Comparison of Antioxidant, Wrinkle Improvement, and Whitening Efficacies of Extracts from Pinus koraiensis Cone Scale Using Extraction Methods (추출방법에 따른 잣나무(Pinus koraiensis) 구과피 추출물의 항산화, 주름개선 및 미백 효능 비교)

  • The present study evaluated the antioxidant activity, skin wrinkle reduction, and whitening activity of Pinus koraiensiscone scale extracts made with three different solvents (ethanol, supercritical fluid, and a mixture of both). Total polyphenol content was 11.03 mg/g GAE in the supercritical fluid extractand 33.79 mg/g GAE in the 70% ethanol extract. Electron donating ability of 1,000 ㎍/mL extract was 13.60% in the supercritical fluid extract, 91.37% in 70% ethanol extract, and 71.62% in mixed solvent extract. SOD-like activities in 100 ㎍/mL extract using supercritical fluid, 70% ethanol, and mixed solvents were 16.49%, 21.84%, and 10.7%, respectively. The ABTS+ radical scavenging activities of 1,000 ㎍/mL extract were 38.19%, 80.23%, and 78.72%for supercritical fluid extract, 70% ethanol extract, and mixed solvent extract, respectively. Tyrosinase inhibitory activities in 1,000 ㎍/mL extract were 24.54%, 36.55%, and 15.23% for supercritical fluid extract, 70% ethanol extract, and mixed solvent extract, respectively. Elastase inhibitory activities in 1,000 ㎍/mL extract were 15.62%, 22.56%, and 26.64% for supercritical fluid extract, 70% ethanol extract, and mixed solvent extract, respectively. Skin astringent activity (81.23% with 5000 ㎍/mL extract) was only detected in the 70% ethanol extract. Supercritical fluid and mixed solvent extracts showed no such activity. Our analysis of Pinus koraiensiscone scale extracts show that the highest aggregate activity was found in the 70% ethanol extract, followed by mixed solvent and supercritical fluid extracts. Therefore, our results oppose the hypothesis stating supercritical extract has the highest total polyphenol content and antioxidant activity.

Supercritical Carbon Dioxide Extraction of Beef and Pork for Low Lipid Sausage Manufacturing (저지방 소시지 제조를 위한 쇠고기와 돼지고기의 초임계 이산화탄소 추출)

  • Lipid and cholesterol were extracted from beef and pork by the supercritical carbon dioxide $(SC-CO_2)$ for the manufacturing of low-lipid and low-cholesterol sausage. The ranges of extraction temperature and pressure were from 35 to $55^{\circ}C$ and from 103 to 375 bar, respectively. $SC-CO_2$ extraction yield of beef lipid increased as extraction pressure increased and/or extraction temperature decreased, while extraction temperature was more influential on the cholesterol extraction than pressure condition. When lipid and cholesterol of freeze-dried beef with varied moisture contents were extracted, their solubilities increased as the moisture content reduced. The extraction of lipid and cholesterol from pork was shown the same tendency as the beef. The chunk type of beef shape was more suitable for the lipid and cholesterol extraction than the powder type of beef. The color of meat after $SC-CO_2$ extraction was lighter than the raw freeze dried meat because of the extraction of color pigments. After $SC-CO_2$ extraction, beef and pork were rehydrated and mixed with raw beef and pork containing lipid and cholesterol. Their mixing ratio up to 50 : 50 did not affect physical properties of the sausage compared with the control sausage.

  • The volatile components of citron peel extracted by three methods, which were SDE (Simultaneous steam distillation & solvent extraction) extraction, solvent extraction and supercritical carbon dioxide extraction were analyzed with gas chromatography. The operation conditions was at the temperature between $30^{\circ}C$ and $45^{\circ}C$ and perssure between 1000 psi and 2500 psi. The aromatic compound was the principal component extracted, the optimum conditions for limonene extraction were 2000 psi and $40^{\circ}C$.

  • Curcuma longa L. of Zingiberaceae family, has been reported to have radical scavenging activity and antiinflammatory effect. On these facts, biological activity and whitening effect were examined to evaluate a bioactivity of the extracts of Curcumma longa L. by extraction conditions as a possible pharmaceutical material. When compared extraction methods, supercritical extraction was the best showing various biological activities and whitening effect. The highest electron donating ability $(89%\;at\;1,000{\mu}g/ml)$ were detected and SOD-like activity was inhibited up to $95%\;at\;500{\mu}g/ml$ in supercritical extraction. To evaluate the whitening effect, tyrosinase inhibitory activity was conducted 89% of tyrosinase inhibitory effect examined at $1,000\;{\mu}g/ml$. From these results, we suggest that the supercritical extracts from Curcuma longa L. is useful for cosmetic ingredients.

Extractions of Surface-Active Substances from Defatted Rapeseed Meal (Brassica napus L.) by Supercritical Carbon Dioxide (초임계 CO2 유체 추출법을 이용한 탈지 유채박 중 표면활성물질 추출의 최적화)

  • In this study, an attempt is being made to extract surface-active substances from defatted rapeseed cakes by supercritical carbon dioxide fluid. Independent variables for the extraction process, being formulated by D-optimal design, are pressure (150~350 bar), temperature ($33{\sim}65^{\circ}C$ and co-solvent (ethanol, 50~250 g). The dependent variables of the extraction yield, the content of neutral lipids, phospholipids and glycolipids in the extracts were analyzed upon the results through the response surface methodology. As for the extraction yield, it was found to increase with increasing independent variables, among which the co-solvent proved to be a major influencing parameter. Similar trends were found for the content of surface-active substances (i.e, phospholipids and glycolipids) in the extracts, except for the content of neutral lipids. Regression equations were suggested to coincide well with the results from the experiments. Extraction conditions are being optimized to maximize the extraction yields, the content of phospholipids, and glycolipids were 350 bar (pressure), $65^{\circ}C$ (temperature) and 228.55 g (co-solvent), respectively.

Concentration of Polyunsaturated Fatty Acids from Anchovy Oil by Supercritical Carbon Dioxide (초임계 이산화탄소에 의한 멸치어유의 고도불포화지방산 농축)

  • To concentrate polyunsaturated fatty acids from anchovy oil by supercritical carbon dioxide $(SC-CO_2)$, effects of different adsorbents, cosolvents and extraction pressures on the extraction yield and fatty acid composition in the extract and the residue fractions were investigated. Anchovy oil mixed with silver nitrate-coated silica gel showed higher extraction yield and concentration of EPA and DHA in the residual fraction than that mixed with only silica gel at $60^{\circ}C/345{\;}bar$. Ethyl acetate was a promising cosolvent for concentrating polyunsaturated fatty acids in the residual fraction from anchovy oil mixed with silica gel. For the extraction pressures tested, 276 bar showed a maximum value in the extraction yield and concentration of EPA and DHA in the residue. Starting with anchovy oil containing 13.3% EPA and 16.2% DHA mixed with a silver nitrate-coated silica gel, the residue fraction containing 28.2% EPA and 38.3% DHA was obtained when ethyl acetate was used as a cosolvent with $SC-CO_2$ at $60^{\circ}C/276{\;}bar$.

Studies on the Recovery of Triglyceride from Used Shortening by Supercritical Fluid Extraction (초임계유체 추출에 의한 폐식용유의 재활용에 대한 기초연구)

  • Supercritical fluid extraction was applied to recycling triglyceride from used shortening. Used shortening and its fractions were analyzed with high performance size exclusion chromatography for their composition in triglycerides, polymer and low molecular weight compounds. Conjugated diene value and color of the fractions were also measured with a UV spectrophotometer and a colorimeter, respectively. Pressure and temperature ranges employed were $15{\sim}30$ MPa and $40{\sim}60^{\circ}C$, respectively. Concentration of fat in supercritical (SC) $CO_2$ ranged from $0.3\;X\;10^{-3}{\sim}7.4\;X10^{-3}(g\;fat/g\;CO_2)$. An exponential relation between concentration of fat in SC $CO_2$ and density was observed. Color of the extracts was light yellow which was very close to that of the fresh shortening. Low molecular weight compounds were preferentially concentrated in the initial fraction, while polymer was extracted in the final fraction. Conjugated diene value of the initial fractions was clearly lower than that of feed. It increased sharply as the polymer content in the fraction became significantly large.

  • This study was carried out to investigate the emulsifying properties of surface-active substances from defatted rapeseed cake by supercritical $CO_2$ extraction. Based on the interfacial tension data, a supercritical fluid extract (SFE) with the lowest value of 14.16 mN/m was chosen for evaluation which was obtained from No. 2 extraction condition (150 bar, $65^{\circ}C$, 250 g). For emulsions with SFE, some physicochemical properties (i.e., fat globule size, creaming stability, zeta potential etc) were investigated according to changes in SFE concentration, pH, and NaCl addition in an emulsion. It was found that fat globule size was decreased with increasing SFE concentration in emulsion, with showing a critical value at 0.5 wt%, thereby resulting in less susceptibility to creaming behavior. The SFE emulsion also showed instability at acidic conditions (pH<7.0) as well as by NaCl addition. This was coincided with zeta potential data of emulsion. In addition, SSL (sodium stearoyl lactylate) found to be suitable as a co-surfactant, as it helped considerably in decreasing fat globule size in emulsions and its optimum concentration to be over 0.03 wt%, based on 0.1 wt% SFE in emulsion.
