The biological activities of Zelkova leaves and the quality characteristics of Zelkova leaf rice cake according to leaf levels (느티잎의 생리활성 및 느티잎 첨가량에 따른 느티떡의 품질특성)

  • The objectives of this study were to determine the antioxidant and cytotoxic activities of Zelkova leaf extract and the sensory and textural characteristics of Zelkova leaf rice cake. The ethyl acetate layer of Zelkova leaf extract showed the most effective DPPH radical and superoxide anion scavenging activities. The residual layer of Zelkova leaf extract exhibited potent cytotoxicity against the HeLa cell line. In the sensory evaluation, the color and taste of Zelkova leaf rice cakes were not significantly different. Zelkova leaf rice cakes with 20%(5.18), 25%(4.94) and 30%(4.88) added Zelkova leaves showed higher overall acceptability than 0%(4.06) added Zelkova leaves. In the textural analysis, the hardness of the Zelkova leaf rice cake increased slowly with increasing leaf content during 3 day storage. The cohesiveness of 25% and 30% Zelkova leaf rice cakes showed no variation with storage time but that of 0% Zelkova leaf rice cakes decreased with increasing storage period. The 'L' value of Zelkova leaf rice cake decreased with increasing leaf content while the 'a' value of 0% Zelkova leaf rice cake was lower than that of 20%, 25% and 30% Zelkova leaf rice cakes.

Antioxidant and Protective Effects of Leonurus sibiricus L. Extract on Ultraviolet B (UVB)-induced Damage in Human Keratinocytes (익모초 추출물의 항산화 효과와 자외선으로 유도된 각질형성세포 손상에 대한 보호효과)

  • In this study, we investigated the anti oxidative potential and protective effects of water extract of Leonurus sibiricus L. leaf (LSLW) against ultraviolet B (UVB)-induced oxidative damage in human keratinocytes (HaCaT cells). To evaluate the anti oxidative activity of LSLW, we measured DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion scavenging activities, lipid peroxidation inhibition, and reducing power of LSLW. For induction of oxidative stress in HaCaT cells, the cells were irradiated with UVB at 40 mJ/㎠. To investigate the protective effects of LSLW against UVB, we measured cell viability and apoptotic bodies using annexin V staining. LSLW showed anti oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion and by reducing lipid peroxidation. In addition, LSLW showed high reducing values. The UVB-induced oxidative conditions led to cell apoptosis. However, treatment with LSLW ameliorated oxidative stress conditions, including inhibition of cell death, apoptotic body. Taken together, LSLW exhibited anti oxidative and protective effects against UVB-induced damage in HaCaT cells. Thus, LSLW could be useful for the development of cosmetics for UVB-induced skin aging.

Antioxidative Activities of Wen-pi-tang-Hab-Wu-ling-san (WHW$^{(R)}$) in vitro (가감온비탕합오산(加減溫脾湯合五散) 완제(完製)(HWW$^{(R)}$)의 항산화 효과에 대한 연구)

  • Objectives: The objective of this study was to investigate the antioxidant effects of manufactured Wen-pi-tang-Hab-Wu-ling-san (WHW$^{(R)}$) in vitro. Methods: WHW$^{(R)}$ was prepared by the pilot manufacture of WHW water extract from a GMP system appointed company. Antioxidative activities were determined by in vitro tests as follows: the scavenging activities of oxygen free radicals including 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide anion, hydrogen peroxide and nitric oxide radicals, as well as ferrous ion chelating capacity and Trolox equivalent antioxidant capacity (TEAC). Results: WHW$^{(R)}$ significantly scavenged oxygen free radicals such as DPPH (IC$_{50}$=115.28 $\pm$ 0.25 $\mu$g/$m\ell$), superoxide anion (IC$_{50}$=8.56 $\pm$ 0.08 $\mu$g/$m\ell$), hydrogen peroxide (IC$_{50}$=240.36 $\pm$ 3.41 $\mu$g/$m\ell$) and nitric oxide (IC$_{50}$=162.28 $\pm$ 0.21 $\mu$g/$m\ell$) radicals. WHW$^{(R)}$ also showed ferrous ion chelating activity (IC$_{50}$=543.19 $\pm$ 4.85 $\mu$g/$m\ell$) and Trolox equivalent effects (IC$_{50}$=45.311 $\mu$g/$m\ell$) in TEAC and ORAC assay, respectively. Conclusion: This study demonstrates that WHW$^{(R)}$ has strong antioxidative properties through free radical scavenging activity. These data suggest that WHW$^{(R)}$ be used as an antioxidant agent.

Antioxidant Activity of Yogurt Supplemented with Red Ginseng Extract (홍삼 추출물을 첨가한 요구르트의 항산화능)

  • The antioxidant properties of red ginseng extracts prepared under different extraction conditions were evaluated by a variety of antioxidant assays, including $DPPH^{\cdot}$ radical scavenging, $ABTS^{+\cdot}$ radical scavenging, superoxide anion scavenging, nitrite scavenging and reducing power activities. The contents of total phenolic compounds and flavonoids were also determined. The various antioxidant activities were compared to positive controls such as Trolox, tannic acid and ascorbic acid. The antioxidant activities of all of the extracts were shown to be the highest in the ethanol extract. The antioxidant activities of the red ginseng powder were the lowest among the samples. The amounts of total phenolic compounds and flavonoids were at a maximum in the ethanol extract. Correlation analysis demonstrated the existence of a linear relationship between free radical scavenging activities and the phenolic compounds contents of extracts. The antioxidant activity of yogurt was increased as the result of the addition of red ginseng extract. The quality characteristics of the yogurt to which red ginseng extract was added were similar to those of yogurt without red ginseng extract. The overall sensory score and color of yogurt made from 0.5% red ginseng was the best of the tested yogurts. In accordance with the antioxidant activity and quality characteristics, the optimal concentration of red ginseng extract was approximately 0.5%.

  • The antioxidant activities of methanol and dichloromethane/acetone extracts of twenty three seaweeds were tested by using 1,1-Diphenyl-2-picryl-hydrazyl(DPPH) at a $100{\mu}g/ml$ concentrations. The dichloromethane/acetone extracts of three seaweeds(Symphyocladia latiuscula, Gloiopoltis furcata, Sagassum thunbergii), were found to be most effective in DPPH radical scavenging activity. The DPPH radical scavenging effect of these seaweeds was Symphyocladia latiuscula(85.82%), Gloiopoltis furcata(82.83%), Sagassum thunbergii(74.05%) in order. These seaweeds were evaluated using the pyrogallol UV-VIS spectrophotometeric method to generate superoxide anion. Among them, the methanol extracts of six seaweeds were showed weak superoxide dismutase-like activities. The dichloromethane/acetone extracts obtained from Symphyocladia latiuscula was fractionated with $CH_2Cl_2$, n-hexane, 15% aq.MeOH, n-BuOH, $H_2O$. The 15% aq.MeOH soluble fraction exhibiting the strongest antioxidative activities was further purified by C18 column flash chromatography and reversed HPLC. The two active principles of Symphyocladia latiuscula were isolated and characterized as 2,3,6-tribromo-4,5-dihydroxybenzyl alcohol(1), 2,3,6-tribromo-4,5-dihydroxybenzyl methyl ether(2).

In Vitro and Intracellular Antioxidant Activities of Brown Alga Eisenia bicyclis

  • The antioxidant activities of a methanolic extract of Eisenia bicyclis and its organic solvent fractions, including dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc), n-butanol (n-BuOH), and water ($H_2O$) fractions, were investigated. Scavenging activities against DPPH, hydroxyl, superoxide anion, and peroxynitrite radicals were evaluated using electron spin resonance spectrometry; intracellular reactive oxygen species (ROS) were evaluated by a 2',7'-dichlorofluorescein diacetate assay using RAW264.7 mouse macrophages. The antioxidant activities of the individual fractions were: EtOAc>n-BuOH>$CH_2Cl_2$ >$H_2O$. The EtOAc fraction exhibited strong radical scavenging activity and a significantly reduced ROS level in RAW264.7 cells. Moreover, the phenolic contents of the extract and fractions followed the same order as their radical scavenging activities. Our results indicate that E. bicyclis is a valuable natural source of antioxidants that may be applicable to the functional food industry.

Total Phenolic Contents, Radical Scavenging Capacities and Inhibitory Effects on Lipid Peroxidation and LDL Oxidation of Prunus persica Branch

  • This study was undertaken to elucidate the antioxidant activity of the ethanol (EEPB) and water (WEPB) extracts of Prunus persica branches. The extracts contained a high phenolic content and revealed a potent hydrogen donating activity in DPPH scavenging assay. Compared to $\alpha$-tocopherol, EEPB (p < 0.001) and WEPB (p < 0.05) significantly inhibited $FeCl_2$-ascorbic acid-induced lipid peroxidation, and also exhibited potent antiradical activities against hydroxyl radical, superoxide anion, nitric oxide and peroxynitrite. In copper- and AAPH-mediated human low-density lipoprotein (LDL) oxidation systems, the extracts demonstrated a strong antioxidant function by metal chelating, rather than direct scavenging, action. Furthermore, EEPB at 5 ${\mu}g/mL$ concentration showed 80.77% inhibition of the electrophoretic mobility of LDL, compared to 77.69% for ascorbic acid and 76.92% for BHT. These results suggest that PB branch extracts may protect against oxidative stress-induced diseases.

Antioxidative Effects of Delphinidin under in vitro and Cellular System

  • This study examined the antioxidative activity of delphinidin, a kind of anthocyanidin from eggplant. Cellular protective potential from oxidative damage by nitric oxide (NO), superoxide anion ($O_2^-$), and peroxynitrite ($ONOO^-$) using epithelial cell line LLC-PK1 cell as well as in vitro radical scavenging effects were investigated. Delphinidin showed strong in vitro radical scavenging effects against NO, $O_2^-$, and hydroxyl radical (${\cdot}OH$) in dose-dependent manners. In addition, delphinidin increased cell viability in LLC-PK1 cells in a concentration-dependent manner when viability was reduced by $ONOO^-$-induced oxidative damage. To elucidate the protective mechanisms of delphinidin from $ONOO^-$, sodium nitroprusside (SNP), and pyrogallol were also employed to generate NO and $O_2^-$, respectively. The treatment of delphinidin recovered reductions in cell viability caused by SNP and pyrogallol, indicating that delphinidin can attenuate oxidative stress induced by NO and $O_2^-$. The present study suggests that delphinidin is a promising anti oxidative agent.

Comparison on Antioxidant Potential of Several Peanut Varieties (땅콩 품종간 항산화 활성 비교)

  • To increase utilities as functional materials, nine peanut varieties were evaluated on antioxidant activity and total phenol content. Peanut varieties were individually divided into three parts such as part without seed coat, seed coat, and seed. Each of the 80% ethanol-extracted solutions from the parts was used for the assay. Inhibitory activities on low density lipoprotein (LDL) oxidation by the part without seed coat, seed coat, and seeds from 9 varieties were respectively 85.3∼91.0%, 77.4∼85.8%, -1.7∼46.1%, and those on linoleic acid oxidation were 94.0∼94.9%, 94.2∼95.0%, 69.4∼80.2%, respectively. Seed coats, parts without seed coat, and seeds from 9 peanut varieties showed scavenging activities as 88.3∼98.7%, 16.7∼56.3%, -28.0∼20.0% on superoxide anion radical and as 89.9∼90.8%, 6.0∼12.0%, 0.6∼2.1% on 1,1-diphenylpicrylhydrazyl (DPPH) radical, respectively. Total phenol contents of seed coat, seed, and part without seed coat were 12.7∼21.0 mg%, 2.46∼4.24 mg%, 0.94∼1.16 mg% as tannic acid equivalent and those of Daekwang, Daesin, Bowon, Saedl, Kipung among the peanut varieties were comparatively high. In conclusion, Daekwang, Daesin, Kipung, and Saedl among the peanut varieties showed comparatively effective antioxidant activities.

Antioxidant and Inhibitory Activities on Angiotensin Converting Enzyme in Lysimachia clethroides Duby (큰까치수영의 항산화 및 안지오텐신 전환 효소 저해 활성)

  • This study was conducted to develop physiologically active plant materials from medicinal plants. Crude extracts and solvent fractions prepared from Lysimachia cletroides Duby were tested for their antioxidant and antihypertensive activities. For ellucidating antioxidant potential, inhibition rate on linoleic acid peroxidation, as well as scavenging activities on superoxide anion and 1,1-dipicrylphenylhydrazyl (DPPH) radical were evaluated. For analyzing antihypertensive effect, inhibitory activity on angiotensin converting enzyme (ACE) was done. Methanol extract of L. cletroides showed potent inhibition activity of 83% on linoleic acid peroxidation, which was more effective than -2% of ${\alpha}-tocopherol$ at $25\;{\mu}g/ml$. Methanol and water extracts exhibited strong scavenging activities of $86{\sim}109%$ and $96{\sim}122%$ on superoxide anion radical which was higher than $-4{\sim}69%$ of ascorbic acid at $5{\sim}200\;{\mu}g/ml$. Hexane, ether and ethylacetate fractions possessed 133, 100 and 88% inhibitory activities on ACE at $4,000\;{\mu}g/ml$, respectively. From the results, it was expected that Lysimachia cletroides could be a new antioxidant and antihypertensive resource.
