Improved HPLC-UV method for determination of five synthetic dyes in Typha orientalis

  • Synthetic azo dyes are used extensively in herbal medicines to render the medicines more visually attractive to consumers. This study developed and validated a rapid high-performance liquid chromatography (HPLC) method to determine whether synthetic colorants such as Tartrazine, Auramine O, Metanil yellow, Sunset yellow, and Orange II are used extensively in Typha orientalis. To increase the recovery of the synthetic dyes, this method employed containing 50 mM ammonium acetate in 70 % methanol at first extraction and 100 mM HCl in 70 % methanol at second extraction. Five synthetic pigments in Typha orientalis were separated by gradient elution with a mobile phase consisting of acetonitrile and 50 mM ammonium acetate in distilled water at ultra-violet (UV) detection 428 nm or 500 nm. Additionally, this study established the liquid chromatography tandem mass spectrometry (LC-MS/MS) method to confirm positive samples suspected by HPLC results. The HPLC-UV method had good linearity, indicating r2> 0.999. The recoveries of the samples spiked with three different concentration ranged from 73.8~91.5 %, and relative standard deviation values indicated 0.2~5.2 %. The established LC-MS/MS could successfully identify the synthetic pigments in herbal medicine samples. The study demonstrates that Typha orientalis adulterated by yellowish synthetic dyes can be successfully distinguished when using the HPLC-UV method.

A Study of Private Development of Long-Term Unexecuted Urban Parks through a Special Act - Difficulties and Solutions of Pohang - (장기미집행 도시공원의 특례법에 따른 민간공원사업 특성연구 - 포항시의 사업추진 어려움과 해결방안 -)

  • This is a case study of private sector's development of long-term unexecuted urban parks in Pohang through a special act, which provides a guideline of 30% of land for non-park while 70% of land for park. The strategy has a lot of validity in many respects along with the urban planning sunset system, but even with the special act it would not easy to be implemented in Pohang, where the guideline has been modified to 20% for non-park, mostly multi-family housing projects while 80% for park. Thus, participation of private companies would be discouraged due to low commercial validity. Also, there would exist various risks because the project would be completed through a long-term decision-making and execution process. Thus, this study argues that it would be better for Pohang to follow the original guideline of the government for better implementation of the projects, along with preparation of a law with which the government be able to recoup excess profits when too much profits would be given to private developers. For the project implemented smoothly, it is also important to understand local housing market and fluctuating economic conditions, and to prepare various incentives for private companies. In addition, to secure publicity, guidelines on the level of publicity of the project should be prepared through negotiation by parties to prevent the project being discouraged too much.

Astronomical Phenomenon Records from Sukjong's Chunbang-Ilgi

  • We investigate the astronomical phenomenon records of Sukjong's Chunbang-Ilgi made by Sigangwon (Royal Educational Office of the Crown Prince) at which King Sukjong was the crown prince (i.e., 1667 January 24-1674 September 22). From the daily records of 2,799 days, we extract the astronomical records of 1,443 days and classify them into 14 categories. Then, we group the records of each category into five phenomena (Atmosphere, Eclipse, Daylight Appearance, Apparition, and Appulse) and compare them with the results of modern astronomical computations wherever possible. Except for Atmosphere group comprising records of meteorological events, such as solar halo, lunar halo, and unusual clouds, the significant findings in every other group are as follows: In Eclipse group, the solar eclipse that occurred on 1673 August 12 was unobservable in Korea, which is in contrast to the record of Joseonwangjo-Sillok (Annals of the Joseon Dynasty), which states that the sun was in eclipse around sunset time, as observed at Nam mountain. From the lunar eclipse records, we verify that the Joseon court did not change the date of the events observed after midnight. In Daylight Appearance group, we confirm that this phenomenon was observed during the daytime and not during twilight. We further suggest that if observation conditions are met, a celestial body brighter than -2.3 mag could be seen during the daytime with the naked-eye. In Apparition group, we find the possibilities that the Orionid meteor shower had influence on the meteor records and the seasonality on the aurora records. We also find that the Korean records in which the coma of comet C/1668 E1 was located below the horizon were overlooked in previous studies. Finally, we find that the records of Appulse group generally agree with the results of modern calculations. The records of Beom (trespass in literal) and Sik (eating in literal) events show average angular separations of 1.2° and 1.0°, respectively. In conclusion, we believe this work helps study the astronomical records of other logs of Sigangwon, such as Sukjong's Chunbang-Ilgi.

Analysis of a forest healing environment based on the thermal comfort and NVOC characteristics of Chungnam National University Experimental Forests

  • The purpose of this study is to provide information on a forest healing environment using the analysis of nature volatile organic compounds (NVOCs) and thermal comfort in Chungnam National University Experimental Forest, with the aim of using the Experimental Forest as a healing environment for health promotion. We analyzed NVOCs and thermal comfort of Chungnam National University Experimental Forest measured on September 12th, 2021. As a result of the NVOC analysis, a total of seven substances were detected, mainly including alpha pinene and beta pinene. The detection amount for each time period was highest at the time of sunset. The thermal comfort was analyzed by time-dependent changes and changes according to clothing and exercise amount. The results showed that the predicted mean vote of the experimental forest is within the range of 'slightly cool' and 'slightly warm' sensation, and thus a comfortable thermal environment could be controlled by the amount of clothing and activity. Based on the analysis, this study provides information on the healing environment of the experimental forest at Chungnam National University. It also indicates that the forest can be used as a health promotion and healing environment with thermal comfort by composing a physical activity program of appropriate intensity for each time period.

The Characteristics and Seasonal Variations of OC and EC for PM2.5 in Seoul Metropolitan Area in 2014 (서울지역의 PM2.5 중 OC와 EC의 특성 및 계절적 변화에 관한 연구)

  • To investigate characteristics and seasonal variations of carbonaceous species for $PM_{2.5}$ in Seoul metropolitan area, Korea, we measured organic carbon (OC) and elemental carbon (EC) from January 2014 to December 2014 using a semi-continuous OC/EC Analyzer (Model-4, Sunset Lab.). Mean concentrations of OC and EC were estimated $4.1{\pm}2.7{\mu}g/m^3$ and $1.6{\pm}1.0{\mu}g/m^3$, respectively. The annual averaged OC/EC ratio was $2.9{\pm}2.7$. Concentrations of OC and EC comprised 13% and 5% of $PM_{2.5}$ and the mass fraction of both was the highest in fall. OC and EC showed similar trend in seasonal variations. Concentrations of those showed a clear seasonal variation with the highest in winter and the lowest in summer. The correlations between the two were the best during the winter ($r^2=0.88$). As results of carbonaceous species analysis, the dominant factor in view of fine particle ($PM_{2.5}$) is primary emission source such as mobile, fossil fuel combustion during commute time(08:00~10:00 or 17:00~21:00) and winter season. Continuous monitoring of atmospheric carbonaceous species is essential to provide the science-based data to policy-maker establishing the air quality improvement policy.

Errors in Net Ecosystem Exchanges of CO2, Water Vapor, and Heat Caused by Storage Fluxes Calculated by Single-level Scalar Measurements Over a Rice Paddy (단일 높이에서 관측된 저장 플럭스를 사용할 때 발생하는 논의 이산화탄소, 수증기, 현열의 순생태계교환량 오차)

  • Using eddy covariance method, net ecosystem exchange (NEE) of $CO_2$ ($F_{CO_2}$), $H_2O$ (LE), and sensible heat (H) can be approximated as the sum of eddy flux ($F_c$) and storage flux term ($F_s$). Depending on strength and distribution of sink/source of scalars and magnitude of vertical turbulence mixing, the rates of changes in scalars are different with height. In order to calculate $F_s$ accurately, the differences should be considered using scalar profile measurement. However, most of flux sites for agricultural lands in Asia do not operate profile system and estimate $F_s$ using single-level scalars from eddy covariance system under the assumption that the rates of changes in scalars are constant regardless of the height. In this study, we measured $F_c$ and $F_s$ of $CO_2$, $H_2O$, and air temperature ($T_a$) using eddy covariance and profile system (i.e., the multi-level measurement system in scalars from eddy covariance measurement height to the land surface) at the Chengmicheon farmland site in Korea (CFK) in order to quantify the differences between $F_s$ calculated by single-level measurements ($F_s_{-single}$ i.e., $F_s$ from scalars measured by profile system only at eddy covariance system measurement height) and $F_s$ calculated by profile measurements and verify the errors of NEE caused by $F_s_{-single}$. The rate of change in $CO_2$, $H_2O$, and Ta were varied with height depending on the magnitudes and distribution of sink and source and the stability in the atmospheric boundary layer. Thus, $F_s_{-single}$ underestimated or overestimated $F_s$ (especially 21% underestimation in $F_s$ of $CO_2$ around sunrise and sunset (0430-0800 h and 1630-2000 h)). For $F_{CO_2}$, the errors in $F_s_{-single}$ generated 3% and 2% underestimation of $F_{CO_2}$ during nighttime (2030-0400 h) and around sunrise and sunset, respectively. In the process of nighttime correction and partitioning of $F_{CO_2}$, these differences would cause an underestimation in carbon balance at the rice paddy. In contrast, there were little differences at the errors in LE and H caused by the error in $F_s_{-single}$, irrespective of time.

A Local Governments' Preferences in Selecting Modern Eight Scenic Landscapes (지자체가 선정한 현대팔경에 나타난 경관 선호 양상)

  • The followings are the landscape preference aspects from the 816 landscapes(景, Kyung), which comprise the 78 modern Palkyungs, presented by the 78 local governments in Korea. First, the natural environment elements selected as Kyung(景), which are topographical landscapes, mostly consist of mountain elements such as mountains, terrace(臺), rocks and stones and water elements classified as rivers, oceans, and lakes. Natural elements also include old-growth and giant trees such as pines, ginkgos, Japanese cornels and fringe trees, tree-lined streets and forests, and plant elements such as azaleas, rhododendrons, lotuses, reeds, and silver grasses which provide seasonal landscapes. Second, more than half of Kyung, selected as human environment elements, are historical and cultural heritages such as graveyards, mountain fortresses, town fortresses, traditional villages, pavilion in villas, and temples. And it is followed by leisure tourism facilities such as traditional markets, exhibition halls, theme parks, beaches, and food streets, green-based structures such as trails, plazas, parks, and botanical gardens, and industrial heritages such as ranches, abandoned coal mines, stations, ports and bridges. Third, modern Palkyungs include objects not related to the views such as local representative facilities, regional products, and festivals. Fourth, although most of the modern Palkyungs consist of eight, some include 20, 38, or 100 in order to increase the number of objects of public relations. Fifth, a certain local government makes two modern Palkyungs with different subjects by introducing traditional Palkyung and modern Palkyung altogether. In this case, it presents several modern Palkyungs like by selecting Palkyungs in a limited area. Furthermore, one Palkyung includes numerous place names at a time in some cases. Sixth, Sosangjeonhyeong(瀟湘典型)-style modern Palkyung uses 'NakAn(落雁)' as the name of Kyung. Sosangyusahyeong(瀟湘類似型)-style modern Palkyung expresses 'Hyojong(曉鐘)' and landscape of glow of the setting sun, sunset, night view, dawn, sunrise and depicts cloud, sunset, moon, and snow. There are many Myeongsocheheomhyeong(名所體驗型)-style Palkyungs exhibiting the behavior of tourism and Myeongseunghyeong(名勝型)-style Palkyungs raising the awareness only by the names of the places. Seventh, modern Palkyung's naming styles are diverse, such as using only four letters instead of specifying Kyungmul(景物) or Kyungsaek(景色) in combination with Chinese characters or adding modifiers specializing in places.

The Private-Initiated Park Development Project in Terms of Securing Publicity Operation Characteristics Analysis - Busan Metropolitan City as a Case - (공공성 확보측면에서 민간공원특례사업 운영특성분석 - "부산광역시를 사례로" -)

  • This study examines the process of project promotion in Busan, which introduced the private participation-type consultative body for the first time in the country in the promotion stage of the private-initiated park development project, and introduced flexible application of the system and differentiated policy elements in the implementation process, and examines the operational characteristics and we tried to analyze performance, etc. As a result of the analysis, first, the preferred bidder was selected by introducing a mixed method in the project method, which is an independent project method that cannot be seen in other local governments. Second, by specifying guidelines considering the characteristics of each park and detailed guidelines such as the location, area, and maximum height of non-park facilities, criteria for establishing a rational development plan utilizing regional identity and the basis of evaluation standards were laid. Third, in the project process, transparency was secured through the delegation-type roundtable, in which the private sector performs the functions and roles of key actors, thereby preventing disputes such as suspicion of preferential treatment. Fourth, in order to improve the quality of park facilities to be donated and to secure design adequacy, after approval of the implementation plan, a general planner was introduced and construction project management (design stage) services were performed to promote efficient implementation and specialization of luxury parks in the region. As a result, the city of Busan carried out the project efficiently by conserving 5 parks from sunset, a park area of 2.25km2, and reducing land compensation and park construction costs by KRW 740 billion. Reinforcement of the public nature of the private-initiated park development project was suggested. However, due to the application of these systems and verification procedures, the project period is prolonged, and park services are delayed along with the financial burden on private operators.

Legal Issues of "Zeroing" Practice Based on the Article 2.4.2 of the WTO Anti-Dumping Agreement (WTO 반덤핑협정 제2.4.2조에 의거한 네거티브 덤핑마진 산정 방식("제로잉")의 법적 문제)

  • This paper intends to analyse some legal issues on "Zeroing" which is based on the article 2.4.2 under the WTO Anti-dumping Agreement. "Zeroing" stands for a specific methodology in calculating a general dumping margin for a product in question under which negative individual dumping margins are treated as zero (thus "zeroed") before aggregating all individual dumping margins. The article 2.4.2 of the Anti-dumping Agreement regulates three types of calculating methodology on dumping margin as first symmetrical method(average-to-average: A-A), second symmetrical method(individual-to-individual: I-I) and asymmetrical method(average-to-individual: A-I). However, this article does not have any provisions about the "Zeroing" practice. In their anti-dumping practices, the EC and the United-States calculated dumping margin based on the "Zeroing", but this methodology has been disputed in the Dispute Settlement Body(DSB) of the WTO. This paper analysed their legal problems with some WTO cases in particular concerning EC-Bed Linen, U.S.-Softwood Lumber Zeroing, U.S.-Zeroing(EC) and U.S.-Sunset Review(Japan) cases. On the basis of theses analysis, we can therefore ask some questions as follows; To begin with, although the article 2.4.2 of the WTO Anti-dumping Agreement does not clearly refer to the "Zeroing", how do some developing countries, as the U.S.A and the E.U. calculate dumping margin as the "Zeroing"? Secondly, what is the relationship between the symmetrical method and asymmetrical method to the dumping margin? And if we adopt the zeroing method, what is the different rate to anti-dumping margin? Thirdly, although the Panel decided that the zeroing methodology of dumping margin used by th U.S.A in administrative review between the U.S.A and the E.U, why does the Appellate Body made the decision that the american methodology is incompatible with the WTO Anti-dumping Agreement? Lastly, what will be affected the upper decision taken by the Appellate Body to the DDA negotiation of anti-dumping matters? Even though the WTO Appellate made a decision that the zeroing method is incompatible with the principles of the WTO law, this methodology contains a lot of problems. Some members of the WTO as the U.S.A and the E.U did not officially declare this methodology to abandon, and the debate concerned is arguing. Therefore this paper tried to present the adequate solution in order to promote the zeroing methodology in the international anti-dumping system and practices.

Biomaterials Inhibiting Biofilm Formation of Staphylococcus aureus (생물소재를 이용한 황색포도상구균의 바이오필름 억제 연구)

  • Biofilms are surface-attached microbial communities with phenotypic and biochemical properties distinct from free-living planktonic cells. Biofilm bacteria show much greater resistance than planktonic counterparts and much higher concentration of biocide is needed to treat biofilms compared to the dosage used for planktonic bacteria. As a result, alternative strategies or more effective agents exhibiting activity against biofilm-producing micro-organisms are of great interest. Therefore, we turned our attention to control of biofilm of S. aureus. The aims of this research are to investigate substances which inhibit the formation of biofilm by S. aureus and to suggest effective materials for controlling skin problems. We coated slide glasses with human placental collagen and the coverslip was incubated with test materials and bacteria. The coverslip was stained with crystal violet and we measured optical density of each sample. The biofilm inhibitory activity was calculated by crystal violet staining degrees. In this study, S. aureus ATCC 6538 was used as test organism. Our results show that both water soluble and insoluble Hinoki cypress polysaccharide strongly inhibited biofilm formation. Whereas, green tea and sunset hibiscus root extract promoted biofilm. Xylitol showed a concentration dependent effect; high concentration (3 % and 5 %) of xylitol reduced biofilm while promoted biofilm formation at a concentration of 1 %. These results support that Hinoki cypress polysaccharide and xylitol have ability to suppress biofilm formation.
