Assessment of Heavy Metal Effects on the Freshwater Microalga, Chlorella vulgaris, by Chlorophyll Fluorescence Analysis (엽록소형광분석을 이용한 담수산 클로렐라(Chlorella vulgaris)에 미치는 중금속의 영향 평가)

  • The response of the freshwater microalga, Chlorella vulgaris, to heavy metal stress was examined based on chlorophyll fluorescence analysis to assess the toxic effects of heavy metals in freshwater ecosystems. When toxic effects were analyzed using regular chlorophyll fluorescence analysis, photosystem II activity($F_v/F_m$) decreased significantly when exposed to $Cu^{2+}$ and $Hg^{2+}$ for 12 h, and decreased in the order of $Hg^{2+}>Cu^{2+}>Cd^{2+}>Ni^{2+}$ when exposed for 24h. The effective photochemical quantum yield(${\phi}{\prime}_{PSII}$), chlorophyll fluorescence decrease ratio($R_{Fd}$), minimal fluorescence yield($F_o$), and non-photochemical quenching(NPQ), but not photochemical quenching(qP), responded sensitively to $Hg^{2+}$, $Cu^{2+}$, and $Cd^{2+}$. These results suggest that $F_v/F_m$, as well as ${\phi}{\prime}_{PSII}$, $R_{Fd}$, $F_o$, and NPQ could be used to assess the effects of heavy metal ions in freshwater ecosystems. However, because many types of heavy metal ions and toxic compounds co-occur under natural conditions, it is difficult to assess heavy metal toxicity in freshwater ecosystems. When Chlorella was exposed to heavy metal ions for 12 or 24h, $F_v/F_m$ and maximal fluorescence yield($F_m$) changed in response to $Hg^{2+}$ and $Cu^{2+}$ based on image analysis. However, assessing quantitatively the toxic effects of several heavy metal ions is challenging.

Analysis of Debris Flow of Chun-cheon Landslide Area using Numerical Methods (수치해석을 통한 춘천 산사태지역 토석류 거동 분석)

  • The characteristic of recent rainfall pattern in Korea is concentrated in summer season and it is very different compare with former characteristic. In 2011, there was heavy rainfall in Chuncheon city of northern part of Korea. Because of rainfall in short time, many landslides were occurred in narrow area and many people were killed by these landslides at that time. The purpose of this study is to calculate run-out distance of debris flow and analyze the movement properties of debris flow according to the elapsed time using numerical analysis method at that time. The debris 2D program, which is developed by prof. Liu in National Taiwan University, was used in this study. Run-out distance of debris flow was calculated under different yield strength conditions which were controlled by rainfall amount. The results reveal that absolute maximum velocity of the debris flow is about 8.1 m/s and maximum depth of debris flow is about 7 m when debris flow was occurred. The run-out distance after 500 sec is about 300 m from end of the valley. It is very well similar with actual debris flow run-out distance. From these results, we can presume the maximum velocity and depth of debris flow at that time.

Growth, Photosynthesis and Zinc Elimination Capacity of a Sorghum-Sudangrass Hybrid under Zinc Stress (고농도 아연 조건에서 수수-수단그라스 교잡종의 생장, 광합성 및 아연 제거능)

  • Plant biomass, photosystem II (PSII) photochemical activity, photosynthetic function, and zinc (Zn) accumulation were investigated in a sorghum-sudangrass hybrid (Sorghum bicolor ${\times}$ S. sudanense) exposed to various Zn concentrations to determine the elimination capacity of Zn from soils. Plant growth and biomass of the sorghum-sudangrass hybrid decreased with increasing Zn concentration. Symptoms of Zn toxicity, i.e., withering and discoloration of old leaves, were found at Zn concentrations over 800 ppm. PSII photochemical activity, as indicated by the values of $F_v/F_m$ and $F_v/F_o$, decreased significantly three days after exposure to Zn concentrations of 800 ppm or more. Photosynthetic $CO_2$ fixation rate (A) was high between Zn concentrations of 100-200 ppm ($22.5{\mu}mol$ $CO_2{\cdot}m^{-2}{\cdot}s^{-1}$), but it declined as Zn concentration increased. At Zn concentrations of 800 and 1600 ppm, A was 14.1 and $1.8{\mu}mol$ $CO_2{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. The patterns of stomatal conductance ($g_s$), transpiration rate (E), and water use efficiency (WUE) were all similar to that of photosynthetic $CO_2$ fixation rate, except for dark respiration ($R_d$), which showed an opposite pattern. Zn was accumulated in both above- and below-ground parts of plants, but was more in the below-ground parts. Magnesium (Mg) and iron (Fe) concentrations were significantly low in the leaves of plants, and symptoms of Mg or Fe deficiency, such as a decrease in the SPAD value, were found when plants were treated with Zn concentrations above 800 ppm. These results suggest that the sorghum-sudangrass hybrid is able to accumulate Zn to high level in plant body and eliminate it with its rapid growth and high biomass yield.

Simulation of the Debris Flow Using FLO-2D According to Curve-shape Changes in Bed Slopes (FLO-2D를 활용한 경사지 형상에 따른 토석류 흐름양상에 대한 수치모의)

  • Due to a high portion of mountainous terrains in Korea, debris flow and its disasters have been increased. In addition, recently localized flash-floods caused by climate change should add frequencies and potential risks. Grasping and understanding the behaviors of debris flow would allow us to prevent the consequent disasters caused by its occurrence. In this study, we developed a number of cases by changing the bottom slopes and their curvatures and investigated their effects on potential damage caused by the debris flow using FLO-2D. As simulating each bed slopes we analyzed for velocity, depth, impact, reach distance, and reach shape. As a result the lower the average slope, the greater the influence of its curvature and the numerical results were analyzed with showed a well-marked difference in impact stress and flow velocity. The result from this study could be referred for protecting from the debris flows when design countermeasure structures in mountainous regions.

Conclusion: Digestive, respiratory-related, but now the recognition of the Dameum changes in diet and lack of exercise, stress, lifestyle diseases due to climate change are increasing asa Dameum and women in the pathogenesis of disease high enough proportion of the treatment for Dameum Applications will be denied a lot of disease is considered.

Suggested and Preferred Amount of Clothing in a Winter Indoor Condition

  • This study proposes the suggested amount of clothing (SAC) and examines the preferred amount of clothing (PAC) for thermal comfort in mild cold conditions. Six male and nine female college students were systematically exposed to mild cold conditions by reducing the amount of clothing (Step I, 1.2clo ${\rightarrow}$ Step II, 1.0clo ${\rightarrow}$ Step III, 0.8clo ${\rightarrow}$ Step IV, 0.7clo). The subjects were then asked to adjust the amount of clothing to attain overall thermal comfort until they maintained thermal comfort for 10 minutes without changing the amount of clothing (Step V). The experiment was carried out in a climatic chamber at $19.5^{\circ}C$, 50%R.H. Body composition was measured and individual cold climate adaptability was surveyed before starting the experiment. Rectal temperature ($T^{re}$), skin temperature ($T_{sk}$), and oxygen consumption ($\dot{V}O_2$) were measured and the overall thermal sensation was voted in each step. PAC was obtained from the garments weight selected by each subject in Step V. SAC was proposed based on the change of oxygen consumption (${\Delta}\dot{V}O_2$). As a result, males showed higher $\bar{T}_{sk}$ and greater $O_2$ than females (p<.01). SAC obtained from $\dot{V}O_2$ were 652.0 (SE 3.9) g/$m^2$ for males and 766.0 (SE 2.5) g/$m^2$ for females and it was significantly different between groups (p<.01). PAC of males and females were 1.6 and 1.5 times heavier than SAC. In conclusion, females were more sensitive to the cold stress and recommended larger amount of clothing than males.

Growth and Yield Responses of Corn (Zea mays L.) as Affected by Growth Period and Irrigation Intensity

  • The frequency and intensity of soil moisture stress associated with climate change has increasing, and the stability of field crop cultivation has decreasing. This experiment was conducted to investigate the effect of soil moisture management method on growth and yield of corn. Soil moisture was managed at the grade of WSM (wet soil moisture, 34.0~42.9%), OSM (optimum soil moisture, 27.8~34.0%), DSM (dry soil moisture, 20.3~27.8%), and ESM (extreme dry moisture, 16.6~20.3%) during V8 (8th leaf stage)-VT (tasseling stage). After VT, irrigation was limited. The treated amount of irrigation was 54.1, 47.7, 44.0 and 34.5% of total water requirement, respectively. The potential evapotranspiration during the growing period was $3.29mm\;day^{-1}$, and upward movement of soil water was estimated by the AFKAE 0.5 model in the order of ESM, DSM, OSM, and WSM. We could confirm this phenomenon from actual observations. There was no significant difference in leaf characteristics, dry matter, and primary productivity depending on the level of soil moisture, but leaf development was delayed and dry weight decreased in DSM. However, dry weight and individual productivity of DSM increased after irrigation withdrawal compared to that of OSM. In DSM, ear yield and number of kernels per ear decreased, but water use efficiency and harvest index were higher than other treatments. Therefore, it is considered that the soil moisture is concentratedly managed before the V8 period, the V8-VT period is controlled within the range of 100 to 500 kPa (20.3~27.8%), and no additional irrigation is required after the VT.

Analysis of Gene Expression in Larval Fat Body of Plutella Xylostella Under High Temperature (고온에서 배추좀나방 유충 지방체의 유전자 발현 변화 분석)

  • BACKGROUND: Insects are ectothermic organisms in terrestrial ecosystems and play various roles such as controlling plant biomass and maintaining species diversity. Because insects are ectothermic, their physiological responses are very sensitive to environmental temperature which determines survival and distribution of insect population and that affects climate change. This study aimed to identification of genes contributing to fitness under high temperature. METHODS AND RESULTS: To identify genes contributing to fitness under high temperature, the transcriptomes of fat body in Plutella xyostella larva have been analyzed via next generation sequencing. From the fat body transcriptomes, structure-related proteins, heat shock proteins, antioxidant enzymes and detoxification proteins were identified. Genes encoding proteins such as structural proteins (cuticular proteins, chitin synthase and actin), stress-related protein (cytochrome P450), heat shock protein and antioxidant enzyme (catalase) were up-regulated at high temperature. In contrast expression of glutathione S transferase was down-regulated. CONCLUSION: Identifications of temperature-specific up- or down-regulated genes can be useful for detecting temperature adaptation and understanding physiological responses in insect pests.

Thermal and light impacts on the early growth stages of the kelp Saccharina angustissima (Laminariales, Phaeophyceae)

  • Anthropogenic disturbances, including coastal habitat modification and climate change are threatening the stability of kelp beds, one of the most diverse and productive marine ecosystems. To test the effect of temperature and irradiance on the microscopic gametophyte and juvenile sporophyte stages of the rare kelp, Saccharina angustissima, from Casco Bay, Maine, USA, we carried out two sets of experiments using a temperature gradient table. The first set of experiments combined temperatures between $7-18^{\circ}C$ with irradiance at 20, 40, and $80{\mu}mol\;photons\;m^{-2}\;s^{-1}$. The second set combined temperatures of $3-13^{\circ}C$ with irradiance of 10, 100, and $200{\mu}mol\;photons\;m^{-2}\;s^{-1}$. Over two separate 4-week trials, in 2014 and again in 2015, we monitored gametogenesis, the early growth stages of the gametophytes, and early sporophyte development of this kelp. Gametophytes grew best at temperatures of $8-13^{\circ}C$ at the lowest irradiance of $10-{\mu}mol\;photons\;m^{-2}\;s^{-1}$. Light had a significant effect on both male and female gametophyte growth only at the higher temperatures. Temperatures of $8-15^{\circ}C$ and irradiance levels of $10-100{\mu}mol\;photons\;m^{-2}\;s^{-1}$ were conditions for the highest sporophyte growth. Sporophyte and male gametophyte growth was reduced at both temperature extremes-the hottest and coldest temperatures tested. S. angustissima is a unique kelp species known only from a very narrow geographic region along the coast of Maine, USA. The coupling of global warming with high light intensity effects might pose stress on the early life-history stages of this kelp, although, as an intertidal species, it could also be better adapted to temperature and light extremes than its subtidal counterpart, Saccharina latissima.

Big Data Model for Analyzing Plant Growth Environment Informations and Biometric Informations (농작물 생육환경정보와 생체정보 분석을 위한 빅데이터 모델)

  • While research activities in the agricultural field for climate change are being actively carried out, smart agriculture using information and communication technology has become a new trend in line with the Fourth Industrial Revolution. Accordingly, research is being conducted to identify and respond to signs of abnormal growth in advance by monitoring the stress of crops in various outdoor environments and soil conditions. There are also attempts to analyze data collected in real time through various sensors using artificial intelligence techniques or big data technologies. In this paper, we propose a big data model that is effective in analyzing the growth environment informations and biometric information of crops by using the existing relational database for big data analysis. The performance of the model was measured by the response time to a query according to the amount of data. As a result, it was confirmed that there is a maximum time reduction effect of 23.8%.
