Cathepsin B 저해물질을 생산하는 Streptomyces aburabiensis의 분리 및 동정 (Isolation and Identification of Streptomyces aburabiensis Producing Cathepsin B Inhibitor)

  • The aim of the present study was to develop strains of actinomycetes producing low molecular weight cathepsin B inhibitor. Among 700 isolates from soil samples, a strain of Streptomyces sp. SMF30 producing cathepsin B inhibitor showing specificity and heat stability was selected by an economical and effective screening method. 50 units characteristics for major cluster analysis and 34 units characteristics for minor cluster were tested and the data were analyzed numerically using the TAXON program. The Isolate SMF30 was identified as a strain of Streptomyces aburabiensis

방선균이 생성하는 단백질 가수분해효소 저해물질의 생산 (Production of Protease Inhibitor from Streptomyces sp. SK-862)

  • A inhibitor acting on substrate proteolytic enzyme was isolated from culture broth of Streptomyces sp. SK-862, which had been isolated from soil in Wonju City, by using the colloidal agar medium. The optimum culture temperature and initial pH for the production of the protease inhibitor was 28$^{\circ}C$ and pH 8.5, respectively. The optimum culture medium was composed of 1.5% glucose, 0.5% peptone, 0.1% K2PHO4, 0.05% CaCO3 and initial pH 8.5. The inhibitor production was maximum when the strain was incubated in shaking incubator at 70 strokes for 60 hours.

  • A strain of Streptomyces sp. SK-862, isolated from soil in Wonju city, was able to prodce a biologically active substance that has a strong inhibitory activity against proteolsis by trypsin. The inhyibitory substance was extracted by n-butanol, and then purified by the adsorption chromatography followed by the reverse-phase high performacne liquid chromatography. The purified substance was stable over the pH range from 2 to 10, but was unstable when treated at 8$0^{\circ}C$ for 60 min. This substance was soluble in water, methanol, ethanol nd butanol, but insoluble in chlorofrom and ethylacetate. The Rf value of the purified substance on the thin layer chromatography were 0.56 in n-butanol : methanol : water(5 : 3 : 1v/v) solvent system compare dto 0.23 in ethanol : ammonium hydoxide : water(8 : 1 : 1v/v) solvent system. This substance has maximum absorption at 259 nm. The chemical reaction of the substance was negative for sugar but positive for ninhydrine and iodine reaction.

Inhibition of DNA Topoisomerase I by Cyclo(L-Prolyl-L-Phenylalanyl) Isolated from Streptomyces sp. AMLK-335

  • Cyclo(L-prolyl-L-phenylalanyl) [cyclo(pro-phe)] was isolated from Streptomyces sp. AMLK-335 and found to inhibit DNA topoisomerase I activity. In a DNA relaxation assay using supercoiled pBR322 DNA, cyclo(pro-phe) inhibited the DNA topoisomerase activity more strongly than camptothecin, a known topoisomerase inhibitor. However, at a concentration of $10{\mu}M$, cyclo(pro-phe) produced a lower degree of DNA relaxation than camptothecin, therefore, the inhibition of topoisomerase I activity by cyclo(pro-phe) was also found to be dose dependent. Accordingly, the current results suggest that cyclo(pro-phe) may be a novel inhibitor of topoisomerase I.

  • Three more unusual macrolides in addition to concnamycin B were isolated from the mycelium of Streptomyces sp. strain Bal6. These four compounds showed a potent cytotoxity to hunian cancer cell lines, SNU-1 (stomach cancer cell line), SNU-354 (liver cancer cell line), MCF- 7 (breast cancer cell line) and KB-3-1 (oral epidermoid carcinoma cell line). Interestingly, these compounds confered slight differential cytotoxity on RHEK-1, a human epidermal keratinocyte cell line immotalized by AD12-SV40 hybrid virus and RHEK-1/pSV$_{2}$ ras which was resulted from H-ras transfomation of RHEK-1. These compounds were determined to be concanamycin A, conca- namycin E and 0-methyl concanamycin B by NMR and other spectral analysis.

Isolation of Marine Actinomycetes with Antimicrobial Activity from East Coast of Korea

  • The marine actinomycetes are very interesting microorganisms for finding of new antibiotics, because they are inhabited in marine environmental conditions, such as salinity, organic material and low concentration of nutrient components that are differ from those of land. In this study, the actinomycetes producing antibiotics from sea waters and bottom deposit samples collected at east coast of Korea were investigated to invent new antibiotics. Nine actinomycetes strains having the antimicrobial activity of 431 actinomycetes were selected from sea waters and bottom deposit samples. Among the 9 strains, the strain having the highest antimicrobial activity was classified to Streptomyces sp. and named Streptomyces sp. NS 13239.

Optimum Condition of Marine Actinomycetes, Streptomyces sp. NS 13239 for Growth and Producing Antibiotics

  • In previous study, marine actinomycetes producing the antibiotics were investigated to invent new antibiotics from east coast of Korea. The optimum growth conditions of Streptomyces sp. NS 13239 were $28^{\circ}C$, pH 7.0 and $3\%$ of NaCl concentration in various media. Streptomyces sp. NS 13239 showed strong antimicrobial activity against gram-positive bacteria, specially Methicillin resistant Staphylococcus aureus (MRSA), but just weak antimicrobial activity against yeasts and mold. On the other hand, it did not show antimicrobial activity against gram-negative bacteria. The optimum conditions for producing antibiotics were almost consistent with optimum growth conditions except carbon source and nitrogen source.

Isolation of Sphinin, an Inhibitor of Sphingomyelinase, from Streptomyces sp. F50970

  • Sphingomyelinase (SMase EC:3.l.4.l2) has been suggested to play important roles in the cell cycle, differentiation, apoptosis, inflammation, and the regulation of eukaryotic stress responses. SMase inhibitors may be a powerful tool to elucidate and regulate these cellular responses in which SMase involves. We first isolated an SMase inhibitor, named sphinin, from a strain of soil actinomycetes, F50970. Sphinin inhibited Mg/sup 2+/ -dependent neutral SMase from chicken embryo at 1.2 ㎍/㎖ of IC/sub 50/ Sphinin also inhibited acidic SMase, but it had no inhibitory activity on PI-PLC and PC-PLC, suggesting that sphinin is a specific inhibitor of SMase. The strain F50970 was identified as a Streptomyces sp. by its spiral spore chain, LL-diaminopimelic acid, menaquinone patterns of MK-9 (H'6) and MK-9 (H'8), FA-2c type of fatty acid pattern, and other morphological, physiological, and cultural characteristics.

Purification and Characterization of Proteinaceous ${\beta}-Lactamase$ Inhibitor from the Culture Broth of Streptomyces sp. SMF-19

  • The aim of this study is to elucidate characteristics of ${\beta}-lactamase$ inhibitor produced by Streptomyces sp. SMF-19 isolated from soil was found to produce proteinaceous extracellular ${\beta}-lactamase$ inhibitor. The ${\beta}-lactamase$ inhibitor was purified through ammonium sulfate fractionation, gel filtration, anion exchange chromatography and fast performace liquid chromatography. The molecular weight of the ${\beta}-lactamase$ inhibitor was estimated to be about 48,000 by SDS-PAGE. The mode of inhibition against penicillin G as a substrate was uncompetitive. The ${\beta}-lactamase$ inhibitor was stable in wide pH range but unstable at high temperature above $50^{\circ}C$.

  • Dehydroaltenusin (1) was isolated from the chloroform extract of the culture filtrate of a Streptomyces sp. and its structure was determined from spectral data as well as by comparison with published values. In an XTT-based in vitro anti-HIV assay, dehydroaltenusin effectively inhibited the cytopathic effects of HIV infection at a concentration of $1-5\;{\mu}g/mL$.
