Separation of Sulfuric Acid from Sulfuric Acid/Glucose Solution by Electrodialysis (황산/글루코스 용액으로부터 전기투석에 의한 황산 분리)

  • Recovery of sulfuric acid is very important after biomass converted to sugar by acid hydrolysis. In this work, the separation of sulfuric acid from sulfuric acid/glucose solution was studied by electrodiaysis. Three chamber method, which requires both anion membrane and cation membrane, is the most commonly used in the electrodialysis process, but two chamber method using only an anion membrane was the focus of this study. Sulfuric acid was perfectly separated from a mixture of 10~30 wt% glucose and 1~3 M sulfuric acid by electrodialysis using two chamber method. The separation rate of sulfuric acid lineary increased with higher current density when the affect of diffusion and convection of the membrane was small. Without electric energy, 45% of sulfuric acid was separated by diffusion and convection only.

A Study on Continuous Alcohol Fermentation with Cell Recycle by Means of Membrane Separation (막분리를 이용한 미생물 재순환 연속 알콜발효에 관한 연구)

  • One of the objectives of this work is to obtain information relevant to the industrial production of alcohol from sugar. The fermentation of alcohol by a strain of saccharomycess cerevisiae ATCC 24858 was studied In a continuous single-stage process with recycle of the cells via tangential flow microfiltration membranes. The experimental results reported in this study pertain to continuous cultures with total cell-recycle by varying the dilution rate (D=0.3, 0.5, and 0.7 $hr^{-1}$) and glucose concentration (50, 100, 150, and 200g/l sugar solution). Productivity using a repeated cell recycle system was found extremely high, 1.e., over 10 to 29 times higher than that of a smile batch system. When a sugar concentration of 200g/1 at dilution rate, 0.7 hr-1 was used, 83.9g/l ethanol was formed with an ethanol yield of 0.42(82% of theoretical) based on sugars utilized.

  • Acid dairy drinks(ADD) are a worldwide product existing in many variations: fruit milk drinks, yogurt drinks, soy milk, butter milk, whey drinks and kefir etc. These drinks are marketed with different shelf lives depending on processing: -Short shelf life(maximum 3 weeks, cold storage) - Long shelf life(2 to 9 months, pasteurized, sterilized or retorted) Acidic protein drinks tend to a separation or destabilization process in the absence of stabilizing system during the shelf life of the ADD. A phase separation results in sedimentation of large particles at the bottom of the package and/or the formation of a serum layer at the top(whey off). These beverages are usually composed of an acid dairy phase (fermented base)or a natural base(milk, soymilk etc.)with an acidic medium (fruit phase: pulp, fruit concentrate etc.) which can be flavored. Sugar and stabilizers are added. It has been proved since the late 1950's that adding high methoxy pectin (HM pectin)to acid milk drinks is the best way to prevent the formation of a sediment and/or the whey off. In this presentation, we explain about stabilization mechanism of ADD induced by pectin. Applications and market trend of ADD in Asia and Europe are explained.

농산폐자원을 이용한 Arabinose의 생산

  • Arabinose is five-carbon sugar that have been used a precursor of chemical synthetic pharmaceutics and an additives for an diet foods. Its contents in the hemicellulose among varied cellulosic materials is approximately $0.2%\;{\sim}\;4.0%$. In previous papers, it was focused in the production of xylose. But this study was subjected to an effective production of arabinose. High arabinose yield and selectivty was achieved at 0.6% sulfuric-acid concentration and $100^{\circ}C$. We will present how to remove an insoluble substances and the process for arabinose separation.

  • Component having antioxidant activity on lard during heating was separated from hexane extract of spinach, and its characteristic chemical structure was speculated through nuclear magnetic resonance spectroscopy, liquid chromatography-mass spectrometry, and Fourier transform infrared spectrophotometry. Lard was heated with hexane-, ethyl ether-, ethyl acetate-, or ethanol extract of spinach at $180^{\circ}C$ for 20 hr. Hexane extract of spinach, having highest antioxidant activity on lard during heating, was fractionated by silicic acid column chromatography (SACC), and SACC fractions having higher antioxidant activity on lard during heating were further separated by thin layer chromatography (TLC). Isolated compound from SACC fractions of hexane extract of spinach by TLC had sugar moieties and benzene ring along with hydroxy, carbonyl, and alkyl groups in the structure.

A Study on Saccharides in Honey via HPLC (HPLC에 의한 벌꿀의 당성분에 관한 연구)

  • Saccharides in Korean honey have been analyzed by traditional wet chemical methods and several chromatographic technics gas, paper, column and thin layer chromatography. A simple, rapid and versatile method for the separation and determination of saccharides via high performance liquid chromatography were used eluting with $H_2O : CH_CN$ at 25:75, at a flow rate of 1.0ml/min. Acasia honey contained 36.8% fructose, 31.4% glucose, 1.7% sucrose and bush clover honey held 34.4% fructose, 32.3% glucose and 30% sucrose. Difference on the regional distribution were not found in the quantity of saccharides (42 species). The quality of inferior honey was assumed to contain much maltose, sugar and glucose than common honey.

  • The use of L-carbohydrates and their corresponding nucleosides in medicinal application has greatly increased. For example L-ribose has been much in demand as the starting material for curing hepatitis B. High performance liquid chromatography (HPLC) method was studied for the analysis of ribose and arabinose fractions from ion exchange chromatography (IEC). Dowex Monosphere 99 Ca/320 resin was packed in IEC to separate ribose and arabinose under various operating conditions. $NH_{2}$ and sugar HPLC columns were then used to analyze the fractions from the IEC column. Pulse input method (PIM) was also used to measure adsorption isotherms of ribose and arabinose in the Dowex column and HPLC columns. Experimental results and simulations by ASPEN chromatography were compared with fair agreement.

Improvement of Alcohol Productivity by Means of Repeated Batch Fermentation (반복적인 회분식 발효공정을 이용한 에탄올 생산성의 향상)

  • The functional relationship between the initial cell concentration and the ethanol productivity was investigated in the repeated batch fermentation of Sacharomyces cerevisiae ATCC 24858. The repeated batch fermentations were performed in the range of 60 to $150g/\ell$ of initial sugar concentration and 17.5g/$\ell$ to $53.1g/\ell$ of initial cell concentration. The time of one batch fermentation was 1 or 2 hours and the batch fermentation was repeated ten times in every repeated formentaction. The functional relationship showed that the productivity increased non-linearly according to the increase of initial cell concentration regardless of initial sugar concentration. When the initial concentration of sugar was $60g/\ell$ and that of biomass was $34.5g/\ell$, the fermentation was completed within one hour and its ethanol productivity was $26.7g/\ell$.hr, the latter including the times of cell separation, pouring the new substrate into a flask and sampling. When the initial sugar concentration was $120g/\ell$ and the initial cell concentration $50.3g/\ell$, the fermentation was also finished within one hour and its productivity was $45.8g/\ell$$.$hr, The maximum ethanol productivity for eight different repealed fermentations in this work was $53g/\ell$.hr.

Concentration of Persimmon Juice by Revers Osmosis System (역삼투 시스템을 이용한 감 과즙의 농축)

  • Membrane separation technology was applied to prepare high quality persimmon juice from persimmon, which is produced in large quantities in Korea. The influences of time, pressure, and temperature on permeate flux were studied during concentration by reverse osmosis. The chemical components of retentate were also analyzed. The permeate flux was higher as the operating temperature and pressure were increased, and was influenced more strongly by operating pressure than temperature. Maximum concentration by reverse osmosis employed in this study in the oBrix scale was about 30. Retention percentage of sugar in the persimmon juice which was concentrated by membrane separation system was more than 90% and was not influenced by operating conditions. Retention percentage of volatile components in the same process was more than 60%; it was increased as operating temperature was decreased and pressure was increased.

Temperature Effect on the Retention Behavior of Sugars and Organic Acids on poly (4-vinylpyridine) Resin (PVP 분리 수지에서 온도에 따른 당과 유기산의 체류 특성 변화)

  • This study was performed to understand temperature effect on retention behavior of fructose and glucose as sugars and lactic acid and acetic acid as organic acids on poly (4-vinylpyridine) resin. The pulse tests were performed to understand temperature effect on retention time of sugars and the results were not shown large change. As it was able to predict with PVP resin not to be used for sugar separation generally, the results were shown poor resolution for separation of sugars and temperature effect on the resolution change of sugars also was not large. On the other hand, in the case of organic acids on PVP resin, the pulse tests were shown temperature effect on the retention behavior was very large. So, the frontal analyses were performed to understand quantitative adsorption behavior of organic acids at 35 and $65^{\circ}C$. These adsorption characteristics of organic acids with PVP resin system can be used to preparative chromatographic process such as SMB (simulated moving bed).
