Studies on Properties of Avian Reovirus Isolated in Korea (국내에서 분리한 닭 레오바이러스의 성상에 관한 연구)

  • Avian reoviruses have been implicated in respiratory disease enteric conditions including Cloacal pasting in young thicks, pericarditis, hydropericardium, anaemia with swollen spleen and liver and petechiation of skeletal muscle and viral arthritis. This study was conducted to examine properties of reovirus field 3 strains isolated from affected broiler from several farms. An infectious agent was isolated from leg tendons and intestine of broiler with clinical tenosynovitis. The agent grew well on the chicken embryo kideny cells(CEK). One of them produced cytopathic effects(CPE) of round type and formed intranuclear inclusions, and the other was characterized by CPE of syncytical type and cytoplasmic inclusion. The properties and serological classification of field strains were examined by hemagglutin test, virus neutralization test, agar gel precipitin, electropherotype. They showed no hemagglutination reactions and not well neutralization and to possess common antigens detectable by AGP test. RNA electropherotype presented 10 segment band as the previous report. These data suggest that the field strains and standard strains (1133, 1733) may be the same group.

A Ultrastructural Study on the Axoneme Formation in the Spermatozoa of the Edible Giant Snail, Achatina fulica (식용 왕달팽이 (Achatina fulica) 정자의 축사형성 (Axoneme formation)에 관한 미세구조)

  • The spermatogenetic process in the edible giant snail is similar to those in the other snails, except for the axoneme formation process. In this study, the axoneme formation process in the giant snail was mainly examined by means of electron microscopy. The tail portion of a spermatozoon is about $160{\mu}m$ long, and extends straight to the rear, surrounded by two large and long mitochondria in spiral forms. A number of glycogen particles $(40\sim70nm)$ are found in the swollen matrix of the mitochodria. The axoneme which composes the tail of a spermatozoon is surrounded by $7\sim10$ lamella-form fibrous sheaths of about $0.2{\mu}m$ in thickness. Most of the mature spermatozoa are found to be clustered into a group of $5\sim7$ ea in syncytial bridges formed by cytoplasmic processes. Sertoli cells contain glycogen particles, endoplasmic reticulum, a lot of mitochondria, and lipids in their cytoplasm. They protrude their filiform pseudopodia and phagocytize abnormal spermatids or spermaozoa.

Cytologic Aspect of Fibrous Osteodystrophy in a Juvenile Siberian Husky (어린 Siberian Husky 견의 섬유성 골형성장애의 세포학적 고찰 증례)

  • A 5-month-old intact female Siberian Husky dog was presented for evaluation of severely enlarged maxilla. Abnormalities in CBC, serum chemistry and urinalysis indicated purulent inflammation and renal failure. Cytologic examination of the swollen maxilla showed a mixed population of multinucleated giant cells and round to polygonal to spindle shaped cells either individualized or aggregated. Both type of cells showed moderate anisokaryosis, and anisocytosis, prominent nucleolus or multiple nucleoli, and coarse chromatin. On histopathology maxilla and turbinate were diffusely expanded and replaced by variably dense fibrous connective tissue, and the kidneys showed changes consistent with renal dysplasia. Based on these findings, the diagnosis of fibrous osteodystrophy due to renal dysplasia and fibrosis was made. Despite the supportive care, the dog continued to decline and was euthanized.

Electron Microscopic Study on the Parafollicular Cells of the Thyroid Gland of the Head and Neck-Irradiated Rats (방사선이 흰쥐 갑상샘 소포곁세포에 미치는 영향에 대한 미세구조적 연구)

  • This experiment was performed to study the morphological responses of the parafollicular cells of rat following X-ray irradiation. Male rats were divided into normal and experimental groups. The head and neck region of the rat, under sodium thiopental anesthesia, was exposed to 3,000 rads or 6,000 rads of radiation in a single dose, respectively. The source was a Mitsubishi Linear Accelerator ML-4MV. The target to skin distance was 80 cm, and the dose rate was 200 rads/min. The rate of experimental groups were sacrificed on the 6th hour, 2nd and 6th day after X-ray irradiation. Pieces of the tissue taken from the thyroid gland were fixed in 2.5% glutaraldehyde-1.5% paraformaldehyde (0.1M Millonig's phosphate buffer, pH 7.3), and in 1% osmium tetroxide (0.1M Millonig's phosphate buffer, pH 7.3), and embedded in araldite mixture. The ultrathin sections stained with uranyl acetate and lead citrate were observed with JEM 100 CX-II electron microscope. The results were as follow; 1. Two types of the parafollicular cells, according to their electron densities, were found, i. e., light cells and dark cells. 2. Three types of the parafollicular cells, according to their sizes of secretory granules were found, i.e., small granule cells ($85nm{\pm}20.1;64{\sim}102nm$), medium granule cells ($120nm{\pm}26.5;77{\sim}179nm$), and large granule cells ($165nm{\pm}25.7;128{\sim}189nm$). 3. The differential ultrastructural changes of the cells according to their cell types, i.e., dark and light cell, or small, medium and large granule cells, were hardly observed in the time and dose range covered by this study. 4. The morphological changes of the parafollicular cells were not pronounced after exposure to 3,000 rads of X-ray. 5. Swollen cisternae of the granular endoplasmic reticulum and partial cytolysis were observed after exposure to 6,000 rads of X-ray. 6. Above results suggest that the parafollicular cells showed the alterations of mitochondrial and granular endoplasmic reticular swelling, and partial cytolysis, but only in doses of 6,000 rads.

Changes in Blood Cell Morphology and Number of Red Spotted Grouper, Epinephelus akaara in Response to Thermal Stress

  • Rising of water temperature due to global warming is a great concern to aquaculturists and fishery biologists. Hence, the present study aimed to investigate the effects of high water temperature on juvenile red spotted grouper, Epinephelus akaara based on the evaluation of stress responses in blood. E. akaara juveniles were exposed to different thermal conditions ($25^{\circ}C$, $28^{\circ}C$, $31^{\circ}C$, and $34^{\circ}C$) for 6 weeks following 2 weeks of acclimation at $25^{\circ}C$. Blood cell morphology and number were examined at three sampling points (2, 7, and 42 days) from a total of 180 fish. Major erythrocytic cellular abnormalities (ECA) observed in blood smears of thermally stressed groups ($31^{\circ}C$ and $34^{\circ}C$) after 6 weeks were echinocytes, teardrop-like cells, swollen cells and vacuolated cells. Both red and white blood cell number (RBC and WBC) were significantly (p<0.05) elevated in $31^{\circ}C$ and $34^{\circ}C$ group after 6 weeks thermal exposure. Differential leucocytes number showed significant increases in neutrophil (N) and decreases in lymphocytes (L) in the highest temperature ($34^{\circ}C$). Different N:L ratio was observed at different thermal conditions which can be used as a reliable alternative to measure stress response. Taken together, these results suggest that higher temperature ($31^{\circ}C$ and $34^{\circ}C$) can interfere the immune system of red spotted grouper by altering the blood cell morphology and number.

Three-dimensional analysis of the arrangement of microtubules of the outer segment in the ciliary-type photoreceptor cell in the Onchidium dorsal eye

  • The inverted retina of the Onchidium dorsal eye (DE) is composed only of ciliary-type photoreceptor cells (CC's). The outer segment (OS) of the CC is a concentric lamellar structure consisting of many modified ciliary membranes and stains positively with anti-$\beta$-tubulin antibody. Near the base of the OS there are about 30 basal bodies each connecting individually to a cilium. The cilia are rod-shaped at the base, progressing upwards to a flattened sheet-like shape with increasing surface area. Three-dimensional analysis on serial sections demonstrates the ladle-shape of a modified cilium. Many modified cilia wrap around each other like the leaves of a cabbage. Nine pairs of microtubules (MT's) are located regularly in a ring at the base of the cilium, gradually losing their regular arrangement towards the periphery, where they separate into two subgroups that are contained within two swollen portions of a modified cilium. Within the CC of the Onchidium DE, MT's in the modified cilium exist as two poles extending longitudinally in a thin expanded ciliary membrane. This arrangement may support the photoreceptive OS and serve to maintain its structural integrity.

Neomycin Sulfate Nephrotoxicosis in Korean Black Goats (한국흑염소에 있어서의 Neomycin Sulfate에 의한 신증유발에 관하여)

  • In order to study the effects of neomycin sulfate on the kidney of Korean black goats after dosing at 36mg/kg(A group) or 18mg/kg of body weight (B group) twice a day, serum chemical values(BUN, SCr), urinary enzyme(GGT) excretion and urinary analysis were checked and renal lesions were observed by light microscope. The proteinuria and granular casts were observed on the 5-6th day and 7th day of treatment with neomycin respectively in the A group. While these changes were observed on the 4th day and the 6th day respectively in one goat of the B group. Urinary GGT excretion began to increase from the 9th day and reached to maximum concentration on the 15th day of treatment with neomycin in the A group. Thereafter, GGT excretion began to decrease and reach to normal level on the 21th day. But GGT excretion increased on the 9th day in only one goat of the B group. Concentrations of BUN and SCr increased on the 12th day in the A group and in one goat of B group. The kidneys were swollen gross pathologically and the renal tubular epithelial cell changes were noticed histologically in the A group and in one goat of the B group. These changes were included degeneration with hyaline droplet formation, flattened epithelial cells and necrotic epithelium. Granular casts were showed in many tubular lumens. Prfesent results indicated that nephrotoxicosis would not occured in Korean black goats dosing with neomycin sulfate less that 36mg/kg of body weight a day.

PEG Distribution in the Cell Walls of PEG-treated Woods using SEM-EDS (SEM-EDS를 이용한 PEG처리재 세포벽내의 PEG분포 분석)

  • This study was carried out to investigate the distribution and penetration of PEG molecules into the cell wall of wood by SEM-EDS method. SEM observation indicated that the cell walls of PEG-treated specimen were swollen, and didn't show any change in their shapes, while the cells of untreated specimen were more or less collapsed. The results of X-ray analyses of potassium stained samples revealed the distribution of the impregnated chemicals within the cell walls, when considering the distribution of potassium ions in the cell walls. Consequently, this study supported the possibility of PEG molecules penetrated into the wood cell wall play an important role in enhancing the dimensional stability of wood.

Effects of Myocardial Protection of Verapamil in Serum: Under Langendorff Apparatus in Ischemic Arrest Heart (Verapamil [anticalcium agent]의 심근 보호작용Langendorff씨 장치하의 심근 보호 작용)

  • This study was evaluated the metabolic, physiologic and histologic effects of myocardial protection of verapamil[isoveratril]on isolated rat hearts to 90 minutes of ischemic arrest. Heart was perfused with a modified Kreb’s Henseleit bicarbonate buffer with glucose and arrested with retrograde coronary perfusion by glucose insulin[GI], potassium and verapamil. Mean aortic systolic pressure, heart rate, coronary flows were measured and morphologic changes were examined during working heart perfusion. Perfusion and arrest were controlled four groups subjected 60 isolated rat hearts. Four groups hearts reperfused during 40 minutes after 90 minutes global ischemia for physiologic recovery. 15 hearts of four groups were assayed to histological morphologic changes. GI treated hearts recovered less than 28% of function and changed more than 80% of mitochondria of control group. Verapamil hearts[0.2, 0.1 gm/kg] recovered more than 88% of function and permitted the maintenance of continuous cellular level of Serum Glutamic Oxalaxetate Transaminase[SGOT], but declined 28% of Phosphate Kinase[CP], GI treated heart showed widespread evidence of extensive damage of mitochondria. The damage was that interstitial huge edema are present and there was contraction band formation within the swollen cells. The verapamil and potassium group were not found morphologic change compared with control group. Their functions were shown that metabolic and physiologic action of verapamil-group lasted 20 minutes longer than potassium group.

Effects of Zinc and Aluminum Hot-dip Galvanized Sheet Steel on the Gill and Hepatopancreas of the Abalone Haliotis discus hannai (아연 및 알루미늄 용융도금 처리된 강판이 북방전복(Haliotis discus hannai)의 아가미와 간췌장에 미치는 영향)

  • We investigated the toxicity of zinc and aluminum hot-dip galvanized sheet steel to abalone Haliotis discus hannai via changes in the gill and hepatopancreas using histological and transmission electron microscopy analysis. Experimental groups were composed of one control and four exposure conditions (direct or indirect exposure to zinc and aluminum hot-dip galvanized sheet steel). In the control group, aluminum exposure groups (direct and indirect), and indirect zinc exposure group, abalone mortality was not observed until the end of the experiment, and no histopathological changes were observed in the gill and hepatopancreas. However, the direct zinc exposure group exhibited 100% mortality. Ultrastructural analysis of the cytoplasm of ciliated and microvilli-bearing epithelial cells from gill filaments revealed electron-dense vesicles near the cell membrane and disruption of the nuclear membrane. We also observed swollen mitochondria and a loss of mitochondrial cristae. The hepatopancreas showed similar changes, and we detected highly electron-dense particles within the vesicles. These results suggest that abalone exposed directly to zinc hot-dip galvanized sheet steel experience acute toxicity, causing damage to cell organelles in the gill and hepatopancreas and, finally, inducing mortality.
