Anti-Biofilm Activity of Origanum Vulgare Supercritical Fluid Extracts and Cosmetic Active Ingredients Development (오레가노 초임계추출물의 황색포도상구균 바이오필름 형성 억제능을 이용한 기능성 화장품 소재의 개발)

  • In this study, oregano was extracted by supercritical extraction and hydrothermal extraction method. In vitro experiments such as antimicrobial and antioxidant activity test were performed. As a result of the disc diffusion method, only the supercritical extracts formed a clear zone. The MIC for S. aureus was found only in the supercritical fluid extracts and it was 1000 ㎍/mL. The hydrothermal extract's MIC is 125 ㎍/mL for C. acnes. Through biofilm inhibition assay, we found that the supercritical fluid oregano extracts inhibit the biofilm of S. aureus by more than 70% even at low concentrations of 125 ㎍/mL. On the other hand, the antioxidant ability of the hydrothermal extract was better than that of the supercritical fluid extracts. Furthermore, we tried to make a skincare ingredient for atopic dermatitis by utilizing the S. aureus biofilm inhibitory ability of oregano supercritical fluid extracts. Liposome was used to overcome the low solubility of the oregano supercritical fluid extracts and increase stability.

Introduction of Modifying Solvents to Carbon Dioxide in Supercritical Extractions

  • A simple apparatus for adding a modifying solvent to supercritical CO2 extractant was described. Small, fixed volumes (typically 100 μL) of liquid modifying solvents were delivered during the extraction process by use of an in-line high pressure loop injector and an air pump. Without disconnecting the extraction cell from the supercritical fluid extraction system, the modifying solvent was repeatedly delivered. The solvent modification device was optimized during the extraction of carbaryl and bis(acetylacetonato) copper(Ⅱ). Extraction recoveries from spiked filter paper and soil samples ranged between 22% and 109%, depending on the analyte and matrix components. The addition of polar modifying solvents were necessary to improve the extractability of the nonpolar CO2.

Quality and characteristics of fermented ginseng seed oil based on bacterial strain and extraction method

  • Background: In this study, the fermentation of ginseng seeds was hypothesized to produce useful physiologically-active substances, similar to that observed for fermented ginseng root. Ginseng seed was fermented using Bacillus, Pediococcus, and Lactobacillus strains to extract ginseng seed oil, and the extraction yield, color, and quantity of phenolic compounds, fatty acids, and phytosterol were then analyzed. Methods: The ginseng seed was fermented inoculating 1% of each strain on sterilized ginseng seeds and incubating the seeds at $30^{\circ}C$ for 24 h. Oil was extracted from the fermented ginseng seeds using compression extraction, solvent extraction, and supercritical fluid extraction. Results and Conclusion: The color of the fermented ginseng seed oil did not differ greatly according to the fermentation or extraction method. The highest phenolic compound content recovered with the use of supercritical fluid extraction combined with fermentation using the Bacillus subtilis Korea Food Research Institute (KFRI) 1127 strain. The fatty acid composition did not differ greatly according to fermentation strain and extraction method. The phytosterol content of ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method was highest at 983.58 mg/100 g. Therefore, our results suggested that the ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method can yield a higher content of bioactive ingredients, such as phenolics, and phytosterols, without impacting the color or fatty acid composition of the product.

  • Performance of a continuous cocurrent supercritical fluid extraction column for processing of anhydrous milk fat (AMF) was determined. The extract loading increased and the extraction yield decreased as the superficial velocity of AMF increased. The maximum solubility of AMF in $CO_2$ at 4$0^{\circ}C$/3,500 psig was 0.0195g/g. The increase of the carbon dioxide density enhanced the solubility of milk fat and decreased the cholesterol concentration in the extract. Three operation conditions, such as recycle , reflux, and temperature gradient of extraction column, were tested. More short-chain fatty acvids were extracted by the reflux operation . Longchain fatty acids and thehighest ratio fo long-chain unsaturated to saturated fatty acids were also obtained in the raffinate phase with refluex.

Supercritical Fluid Extraction of Catecnin Compounds from Green Tea (녹차에서 카테킨 화합물의 초임계 유제 추출)

  • Catechin compounds from the Green tea cultivated in Bosung (Chollanamdo) were extracted using supercritical fluid and various additives, and analyzed by reversed-phase high performance liquid chromatography (RP-HPLC). The chromatographic column was packed with LiChrospher 100RP-18(15 $\mu$m), and water was used as the mobile phase with 0.05% phosphoric acid and acetonitrile. Gradient election was applied to separate EGCG by changing the mobile phase compositions. Comparing the extraction yield of three different types of supercritical fluids, pure CO$_2$, with additives of water and ethanol (5 wt,%), the extraction amount of EGCG was most abundant in the suprcritical CO$_2$with ethanol. However, more was extracted and pure higher purity was achieved by solvent extraction using ethanol.

  • Based on process analysis as well as extensive operation experience, two fuzzy expert control algorithms, for startup and control, are proposed for a supercritical fluid extraction process which has high interacting multivariable structure. In the proposed algorithms, a new simple defuzzification method which only requires four fundamental arithmetic rules is also presented. Through numerical simulations, control performance using the proposed control algorithm is compared with that of a different fuzzy algorithm by an other researcher and that of conventional PID-type controllers which are tuned by well-known optimal criteria. Also, the proposed control algorithm has been tested to the bench scale supercritical fluid extraction process. As a consequence, the proposed fuzzy expert controller has shown fast and robust control performance while the other controllers show sluggish and/or highly oscillatory responses.

Characteristics of Extraction of Daidzein and Genistein in Soybean Using Sub/Supercritical Fluids (아임계/초임계 유체를 이용한 콩에 포함된 Daidzein과 Genistein의 추출특성)

  • Daidzein and genistein were extracted from Korean soybean by supercritical $CO_2$ and sub/supercritical water. The extracted sample was analyzed by reversed-phase high performance liquid chromatography (RP-HPLC). The retention time, retention factor, column efficiency, column selectivity and resolution of aglycons were compared with the change in the temperature and pressure of supercritical fluid and ethanol concentration. The characteristics of extraction of daidzein and genistein were more affected by ethanol concentration using supercritical $CO_2$. The most desirable extraction yield was obtained by supercritical $H_2O$ with $400^{\circ}C$ and 250 bar. Generally, the extraction yield of aglycons increased over 10 times using supercritical $CO_2$ than sub/supercritical $H_2O$.

Extraction of Phenolic Compounds from grape Seed Using Supercritical $CO_2$ and Ethanol as a Co-solvent (초임계 이산화탄소와 에탄올 보조용매를 이용한 포도씨로부터의 페놀성 화합물의 추출)

  • A supercritical fluid extraction was performed for the extraction of phenolics from grape seeds which up to now have been discarded. The optimum condition for extraction process was predicted through response surface methodology using central composit experimental design. The extraction amount of grape seed phenolics was increased by increasing extraction temperature, pressure, and concentration of co-solvent (ethanol). The optimum extraction conditions were 84.83$^{\circ}$C, 51.50MPa and 1.27% ethanol. The yield of phenolics using SFE was higher with 3 folds than ethanol and 4 folds than hexane but less than 80% methanol. In the respects of food poisoning, the approved solvents were restricted to ethanol and hexane. So, SFE for extraction of phenolics could be powerful alternative method for solvent extraction.

Comparisons of Effectiveness of the Supercritical Fluid Extraction Dewaxing on the Beeswax-Treated Paper with Different Aging Degrees (열화율이 상이한 밀랍지의 초임계유체추출 탈랍처리효과 비교분석)

  • This study aims to investigate the applicability of supercritical fluid extraction as a dewaxing technique to restore the beeswax-treated volume of the Annals of the Joseon Dynasty in various deterioration and damage conditions. Thus, this study analyzed the dewaxing efficiency and changes in physical and morphological properties before and after dewaxing, by applying the optimal supercritical fluid extraction dewaxing condition ($70^{\circ}C$, 40 MPa, $CO_2$, Co-solvent 20% DCM, 2 hour) to 3 kinds of beeswax-treated paper with different deterioration rates (dry artificial aging of 10, 20 and 30 days at $120^{\circ}C$). After dry artificial aging at $120^{\circ}C$ for 30 days, the average molecular weight of the beeswax-treated specimen was $1.856{\times}10^5g/mol$, showing deterioration about 80% of the beeswax-treated paper before dewaxing. It was a similar level to the molecular weight of the bees-waxed volume of the Annals of the Joseon Dynasty that has a higher degree of damage. As a result of analyzing the dewaxing efficiency through FT-IR analysis, this study discovered that it was possible to effectively dewax beeswax-treated paper in the range of deterioration 20 to 80% with this supercritical fluid extraction dewaxing technique applied. As a result of analyzing changes in the physical and morphological properties before and after dewaxing, the viscosity tended to decrease to a slight degree, and since no morphological deformation or damage of stencil fibers was found, it was concluded possible to conduct stable dewaxing through this supercritical fluid extraction technique.
