This study was initiated to evaluate ethanol production by a Korean isolate of white rot fungus Irpex consors. It was found that the fungus could produce ethanol by converting glucose, mannose, xylose, and cellobiose under semi-aerobic condition with yields of 0.23, 0.19, 0.21, and 0.17 g ethanol per g sugars, respectively. Furthermore, the strain produced ethanol by simultaneous saccharification and fermentation of rice straw treated with steam pressured boiling water, 3% NaOH, and 3% $H_2SO_4$ with maximum yields of 0.12, 0.15, and 0.19 g ethanol per g rice straw, respectively. These results suggested that I. consors could produce ethanol from the components of cellulose and hemicellulose including glucose, mannose, xylose, cellobiose as well as rice straw treated with steam pressured boiling water, dilute sodium hydroxide, and dilute sulfuric acid. This is the first report that I. consors mycelia produce ethanol from various sugars and lignocellulosic substance including rice straw.
The use of renewable energy sources is becoming increasingly necessary to minimize the problems derived from the global warming impacts caused by the utilization of fossil fuels as well as their limited supply and reservoir. Also, localized heavy rain has occurred in many areas. As a result, suspended wood waste is being inflow into the dam and the problem of waste disposal has occurred. It is a unique renewable and alternative source for the production of energy. The experiment using wood waste (dry weight 25.0g) was conducted for extraction sugars such as xylose, lactose and glucose. For the sugar extraction from wood waste, hydrolysis experiment using wood waste was conducted by two steps. First step was reacted with 72% sulfuric acid (24.0N and 37.5 ml) for 1hr at $30^{\circ}C$ and second step was reacted at $105^{\circ}C$ for one hour after adding 2.45times of hot water. Extracted sugar was used in the experiment of sugar consumption to estimate feasibility of ethanol production using yeast(P. Stipitis and S cerevisiae). As a result, sugar extracted from wood waste was effective consumed by yeast(P. Stipitis and S cerevisiae). The consumption rate by yeast was S. cerevisiae was faster than that of P. stipitis. It can be confirmed that resource as ethanol production using wood waste was available.
The study was conducted to investigate the effects of diatomearth(pH 3 and CEC 20me/100g) on the adjustment of nursery soil pH and on the growth of rice seedling in the tray. The results are as follows ; 1. The soil pH and damping-off of rice seedling were decreased with increase in diatomearth application. 2. Ten to fifteen percent of diatomearth was required to adjuse the optimum soil pH range of 4.5-5.5. 3. Changes in nursery soil pH adjusted by diatomearth were less than those by sulfuric acid during the incubation period. 4. Diatomearth application increased dry weight and height of rice seedling.
To determine levels of PCBs in food, beef, pork, chicken, egg, mackerel, yellow croaker, anchovy, common squid and little neck clam were chosen and collected at markets in Seoul, Busan and Kwangju. Among 209 PCB congeners, 7 congeners (#28, #52, #101, #118, #138, #153 and #180) were selected as target compounds that were known as indicator congeners. Samples were homogenized, treated in alkali solution for 1 hour, and extracted with organic solvents. After extraction, extracts were cleaned up by sulfuric acid, purified on silica gel column chromatography, analyzed by dual column-HRGC/ECD and then confirmed by HRGC/MSD. As results, PCBs were detected in fish samples ranged from 0.0002 to 0.001 mg/kg. Both PCB #101 and PCB #118 were the major contributors among 7 congeners.
The concentration and determination of trace phosphate ion was studied by $La(OH)_3$ coprecipitaiton. Phosphate ions in 1.0L samples were coprecipitated with lanthanium hydroxide at pH 9.5 adjusted with ammonia solution. The precipitates were floated with the aid of mixed surfactant(1:8 sodium oleate/sodium dodecyl sulfate) and nitrogen gas bubbles. The floated precipitate was collected in suction flask from the solution. The precipitate were washed with dil. ammonia solution and dissolved in sulfuric acid. The phosphate ion in the concentrated solution was finally determinated by UV/VIS spectrophotometry using the molybdenium blue method. The proposed method could be applied to the determination of phosphate ion in tap water and river water.

For the rapid and reliable detection of toxic compounds in water, a novel toxicity detection methodology based on sulfur-oxidizing bacteria (SOB) has been developed. The methodology exploits the ability of SOB to oxidize elemental sulfur to sulfuric acid in the presence of oxygen. The reaction results in an increase in electrical conductivity (EC) and a decrease in pH. Using a synthetic stream water (EC=0.12 mS/cm and pH=7.2), the baseline steady-state EC and pH values were 0.5~1.2 mS/cm and ~2.5 over 7 days of testing at HRT 30 minutes. When nitrite compounds were added to the system, the effluent EC decreased and the pH increased due to the inhibition of the SOB. Optimum HRT was 30 min and this HRT could be decresed by using smaller sulfur particles.

In this study, we developed the program to calculate the corrosion evaluation index for examining the corrosivity of raw water. When it was applied to the Han river and Nakdong river system, sulfuric acid ion, which accelerated corrosion, was higher in Nakdong river system than Han river system while calcium and hardness, which restrained corrosion, was the same way. Summarization of the LI and CCPP calculation result by the developed corrosion evaluation model showed that water quality of Han river system had strong tendency to corrode (is strongly corrosive). Moreover, this study evaluated the corrosivity of calcium carbonate saturation index by adding the chemicals to tap water. Saturation status was maintained in the order of $Ca(OH)_2$ > NaOH > ${Na_2}{CO_3}$ > $CaCO_3$ in the case of LI and RI.
Alcohol fermentation of uncooked naked barley was carried out by the combined action of the maceration enzyme from black Aspergillus niger and the glucoamylase from Rhizopus sp. The combined enzyme preparation was found to be effective in maceration and saccharification of the raw naked barley starch. The Hydrolysis rate measured by the amount of glucose liberated reached more than 70% at pH 4.5 and 3$0^{\circ}C$ after 76 hrs. For alcohol fermentation without cooking, the naked barley mash of 18% initial total sugar was pretreated with concentrated sulfuric acid (0.15 weight % of the mash volume) at 55$^{\circ}C$ for 2hr, and used for alcohol fermentation. A simultaneous saccharification and fermentation was carried out at pH 4.8 and 3$0^{\circ}C$ for 96 hrs. Under this fermentation condition, 3.5% increase in alcohol yield together with 2.0% increase in alcohol concentration were obtained when compared with the conventional cooking fermentation.
Glutathione reductase (GR, E.C. 1.6.4.2) is an important enzyme that reduces glutathione disulfide (GSSG) to a sulfydryl form (GSH) in the presence of an NADPH-dependent system. This is a critical antioxidant mechanism. Owing to the significance of GR, this enzyme has been examined in a number of animals, plants, and microbes. We performed a study to evaluate the molecular properties of GR (OsGR) from rice (Oryza sativa). To determine whether heterologous expression of OsGR can reduce the deleterious effects of unfavorable abiotic conditions, we constructed a transgenic Saccharomyces cerevisiae strain expressing the GR gene cloned into the yeast expression vector p426GPD. OsGR expression was confirmed by a semiquantitative reverse transcriptase polymerase chain reaction (semiquantitative RT-PCR) assay, Western-blotting, and a test for enzyme activity. OsGR expression increased the ability of the yeast cells to adapt and recover from $H_2O_2$-induced oxidative stress and various stimuli including heat shock and exposure to menadione, heavy metals (iron, zinc, copper, and cadmium), sodium dodecyl sulfate (SDS), ethanol, and sulfuric acid. However, augmented OsGR expression did not affect the yeast fermentation capacity owing to reduction of OsGR by multiple factors produced during the fermentation process. These results suggest that ectopic OsGR expression conferred acquired tolerance by improving cellular homeostasis and resistance against different stresses in the genetically modified yeast strain, but did not affect fermentation ability.

Ion exchange fibers, high functionalized onto hybrid polyolefine fiber's surface, were synthesized by $\gamma-ray$ mutual radiation. Degree of grafting (DG) of copolymer increased with increasing GMA monomer concentration and the maximum rate of DG was 355% at 50 GMA. The graft reaction occurred in polar solvent and DG was 190% maximum value in $1.0\times10^{-3}$ Mohr's salt and 0.1 M sulfuric acid, respectively. The amination for graft copolymers varied depending on amine reagents, and the reactivity for copolymers was highest for methylamine, and that of triethylamine lowest. It was shown that water uptake and ion exchange capacities increased with increase in the rate of amination while surface area decreased rapidly as proceeding for graft reaction and amination.
