훈련방법의 차이가 SOD, Neutrophils 및 T세포에 미치는 영향 (The Effect of Different Type of Exercise on SOD, Neutrophils and T Lymphocytes)

  • Background: A physically active lifestyle and regular exercise training incurs many health benefits. One recently recognized benefit of regular moderate exercise is stress reduction and immune enhancement. Thus, a physical stress such as exercise may act at any number of points in the complex sequence of events collectively termed the immune response. Although exercise causes many propound changes in parameters of immune function, the nature and magnitude of such changes rely on several factors including the immune parameters of interest; type, intensity, and duration of exercise; fitness level or exercise history of the subject; environmental factors such as ambient temperature and humidity. Methods: This study was undertaken to investigate the effect of different type of exercise on superoxide dismutase (SOD), neutrophils, and T lymphocytes of Sprague-Dawley rats. Sprague-Dawley rats were randomly divided into three groups; a non-Trained group (NTG, n=6), a swim-Trained group (STG), and a treadmill-Trained group (TTG). The exercise regimen was designed in a treadmill (5 times/5 days/week) during 8-weeks for TTG, and swim training (5 times/5 days/week) during 8-weeks for STG, and the volume of exercise training was the same in both groups. Results: 8 weeks of regular swim and treadmill training significantly increased liver SOD concentration however, muscle SOD concentration was not statistically significant. In the level of neutrophils, TTG and STG showed significant difference, compared to NTG. TTG was the highest level of neutrophils. In the level of immune cell counts, there was significant difference among TTG, STG, and NTG both in the spleen and thymus. Conculsion: In conclusion, it can be stated that eight weeks swim and treadmill exercise training has beneficial effect in improving immune response and antioxidant defence capacity by augmenting immune cells and SOD activities of SD rats.

  • This study was designed to investigate the changes in energy substrates, glucose and non-esterified fatty acid(NEFA), and fatty acid compositions in serum, following physiolgical stress in rats fed diets containing various fatty acids. Forty two Sprague-Dawley strain male rats, weighing 108$\pm$2.1g, were fed 3 different experimental diets for 4 weeks. The diets were composed of 105 fat(w/w) of either corn oil(CO;18:2 n6:57%), plant perilla oil(PO;18:3 n3:59%), or tuna fish oil(FO;20:5 n3:17%%, 22:6 n3:19%). After 4 weeks of feeding, each group wa subdiveided into (a) control, (b) 2 min swim in ice-cold water. Animals wer decapitated 20min after commencing the swim; trunk blood, brain, liver and epididymal fat pad were obtained. The levels of serum corticosterone, glucose, NEFA, triglyceride, fatty acid compositions, brain serotonin and 5-hydroxyindoleacetic acid were determined. Basal levels of corticosterone na NEFA of serum were significantly lower in fish oil fed animals than those of any other oil fed animals. Compared to either perilla oil-fed or corn oil-fed rats, cold swim stress in fish oil fed rats produced significantly smaller NEFA and larger corticosterone responses. However, there was no significant difference in basal levels of serum glucose. Stress increased serum glucose levels slightly, and the amount of increment was larger in fish oil rats than those of any other oil fed rats than those of any other oil fed rats, although all the values were normal level. Dietary fats and stress did not affect serotonin metabolism. In additions, the composition of fatty acids in serum was significantly affected by the dietary compostion of fatty acids and stress. Stress induced decreases in monounsaturated fatty acid and non-polyunsaturated fatty acid concentration in either perilla oil fed or fish group, but did not in corn oil fed group. Stress resulted in changes in fatty acid metabolism similar to that associated with essential fatty acid(EFA) dificiency, when feeding animals n-3 fatty acids in diet. In conclusion, feeding fish oil was more effective to decrease NEFA in serum than feeding perilla oil or corn oil and improved lipid metabolism, when the rats were maintained in normal or exposed to stressful environment. However, the fact that feeding diet containing n-3 fatty acids decreased EFA status under stress suggests that the requirement of n-6 PUFA should be increased in these groups.

자유로운 신체운동과 멜라토닌이 우울장애 동물모델에 미치는 효과 (Effects of Physical Activity and Melatonin in a Rat Model of Depression Induced by Chronic Stress)

  • Purpose: Stress, depending on its intensity and duration, results in either adaptive or maladaptive physiological and psychological changes in humans. Also, it was found that stressful experiences increase the signs of behavioral despair in rodents. On the other hand, exercise and melatonin treatment is believed to have many beneficial effects on health. Thus, this study was designed to evaluate the anti-depressant effects of physical activity and melatonin against chronic stress-induced depression in rats. Methods: Adult male Sprague-Dawley(SD) rats(200-250g, 7 weeks of age) were subjected to depression induced by chronic stress. Chronic depression was induced with forced-swim stress (FSS) and repeated change of light-dark cycle for 4 weeks. In the last 2 weeks, some rats were confined in a cage enriched with a running wheel, seesaw and chewed a ball from 19:00 to 07:00 every day. Melatonin was injected intra-peritoneally (I.P), and the rats received intraperitoneal injections of melatonin (15 mg/kg). The Forced Swim Test (FST) was performed to evaluate the immobility behaviors of rats for a 5 min test. Results: It was found that, the immobility time in FST was significantly (p<.05) lower in physical exercise ($M=58.83{\pm}22.73$) and melatonin ($M=67.33{\pm}37.73$) than in depressive rats ($M=145.93{\pm}63.16$) without physical activity. Also, TPH positive cell in dorsal raphe was significantly (p<.05) higher in exercise ($M=457.38{\pm}103.21$) and melatonin ($M=425.38{\pm}111.56$) than in depressive rats ($M=258.25{\pm}89.13$). Conclusion: This study suggests that physical activity and melatonin produces antidepressant-like effect on stress-induced depression in rats. So, physical exercise and melatonin may be a good intervention in depression patients.

수영(水泳)-스트레스에 의한 혈장 Corticosterone 함량 및 뇌(腦) Catecholamine대사(代謝)의 변동(變動)에 미치는 Thyroxine 및 Propylthiouracil의 영향 (Effect of Thyroxie and Propylthiouracil on the Responses of Plasma Corticosterone and Brain Norepinephrine to Swim-Stress)

  • 수영-스트레스로 나타나는 뇌 catecholamine대사 및 혈장 corticosterone 함량의 변동에 대한 clonidine(500 ug/kg)의 억제작용을 propylthiouracil(0.01% 용액으로 5주간 마시게함) 및 1-thyrorine(4 mg/kg/day로 5일간 복강내에 주사)로 처치한 웅성-마우스에서 실험관찰하여 다음과 같은 성적을 얻었다. 마우스의 일과성 자발운동량의 변동은 갑상선-홀몬의 변동에 영향을 받지 않았고, 수영스트레스(SS)로 나타나는 혈장 corticosterone(CS)의 증가가 propylthiouracil 전처치 (PTU) 및 1-thyroxine 전처치 (T4)로 각각 다소의 감소 및 증강됨을 보였으나, SS에 의한 혈장 CS증가에 대한 clonidine의 억제작용은 PTU 및 T4의 영향을 받지 않았다. SS부하로 뇌 3-methoxy-4-hydroxyphenylglycol 함량(MHPG)가 유의하게 증가되고 clonidine에 의하여는 MHPG가 현저히 감소되었으나 뇌 norepinephrine 함량(NE)은 별 변동을 보이지 않아서 MHPG/NE비는 SS와 clonidine에 의하여 각각 현저한 증가 및 감소를 나타내었다. 아울러, PTU및 T4은 각각 뇌NE을 유의하게 감소 또는 증가시켰으나 뇌 MHPG에는 별 영향을 미치지 않았다. Clonidine은 SS에 의한 뇌 MHPG 및 MHPG/NE비의 증가를 모두 현저히 억제하였으며 그 억제작용은 PTU 및 T4에 의하여 별 영향을 받지 않았다. 이상의 성적으로 미루어서, 마우스의 일과성 자발운동양상 및 스트레스반응으로 나타나는 혈장 corticosterone의 증가현상등이 갑상선-홀몬의 변동에 별 영향을 받지 않으며, 시상하부-뇌하수체-부신계의 활성화가 시상하부의 norepinephrine성 신경-흥분에 매게되어 나타나는 바, 스트레스성 혈장 corticosterone 증가에 대한 clonidine의 억제작용이 그의 절전-${\alpha}_2-adrenoceptor\;agonist$ 작용에 기인되는 것으로 사료된다.

Boophone disticha attenuates five day repeated forced swim-induced stress and adult hippocampal neurogenesis impairment in male Balb/c mice

  • Depression is one of the most common neuropsychiatric disorders and is associated with dysfunction of the neuroendocrine system and alterations in specific brain proteins. Boophone disticha (BD) is an indigenous psychoactive bulb that belongs to the Amaryllidacae family, which is widely used in Southern Africa to treat depression, with scientific evidence of potent antidepressant-like effects. The present study examined the antidepressant effects of BD and its mechanisms of action by measuring some behavioural parameters in the elevated plus maze, brain content of corticosterone, brain derived neurotropic factor (BDNF), and neuroblast differentiation in the hippocampus of Balb/c mice exposed to the five day repeated forced swim stress (5d-RFSS). Male Balb/c mice were subjected to the 5d-RFSS protocol to induce depressive-like behaviour (decreased swimming, increased floating, decreased open arm entry, decreased time spent in the open arms and decreased head dips in the elevated plus maze test) and treated with distilled water, fluoxetine and BD. BD treatment (10 mg/kg/p.o for 3 weeks) significantly attenuated the 5d-RFSS-induced behavioural abnormalities and the elevated serum corticosterone levels observed in stressed mice. Additionally, 5d-RFSS exposure significantly decreased the number of neuroblasts in the hippocampus and BDNF levels in the brain of Balb/c mice, while fluoxetine and BD treatment attenuated these changes. The antidepressant effects of BD were comparable to those of fluoxetine, but unlike fluoxetine, BD did not show any anxiogenic effects, suggesting better pharmacological functions. In conclusion, our study shows that BD exerted antidepressant-like effects in 5d-RFSS mice, mediated in part by normalizing brain corticosterone and BDNF levels.

만성스트레스 모델에서 하엽추출물의 항우울 효과 (Antidepressant-like effects of Nelumbo nucifera leaves extract in chronic mild stress model)

  • Objectives : Chronic mild stress (CMS) model is currently recognized as a better animal model of depression. The purpose of this study was to investigate the antidepressant-like effects of the Nelumbo nucifera leave extract using CMS model. Methods : The antidepressant-like effects of Nelumbo nucifera leaves extract was determined by using animal models of depression. Male ICR mice were divided into four groups: saline-treated normal, without CMS; saline-treated stress control; CMS+ Imipramine(20mg/kg); CMS+Nelumbo nucifera leaves extract(200mg/kg). All mice except the normal group exposed an unpredicted sequence of chronic mild stressors for 5 weeks. The behavior of mice were detected by sucrose preference test, forced swim test and tail suspension test. Then concentration of corticosterone in serum was detected by enzyme immunoassay. Results : Nelumbo nucifera leaves extract administration by daily gavage from the 3rd week exhibited an antidepressant-like effect on CMS-induced depression in mice. Nelumbo nucifera leaves extract administration at dose of 200 mg/kg significantly increased the sucrose consumption, and decreased the immobility durations in forced swim test and tail suspension test. Furthermore the corticosterone level decreased than control group. Conclusions : Chronic mild stress can affect mouse behavior and corticosterone level and cause depression. The present experiments not only further confirm the antidepressant-like effects of Nelumbo nucifera leaves extract in the sucrose preference test, forced swimming test and tail suspension test, but also the improving effects of Nelumbo nucifera leaves extract on the depression-like symptoms in the CMS model. Nelumbo nucifera leaves extract has the antagonism on CMS and produce antidepressive effects.

구속 스트레스 쥐 모델에서 스트레스 반응 감소에 대한 사카린 섭취의 효과 (Effect of Saccharin Intake in Restraint-induced Stress Response Reduction in Rats)

  • Purpose: Stress activates the sympathetic nervous system and hypothalamic-pituitary-adrenal (HPA) axis and induces the release of glucocorticoids. Saccharin is 300 times sweeter than sucrose, but does not increase blood insulin levels. Thus, this study was designed to evaluate the effect of saccharin intake in restraint-induced stress response reduction in rats. Methods: Adult male Sprague-Dawley (SD) rats had stress induced by restraint for 2 hours/day for 1 week. Saccharin was provided in sufficient amounts to allow them to intake it voluntarily at 0.1% diluted in water. The Y-maze test and forced swim test (FST) were performed to evaluate cognitive function and the depressive behavior of the rats. The protein expression of the glucocorticoid receptor (GR) in hippocampal cornu ammonis (CA) 1 was investigated by using immunohistochemistry. Results: It was found that, the percentage of alternation in the Y-maze test was significantly (p<.01) higher in the Stress + saccharin group than in the Stress group. Immobility time in the FST was significantly (p<.01) lower in the Stress + saccharin group than in the Stress group. Also, the positive cells of GR in hippocampus CA1 were significantly (p<.05) lower in the Stress + saccharin group than in the Stress group. Conclusion: This study showed that there was an effect of saccharin intake in restraint-induced stress response reduction in rats.

구속 스트레스로 인한 동물모델의 행동학적 변화와 간 손상 (Analysis of Restraint Stress-induced Behavioral Alteration and Liver Injury in Mice)

  • Background : Recent studies have shown that stress fundamentally influences the functional modulation of organ and stress-related disease causes high morbidity and mortality rates. Objective : The present research investigated the effect of restraint stress on psychological and physiological responses. Results : Body weight and food intake were changed in stress group. Body weight has continuously decreased, and food intake has been slightly altered. As a result of measuring each tissue's weight, the liver and kidney's weight loss was greater than that of other organs. The lipid profile of stressed animals showed significant increases in cholesterol (TC), triglyceride (TG), low-density lipoprotein (LDL), and high-density lipoprotein (HDL) levels compared to control. As hepatic marker enzymes, serum glutamic pyruvic transaminase (GPT; alanine aminotransferase), glutamic oxalacetic transaminase (GOT; aspartate aminotransferase), and lactic dehydrogenase (LDH) were increased in the stress group. However, levels of serum cortisol and corticosterone did not affect. Results of the behavioral tests show that the stress group has increased activity, sluggish movements, and anxiety in the central part compared with the control group through the open field test. In the forced swim test, the stress group models had a longer duration of slowing movement, and its rate also increased. Also, in immunoblotting, stress increased the inflammatory factors Inducible Nitric Oxide Synthase (iNOS), cyclooxygenase-2 (COX-2) and activated the mitogen-activated protein kinase (MAPK) pathway. Conclusions : We observed that mouse model were affected behavioral response and liver injury when exposed to restraint stress, indicating the importance of the restraint stress in the development of psychological and physiological processes.

Effects of Swim Training and Vitamin C Supplementation on the Antioxidant System Following Exhaustive Exercise Stress

  • This study was intended to investigate the effects of regular swimming exercise and vitamin C supplementation on the antioxidant system following exercise stress. For the swimming exercise experiment, a swimming adaptation exercise of 1 week was given to a group of 6-week-old mice. Following this, a swimming exercise for 8 weeks was conducted. The experimental group was divided into 3: a control group (C), a swimming exercise trained group (T), and a group of swimming + vitamin C supplementation (TC: vitamin supplementation: 1.3 mg/l00 g diet). After the swimming exercise, these group were further divided into those that had received the exercise stress for 2 hours and those that had not experienced exercise stress group. Then, the activities of the superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) concentrations were measured. There was a lower weight increase in the T and TC groups than in the C group, and there was no significant difference between T and TC group. When exercise stress was not experienced, the activity of SOD was significantly increased in the TC group than in the T group, but there was no significant difference between C and T groups. The groups that had experienced a 2-hour exercise stress showed the SOD activity levels according to the following order, C < T < TC, with a significant difference between the three groups (p<0.05). There was no difference in MDA concentration amongst the experimental groups in non-exercise stress group. As well, there was no differences in MDA concentration between the C group and T group in the 2 hour exercise stress group. However, the TC group showed a MDA concentration level significantly lower than that of the T group. A significant increase in MDA concentration was observed in C group, when exercise stress was provided with no significant difference in the T and TC groups. As a result, regular exercise and vitamin C supplementation can be considered important in controlling the formation of lipid peroxides in exercise stress.

청소년기 동물모델에서 구속 스트레스로 유발된 불안, 우울행동에 미치는 영향 (The Alleviating Effects of Sweet Drinks on Restraint Stress-Induced Anxiety and Depressive Behavior in Adolescent Rats)

  • Purpose: Some of the adolescent drinks more sugar-sweetened beverages. However, there is little evidence on the effect of eating behavior on emotional state and neurochemical changes under stress, especially on the levels of typical inhibitory neurotransmitters and gamma-aminobutyric acid. This article demonstrates that sucrose or saccharin drink reduces stress-related behavior responses and GABAergic deficits in adolescent rats. Methods: We randomly assigned 7-weeks-old Sprague-Dawley male rats to three groups: control group (Control), restraint stress only group (Stress), and restraint stress with unrestricted access to saccharin solution (Saccharin) and sucrose solution (Sucrose) as a positive control. We evaluated both anxious and depressive moods using an open field test and forced swim test, respectively. Using western blot analyses, the expression of a GABA-synthesizing enzyme, glutamate decarboxylase-67 (GAD67) and GABAergic markers, including calbindin and parvalbumin was assessed in the prefrontal cortex and the amygdala. Results: We found that both the drinks alleviated anxiety and depressive moods, induced significant attenuation in GAD67 level, and reduced calbindin level under stress in the prefrontal cortex and the amygdala. Conclusion: The results provide an understanding of the effect of sucrose or saccharin drink on stress-related responses. We propose the consumption of sweet drinks as a plausible strategy to alleviate stress-related alterations in adolescents.
