The present study was performed to determine the current infecion status of Metagonimus yokogawai metacercariae in sweetfish, Plecoglossus altivelis, collected from several streams in eastern and southern coastal areas of Korea. The sweetfish collected were artificially digested with pepsin-HCI solution and examined under a stereomicroscope in August and September, 2007. Out of 145 sweetfish collected from 10 streams in eastern coasts (Gangwon-do and Gyeongsangbuk-do), 88 (60.7%) were infected with Metagonimus metacercariae. The average metacercarial density was 61 per infected fish. Among 141 sweetfish collected from 10 streams in southern coasts (Gyeongsangnam-do, Ulsan Metropolitan city, and Jeollanam-do), 140 (99.3%) were infected with Metagonimus metacercariae, and their average density was 949 per infected fish. The present study confirmed that M. yokogawai metacercariae are still prevalent in the sweetfish from several eastern and southern coastal localities. The prevalence and metacercarial density were much higher in the sweetfish from streams of southern coasts than in those of eastern coasts. Therefore, attention should be paid to this small fluke infection, and consumption of raw sweetfish naturally produced in these areas should be prohibited.
This study investigated the possibility of salinity acclimation of freshwater rotifers (Brachionus calyciflorus) as live food for sweetfish (Plecoglossus altivelis) larvae, and also examined the optimal salinity for the growth of sweetfish. Freshwater rotifers cultured in 0 and 4 PSU and seawater rotifers (B. rotundiformis) cultured in 33 PSU were supplied to the larvae with four kinds of enrichment material (condensed freshwater Chlorella, $\omega-yeast,$ baker's yeast, Super Selco) and larval growth at 4 PSU was examined. Growth of the freshwater rotifers positively increased from 0 PSU to 6 PSU, but decreased when over 8 PSU was reached. Growth and survival of the sweet fish larvae reared in 0 PSU were significantly lower than those reared in either 4 PSU or 33 PSU. This indicated that the freshwater rotifers (B. calyciflorus) could be used as live food for sweetfish larvae reared in 4 PSU. The body weight of sweetfish larvae fed on freshwater rotifers enriched with Super Selco was the highest at 0.163 mg, but there was no significant difference in survival and body length of the fish fed with the other enrichment materials. The content of n-3 HUFA of the sweetfish larvae fed on the freshwater rotifers enriched with Super Selco and the condensed freshwater Chlorella was higher than that enriched with $\omega-yeast$ and baker's yeast. These results indicated that B. calyciflorus cultured with the condensed freshwater Chlorella could be used for the sweetfish larvae without enrichment, and the most efficient enrichment material for B. calyciflorus is Super Selco.
섬진강산 은어의 식품영양학적 평가의 기초 자료를 마련하고자 은어를 대소의 체형과 개체부위별로 구분하여 일반성분과 지질조성 및 총지질의 구성지방산을 분석${\cdot}$비교하였다. 1. 은어의 일반성분 중 조지방 함량은 수분과는 대조적으로 체형이 작은 것에 비하여 큰것에서 그 함량이 많았고, 같은 체형일 때는 체부위 중 내장에서 가장 높았고(소형, $43.2\%$; 대형, $52.8\%$), 근육은 가장 낮은 함량을 보였다(소형, $5.7\%$; 대형, $6.9\%$). 2. 은어의 지질은 $81.7{\sim}91.9\%$가 중성지질이었고, 인지질은 $10.9{\sim}18.3\%$로서, 나머지 지질의 대부분을 차지하였다. 그리고 이같은 지질의 조성은 어체가 클수록 그 함량이 높은 차이를 보였다. 3. 총지질의 구성지방산은 어체의 크기, 부위에 관계없이 포화산이 총지질 구성지방산의 약 $45\%$였고($43.78{\sim}48.29\%$), 다음으로 monoene산($37.41{\sim}39.58\%$), polyene산($13.13{\sim}17.10\%$)의 순으로 함유되어 있었다. 4. 지질 구성지방산 중 높은 비율을 차지하는 지방산은 16:0, 18:1n-9, 16:1n-7, 18:2n-6 등이었다.
은어의 수분 함량은 72.5%, 지방 함량은 5.3%, 회분 함량은 1.1%였다. 은어 지방 중에 단일 불포화지방산은 34.0%였다. 그중 oleic acid가 21.1%, palmitoleic acid가 7.8%였다. 다가 불포화지방산은 28.2%였는데, 그중 DHA가 12.1%, linoleic acid가 8.1%, EPA가 5.7%였다. 통째인 은어를 12일 동안 냉장($2^{\circ}C$) 보관하며 TBA 값과 PV를 측정한 결과, ascorbic acid를 처리한 군과 진공포장한 군이 대조군과 비교하여 유의적인 차이가 없었다(p>0.05). FFA는 12일 동안 세 그룹간의 유의적인 차이가 없었다(p>0.05). 잘게 다진 상태의 은어를 12일 동안 냉장($2^{\circ}C$) 보관하면서 TBA와 PV를 측정한 결과, ascorbic acid를 처리한 군과 진공포장한 군이 대조군에 비하여 지방산화 정도가 유의적으로 낮았다(p<0.05). 이러한 연구 결과로 볼 때, 은어가 불포화지방산과 DHA와 EPA의 좋은 급원 식품이며, ascorbic acid 처리와 진공포장이 은어 지방의 산화를 억제하는데 도움이 될 것으로 판단된다.
The effect of live food and commercial diet on sweetfish Plecoglossus altivelis culture was assessed by determining the daily growth rate and feed conversion ratio. After feeding for 8 weeks, the Moina macrocopa diet resulted in a higher growth rate of 5.101 compared to commercial (3.441) and Artemia (2.729) diets. In addition fish fed the M. macrocopa diet had the weight gain, averaging 41.29 g at the end of the experiment. The content of n-3 highly unsaturated fatty acids (HUFA) was highest in the commercial diet, while free fatty acids were not detected in the M. macrocopa diet. Triglycerides, the energy source in fishes, constituted 40.0% of the M. macrocopa diet, which was significantly higher than in the commercial and Artemia diets at 20.0 and 12.0%, respectively These results indicate that freshwater M. macrocopa can be used as feed for sweetfish larvae, even without docosahexaenoic acid (DHA) enrichment from Artemia.
기존의 Cx 배열을 참고로 종내${cdot}$종간을 통하여 잘 보존되어져 있는 영역에서 primer를 설계하고, 은어의 난소를 재료로 하여 PCR을 실시하였다. 증폭된 cDNA 단편을 이용하여, 5'RACE 및 3'RACE법에 의해 미지의 영역을 cloning하여 난소에서 발현하는 Cx cDNA의 전염기배열을 결정하였다. 기존의 Cx 배열과 상동성을 비교한 결과, 대서양산 민어의 Cx32.7과 $63.8{\%}$, bovine의 Cx44와 $61.6{\%}$ 및 대서양산 민어의 Cx32.2와는 $56.7{\%}$의 상동성이 나타났다. 본 cDNA는 35,028 Da의 분자량을 code하는 open reading frame (ORF)으로 구성되어 있어, 은어 Cx35로 명명되었다. 또한 아미노산 배열의 친수성${\cdot}$소수성 영역의 분포예측 결과, 4곳의 소수성 영역과 4곳의 친수성 영역을 교차하는 전형적인 Cx의 구조와 일치하였으며, Cx family의 공통${\cdot}$필수적인 배열인 제1세포외 domain의 consensus 배열 및 제2세포외 domain의 consensus 배열도 존재하였다.
본 연구는 어도 생태수리실험에 의하여 계단식 어도에서 회유성 어류의 소상능력을 평가하기 위한 것으로서 은어와 송어를 대상으로 실험하였으며, 기존 계단식 어도 설치 하천의 회유성 어류 이동생태환경을 분석하였다. 계단식 어도에서 은어의 소상능력은 낙차가 16.0cm미만으로 작을 때는 송어보다 크고, 20.0cm이상으로 낙차가 클 때는 송어의 소상능력이 은어보다 크다. 은어는 송어보다 도약을 선호하며 도약 후의 지구력은 송어가 은어에 비하여 크다. 수계별 기존 설치된 계단식 어도의 낙차가 매우 크기 때문에 은어와 송어의 이동에 관한 생태환경이 매우 불량하다. 본 연구 결과를 통하여 볼 때 고전적인 계단식 어도의 설치는 하천에 서식하는 회유성 어종의 다양성 측면을 고려하여야 하며 설계시 어도내 상류와 하류 웅덩이간 낙차는 20cm 미만으로 하여야 할 것이다.
은어의 효과적인 인공 종묘 생산을 위하여 자어의 성장과 생존율에 대한 먹이생물의 영양 가치에 대해 실험하여 다음과 같은 결과를 얻었다. 자어의 rotifer type에 따른 먹이효율을 조사하기 위해 3개 rotifer type (L-type, S-type, Us-type rotifer)을 각각 단독 또는 혼합하여 공급하였을 때 자어의 생존율은 모든 실험구에서 차이가 없었으나, 성장은 3 개 type rotifer를 모두 혼합하여 공급한 실험구에서 가장 높았다. Rotifer와 Artemia nauplius를 Chiorefla effipsoidea, 유지효모, 건조분말 Spimlina Platenis, Super Selco로 영양 강화시켜 자어에 공급한 영양 강화 실험에서 건조분말 S. platenis와 Super Selco를 혼합하여 영양 강화시킨 실험구에서 가장 높은 성장과 생존율을 보였다. Rotifer와 Arremia nauplius 공급시 자어의 사육 수조에 green water의 효과를 조사하기 위해 Chforefia ellipsoidea, 건조분말 Spimfina platensis, 광합성 세균(Rhodopseudomonas capsulatus)을 green water 원으로 사용하였을 때 건조분말 Sprinlina platenis와 광합성 세균을 혼합하여 공급한 실험구에서 가장 높은 성장과 생존율을 나타냈다.
In order to investigate the effect of salinity on growth and osmoregulation of the sweetfish, Plecoglossus altivelis, the fish (post-larva, juvenile, adult) reared in seawater were directly transferred into the experimental rearing water of the different salinities (post-larva : $0\textperthousand$, $5\textperthousand$, $10\textperthousand$, $15\textperthousand$, $20\textperthousand$, $30\textperthousand$. juvenile : $0\textperthousand$, $\10textperthousand$, $20\textperthousand$, $30\textperthousand$. adult : $0\textperthousand$, $15\textperthousand$, $35\textperthousand$, respectively). Fish samples were taken in time courses after transfer for blood and body fluid collection. The changes of sodium, potassium concentration and osmolality of the body fluid and plasma, muscle water content, gonadosomatic index (GSI) and hepatosomatic index (HSI) in the experimental fish were observed in time courses. Growth rate of the post-larva in $\5~25\textperthousand$ were faster than in $0\textperthousand$ (P<0.05). Survival rate was very low in $0\textperthousand$ (P<0.05). No fish survived more than 50 days after transferred into $0\textperthousand$. The concentration of sodium, potassium and osmolality in body fluid of the juvenile sweetfish were slightly decreased at 0.25 day after the beginning of the maintainable level of sodium, potassium and osmolality were $36.4\pm6.2 mM/\ell, 47.5\pm0.7 mM/\ell, 366.3\pm19.1 mOsm/kg, respectively. Except potassium concetration, fish in $0\textperthousand$ and $10\textperthousand$ displayed the pattern of hypoosmotic regulation. Growth rate and survival rate of juveniles of $30\textperthousand$ are significantly low than other groups (P<0.05). The concentration of sodium, potassium and osmolality in plasma of adult sweetfish were slightly decreased at 1 day after the beginning of the maintainable level of sodium, potassium and osmolality were $159.0\pm2.5 mM/\ell, 5.8\pm0.9 mM/\ell, 338.1$\pm13.3 mOsm/kg respectively. Compared with these values, fish in $0\textperthousand$ and $15\textperthousand$ displayed hypoosmotic regultion. Growth and survival rate of adult in $35\textperthousand$ are significantly low than other experimental groups. GSI was the highest in $15\textperthousand$ after 60 days. HSI was the lowest in $35\textperthousand$. With decreased salinities, oxygen consumption was found to be decreased in the other experimental groups compared to the more than $30\textperthousand$.
