Transport of Selected Veterinary Antibiotics (Tetracyclines and Sulfonamides) in a Sandy Loam Soil: Laboratory-Scale Soil Column Experiments (토양컬럼을 이용한 테트라사이클린계 및 설폰아마이드계 항생물질의 이동특성 평가)

  • Antibiotics are biologically active substances and commonly used for therapeutic treatment of infectious disease in humans and for treating and protecting the health of animals. In recent years, antibiotics have attracted worldwide attention because of their side effects on the environment. Consequently, efforts have been made to monitor the residual of antibiotics in the environment. This study tested the mobility of tetracyclines and sulfonamides in soil and leachate through column experiments. The three tetracycline antibiotics showed higher mass recovery rates in all kinds of soils(28.00~44.11%) than in leachate(10.54~27.43%). This seems attributable to the high adsorption coefficient values($K_d$) of tetracyclines representing strong and active adsorbability to organic and mineral phases in soil, ending up relatively small amount being detected in surface water. By contrast, the sulfonamides(sulfamethazine and sulfathiazole) showed higher mass recovery rates in leachate(23.19~26.20%) compared to in soil(10.41~14.21%) due to lower adsorption coefficient values and higher mobility of sulfonamides, enabling easier movement to surface water through the runoff in the environment.

  • Betacyclodextrin 과 수종의 sulfonamide와의 상호반응을 solubility method로 고찰하였다. 본실험에서 사용한 betacyclodextrin의 농도 범위에서는 sulfonamide의 solubility 증가와 batacyclodextrin의 농도와의 관계 함수관계에 있음을 나타내었다.열역학적인 해석을 시도하여 본반응의 formation constant, free energy, enthalpy 및 entropy를 개측하였다.

A study on simultaneous determination of residual sulfonamides in livestock productions by high performance liquid chromatography (HPLC를 이용한 축산식품중 잔류 설폰아미드제의 동시분석법 연구)

  • This study was carried out to explore the most sensitive and useful method for simultaneous determination of five sulfa drugs(sulfamethazine, sulfamerazine, sulfamonomethoxine, sulfadimethoxine, sulfaquinoxaline) in livestock productions(pork muscle, bovine muscle, chicken muscle, milk ) by HPLC with UV detector and reverse phase column. The results obtained were as follows:1. For mobile phase acetonitrile-0.01M ammonium acetate (23:77) showed more applicable sensitivity and retention times than acetonitrile-1% acetic acid(23:77). Thus acetonitrile-0.01M ammonium acetate(23:77) selected and applied to the modification test, from which it was found pH 6.75 was the most adequate. 2. Optimal wavelength of UV for SMT(sulfamethazine), SMR(sulfamerazine), SMM(sulfamonomethoxine), SD(sulfadimethoxine), and SQ(sulfaquinoxaline) were 266, 266, 265, 269 and 250nm, respectively, and that for simultaneous application it was 263nm. 3. The average recovery rate by extractant(chloroform, dichloromethane, chlorform+dich-loromethane) in the classic method was not significantly different(p>0.05) but that by chloroform higher than the others. Thus chloroform was found to be adequate as extractant in this classic method. 4. The average recovery rate was 86.5% by the MSPD(matrix solid phase disperse) method, which was significantly higher than that by the classic method(p<0.05). Also the recovery rates by method were significantly different(p<0.05) in accordance with sample and type of drug. The MSPD method was much superior to classic method on clean-up.

Studies on the detection of sulfonamide residues in swine edible tissues (돈육내 sulfonamides의 잔류물질 검출에 관한 연구)

  • The study was carried out to determine the residues of sulfonamides in swine edible tissues with high performance thin layer chromatography. For this purpose, the Rf values of sulfonamides in various solvent systems and the recovery rate of sulfameathazine from sampike saples were obtained. Thirty-four samples collected from meat market in Seoul were analyzed. The results obtained from the present study were followings: 1. The average recovery rate of sulfamathazine residues from spiked tissues 0.05, 0.1, 0.5 and 1mg/kg sample weight was 85%. 2. Two of 34 samples of pork for domestic consumption were reported to have been exceeded 0.05 ppm in sulfamethazine residues degree. 3. On the basis of the results, the degree of residues of sulfamethazine in swine meat for domestic consumption is seemed not to be dangerous for public health.

Simultaneous Determination of Sulfonamides in Porcine and Chicken Muscle Using High Performance Liquid Chromatography with Ultraviolet Detector

  • The present study used the liquid extraction pretreatment method and developed a liquid chromatographyultraviolet detector (LC-UV) for the simultaneous determination of 14 sulfonamides (SAs) residues in porcine and chicken muscle. Linearity within a range of $50-150\;{\mu}g/kg$ was obtained with the correlation coefficient ($r^2$) of 0.9673-0.9997. The mean recovery of SAs was 55.9-109.7% (relative standard deviations; RSDs 1.7-17.3%) in porcine muscle and 52.8-112.4% (RSDs 2.3-16.9%) in chicken muscle. The limits of detection (LODs) and limits of quantification (LOQs) were 2-32 and $7-96\;{\mu}g/kg$ in porcine muscle, and 4-32 and $13-97\;{\mu}g/kg$ in chicken muscle, respectively. These values were lower than the maximum residue limits (MRLs) established by the European Union. The sum of all SAs residues present should be less than $100\;{\mu}g/kg$.

Cytotoxic Activity and Quantitative Structure Activity Relationships of Arylpropyl Sulfonamides

  • B13 is a ceramide analogue and apoptosis inducer with potent cytotoxic activity. A series of arylpropyl sulfonamide analogues of B13 were evaluated for their cytotoxicity using MTT assays in prostate cancer PC-3 and leukemia HL-60 cell lines. Some compounds (4, 9, 13, 14, 15, and 20) showed stronger activities than B13 in both tumor cell lines, and compound (15) gave the most potent activity with $IC_{50}$ values of 29.2 and 20.7 ${\mu}M$, for PC-3and HL-60 cells, respectively. Three-dimensional quantitative structure-activity relationship (3D-QSAR) analysis was performed to build highly reliable and predictive CoMSIA models with cross-validated $q^2$ values of 0.816 and 0.702, respectively. Our results suggest that long alkyl chains and a 1R, 2R configuration of the propyl group are important for the cytotoxic activities of arylpropyl sulfonamides. Moreover, the introduction of small hydrophobic groups in the phenyl ring and sulfonamide group could increase biological activity.

Analysis of antimicrobial residues in meat samples from slaughtered livestock in 2006 (2006년 도축장 출하 지육에 대한 항생물질 잔류현황 분석)

  • A total of 3,943 samples (beef 698, pork 2,178 and chicken 1,067)were collected from slaughter houses in Daegu from January to December 2006 for analysis of antimicrobial residues. The results were also comparatively analyzed with those carried out from 16 veterinary service laboratories nationwide as a part of national residue program in 2006. The samples from 11 farms (cattle 2, pig 9) were found to exceed the legal limit. Among them 5 were classified as tetracyclines, 3 as $\beta$-lactams, 2 as sulfonamides and 1 as quinolone. Nationally a total of 222 farms (241 cases) were found to violate the residual limits, of which 47.3% were detected by screening test, followed by surveillance (38.3%). Failure to observe withholding periods of antimicrobial drugs after treatment was the most significant cause of the non-compliance.

A DFT and QSAR Study of Several Sulfonamide Derivatives in Gas and Solvent

  • The activity of 34 sulfonamide derivatives has been estimated by means of multiple linear regression (MLR), artificial neural network (ANN), simulated annealing (SA) and genetic algorithm (GA) techniques. These models were also utilized to select the most efficient subsets of descriptors in a cross-validation procedure for non-linear -log (IC50) prediction. The results obtained using GA-ANN were compared with MLR-MLR, MLR-ANN, SA-ANN and GA-ANN approaches. A high predictive ability was observed for the MLR-MLR, MLR-ANN, SA-ANN and MLR-GA models, with root mean sum square errors (RMSE) of 0.3958, 0.1006, 0.0359, 0.0326 and 0.0282 in gas phase and 0.2871, 0.0475, 0.0268, 0.0376 and 0.0097 in solvent, respectively (N=34). The results obtained using the GA-ANN method indicated that the activity of derivatives of sulfonamides depends on different parameters including DP03, BID, AAC, RDF035v, JGI9, TIE, R7e+, BELM6 descriptors in gas phase and Mor 32u, ESpm03d, RDF070v, ATS8m, MATS2e and R4p, L1u and R3m in solvent. In conclusion, the comparison of the quality of the ANN with different MLR models showed that ANN has a better predictive ability.

Detection of tetracyclines in beef and pork (식육중 테트라사이클린계 항생물질 잔류조사)

  • In an effort to Improve the quality of meat and to monitor farms, residual tetracyclines in local beef and pork produced in the province of North Chung-cheong were determined by a EEC 4-plate, Charm II and HPLC, respectively. The results were summarized as follows ; 1. Of the 547 samples, 4 beef and 13 pork samples were judged to be positive by EEC 4 - plate method. 2. Detection rates of tetracyclines by type 1(++--) and type 2(+++ or ++$\pm$-) micro-bial growth Inhibition In EEC 4-plate method were 100% and 71%, respectively. 3. Of 17 positive samples, 6 were positive for tetracyclines, 4 were positive for tetracyclines and sulfonamides, 1 was positive for sulfonamides, and 2 were positive for others by Charm II test. 4. The best eluents were 0.01M methanolic oxalic acid, and the ideal temperature for stable concentration was $40^{\circ}C$ as optimal HPLC analytical conditions for the detection of tetracy-clines. 5. Of the 10 positive samples for tetracyclines by Charm II test, tetracyclines were confirmed in 2 beef and 6 pork samples, using HPLC, at levels ranging from 3.64~4.22 ppm and 0.2~1.20 ppm, respectively.
