Streptomyces sp. YJB-599가 생산하는 Genistein의 분리 및 정제

  • A cytotoxic material was produced by strain No. 5-99 which was isolated from soil. Analyzing the cell wall components, LL-diaminopimelic acid was identified. From the existance of glycine in the cell wall, this strain was identified to Streptomyces sp. which has cell wall chemotype I and peptidoglycan type A3 connected by glycine. So, we named this strain to Streptomyces sp. YJB-599. The Active material was purified through solvent extraction, silica gel column chromatography and crystallized to needle-shaped white -crystal. Analyzing the structure of this crytal by instrumental analysis and database, it was determined to genistein.

  • Genistein, a inhibitor of the progression of G$_{1}$ and G$_{2}$ phase of the mammalian cell cycle, was discovered through a unique screening system, in which effects of microbial metabolites on the cycle progression of the cultured mouse mammalian carcinoma cell were monitored by flow cytometry. The inhibitor was extracted from the fermentation broth of Streptomyces sp. ZF10 with ethyl acetate, and purified by silica gel column chromatography and HPLC.

  • In fermentation studies it revealed that Streptomyces sp. SMF 3001 started to synthesize extracellular alkaline protease from early exponential phase of cell growth. The biosynthesis of the alkaline protease was greatly induced by skim milk as a sola nitrogen source and further stimulation was observed under inorganic sulphur limited culture. However, it was found that the biosynthesis was apparently repressed by $NH_4^+$ and free amino acids, specially by cysteine. It was considered that the strain SMF 301 of Streptomyces sp. would produce the alkaline protease for the uptake of sulphur compounds from protein contained in the culture broth.

  • The limited effectiveness of current plant disease management treatments necessitates the development of new methods for controlling diseases using beneficial microbes. Demanding sustainable agriculture is increasingly highlighted as a biocontrol approach, particularly Streptomyces species known to produce a variety of antibiotic compounds and secondary metabolites. The Streptomyces globisporus SP6C4 strain and Streptomyces sp. S8 have been reported as potent antifungal agents and are gaining attention for improving crop growth in sustainable agriculture. In this study, we investigated the use of Streptomyces species formulations to enhance bacterial growth with nitrogen sources. Specifically, the addition of L-glutamic acid and L-cysteine resulted in earlier sporulation and bacterial growth in Streptomyces strains, respectively. This approach could expand the range of fermentation techniques in agriculture and be useful for controlling plant growth-promoting bacteria.

Control of Ginseng Damping-off by Streptomyces sp. A75 and A501 (Streptomyces sp. A75와 A501 균주의 인삼 잘록병에 대한 방제효과)

  • Streptomyces sp. A75 and A501 inhibited the mycelial growth of pathogenic Rhizoctonia solani and Pythium sp., which cause the ginseng disease known as damping-off. Three methods were evaluated for the control of these pathogens, using a mixture of the culture broths from Streptomyces sp. A75 and A501. The methods tested were seed dipping with 50-fold diluted broth, drenching of soil with 100-fold diluted broth after sowing, and combined seed dipping and drenching. These methods reduced the incidence of ginseng damping-off caused by R. solani by 81.3%, 84.8%, and 32.2% and that caused by Pythium sp. by 51.0%, 52.1%, and 75.3%, respectively. Based on these results, the combination of seed dipping and soil drenching after sowing using a mixture of the culture broths from Streptomyces sp. A75 and A501 effectively reduced the incidence of damping-off in ginseng.

Analysis of Antifungal Proteins in Streptomyces sp. SAR01 (Streptomyces sp. SAR01 균주에서의 항진균 관련 단백질 분석)

  • To analyze proteins related to antifungal activity, SAR01 strain was isolated from seaweed and identified as Streptomyces sp. from the result of FAME (fatty acid methyl ester) analysis. The isolated strain had antifungal activities against T species of plant pathogenic fungi. Antifungal activity deficient mutant (SAR 535) of Streptomyces sp. SAR01 was induced by gamma radiation $(^{60}Co,\;5kGy)$. By 2 D electrophoresis analysis, 6 protein spots were found in wild strain (SAR01) but these spots disappeared in mutant strain (SAR535). Among them, 5 proteins showed similarities to heat shock protein 70(HSP70), Fe-containing superoxide dismutase II (Fe- SODII), ribosome recycling factor (RRF), 10 kDa chnperonin (GroES) and inorganic pyrophosphatase (PPAse), respectively. It suggested that the above 6 proteins could be closely related to the antifungal activity of Streptomyces sp. SAR01.

  • Four actinomycetes which produces extracellular chitinase were isolated from soil and organic matter all over the Chonnam provincial area. The chemical composition, morphological, cultural and physiological properties of isolated strain S-25, S-42, S-172 and S-267 were studied in relation to the toxonomical properties. All of strains contained phospholipids such as phosphatidyl ethanolamine, phosphatidyl inositol and diphosphatidyl glycerol. The components of cell wall in all strains have L, L-DAP, Glutamic acid, Alanine, Glycine and Glucosamine. The surface of spore is smooth and colony is grey in all strains, Based on the results obtained in these experiments all of strains are identified as Streptomyces sp.S-25, Streptomyces sp.S-42, Streptomyces sp.S-l72 and Streptomyces sp. S-267.

  • Three isoflavonoids having tyrosinase-inhibiting activity were isolated from the culture filtrate of Streptomyces sp. 20747. Their structures were determined by UV, EI-MS, 1H-NMR, 13C-NMR to be daidzein, daidzein 7-rhamnoside, and genistein 7-rhamnoside, which were competitive with substrate and had IC50 value of 14, 19, and 16 ng/ml, respectively to mushroom tyrosinase and did not inhibit melanin production of Streptomyces bikiniensis. Soybean meal as well as peptone were found to be a good nitrogen source for tyrosinase-inhibiting isoflavonoids production, susgesting that soybean meal is not the origin of tyrosinase inhibiting isoflavonoids formation in Streptomyces sp. 20747 strain.

  • A potent actinomycetes strain was selected to digest raw starch, which was classified as a strain of Streptomyces sp.. Its amylase production was maximized when it was grown on wheat bran extract media added 4% of cooked corn starch and 0.16% of potassium nitrate for 6 days at 3$0^{\circ}C$ and initial pH 6.2.
