  • 제주지역 양식 넙치의 주요 세균성 질병의 일종인 연쇄구균증 원인균 Streptococcus iniae 및 Streptococcus parauberis에 대한 생물학적 특성을 시험하였다.온도별 성장 시험결과 S. parauberis 균주는 10℃ 배양조건에서 성장이 이루어졌으나 S. iniae 균주의 경우는 10℃에서는 성장이 확인되지 않았으며, 용혈성 시험결과에서는 S. iniae 균주의 경우에만 β용혈성을 나타내었다.생화학적특성 시험결과 acetoin production test (VP test), hippurate hydrolysis test (HIP test)에서는 S. parauberis 균주의 경우에만 양성반응을 보여 두 균주사이에 뚜렷한 차이를 보였다.또한 α-D-glucose, D-mannose, D-psicose, D-trehalose, pyruvatic acid methyl ester, glycerol은 두 균주 모두 기질로 이용하였으나 L-lactic acid는 S. iniae 균주만이 이용하였고, β-methyl-D-glucoside의 경우는 S. parauberis 균주만이 기질로 이용하는 것으로 나타났다.병원성 시험결과, S. iniae는 급성감염 형태를 보인 반면에 S. parauberis는 만성 감염형태를 보임으로써 차이를 나타내었다.

  • Here, we have cloned and sequenced Streptococcus parauberis pgm gene, encoding the enzyme phosphoglucomutase (PGM), which is known to be in association with virulence in other streptococcal species. The PGM of S. parauberis is the most closely related to that of S. iniae based on their amino acid sequences.

발효배양액에서 Streptococcus parauberis Z49균주가 생산하는 Bacteriocin의 간편한 추출 (Simple and Rapid Extraction of a Bacteriocin Produced by Streptococcus parauberis Z49 from Fermented Cultures)

  • Streptococcus parauberis Z49가 생산하는 새로운 bacteriocin의 특성을 조사하고 수성 이상계를 사용하여 발효배양액에서 효율적으로 추출하였다. Nisin과 유사한 S. parauberis Z49의 bacteriocin은 $121^{\circ}C$에서 15 min 열처리해도 활성이 있었으며, 넓은 범위의 pH (pH 2-12)에서 안정하며, M. luteus, Lactobacillus sp., L. fermentum, E. faecium, L. monocytogenes, and P. fluorescens의 생육을 억제하였다. S. parauberis Z49의 발효배양액에 있는 bacteriocin을 간편하게 분리하기 위한 PEG 600/$Na_2SO_4$ 수성 이상계의 최적조건은 PEG 600 15%, $Na_2SO_4$ 30%, NaCl 8% 이었으며, bacteriocin은 PEG층에서 농축이 되었다.

  • Edwardsiella tarda, Streptococcus iniae and S. parauberis are main bacterial pathogens in aquaculture farms of olive flounder, Paralichthys olivaceus. We have discussed the efficacy and safety of 3 type-combined vaccines (A: S. iniae 1mg + S. parauberis 1mg + E. tarda 1mg, B: S. iniae 1mg + S. parauberis 1mg + E. tarda 0.5mg, C: S. iniae 1.5mg + S. parauberis 1.5mg + E. tarda 1mg) through intraperitoneal injections in olive flounder. None of the vaccines gave rise to any signigicant side effects on histopathology and blood chemistry. The antibody titers and lysozyme activities of A type were higher than those of B, C and control. Four weeks after vaccination, RPS (relative percent survival rates) was 62.5~75% (A type), 50~66.7% (B type) and 55.6~62.5% (C type) respectively. As the results, the combined vaccines are possible to prevent edwardsiellosis and streptococcosis, and A type : S. iniae 1mg + S. parauberis 1mg + E. tarda 1mg, is the most effective out of them.

  • Streptococcosis, caused by Streptococcus iniae and Streptococcus parauberis is an important bacterial disease that affects in olive flounder in Korea. In Korea, multivalent bacterial vaccines are used to prevent streptococcal diseases in aquaculture. In this study, commercial vaccines containing formalin-inactivated bacterial cells of S. iniae and S. parauberis were administered at six fish farms and one unvaccinated fish farm were designated for investigation (Wando; 4 sites and Jeju; 3 sites). Blood was collected from vaccinated and unvaccinated olive flounders, and titers of antibodies against S. iniae and S. parauberis in serum were analyzed using ELISA. After a one shot vaccination in the farms at Jeju (farm A) and Wando (farm D), the proportion of individuals with specific antibodies against S. parauberis OD values of 0.4 or higher was 60% and 53.5%, respectively. But after booster vaccination, the proportion of individuals with serum OD values of 0.4 or higher was higher substantially increased to 96.6% (farm A) and 100% (farm D). The levels of S. parauberis specific antibodies of olive flounder were increased after vaccination in three fish farms (farm D, E, and F), but not S. iniae specific antibodies.

Gliotoxin is Antibacterial to Drug-resistant Piscine Pathogens

  • By activity-guided fractionation, gliotoxin was isolated as an antibacterial metabolite of the fungus Penicillium decumbens which was derived from the jellyfish Nemopilema nomurai. Gliotoxin was further evaluated for antibacterial activity against several piscine and human MDR (multidrug resistance) pathogens. Gliotoxin showed significant antibacterial activity against Gram-positive piscine pathogens such as Streptococcus iniae FP5228, Streptococcus iniae FP3187, Streptococcus parauberis FP3287, Streptococcus parauberis SPOF3K, S. parauberis KSP28, and Lactococcus garvieae FP5245. Gliotoxin showed strong activity especially against S. parauberis SPOF3K and S. iniae FP5228, which are resistant to oxytetracycline. It is noteworthy that gliotoxin effectively suppressed streptococci which are the major pathogens for piscine infection and mortality in aquaculture industry. Gliotoxin also showed strong antibacterial activity against multidrug- resistant human pathogens (MDR) including Enterococcus faecium 5270 and MRSA (methicillin-resistant Staphylococcus aureus) 3089.

Efficacy of a vaccine against Streptococcus parauberis infection in starry flounder Platichthys stellatus Pallas

  • Starry flounder, which are recently increasingly cultured in Korea, are known to highly vulnerable to Streptococcus parauberis infection. Five groups of starry flounder (n=30 for each group) were vaccinated with S. parauberis formalin-killed whole cells by intraperitoneal injection at a final concentration of 0, 0.01, 0.1, 1 and 10 mg $fish^{-1}$. Specific antibody production of 1 and 10 mg $fish^{-1}$ administered groups significantly increased at four weeks post immunization. All vaccinated groups showed higher survival rates than a control group when five groups of fish were challenged with S. parauberis at a dose of $1.14{\times}10^4$ cfu $fish^{-1}$ and $1.14{\times}10^2$ cfu $fish^{-1}$, respectively. In particular, 0.1 or higher concentrations of formalin killed bacterial cells are able to confer the fish high protection against S. parauberis infection.

제주도 양식넙치 (Paralichthys olivaceus)로부터 분리한 비 용혈성 연쇄구균의 동정 (Characterization of Streptococcus parauberis isolated from cultured Olive flounder, Paralichthys olivaceus in the Jeju Island)

  • 본 연구에서는 국내 연구보고는 아직 없는 실정이나 양식터봇의 연쇄구균 병원체로 보고 된 바가 있는 제주 양식넙치에서 분리되는 γ 용혈성의 연쇄구균인 Streptococcus parauberis의 특성을 제시하고자 하였다. S. parauberis에 대한 분리균주의 탄소원을 기질로 이용하는 특성은 Annette and Collllins (1990)가 보고한 결과와 유사하였다.또한 분리된 균주들을 계통분류학적 분석 결과 Accesion number AY584477 하나의 그룹으로 묶였지만 S. iniae와는 다른 그룹으로 묶였다. 그러나 Collins가 Streptococcus iniae (X58306)와 S. parauberis (X89967)이 한 그룹으로 묶인다고 발표한 것과는 다른 결과를 나타냈다. 16S rRNA gene에 대한 염기서열 분석 결과는 S. parauberis (AY942572)와 98%이상의 상동성을 나타내어 S. parauberis로 동정하였다. S. parauberis는 S. uberis type Ⅱ로 분류되었으나 Williams and Collins (1990)에 의해서 S. parauberis라는 종명이 제시되었다. 이러한 결과로 볼 때 제주도 양식넙치의 연쇄구균증 병원체로 S. parauberis로 판단되나 이번 실험에서 감염실험이 수행되지 않아 앞으로 본 병원체에 대한 감염 실험과 병리학적인 연구가 필요하다고 사료되어진다.

넙치(Paralichthys olivaceus)의 Streptococcus parauberis 인위감염을 위한 공격실험 방법에 관한 연구 (An Experimental Study on Artificial Infection of Olive Flounder Paralichthys olivaceus by Streptococcus parauberis Using Different Injection Sites)

  • Streptococcosis in the olive flounder Paralichthys olivaceus can be caused by Streptococcus parauberis. We compared three routes of administration for experimental injections of the S. parauberis 19FBSPa0003 strain in the olive flounder. Pathological changes were observed during the experimental infection. Inflammation of the serous membrane in the liver, intestine, spleen and heart was the major pathological change found in the infected olive flounder. No mortality was observed in fish that received intraperitoneal (IP) injection at less than 1×104 colony-forming unit (CFU)/fish. The lethal dose 50 for olive flounder, given an intravenous (IV) injection, was 7.94×104 CFU/fish. Fish with a higher concentration of IV injected S. parauberis (1×108 CFU/fish) died within a maximum of two days. However, serious necrosis and bacterial proliferation in ellipsoidal cells of the spleen and heart tissues were found in moribund or dead fish, 1-2 days after IV injection. Similar histopathological signs were observed in olive flounder inoculated by subcutaneous (SC) infected and naturally infected. In addition, SC was also strongly associated with bacteria concentration and cumulative mortality rate. Based on these results, SC is the recommended method for artificial infection by S. parauberis in the olive flounder.

In vitro Antibacterial and Synergistic Activity of an Ecklonia cava Extract against Anti biotic-Resistant Streptococcus parauberis

  • In an effort to discover alternative phytotherapeutic antimicrobial agents to combat Streptococcus parauberis, a fish pathogenic bacterium, we evaluated the antibacterial activity of seaweed extracts in vitro. A methanolic extract of Ecklonia cava exhibited strong antibacterial activity against S. parauberis isolated from olive flounder Paralichthys olivaceus. Furthermore, the n-hexane soluble (Hexane) fraction of the E. cava methanolic extract exhibited the greatest antibacterial effect on S. parauberis strains with a minimum inhibitory concentration (MIC) ranging from 256 to $1,024{\mu}g/mL$. In addition, the MIC values of oxytetracycline against antibiotic-resistant S. parauberis were markedly reduced up to 64-fold in combination with the Hexane fraction, suggesting that the antibacterial activity of the antibiotic was restored when combined with the Hexane fraction. The interaction between both antibiotics and the Hexane fraction was assessed by the fractional inhibitory concentration (FIC) index. The Hexane fraction and oxytetracycline combination against antibiotic-resistant S. parauberis strains resulted in a median ${\sum}FIC$ range of 0.502 to 0.516. Thus, the synergistic ranges of median ${\sum}FIC$ < 1 were observed for all combinations of the Hexane fraction and oxytetracycline against S. parauberis. To the best of our knowledge, this is the first report indicating the efficacy of an E. cava extract against fish pathogenic bacterium S. parauberis.
