Changes in the Contents of Prosapogenin in the Red Ginseng (Panax ginseng) Depending on Steaming Batches

  • This study compared the contents of ginsenosides depending on steaming conditions of red ginsengs to provide basic information for developing functional foods using red ginsengs. The red ginseng steamed eight times at $98^{\circ}C$ ranked atop the amounts of prosapogenins ever detected in red ginsengs (ginsenoside $Rg_2$, $Rg_3$, $Rg_5$, $Rg_6$, $Rh_1$, $Rh_4$, $Rk_1$, $Rk_3$, $F_1$, $F_4$, 1.15%) among red ginsengs steamed more than twice. When steamed eight times at $98^{\circ}C$, 2.7 times as much prosapogenins such as ginsenosides $Rg_2$, $Rg_3$, $Rg_5$, $Rg_6$, $Rh_1$, $Rh_4$, $Rk_1$, $Rk_3$, $F_1$, and $F_4$ as those steamed just once at $98^{\circ}C$ was collected. In addition, the red ginsengs steamed eight times at $98^{\circ}C$ contained more amounting ginsenoside $Rg_3$ (0.28%) than that in the red ginseng steamed several times at random. Accordingly, it is recommendable that red ginsengs steamed 8 times, which proved to be the optimal steaming condition, be used rather than those steamed 9 times (black ginsengs), in order to develop red ginseng products of high prosapogenin concentration and high functions.

KRG and its major ginsenosides do not show distinct steroidogenic activities examined by the OECD test guideline 440 and 456 assays

  • Background: Ginseng has been used as a traditional medicine for treatment of many diseases and for general health maintenance. Previously, we showed that ginseng did not demonstrate estrogenic property in ovariectomized mouse model. However, it is still possible that disruption of steroidogenesis leading to indirect hormonal activity. Methods: The hormonal activities were examined in compliance with OECD guidelines for detecting endocrine disrupting chemicals: test guideline (TG) No. 456 (an in vitro assay method for detecting steroidogenesis property) and TG No. 440 (an in vivo short-term screening method for chemicals with uterotrophic property). Results: Korean Red Ginseng (KRG) and ginsenosides Rb1, Rg1, and Rg3 did not interfere with estrogen and testosterone hormone synthesis as examined in H295 cells according to TG 456. KRG treatment to ovariectomized mice did not show a significant change in uterine weight. In addition, serum estrogen and testosterone levels were not change by KRG intake. Conclusion: These results clearly demonstrate that there is no steroidogenic activity associated with KRG and no disruption of the hypothalamic-pituitary-gonadal axis by KRG. Additional tests will be performed in pursuit of cellular molecular targets of ginseng to manifest mode of action.

Ginseng total saponin modulates podocyte p130Cas in diabetic condition

  • Proteinuric conditions demonstrate structural and compositional changes of the foot processes and slit diaphragms between podocytes. p130Cas in podocytes serves as an adapter protein anchoring glomerular basement membrane to actin filaments of podocyte cytoskeleton. To investigate the effect of ginseng total saponin (GTS) on the pathologic changes of podocyte p130Cas induced by diabetic conditions, we cultured mouse podocytes under: 1) normal glucose (5 mM, control); 2) high glucose (HG, 30 mM); 3) advanced glycosylation endproducts (AGE)-added; or 4) HG plus AGE-added conditions and treated with GTS. In confocal imaging, p130Cas colocalized with zonula occludens-1 and synaptopodin connecting to F-actin. However, diabetic conditions relocalized p130Cas molecules at perinuclear cytoplasmic area and reduced the intensity of p130Cas. In Western blotting, diabetic conditions, especially HG plus AGE-added condition, decreased cellular p130Cas protein levels at 24 and 48 h. GTS improved such quantitative and qualitative changes. These findings imply that HG and AGE have an influence on the redistribution and amount of p130Cas of podocytes, which can be reversed by GTS.

Change of Neutral Ginsenoside Contents in Red and Fresh Ginseng (Panax ginseng C. A. Meyer) by Hydrolysis (가수분해 처리에 의한 홍삼과 인삼의 중성 Ginsenoside 함량 변화)

  • This study was carried out to investigate change of ginsenoside contents in red and fresh ginseng according to root part and age by hydrolysis. Neutral total ginsenoside contents by hydrolysis in 6-year main root and lateral root were significantly increased than those by non-hydrolysis, as 41.6 and 32.8%, respectively. However, there was no significant difference in red ginseng. In fresh ginseng, ginsenoside contents of the protopanaxatriol group such as Re, Rf, $Rg_1$, $Rg_2$, and $Rh_1$ were not significantly different, but $Rb_1$, $Rb_2$, $Rb_3$, Rc, and Rd showed significant difference. The increase rate of neutral total ginsenoside content by hydrolysis was higher in epidermis-cortex than stele. Also, the neutral total ginsenoside content was fine root > rhizome > lateral root > main root, respectively. While there was no tendency towards the increase of ginsenoside by hydrolysis with the increase of root age in fine root and rhizome, there was significant decrease in main root and lateral root.

  • This study was conducted to investigate the difference of general feature and ginsenoside content of 6 years old ginseng root among different grade of roots. Total weight of a 1st grade-6 years old ginseng root was 115.1g and weight, length, diameter and specific gravity of main root were 64.68g, 8.39cm, 3.31cm and 0.96, respectively. Main root of 1st grade ginseng root was larger in size and specific gravity and more heavy than that of End or 3rd grade of the roots. Though crude saponin contents were not so different among the different grade of roots, but ginsenoside Rb1, Rg1 and Re content were higher in 1st grade of root than that of 2nd or 3rd grade of root. Those ginsenosides were located mainly in periderm and cortex.

  • To obtain useful data on root rot in Korean ginseng, we performed phylogenetic analysis and pathogenicity test for Cylindrocarpon destructans isolated from peony. Cylindrocarpon destructans isolates from peony were proven to cause ginseng root rot. The isolate KACC44663 was identified as Ilyonectria robusta under the new classification system, which belongs to the I. radicicola species complex. This is the first report of the pathogenic isolate, which was isolated from another host plant, but not ginseng, that can cause root rot disease on ginseng in Korea.

  • Ginseng saponins (a secondary metabolite, termed ginsenosides) are the principal bioactive ingredients of ginseng, and modification of the sugar chains may markedly change the its biological activity. One of soil bacteria having $\beta$-glucosidase (to transform ginsenoside $Rb_1$) activity was isolated from soil of a ginseng field in Daejeon. 16S rRNA gene sequence analysis revealed that the isolate belonged to the genus Cellulosimicrobium, with highest sequence similarity (99.7%) to Cellulosimicrobium funkei ATCC BAA-$886^T$. The strain, Gsoil 235, could transform ginsenoside $Rb_1$ into Rd, $Rg_3$ and 3 of un-known ginsenosides by the analyses of TLC, HPLC. By investigating its deglycosylation progress, the optimal broth for, $\beta$-glucosidase was nutrient broth (In 48 hours, almost ginsenoside $Rb_1$ could be transformed into minor ginsenosides). On the contrary, the optimal broth for growth was determined as trypic soy broth (TSB).

  • In order to facilitate the quality control of betaine from the fruits of Lycium chinense, we have developed a rapid and simple method for quantitative determination. Determination was achieved on a Discovery C18 column with an isocratic solvent system of 0.32% perfluoropentanoic acid aqueous-acetonitrile at a flow-rate of 0.5 mL/min and detected an ELSD. The method was reproducible with intra- and inter-day variations of less than 6% (R.S.D). The recoveries were in the range of 90.01~100.05%. The method turned out to be fast and simple, furthermore, to have a good selectivity and sensitivity for the quantity determination of betaine in the fruits of L.chinense.

  • To elucidate the effect of bioconversioned ginseng(Sun ginseng) and its butanol fraction on adenine-induced renal failure. rats were fed ad libitum on diet containing 0.75% adenine for 20 days to induce renal failure, and bioconversioned ginseng was orally administrated during the feeding period. On days 10 and 20, BUN, Creatinine, Ca and P contents were analyzed in serum and urine, and on days 20, BUN, Creatining, Ca and P contents were analyzed in serum and urine, and on days 20, blood pressure, hear pulse and relative kidney weight were measured.
