Occurrence of the strA-strB Streptomycin Resistance Genes in Pseudomonas Species Isolated from Kiwifruit Plants

  • The occurrence of strA-strB streptomycin-resistance genes within transposon Tn5393 was examined in Pseudomonas syringae pv. actinidiae, P. syringae pv. syringae, and P. marginalis, isolated from kiwifruit plants in Korea and Japan. PCR amplification with primers specific to strA-strB revealed that three of the tested Pseudomonas species harbored these genes for a streptomycin-resistance determinant. Tn5393, containing strA-strB, was also identified with PCR primers designed to amplify parts of tnpA, res, and tnpR. No IS elements were detected within tnpR, nor were they found in the intergenic region between tnpR and strA. Nucleotide sequence analysis indicated that the strA sequence of P. syringae pv. actinidiae contained a single nucleotide alteration at position 593 (CAA $\rightarrow$CGA), as compared to Tn5393a in P. syringae pv. syringae. This resulted in an amino acid change, from Gin to Arg.

Antibiotic resistance pattern of E. coli and Salmonella spp. isolated from chicken feces (닭 분변유래 E. coli 및 Salmonella spp.의 항생제 내성패턴)

  • The purpose of this study was to investigate the antibiotic resistance pattern of E. coli and Salmonella spp. isolated from chicken feces. One hundred and forty-seven E. coli isolates showed resistance to tetracycline (95.2%), erythromycin (89.2%), ampicillin (70.1%), streptomycin (59.2%), cephalothin (56.5%), sulfamethoxazole/trimethoprim (53.7%), ciprofloxacin (57.1%), enrofloxacin (59.2%) and norfloxacin (57.1%). The multiple resistance was seen in 144 isolates (97.9%) and the rate of five, six and seven drugs resistance pattern were 20.4%, 18.4% and 16.3%, respectively. Also, the multiple resistance of E. coli to twelve drugs were seen in 1 isolates (0.7%). Fourteen Salmonella spp. showed resistance to ampicillin (50.0%), streptomycin (57.1%), erythromycin (64.3%) and tetracycline (57.1%) and the rate of two and three drugs resistance pattern were 4 isolates (28.6%), respectively. The prevalence of resistant organisms in Korea probably reflects lack of proper antibiotic policy resulting in prolonged and indiscriminate use of antimicrobial agents.

Sensitivity of Staphylococcus aureus Isolated From Bovine Udders to Various Chemotherapeutic Agents (유우유방원(乳牛乳房源) Staphylococcus aureus 의 각종 화학요법제(化學療法劑)에 대한 감수성시험(感受性試驗))

  • The minimum inhibitory concentration (MIC) of five chemotherapeutic agents (penicillin, streptomycin, tetracycline, oxytetracycline and furazolidone) was measured for 126 strains of Staphylococcus aureus isolated from the udder of dairy cattle. The results obtained were as follows: 1. The MIC of penicillin, streptomycin, tetracycline, oxytetracycline and furazolidone ranged from 0.03 to 32 ug/ml, 0.06 to 128 ug/ml, 0.06 to 128 ug/ml, 1.0 to 512 ug/ml, and 0.06 to 32 ug/ml, respectively. The most frequent MIC of the above drugs were; penicillin 0.5ug/ml, streptomycin 1.0ug/ml, tetracycline 0.5ug/ml, oxytetracycline 4.0ug/ml, and furazolidone 2.0ug/ml. 2. The number of strains resistant to penicillin. streptomycin, tetracycline and oxytetracycline were 89(70.6), 9(7.1%), 10(7.9%), and 26(20.6%), respectively. Twenty-eight (29.2%) strains showed multiple resistance to more than two antibiotics tested.

Surveillance of antimicobial resistance ratio of E. coli and Enterococcus spp. isolated from fecal and carcasses of pigs in slaughterhouse (돼지 분변 및 도체에서 분리한 대장균, 장구균의 항생제 내성율 조사)

  • The present study was conducted to investigate isolation and antimicrobial resistance ratio of E. coli, E. faecium and E. faecalis from feces(l50 samples) and carcasses (150 samples) on slaughtered pigs from 6 slaughterhouse of 13 cities in the Gyeongnam during the period from January 2009 to December 2009. Isolation ratio of E. coli from feces and carcasses were 98 (65.3%) and 110(73.3%), respectively, and simultaneously, E. faecalis and E. faecium from feces and carcasses were isolated 21 (14%), 52(34.7%) and 18(12%), 14 (9.3%), respectively. All E. coli isolated from feces and carcasses except cefepime (0%) and ceftiofur (0%) were exhibited 2.4~83.6% of resistance to teteracycline (83.6%), ampicillin (68.2%), streptomycin (60%), chloram-phenicol (53.8%) and cephalothin (2.4%). All E. faecalis isolated from feces and carcasses except penicillin(0%) and vancomycin (0%) were exhibited 2.7~80.8% of resistance to teteracycline (80.8%), quinupristin/dalfopristin (78%), erythromycin (56.1%), streptomycin (43.8%) and bacitracin (2.7%). All E. faecium isolated from feces and carcasses except gentamicin (0%), vancomycin (0%), florfenicol (0%), linezloid (0%) and bacitracin (0%) were exhibited 3.1~53.1% of resistance to rifampin (53.1%), erythromycin and tetracycline (25%), penicillin (15.6%), ciprofloxacin (9.3%), and streptomycin, chloramphenicol, and quinupristin/dalfopristin (3.1%). According to the heard size, resistance ratio of E. coli strains isolated from feces and carcasses in slaughtered pigs-breeding farms over 1,500 heard to tetracycline, ampicillin, streptomycin and chloramphenicol showed higher resistance ratio (1.0~16.8%) than those of farms-breeding under 1,500 heard. From the our results, we suggest that a few of antimicrobials were used in the Gyeongnam than the other cities.

  • Clinically isolated Salmonella Enteritidis strain has a multi drug resistance plasmid, which confers ampicillin, chloramphe-nicol, sulfonamide, streptomycin and tetracycline, named pCAST2. We cloned a 7 kb Sacl fragment of pCAST2 which has sulfonamide, streptomycin and tetracycline resistance genes. The 7 kb SacI fragment showed the organization of sulII-strA-strB-tetR-tetA gene cluster which is different from the other clusters reported previously. In this study, we presented the method to detect this cluster by PCR analysis and showed that this cluster was found in Salmonella strains occurred sporadically at Kyungpook province in 2002.

  • A total of 41 Pseudomonas syringae pv. syringae, the causal agent of bacterial blossom blight, were isolated from kiwifruit plants in Korea. Among them, two strains showing streptomycin resistance were examined to investigate the structure of resistant determinants by PCR and nucleotide sequence analysis. PCR results suggested that the streptomycin resistance is mediated by strA-strB genes carried on Tn5393a. Insertion sequences, IS6100 and IS1133, which were located within or downstream of tnpR gene in Xanthomonas campestris and Erwinia amylovora were not found. Nucleotide sequences of strA-strB were 100% identical with Tn5393a. Two stretomycin resistant strains had three plasmids. Southern blot hybridization using strA-strB probe indicated that the resistant genes were carried on a 100kb plasmid.

  • 1993년과 1994년에 걸쳐 한국의 고추 재배 지역에서 분리한 고추 더뎅이병균은 66개의 균주중 24개가 race 1이었으며, 나머지는 42균주가 race 3이었다. Race 2균주는 발견되지 않았다. 총 66개의 균주중 8%가 200 $\mu\textrm{g}$/ml의 황산구리에 저항성을 보였으며, 100 $\mu\textrm{g}$/ml의 streptomycin sulfate에는 단지 한 균주만이 저항성을 나타냈다. Race 1균주중 21%가 구리에 저항성을 보였으나 race 3균주는 모두 구리에 감수성을 나타냈다. 모든 감수성 균주는 구리 농도 1 $\mu\textrm{g}$/ml에서 죽거나 아주 낮은 수준만이 생존하였으나 저항성 균주는 128 $\mu\textrm{g}$/ml에서도 생존하였다.

Isolation and Resistance Patterns of E. coli from Domestic Animals and Dranage in 1987 (가축 및 하천에서 분리한 대장균의 항균제내성)

  • One hundred and sixteen strains of E. coli isolated from drainage in Seoul city in 1987 and 104 strains of E. coli isolated from stools of domestic animals in suburb of Seoul in 1987 were examined for susceptibilities to 10 antimicrobial agents by the agar dilution method. The susceptibilities of the two groups to each antimicrobial agent were compared and correlations in the antimicrobial susceptibility of the 220 strains of E. coli among the 10 antimicrobial agents were also analyzed. The frequency of resistance strains was highest to tetracycline with 77%, and followed by chloramphenicol, streptomycin, ampicillin, kanamycin and cephalosporin in the descreasing order, tanging from 62% to 10%. Strains resistant to tobramycin, nalidixic acid, gentamicin and amikacin occupied 3 strains, 2 strains, 2 strains and 1 strain respectively.

  • Decreased porin-mediated outer membrane penetration of hydrophilic antibiotics is a common mechanism of antibiotic resistance in Gram-negative bacteria. This study was undertaken to determine whether a null mutation in Pseudomonas putida would suppress porin synthesis, and therefore reduce the susceptibility of the organism to streptomycin, norfloxacin, and tetracycline. Inverse PCR amplification and double-stranded DNA sequencing were used to identify chromosomal genes carrying TnphoA'-1 inserts. Genome database available was used to identify putative homologue genes, one of which encodes protein with homology to domains of the MutS of P. putida, suggesting a crucial role in the multidrug resistance. Increased resistance to streptomycin, norfloxacin, and tetracycline might be due to accumulation of compensatory mutations. Either no growth or slow growth was observed in P. putida KH1027 when grown in minimal medium containing gluconate, glucose, or citrate; however, it is not clear whether the growth patterns contributed to the multidrug resistance.

Investigation on antimicrobial resistance genes of Salmonella spp. isolated from pigs and cattle (소와 돼지유래 살모넬라속균의 약계내성유전자의 특성에 관한 연구)

  • At the present study, it was aimed to detect virulence genes and antimicrobial resistance genes among 102 strains of 12 Salmonella serotypes isolated from pigs and cattle. In polymerase chain reaction (PCR), invA was detected from all strains of Salmonella spp., spvC was detected from Salmonella enterica serotype Enteritidis (S. Enteritidis) (100%), S. Bradenburg (75%), and S. Typhimurium (20.4%). Drug resistance related genes of 12 types were detected from all strains. TEM ($bla_{TEM}$) gene was detected from 51 (92.7%) of 55 $\beta$-lactams (54 ampicillin or 1 amoxicillin) resistance strains. 55 (100%) of 55 chloramphenicol resistance strains, 3 (100%) of 3 gentamicin resistance strains and 5 (100%) of 5 kanamycin resistance strains did contain cml, aadB, and aphA1-Iab, respectively. strB (89.9%), strA (88.4%), aadA2 (84.1%) and aadA1 (72.5%) were detected from 69 streptomycin resistance strains. sulII and dhfrXII were detected from 49 (100%) of 49 sulfamethoxazole/trimethoprim resistance strains, but sulI was not detected. tetA (97.9%) and tetB (21.6%) were detected from 97 tetracycline resistance strains. int gene was detected from 58 (56.9%) of 102 strains. 54 S. Typhimurium of 102 Salmonella spp. were attempted to detect drug resistance genes. TEM was detected from 44 (95.7%) of 46 $\beta$-lactams (45 ampicillin or 1 amoxicillin) resistance strains. cmlA was detected from 51 (100%) of 51 chloramphenicol resistance strains. aadA2 (100%), strA (100%), strB (100%), and aadA1 (79.6%) were detected from 54 streptomycin resistance strains. sulII (100%) and dhfrXII (100%) were detected from 49 sulfamethoxazole/trimethoprim resistance strains. tetA was detected from 54 (100%) of 54 tetracycline resistance strains. int gene was detected from 54 (100%) of 54 strains. The major drug resistance pattern and resistance gene profile were ampicillin, chloramphenicol, streptomycin, sulfamethoxazole/trimethoprim and tetracycline (ACSSuT) and TEM, cmlA, aadA1, aadA2, strA, strB, sulII, dhfrXII, tetA and int, respectively.
