristics of a 33-armed, star polystyrene (PS-33A) and two $3^{rd}$-generation, dendrimer-like, star-branched poly(methyl methacrylate)s with different architectures (pMMA-G3a and PMMA-3Gb) and 32 end-branches under good solvent and theta ($\Theta$) solvent conditions by using synchrotron small angle X-ray scattering (SAXS). The SAXS analyses were used to determine the structural details of the star PS and dendrimer-like, star-branched PMMA polymers. PS-33A had a fuzzy-spherical shape, whereas PMMA-G3a and PMMA-G3b had fuzzy-ellipsoidal shapes of similar size, despite their different chemical architectures. The star PS polymer's arms were more extended than those of linear polystyrene. Furthermore, the branches of the dendrimer-like, star-branched polymers were more extended than those of the star PS polymer, despite having almost the same number of branches as PS-33A. The differences between the internal chain structures of these materials was attributed to their different chemical architectures.

An Assay Method for Screening Inhibitors of Prolyl 4-hydroxylase in Immortalized Rat Hepatic Stellate HSC-T6 Cells

  • Hydroxyproline (HYP) is a post-translational product of proline hydroxylation catalyzed by an enzyme prolyl 4-hydroxylase (P4H) which plays a crucial role in the synthesis of all collagens. Considering the role of collagen and its significance in many clinically important diseases such as liver fibrosis, a great deal of attention has been directed toward the development of an assay at cell-based system. The reason is that cell-based assay system is more efficient than enzyme-based in vitro system and takes much less time than in vivo system. Several assay procedures developed for P4H are laborious, time-consuming and not feasible for the massive-screening. Here, we report the cell-based assay method of prolyl 4-hydroxylase in immortalized rat hepatic stellate HSC-T6 cells. To optimize the cell culture condition to assay for HYP content, various concentrations of reagents were treated for different times in HSC-T6 cells. Our data showed that the treatment with ascorbate in a hypoxic condition for 24 h resulted in the maximal increase of HYP by 1.8 fold. Alternatively, cobalt chloride ($5\;{\mu}M$) and ascorbate ($50\;{\mu}M$) in normoxic states exhibited similar effect on the production of HYP as in a hypoxic condition. Therefore, cobalt chloride can be substituted for a hypoxic condition when an anaerobic chamber is not available. Rosiglitazone and HOE077, known as inhibitors of collagen, synthesis decreased P4H enzyme activity by 32.3% and 15%, respectively, which coincided with previous reports from liver tissues. The level of the smooth muscle ${\alpha}$-actin, a marker of activated stellate cells, was significantly increased under hypoxia, suggesting that our experimental condition could work for screening the anti-fibrotic compounds. The assay procedure took only 3 days after treatment with agents, while assays from the primary stellate cells or liver tissues have taken several weeks. Considering the time and expenses, this assay method could be useful to screen the compounds for the inhibitor of prolyl 4-hydroxylase.

Hypoxic condition enhances chondrogenesis in synovium-derived mesenchymal stem cells

  • Background: The chondrogenic differentiation of mesenchymal stem cells (MSCs) is regulated by many factors, including oxygen tensions, growth factors, and cytokines. Evidences have suggested that low oxygen tension seems to be an important regulatory factor in the proliferation and chondrogenic differentiation in various MSCs. Recent studies report that synovium-derived mesenchymal stem cells (SDSCs) are a potential source of stem cells for the repair of articular cartilage defects. But, the effect of low oxygen tension on the proliferation and chondrogenic differentiation in SDSCs has not characterized. In this study, we investigated the effects of hypoxia on proliferation and chondrogenesis in SDSCs. Method: SDSCs were isolated from patients with osteoarthritis at total knee replacement. To determine the effect of oxygen tension on proliferation and colony-forming characteristics of SDSCs, A colony-forming unit (CFU) assay and cell counting-based proliferation assay were performed under normoxic (21% oxygen) or hypoxic (5% oxygen). For in vitro chondrogenic differentiation, SDSCs were concentrated to form pellets and subjected to conditions appropriate for chondrogenic differentiation under normoxia and hypoxia, followed by the analysis for the expression of genes and proteins of chondrogenesis. qRT-PCR, histological assay, and glycosoaminoglycan assays were determined to assess chondrogenesis. Results: Low oxygen condition significantly increased proliferation and colony-forming characteristics of SDSCs compared to that of SDSCs under normoxic culture. Similar pellet size and weight were found for chondrogensis period under hypoxia and normoxia condition. The mRNA expression of types II collagen, aggrecan, and the transcription factor SOX9 was increased under hypoxia condition. Histological sections stained with Safranin-O demonstrated that hypoxic conditions had increased proteoglycan synthesis. Immunohistochemistry for types II collagen demonstrated that hypoxic culture of SDSCs increased type II collagen expression. In addition, GAG deposition was significantly higher in hypoxia compared with normoxia at 21 days of differentiation. Conclusion: These findings show that hypoxia condition has an important role in regulating the synthesis ECM matrix by SDSCs as they undergo chondrogenesis. This has important implications for cartilage tissue engineering applications of SDSCs.

  • A novel and convenient route for the synthesis of a series of thalidomide derivatives is described. Compound 2 was cyclized with different amines under alkaline condition to obtain 4-nitro substituted phthalimidines 3a-d. Hydroxylactams 4a-d were produced via bromination and hydroxylation. Different acyl chlorides were reacted with hydroxylactams to provide the desired esters 5a-d. All compounds were evaluated by MTT assay for their inhibitory activities against HCT-116, MG-63, MCF-7, HUVEC and HMVEC cell lines in vitro. Most of them showed no obvious cytotoxic effect on normal human cells, compounds 4a-d, $5a_2$, $5a_4$, $5a_5$, $5b_2$, $5c_2$ and $5d_2$ exhibited potent antitumor activities, among which compounds $5a_2$ and $5b_2$ were more effective than 5-FU.

  • The synthesis of high purity and ultra-fine $BaTiO_3$ by precipitation with gaseous $NH_3$ as precipitator was investigated to find an alternative process to solve various problems of recent wet methods. A synthesis process for $BaTiO_3$ powder using $NH_3$ gas as a precipitator in a bubble column reactor was experimentally successful in developing the production process of piezoelectric ceramic $BaTiO_3$ powder. And a 2.33m1/sec is approprite for the feed flow rate, $BaTiO_3$ powder produced under above the condition is spherical type, its particle size was about $0.2{\mu}m$.

  • The application of microwave technology to synthesis of polymer in solvent media has been shown by the synthesis of copper phthalocyanine (CUPc). The increase of synthetic yield-was demonstrated successfully in this study. A power variable microwave synthetic system has been developed with modifying cavity of domestic microwave oven and attaching microwave power controller. The properties of the specimen synthesized at various condition under the conventional thermal processing and microwave processing has been characterized by the means of chemical analysis, X-ray diffractometry (XRD), scanning electron microscopy (SEM), and particle size analysis (PSA).

  • (Ti.W)C complex carbide was synthesized by self-propagating high temperature synthesis (SHS) chemical furnace. Attempt to find the optimal condition for synthesis of (Ti.W)C the effects of molar ratio of Ti:W:C on the synthesized powders and mechanical properties were investigated, Optimum molar ratio of these synthe-sized powder was Ti:W:C=0.7:0.2:1.0 The bulk density M,O.R Hardness Fracture toughness of (Ti.W)C complex carbide sintered at 200$0^{\circ}C$ for 60 min by hot-pressing under the pressure of 20 MPa were 7.6g/cm3, 475 MPa, 17,.7 GPa respectively.

Heat-Shocked Drosophila Kc Cells Have Differential Sensitivity to Translation Inhibitors

  • The heat shock response is a universal stress response observed in all organisms and cultured cells. The response is regulated at both the transcriptional and translational level. Heat shocked Drosophila melanogaster Kc cells are used as the system for the study of translational regulation. In this system non-heat shock messages are associated with polysome but are not translated in a heat shocked condition. To figure out the change in the translation machinery. the effects of translation elongation inhibitors were tested on Kc cells. The result showed that the sensitivity of translation to these drugs changed in heat shocked cells. The significant changes were the decreased inhibition of heat shock protein synthesis by cycloheximide, emetine. and puromycin. and the increased inhibition of heat shock protein synthesis by verrucarin A. implying that the translation elongation mechanism in heat shocked cells changed.
