This study describes an efficient protocol for Agrobacterium tumefaciens-mediated transformation of two subgroups of genotype AAA bananas (Musa acuminata cv. Pei Chiao and Musa acuminata cv. Gros Michel). Instead of using suspension cells, cauliflower-like bud clumps, also known as multiple bud clumps (MBC), were induced from sucker buds on MS medium containing $N^6$-Benzylaminopurine (BA), Thidiazuron (TDZ), and Paclobutrazol (PP333). Bud slices were co-cultivated with A. tumefaciens C58C1 or EHA105 that carry a plasmid containing Arabidopsis root-type ferredoxin gene (Atfd3) and a plant ferredoxin-like protein (pflp) gene, respectively. These two strains showed differences in transformation efficiency. The EHA105 strain was more sensitive in Pei Chiao, 51.3% bud slices were pflp-transformed, and 12.6% slices were Atfd3-transformed. Gros Michel was susceptible to C58C1 and the transformation efficiency is 4.4% for pflp and 13.1% for Atfd3. Additionally, gene integration of the putative pflp was confirmed by Southern blot. Resulting from the pathogen inoculation assay, we found that the pflp transgenic banana exhibited resistance to Fusarium oxysporum f. sp. cubense tropical race 4. This protocol is highly advantageous to banana cultivars that have difficulties in setting up suspension cultures for the purpose of quality improvement through genetic transformation. In addition, this protocol would save at least 6 months in obtaining explants for transformation and reduce labor for weekly subculture in embryogenic cell suspension culture systems.
The objectives in the present study were to evaluate the therapeutic effect of dioctahedral smectite (smectite) against piglet diarrhea caused by E. coli and Salmonella. Of this study, twenty piglets (body weight, $6.60{\pm}0.34\;kg$) with diarrhea were used to estimate the efficacy of smectite on piglet diarrhea with 20% smectite suspension in PBS. Pigs were administered with 1 ml smectite suspension three times a day after feeding, and fecal samples were collected at the gate of treatment and on 1st, 2nd, 3rd, and 4th day after administration. Changes in body weight, diarrhea index, and shedding of E. coli and Salmonella were monitored during the experimental period. The group treated with smectite reduced diarrhea index (p < 0.01), and increased body weight compared to control group. In the treated group, the number of E. coli and Salmonella was significantly decreased compared to each control group (p < 0.01). From results of the present study, 20% smectite suspension had therapeutic effect on diarrhea caused by E. coli and Salmonella in piglets.
The aim of this study was to identify the effect of suspension unloading (SU) and electrical stimulation upon the development of neonatal muscular system. For this study, the neonatal rats were randomly divided into three groups: a control group, an experimental group I, and an experimental group II. The SU for experimental group I and II was applied from postnatal day (PD) 5 to PD 30. The electrical stimulation for soleus muscle of experimental group IIwas applied from PD 16 to PD 30 using neuromuscular electrical stimulation (NMES), which gave isometric contraction with 10 pps for 30 minutes twice a day. In order to observe the effect of SU and ES, this study observed myocyte enhancer factor 2C (MEF2C) and vascular endothelial growth factor (VEGF) immunoreactivity in the soleus muscles at PD 15 and PD 30. In addition, the motor behavior test was performed through footprint analysis at PD 30. The following is the result. At PD 15, the soleus muscles of experimental group Iand II had significantly lower MEF2C, VEGF immunoreactivity than the control group. It proved that microgravity conditions restricted the development of the skeletal muscle cells at PD 15. At PD 30, soleus muscles of the control group and experimental group II had significantly higher MEF2C, VEGF, immunoreactivity than experimental group I. It proved that the NMES facilitated the development of the skeletal muscle cells. At PD 30, it showed that SU caused the decrease in stride length of parameter of gait analysis and an increase in toe-out angle, and that the NMES decreased these variations. These results suggest that weight bearing during neonatal developmental period is essential for muscular development. They also reveal that NMES can encourage the development of muscular systems by fully supplementing the effect of weight bearing, which is an essential factor in the neonatal developmental process.

Railway track alignment Standards set a minimum lenght value for straight and circular alignments (art. 5.2.9.), in order to ensure passenger ride comfort in railway vehicles of which dynamic oscillations will thus have to be limited. The transitions between alignments can cause abrupt changes (usually called discontinuities or singular points of the alignment) of curvature, of rate of change of curvature or of rate of change of cant. A passenger is likely to experience effects due to the excitation of the elastic suspension of the vehicle which generates oscillations that are damped as the vehicle moves away from the singularity. The amplitude of these oscillations should be adequately attenuated by the damping of the suspension system within the interval between two successive singular points, especially to avoid resonances. Therefore minimum lengths between two successive singular points are stated in alignment standards. Nevertheless, these nonnative values can be overly conservative in some cases. As an alternative, track alignment designers could try to assess how much the excitation has been attenuated between two successive singular points and thus assess at which point a new singularity may be present without affecting ride comfort. Although such assessment can be made with commercial SW packages which simulate the dynamic behavior of a vehicle considered as a set of rigid bodies interconnected with elastic elements simulating the suspension systems (such as SIMPACK, ADAMS or VAMPIRE), a simplified and user-friendly computation method (based upon the analytical solution of differential equations governing the phenomenon) is made available in this paper to track design engineers, not always used to working with full dynamic models.

In this paper, we develop the so-called functional test model for magnetic bearing reaction wheels. The functional test model has three degree of freedom, which consists of one axial suspension from gravity and the other two axes gimbaling capability to small angle, and does not include the motor. For the control of the functional test model, we derive the optimal electromagnetic forces based on the least squares method, and use the proportional-integral-derivative controller. Then, we develop a hardware setup, which mainly consists of the digital signal processor and the 12-bit analog-to-digital and digital-to-analog converters, and show the experimental results.

this paper develops a computational model for the turning maneuver analysis of a cabover type heavy truck. The model having 42 degree-of-freedom is developed using ADAMS. Leaf springs used in the front and rear suspension systems are modeled by dividing it three links and joining them with joints. Force and displacement relationship showing nonlinear hysteric characteristics of the leaf spring is measured and modeled with an exponential function. A velocity and force relationship of a shock absorber is measured and modeled with a spline function. And a stabilizer bar is modeled using ADAMS beam element to consider a twisting and bending effect. To verify the developed model an actual vehicle test is performed in the double lane change course with 50kph and 60kph vehicle velocity. In the actual vehicle test lateral acceleration roll angle and yaw rate are measured, The tendency and peak-to-peak values of the actual vehicle test and simultion results are compared each other.

Load/Unload(L/UL) technology includes the benefits, that is, increased areal density, reduced power consumption and improved shock resistance contrary to contact start stop(CSS). It has been widely used in portable hard disk drive and will become the key technology for developing the small form factor hard disk drive. The main objectives of L/UL are no slider disk contact or no media damage. For realizing those, we must consider many design parameters in L/UL systems. In this paper, we focus on the effect of the ramp profile. We can find out the lateral velocities in L/UL process through experiments and simulations for force of voice coil motor and friction force on ramp. And then, we will gain the optimal design of ramp slope to maintain the minimum clearance of suspension dimple and slider with FE model. In special, after finding the point at which air bearing breaks and designing the ramp, we will identify the results for improving unload performance.
Objectives : A growing body of evidence has suggested that the dysfunction of glutamatergic systems plays a pivotal role in major depressive disorder (MDD). This study was performed to investigate the antidepressant-like effects of the ethanolic extract of Gastrodia elata Bl (GE) in mouse models and to investigate the role of ${\alpha}$-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA) receptors in producing these antidepressant-like effects. Methods : The forced swim test (FST) and tail suspension test (TST) were used to investigate GE's behavioral effects in mice. Additional biochemical and behavioral experiments with NBQX, an AMPA receptor antagonist, were undertaken to determine whether the antidepressant-like properties of GE are involved in AMPA receptor throughput. Results : Oral administration of GE extract (1,600 mg/kg) 1h prior to testing significantly reduced the immobility times in the FST and TST. These antidepressant-like effects of GE extract were increased dose-dependently. Pre-treatment with NBQX significantly attenuated the reduction in immobility time induced by the GE extract in the FST and TST. Conclusions : The ethanolic extract of GE may exert antidepressant-like effects with involvement of AMPA receptor.
Microcystin-LR(MC-LR), a cyanobacterial toxin produced by Microcystis aeruginosa, causes severe hepatotoxicity. Here we investigated the morphologic changes of rat hepatocyte spheroid induced by exposure of MC-LR($10^{-6}M$) in vitro. In addition, to determine the effects of such toxin in the process of hepatocyte spheroid formation, primarily isolated hepatocytes were incubated with MC-LR and the process of spheroid formation was observed. In both hepatocyte spheroid and suspension culture systems, the morphologic changes caused by MC-LR were noticible at 5 min post exposure and were characterized by the loss of microvilli, cytoplasmic vacuolation, the accumulation of lipid droplets, and blob formation. Especially, the size and numbers of blob on the cell surface were increased as the incubation time prolonged and the appearance of electron dense bodies were observed in the cytoplasm of hepatocyte at 20 min post exposure. Furthermore, bile canaliculi-like structures in the hepatocyte spheroids were slightly widened and the process of spheroids formation was inhibited in the isolated hepatocytes incubated with MC-LR. These results indicate that morphologic changes in. the hepatocyte membrane and organelles seem to be typical events in showing the MC-LR induced hepatotoxic effects and the spheroid culture method might be a useful experimental tool to evaluate hepatoxicity since it reflects the in vivo status of hepatocytes.
The modal parameters of the deck of Runyang Suspension Bridge (RSB) as well as their relationships with wind and temperature are studied based on the data recorded by its Structural Health Monitoring System (SHMS). Firstly, frequency analysis on the vertical responses at the two sides of the deck is carried out to distinguish the vertical and torsional vibration modes. Then, the vertical, torsional and lateral modal parameters of the deck of RSB are identified using Hilbert-Huang Transform (HHT) and validated by the identified results before RSB was opened to traffic. On the basis of this, the modal frequencies and damping ratios of RSB during the whole process of Typhoon Masta are obtained. And the correlation analysis on the modal parameters and wind environmental factors is then conducted. Results show that the HHT can achieve an accurate modal identification of RSB and the damping ratios show an obvious decay trend as the frequencies increase. Besides, compared to frequencies, the damping ratios are more sensitive to the environmental factors, in particular, the wind speed. Further study on configuring the variation law of modal parameters related with environmental factors should be continued.
