Kinetic Mechanism in the Absence of Metal of Hafnia alvei Aspartase in the Amination Direction

  • The kinetic mechanism of Hafnia alvei aspartase in the amination direction has been determined in the absence of metal. The initial velocity pattern obtained by varying the concentration of fumarate at several fixed concentrations of NH4+ , shows an intersection on the left of the ordinate at pH 7.0, indicating that the kinetic mechanism is a sequential mechanism in which substrate inhibition by fumarate is observed. The dead-end inhibition pattern by varying the concentration of NH4+ at several fixed concentration of succinate shows an intersection on the left of the ordinate. These data are consistent with random addition of NH4+, or fumarate. The Haldane relationship gives a Keq of 1.18 ${\times}$10-3 M at pH 7.0, which is in agreement with the values obtained from the direct determination of reaction concentrations at equilibrium (6.0 $\pm0.2$ ${\times}$10-3 M).

Degradation Properties of n-Alkane Assimilating Pseudomonas putida 3SK Carrying $CAM::TOL^{*}$ Plasmid and NAH Plasmid

  • Pseudomonas putida 3SK, which was constructed by the conjugal transfet of the $CAM::TOL^{*}$ plasmid of Pseudomonas putida CSnA and the NAH plasmid of Pseudomonas putida KCTC 2403 into n-alkane assimilating Pseudomonas putida KCTC 2405, showed a broad degradation spectrum and floc-forming ability. This strain degraded m-toluic acid, naphthalene, camphor and decane simultaneously. $Hg^{2+}$ at the concentration of 1 ppm in the minimal medium could not inhibit the growth of this strain. The degradation of m-toluic acid by Pseudomonas putida 3SK was not repressed by the easily utilizable compounds, such as glucose and succinate. But, the addition of formalin inhibited the growth of Pseudomonas putida 3SK. After the cultivation of this strain on the artificial wastewater containing m-toluic acid, naphthalene, camphor and decane for 24 hr, the initial COD value (1500) of the artificial wastewater was declined to 300.

  • Botanical antimicrobial agent-grapefruit seed extract mixture (BAAG) have an unknown compounds which exhibit the antibiotic activities aganist microorganisms including bacteria and fungi. We have examined the effects of BAAG on the physiological function of Botrytis cinerea which was isolated from necrotic lesions of decayed fruits and vegetables such as cucumbers, grapes, tomatoes, and red peppers during storage. In the results of enzymatic activities related to the energetic metabolism there was no inhibitory effect of BAAG on the activities of several enzymes in vitro including glucose 6-phosphate dehydrogenase and malate dehydrogenase, while there was inhibitory effect of BAAG on the activities of hexokinase and succinate dehydrogenase. O-nitrophenyl-$\beta$-D-galactopyranoside(ONPG), the artificial substrate of $\beta$-galactosidase was hydrolyzed in the presence of BAAG, indicating that the membrane was pertubated by the BAAG. From the results we suggested that the antibiotic activity of BAAG is due to the change of membrane permeability of the cell. BAAG was fractionated and purified by silica gel and sephadex column chromatography. Among active fractions two peaks were identified as naringin and limonin when they were analyzed by by NMR and Fast atomic bombardment.

Analysis of Gene Encoding the PBSA Degradation Enzyme (PBSA 분해효소 유전자의 분석)

  • Burkholderia cepacia PBSA-7, Bacillus licheniformis PBSA-8 and Burkholderia sp. PBSA-9 previously collected from Korea soil (Joo and Kim, 2009) were analyzed for the presence of genes encoding proteins operative in the degradation of poly(butylene succinate-co-butylene adipate; PBSA). Polymerase chain reaction analyses revealed a 1.5 kb fragment of the lipase gene (lip A) in B. cepacia PBSA-7 and Burkholderia sp. PBSA-9, while B. licheniformis PBSA-8 harbored the same gene fragment at 600 bp. The three strains possessed "Gly-X1-Ser-X2-Gly" and "Ala-X1-Ser-X2-Gly" lipase sequence regions. Burkholderia sp. PBSA-7 lip A displayed 36~40% homology with the family 1-1 lipases and 82~92% homology with the family 1-5. Burkholderia sp. PBSA-8 lip A was 64~65% homologous with the subfamily 1-4 lipases, but displayed no homology with the subfamily 1-5 lipases. Burkholderia sp. PBSA-9 lip A displayed 35~37% homology with the family I1 lipases and 83~94% homology with the family I2 lipases, similar to Burkholderia sp. PBSA-7.

Determination and Multivariate Analysis of Flavour Components in the Korean Folk Sojues Using GC-MS (GC-MS 를 이용한 전통민속소주의 향기성분 분석과 다변량통계해석)

  • Flavour components of seven Korean folk sojues, five Chinese kaoliangchiews and Japanese shochu were determined by GC and GC-MS after solid phase extraction with polydivinyl benzene. Less volatile ethyl succinate and ethyl pelargonate were present in Korean folk sojues while volatile ethyl acetate and ethyl butyrate in Chinese kaoliangchiews. In the case of alcohols, the amount of isopentyl alcohol was relatively higher than that of isobutyl alcohol or n-propyl alcohol in Korean folk sojues. On the contrary, less volatile n-propyl alcohol was present more than isopentyl alcohol in Chinese kaoliangchiews. Multivariate statistical analyses involving principal components analysis (PCA) and discriminant analysis (DA) were applied to the GC data. The results of PCA clearly demonstrate that the first principal scores of Korean folk sojues were similar but the second principal scores were different from each other. Classification of Korean sojues and Chinese kaoliangchiews into two groups could be conducted by DA. These results suggested that the common charateristics and identities as a distilled liquors was found in Korean folk sojues.

Production of Cyclodextrin by Bacillus sp. I-5 Cyclodextrin Glucanotransferase (Bacillus sp. I-5 Cyclodextrin Glucanotransferase에 의한 Cyclodextrin의 영향)

  • A cyclodextrin glucanotransferase(CGTase)-producing Bacillus sp. I-5 was isolated from soil and the enzyme exhibited the maximum reaction rate at pH 8.0 and $50^{\circ}C$. It was found that CGTase of I-5 produced ${\beta}-$ and ${\gamma}-CD$ mainly but the production ratio of cyclodextrins (CDs) was influenced by the buffer solution. Sodium acetate significantly stimulated the formation of ${\gamma}-CD$, increasing the content by 35%. The production of CDs was influenced by DE value of starch. The results indicated that DE value in the range of $3.5{\sim}6.0$ were most effective for the CD formation. CGTase was immobilized on the reversibly soluble-insoluble carrier, hydroxypropyl mothylcellulose acetate succinate. The immobilized CGTase was soluble at pH 7.5, and precipitated easily at pH 6.0. Enzyme reactor was designed to produce CD continuously. It was composed of three major stages-CD produttion by immobilized CGTase, conversion of the residual dextrin to glucose by amylase and glucoamylase and alcohol fermentation by yeasts to remove the glucose into alcohol. The yield of total CDs was 3.65g from 10g soluble starch.

Analysis of Aliphatic Carboxylic Acids Using Ion-Exchange Chromatography: Application to Groundwater Affected by Landfill Leachates (이온-교환 크로마토그래피를 활용한 유기산 분석: 매립지 침출수의 영향을 받은 지하수에 대한 적용)

  • An analytical method using ion-exchange chromatography was developed for simultaneous quantification of low-molecularweight organic acids ($C_1-C_6$ aliphatic carboxylic acids) and inorganic anions, and then applied to the assessment of ground water contaminated by leachates from a municipal solid waste landfill. Peak interferences of halide ions to organic acids were removed by pretreatment of water samples with Ag-containing cartridges. This method allowed accurate detection of low-molecular weight organic acids (i.e., formate, acetate, propionate, pyruvate, succinate, and oxalateas) low as 0.5 mg/L with a linear dynamic range up to 20 mg/L within 11 min run time along with typical inorganic anions. High level of pyruvate and low level of formate and acetate were detected in groundwater and landfill leachates using the analytical method. Pyruvate concentration in groundwater showed a significant correlation with concentrations of $Cl^-$ and $HCO_3^-$, and pyruvate levels decreased along the downgradient from the landfill, indicating the sources of pyruvate are landfill leachate.

Cell Biological Studies on Brain Formation at the Early Stage of Chick Embryogenesis (초기계배의 뇌 형성에 관한 세포 생물학적 연구)

  • The effect of tryptophan on brain formation at the early stage of chick embryo has been investigated morphologically using electron microscope. The electron micrographs of cerebral cortex cells of $5\\sim10$ day old chick embryo, which received 1.0mg of tryptophan showed that the irregularity, evagination and disruption of nuclear membrane and nuclear chromatin condensation, nucleolar chromatin margination and segragation. Hypertrophy of stalks, vesicles, and vacuoles can be seen and dilation and vesiculation of rough endoplamic reticulum and polysome disaggregation occured. Protein and RNA levels and the activity of several enzymes such as lactate dehydrogenase, succinate dehydrogenase, malate dehydrogenase and glucose 6-phosphate dehydrogenase of tryptophan administered group were significantly lower than those of control group suggesting that the tryptophan administration depressed protein biosynthesis resulting in the decrease of enzyme activity. It was found that serotonin content of egg yolk which has been incubated for 10 days were as much as three times that of control egg yolk. It is not clear whether the increase of serotonin content might inhibit intracellular yolk granule degradation which might result in malformation of chick embryo, but it is likely that tryptophan administration might depress protein biosynthesis, consequently, the enzyme biosynthesis would be impaired. This might give rise to improper development of chick embryo.

Carboxydobacteria 를 위한 재조합 Plasmid 백터와 형질전환방법 개발

  • Recombinant plasmid shuttle vectors were constructed for genetic studies on the oxidation of carbon monoxide by carboxydobacteria. Two vectors. pYK322 (7.2 kb, Ap'. Tc') and pYK 324 (7.2 kb, Ap', Tc'), were constructed using pBR322 and pYK100. a small plasmid in Pseudomonas carbo,xydovorans. Four plasmids. pYK2IO (5.2 kb. Cm'), pYK220 (5.2 kb, Cmr), pYK230 (5.2 kb, Cm'), and pYK232 (5.2 kb. Cm'), were constructed using pACYC184 and pYK100. Transformation of several carboxydobacteria with pYK322 and pYK220 was round to be efficient when the cells were transformed by the methoti of Bagdasarian and Timmis (Curr. Top. Microbiol. Immunol. 96:47-67. 1982) with several modifications; cells growing on 0.2% succinate were harvested at the mid-exponential phase. 10 mM RbCl in transformation solution was substituted with 100 mM KCI. cclls in transformation solution were incubated for 12 h at 4'C before addition of DNA and heat shock was carried out for 3 min at 45$^{\circ}$C. Plasmid vectors used for transformation, however. were not detected from antibiotics-resistant transformants, suggesting that the vectors may be integrated into the chromosomal DNA.

Isolation and Identification of Low Molecular Volatile Compounds from Ethyl Acetate Layer of Korean Black Raspberry (Rubus coreanus Miq.) Wine (복분자(Rubus coreanus Miquel) 와인의 ethyl acetate 획분으로부터 저분자 휘발성 화합물들의 분리 및 구조해석)

  • In the course of our investigation for chemical constituents in the ethyl acetate layer of Korean black raspberry wine, five compounds were isolated and purified by silica gel column chromatography and high-performance liquid chromatography. The isolated compounds were identified as ethyl succinate (1), vanillic acid (2), ethyl 3,4-dihydroxybenzoate (3), furan-2-ol (4), and 4-(4-hydroxyphenyl)butan-2(S)-ol (5) based on the spectroscopic data of electrospray ionization tandem mass spectrometry and nuclear magnetic resonance. The presence of 2 in Korean black raspberry has previously reported. However, 1 and 3-5 in Korean black raspberry and its wine were isolated for the first time.
