Research fecal metabolite according to fed different ratios of roughage to concentrate on lactating cow using 1H-NMR analysis (조사료와 농후사료 급여비율에 따른 젖소 우분의 대사체 탐색)

  • This study examined the metabolites in different roughage to concentrate ratios using proton nuclear magnetic resonance spectroscopy (1H-NMR). Six lactating cows were divided into two groups that were fed different roughage to concentrate ratios (HR group = 8:2, HC group = 2:8). Feces samples were collected individually at one time, and the metabolites were analyzed using an SPE-800 MHz NMR-MS system. The metabolites were identified and quantified using a Chenomx NMR suite 8.4. Metabolic pathway analysis and principal component analysis were conducted using a Metaboanalyst 4.0. Statistical analysis was performed using a Dunnett's test on the SAS program. As a result, several metabolites were identified, and among them, 77 metabolites were used in statistical analysis. The levels of twelve metabolites were significantly higher in the HC group: succinate, dimethylamine, histamine, homovanillate, thymol, acetate, propionate, butyrate, isovalerate, valerate, imidazole, N-nitrosodimethylamine, and O-acetylcholine. In the HC group, the concentrations of all metabolites were higher than in the HR group, and the metabolic pathway was also different. This study is expected to be useful for a variety of livestock studies by 1H-NMR because it examined the change in metabolites in the body metabolism and microorganisms.

In-vivo $^1H$ MR Spectroscopy in Abscessess Induced in Rabbit Thighs: Evaluation of Treatment Effect (토끼 대퇴부에 유발한 농양의 생체내 수소 자기공명분광법: 치료효과 평가)

  • Purpose : To analyze the serial changes of proton magnetic resonance (MR) spectra in the abscess and to determine the effect of the antibiotic treatment on the metabolite patterns. Materials and Methods : MR imaging and MR spectroscopy of an experimentally induced abscess were performed sequentially for four weeks at interval of one week in both the control group (n=5) and the antibiotic treatment group (n=5). On MR imaging, the shape and the size of the abscess were analyzed. On MR spectroscopy, the resonance peaks of metabolites were assigned on the basis of reported peaks in the literature. The metabolite ratios measured by using N-acetyl alanine as an external reference and by using lipid as an internal reference were compared in both the control and treatment g roups. Results : The abscesses were seen as cystic masses on MR imaging. On MR spectroscopy, the variable peaks of acetate, succinate and various amino acids, which are the metabolites of infection, were identified in the control and antibiotic treatment groups. The most frequent peak was that of acetate at 1.92ppm (70%). Both the peak ratios of acetate to lipid and acetate to external reference tended to decrease in the treatment group while the ratios did not change significantly in the control group. Conclusion : MR spectroscopy is useful not only for the diagnosis of abscess but also for monitoring the evolution of the abscess by using the acetate peak.

Growth Characteristics of a Pyruvate Decarboxylase Mutant Strain of Zymomonas mobilis (Pyruvate decarboxylase 돌연변이 Zymomonas mobilis 균주의 생장 특성 연구)

  • Studies of the inactivation of a gene encoding pyruvate decarboxylase, pdc, in an ethanol-producing bacterium, Zymomonas mobilis, identified a mutant strain with 50% reduced PDC activity. To evaluate the possibility of a carbon-flux shift from an ethanol pathway toward higher value fermentation products, including pyruvate, succinate, and lactate, fermentation studies were carried out. Despite attempts to silence pdc expression in the wild-type strain ZM4 using cat-inserted pdc and pdc-deleted homologs by electroporation, the strain isolated showed partial gene activation. Fermentation experiments with the PDC mutant strain showed that the reduced expression level of PDC activity resulted in decreased rates of substrate uptake and ethanol production, together with increased pyruvate accumulation of 2.5 g l-1 , although lactate and succinate concentrations were not significantly enhanced in these modified strains. Despite numerous attempts, no strains were isolated in which complete pdc inactivation occurred. This result indicates that the ethanol fermentation pathway of this bacterium is totally dependent on the activity of the PDC enzyme. To ensure a redox balance of intracellular NAD and NADH levels, other enzymes, such as lactate dehydrogenase for lactate, and enzymes involved in the production of succinic acid, such as pyruvate dehydrogenase (PDH) and malic enzymes, may be needed for their increased end-product production.

Fermentation Characteristics of Wine Yeast Strains for White Wine Making (백포도주 양조에 있어서 포도주 효모의 발효 특성)

  • The characteristics of used wine yeast strains were as follows, S6U showed low fermentation speed than those of other yeast strains, but this strain fermented completely later. The wine fermented by W-3 was very low contents of total acid, 0.75% and the ones fermented by UCD530 and AC- contained much extract, 3.26 mg/L and 3.22 mg/L respectively. The wine fermented by CEG and CS2 were predominant in yellowness, and EC1118 produced large amount of acetaldehyde, 49.9 mg/L than those of other strains. EC1118 and CY3079 displayed low methylene blue dyeing ratio, below 15%, meaning high alcohol tolerance yeast. UCD530 produced extremely high contents of glycerol, succinate and lactate compared with other strains. These properties revealed that UCD530 was a typical Saccharomyces bayanus species. The main organic acids produced by wine yeasts were pyruvate, lactate, succinate and acetate. The concentration of acetate in experimental wine could be divided into two parts, one group had concentration below 170 mg/L (UCD530, EC1118, AC-, CY3079, W-3), and the other had concentration up to 350 mg/L (S6U, CEG, CS2).

  • Twenty esters, 14 alcohols, 5 aldehydes, 5 ketones, 5 lactones, 2 S-containing compounds and 1 hydrocarbon are identified by GC-MS from volatile compounds in a glucose solution containing Yeat-Carbon-Base medium fermented for 64 hrs by Saccharomyces bayanus at pH 3.5, $25^{\circ}C$, 400rpm and 35L/h of aeration for 24hrs. Under the different conditions of conservation(1~4), ethyl 2-hydroxy-4-metjhylpentanoate, ethyl succinate, nonanol and phenylacetaldehyde are produced during conservation of fermented solution. 17 esters increased during conservation at $13^{\circ}C$ for 12 weeks and the increase of ethyl 9-hexadecenoate is important among 13 esters increased during conservation at $35^{\circ}C$ for 24hrs. During conservation, aldehydes increased at $35^{\circ}C$, but decreased at $13^{\circ}C$ and the great increase of isobutanal, benzaldehyde and phenylacetaldehyde is observed at $35^{\circ}C$. Alcoho and lactones increased but ketones decreased during conservation.

Hydrogen Production by Purple Sulfur Bacteria, Thiocapsa roseopersicina in Photoheterotrophic Culture Condition (홍색유황세균, Thiocapsa roseopersicina의 photoheterotrophic 조건에서의 수소생산)

  • The purple sulfur phototrophic bacterium, Thiocapsa roseopersicina NCIB 8347 has been studied on hydrogen production and cell growth under different culture conditions, such as light source, light intensity, and growth temperature. T. roseopersicina showed maximum cell growth of 1.38 and 1.42 g-DCW/L under 7.5-10 klux of halogen and fluorescent light, respectively, and produced maximum amount of hydrogen with values of 0.90 and 0.48 $mL-H_2/mg$-DCW under the irradiation of 10 klux of halogen and fluorescent light, respectively. The optimum growth temperature for hydrogen production was $26^{\circ}C$, and hydrogen production rate was lowered over $30^{\circ}C$. When T. roseopersicina was grown photoheterotrophically under irradiation of 8-9 klux of halogen lamp, the generation time was 4.2 hr. The strains started producing hydrgen from the middle of the logarithmic growth phase and continued until succinate concentration leveled out.

Effect of Various Biodegradable Chelating Agents on Root Growth of Plants under Copper Stress (생분해 되는 다양한 킬레이트가 구리에 노출된 식물의 뿌리성장에 미치는 영향)

  • Phytoextraction is a method of phytoremediation using plants to clean up metal-contaminated soils. Recently, various chelating agents were used in this method to increase the bioavailability of metals in soils. Even though phytoextraction is an economic and environmentally friendly method, this cannot be applied in highly metal-contaminated areas because plants will not normally grow in such conditions. This research focuses on identifying chelating agents which are biodegradable and applicable to highly metal-contaminated areas. Copper (Cu) as a target metal and cysteine (Cys), histidine (His), citrate, malate, oxalate, succinate, and ethylenediamine (EDA) as biodegradable chelating agents were selected. Ethylenediamine tetracyclic acid (EDTA) was used as a comparative standard. Plants were grown on agar media containing various chelating agents with Cu to analyze the effect on root growth. Cys, His, and citrate strongly diminished the inhibitory effect of Cu on root growth of plants. The effect of oxalate was weak, and malate and succinate did not show significant effects. EDTA diminished and EDA promoted the inhibitory effects of Cu on root growth. These effects of chelating agents are correlated with Cu uptake into the roots. In conclusion, as biodegradable chelating agents, Cys, His, and citrate are good candidates for highly Cu-contaminated areas, while EDA can be useful in phytoextraction for Cu.

The Inhibitory Effect of Grapefruit Seed Extracts on the Physiological Function of Enterobacter pyrinus (Grapefruit 종자추출물이 Enterobacter pyrinus의 생리기능에 미치는 영향)

  • Grapefruit seed extracts(GFSE) have some unknown compounds which exhibit the antibiotic activities aganist microorganisms including bacteria and fungi. We have examined the effects of GFSE on the growth of Enterobacter pyrinus which was isolated from necrotic lesions of pear trees. During the cultivation, the growth of the bacteria was strongly inhibited at the low concentration(0.01%, w/w) of GFSE. Hydrophobic fraction extracted from GFSE by mixed solvents (chloroform : methanol : water, 1 : 2 : 0.8, v/v/v) had components which inhibited the growth of bacteria. There was, however, no inhibitory effect of GFSE on the activities of several enzymes including hexokinase, glucose 6-phosphate dehydrogenase, malate dehydrogenase and succinate dehydrogenase. $O-nitrophenyl-{\beta}-D-galactopyranoside(ONPG)$, the artificial substrate of ${\beta}-galactosidase$ was hydrolyzed in the presence of GFSE, indicating that the membrane was pertubated by the GFSE. From the results it was suggested that the antibiotic activity of GFSE is due to the change of membrane permeability of cell. GFSE was fractionated by high performance liquid chromatography equipped with $C_{18}$ reverse phase column. Among active fractions, three peaks were identified as 1-chloro-2-methyl-benzene (o-toluene), N,N-dimethyl-benzenemethaneamine, 1-[2-(2-ethylethoxy)ethoxy]-4- (1,1,3,3-tetramethyl)-bezene, respectively, while the other three remained unidentified.

Volatile Flavor Components in Mash of Takju prepared by using Rhizopus japonicus Nuruks (Rhizopus japonicus 누룩으로 담금한 탁주 술덧의 발효 과정 중 휘발성 향기성분)

  • Volatile flavor components in the mash of takjus prepared by using Rhizopus japonicus nuruk were identified by using GC and GC-MS. Twenty-four esters, 19 alcohols, 9 acids, 10 aldehydes and 4 others were found in the mash of takju. Thirty nine components including 14 esters and 12 alcohols were detected in the beginning of fermentation. Seventeen components were more detected after second day of fermentation and 66 components were detected after 12 days of fermentation. Thirty eight flavor components including 12 alcohols such as ethanol, 2-methyl-1-propanol and 3-methyl-1-butanol, 14 esters such as ethylacetate, ethylcaprylate and isoamylacetate, 6 aldehydes and 5 acids were usually detected in the fermentation process. Ethanol was predominantly found in the range of 76.2149-92.1155% as a major component by using relative peak area. 3-Methyl-1-butanol, 2-methyl-1-propanol, ethyl caprylate, 2,3-butanediol and benzeneethanol were some of the major volatile components through the fermentation. Peak area of ethylacetate, diethyl succinate, octanoic acid, acetic acid and isobutylaldehyde among the same group were higher than other component depending upon fermentation time.

The Predictive Factors of the Serum Creatine Kinase Level Normalization Time in Patients with Rhabdomyolysis due to Doxylamine Ingestion (독시라민 중독으로 발생한 횡문근융해증 환자에게서 혈중 크레아틴인산활성화효소 수치가 정상화되는 시기를 예측할 수 있는 인자)

  • Purpose: Doxylamine succinate (DS) is frequently used to treat insomnia and it may induce rhabdomyolysis in the overdose cases. The purpose of this study is to evaluate the factors that can predict the serum creatine kinase (CK) level normalization time for patients with rhabdomyolysis due to DS ingestion. Methods: This study was conducted on 71 patients who were admitted with rhabdomyolysis after DS ingestion during the period from January 2000 to July 2009. Rhabdomyolysis was defined as a serum CK level over 1,000 U/L. The collected data included the general characteristics, the anticholinergic symptoms, the ingested dose, the peak serum CK level, the time interval (TI) from the event to the peak CK level and the TI from the event to a CK level below 1,000 U/L. We evaluated the correlation between the patients' variables and the TI from the event to the peak CK level time and the time for a CK level below 1,000 U/L. Results: The mean ingested dose per body weight (BW) was $30.86{\pm}18.63\;mg/kg$ and the mean TI from the event to treatment was $4.04{\pm}3.67$ hours. The TI from the event to the peak CK level was longer for the patients with a larger ingestion dose per BW (r=0.587, p<0.05). The CK normalization time was longer for the patients with a larger ingested dose per BW (r=0.446, p<0.05) and a higher peak CK level (r=0.634, p<0.05). Conclusion: The ingested dose per BW was correlated with the TI from the event to the peak CK level, and the ingested dose per BW and the peak CK level have significant correlations with the CK normalization time. These factors may be used to determine the discharge period of patients who had rhabdomyolysis following a OS overdose.
