Plant Extracts Inhibiting Biofilm Formation by Streptococcus mutans without Antibiotic Activity

  • Streptococcus mutans causes oral diseases, including tooth decay, by producing a biofilm called plaque. Therefore, inhibition of biofilm formation is essential for maintaining oral health. Plants produce a variety of secondary metabolites, which act as starting sources for the discovery of new bioactive chemicals that inhibit biofilm formation of S. mutans. Previous studies have reported on chemicals with antibiotic activity for the inhibition of biofilm formation by S. mutans. In this study, nine plant extracts from Melonis Pedicellus, Agastachis Herba, Mori Cortex Radicis, Diospyros kaki leaves, Agrimoniae Herba, Polygoni Multiflori Radix, Lycopi Herba, Elsholtziae Herba, and Schizonepetae Spica were screened for the inhibition of biofilm formation from a plant extract library. The water-soluble compounds of the extracts did not affect cell growth but selectively inhibited biofilm formation. These results suggest that the selected plant extracts constitute novel biofilm formation inhibitors, with a novel biological mechanism, for improving oral hygiene.

Anticariogenic Activity of Callistemon citrinus Extract against Streptococcus mutans

  • Streptococcus mutans is known as a strong cariogenic bacterium and produces dental caries. This study was carried out to develop anticariogenic agents from Callistemon citrinus. Crude extracts and thier various solvent fractions of the plant extracts were tested for the antibacterial activity against S. mutans. The anticariogenic activity was evaluated with by paper disc diffusion method and minimal inhibition concentration (MIC). The extract from the fruit and its hexane fraction showed a strong inhibitory effect on the growth of S. mutans and the MICs were 31.2 and $3.9{\mu}g/m{\ell}$, respectively.

Conditions for Preparing Glycyrrhiza uralensis Extract for Inhibiting Biofilm Formation of Streptococcus mutans

  • Licorice, which has an extensive history of use as an herbal medicine, has been suggested to have oral health benefits. However, to date, no systematic study has been conducted on the preparation method of licorice extracts for oral health. In this study, licorice extracts prepared using water and ethanol were investigated for its ability to inhibit the biofilm formation of Streptococcus mutans. The licorice extract prepared with around 60% ethanol effectively inhibited the biofilm formation of S. mutans. Licorice extracted with 50% ethanol almost completely inhibited the biofilm formation at 1.5 g/L of licorice extract. This inhibitory activity was confirmed in a microplate assay and a flow cell system. Glycyrrhetic acid was extracted from licorice effectively with 60% ethanol concentration. The strong inhibitory activity of glycyrrhetic acid and the synergistic inhibition with glycyrrhizin on biofilm formation were suggested as major reasons for a concentration-specific extraction. These results suggest that licorice extract prepared using around 60% ethanol effectively inhibits the biofilm formation of S. mutans.

혼합치열기 어린이에서 Streptococcus mutans와 Streptococcus sobrinus의 분포도 조사 (PREVALENCE OF STREPTOCOCCUS MUTANS AND STREPTOCOCCUS SOBRINUS IN CHILDREN WITH MIXED DENTITION)

  • 본 연구는 125명의 6-11세 혼합치열기 아동을 대상으로 연령에 따라 초기 혼합치열기 A군(6-8세; 62명)과 후기 혼합치열기 B군(9-11세; 63명)으로 분류하고 유치의 dfs, 영구치의 DFS를 기록한 후 자극성 타액을 채취하여 TYCSB 배지에 배양해서 S. mutans와 S. sobrinus를 구분하였으며 PCR을 시행하여 확인하였다. S. mutans와 S. sobrinus의 분포도와 치아우식증과의 상관관계를 조사하여 다음과 같은 결과를 얻었다. 1. S. mutans와 S. sobrinus의 상관계수는 A군 0.70, B군 0.50로 두 군 모두에서 높은 상관관계를 보였다. 2. S. mutans와 치아우식증과의 관계는 A군(r=0.25)과 B군(r=0.34) 모두에서 약간의 관련성이 나타났다. 3. A군에서 S. sobrinus와 치아우식증간에 상관관계가 없는 것으로 나타났으며, B군(r=0.21)에서는 미약한 상관관계가 성립되었다. 4. S. mutans와 나이의 상관관계는 A군과 B군 모두에서 관련성이 없는 것으로 나타났다. 5. S. sobrinus와 나이의 상관관계는 A군(r=0.32)에서는 약간의 관련성이 나타났고, B군에서는 관련성이 없는 것으로 나타났다.

AP-PCR을 이용한 다발성 우식아동의 구강내 Streptococcus mutans의 유전자형 분류 (GENOTYPING OF STREPTOCOCCUS MUTANS USING AP-PCR IN CHILDREN WITH RAMPANT CARIES)

  • For the purpose of evaluating the appropriateness of AP-PCR as a facile, rapid and reproducible method for genotyping Streptococcus mutans, and selecting the discriminant primer for it, a DNA fingerprinting was performed on the microorganisms isolated from caries-free children and children with rampant caries, respectively. In the course of selecting appropriate primer for S. mutans genotyping, we chose S2 primer from 6 different primers which shows highest resolution on the agarose gel as well. Nineteen kinds of fingerprint patterns were observed in caries-free children and children with rampant caries which were produced by combination of 7 different fragments. Interestingly, the number of types observed in caries-free children was greater than that in children with rampant caries. And we observed Type 2 was predominant in children with rampant caries (about 80%) and relatively even distribution of each types in caries-free children. Furthermore, it was appeared that the major types in normal control were not or rarely found in children with rampant caries. In conclusion, we could establish simple, rapid and highly reproducible AP-PCR method for genotyping S. mutans. We also found differences in distribution of S. mutans between normal and patient, which suggested that cariogenicity is also dependent on qualitative aspects which is caused by the difference in genotypes of S. mutans in oral cavity.

나노컴포지트에서 Acidulated Phosphate Fluoride 적용에 따른 Streptococcus mutans 부착량 변화 (CHANGES IN ADHESION OF STREPTOCOCCUS MUTANS TO NANOCOMPOSITE RESINS AFTER ACIDULATED PHOSPHATE FLUORIDE GEL APPLICATION)

  • 복합레진 표면에 대한 APF gel 도포는 표면을 거칠게 하고 세균 부착을 증가시킨다. 본 연구는 Filtek Z250(FZ), Filtek Supreme Universal(FS)과, 실험적으로 나노충전재를 각각 0%, 3%, 6% 포함시켜 만든 복합레진(E0, E3, E6)으로 제작한 레진 시편을, APF gel을 적용한 군과 적용하지 않은 군으로 나누어 Streptococcus mutans의 부착량과 표면 조도를 측정하고 비교, 평가하여 다음과 같은 결과를 얻었다. 1. APF gel을 적용하지 않은 레진 시편에 대한 S. mutans 부착량은 FS에서 가장 적었고, FZ, E3, E6에 대한 부착량보다는 유의하게 낮았다(p<0.05). 2. 모든 레진군에서 APF gel을 적용한 레진 시편에 대한 S. mutans 부착량은 APF gel을 적용하지 않은 시편에 대한 부착량보다 유의하게 높았다(p<0.05). 3. APF gel을 적용한 레진 시편에 대한 S. mutans 부착량은 레진군간 유의한 차이를 보였으며(p<0.05), FS, FZ, E0, E3, E6의 순서로 높은 값을 보였다. 4. APF gel 적용 전이나 후에, 레진군간 표면조도의 차이는 유의하지 않았다(p>0.05).

  • Mutans streptococci는 치아우식증과 관련이 있다고 보고되고 있으며, 이 streptococci 종 중에서 S. mutans나 S. sobrinus가 인간의 치아우식증에서 가장 일반적으로 나타난다. 치아우식증에서 이 세균종의 분포는 인종과 지역에 따라 다양하다. 본 연구에서는 2-6세인 52명의 치아우식증 환자에서 비우식부위와 우식부위로부터 치태를 채취하여 DNA를 추출한 후 dex와 gtf 유전자에 기초한 S. mutans와 S. sobrinus에 특이한 primer를 제작한 후 PCR을 시행하여 S. mutans와 S. sobrinus의 분포도를 조사하여 다음과 같은 결과를 얻었다. 1. 어린이 치아우식증 환자에서 S. mutans나 S. sobrinus가 발견된 어린이는 37명(71.2%)이었다. 2. 이들 어린이 중에서, S. mutans 또는 S. sobrinus가 비우식부위에서만 발견된 어린이는 3명(5.8%), 우식부위에서만 발견된 어린이는 22명(42.3%), 두 부위 모두에서 발견된 어린이는 12명(23.1%)이었다. 3. S. mutans나 S. sobrinus가 나타난 비우식부위 치태는 15개(28.8%), 우식부위치태는 34개(65.4%)였다. 4. 비우식부위의 경우 S. mutans만 출현한 어린이는 52명 중 8명(15.4%), S. sobrinus만 출현한 어린이는 6명(11.5%), 두 세균종이 같이 출현한 어린이는 1명(1.9%)이었다. 5. 우식부위에서 S. mutans만 출현한 어린이는 52명 중 24명(46.2%), S. sobrinus만 출현한 어린이는 2명(3.8%), 두 세균종이 같이 출현한 어린이는 8명(15.4%)이었다. 6. S. mutans는 gtf에 기초한 primer, S. sobrinus는 dex에 기초한 primer가 PCR에서 효과적으로 각각의 세균을 검출하였다. 이상의 결과로 미루어 우리나라 어린이의 경우 치아우식증은 MS와 밀접한 연관성이 있으며, MS중에서도 S. mutans가 중요하다고 판단된다. 한편 S. mutans와 S. sobrinus의 동반출현은 치아우식증과 더욱 밀접한 관련이 있는 것으로 보인다.

Pulsed Nd : YAG Laser 조사의 Streptococcus mutans에 대한 효과 (A Study on the Antibacterial Effects of Pulsed Nd: YAG Laser Irradiation to Streptococcus Mutans)

  • In order to investigate the antibacterial effects of pulsed Nd:YAG Laser concerning to Streptococcus mutans, Streptococcus mutans was cultured and standardized and plated in 96 well plates. After that, black dye and chlorhexidine were applied and pulsed Nd:YAG Laser was irradiated to plates. Experimental groups which was applied with black dye and chlorhexidine were divided into 5 groups by concentration. And experimental groups and control group were subdivided as unlased and lased of 20, 40, and 60J. After laser irradiation, All groups were plated in agar plates and incubated for 18 hours. Bacterial colonies were counted and analysed comparitively, and the temperature of culture medium was measured on every laser irradiation. The results were obtained as follows; 1. In case of laser irradiation only, there is no significant reduction in number of Streptococcus mutans by irradiated energy of laser. 2. In case of black dye application, there is no difference by concentration of black dye but there is decrease when applied Laser and black dye simultaneously. 3. When applied chlorhexidine, the number of Streptococcus mutans was decreased according to increase of concentration, but there is no difference by irradiated energy of laser. 4. The temperature of culture medium was increased according to increase of irradiated energy of Laser, and the number of Streptococcus mutans was reduced remarkably just above 50 centigrade and killed above 70 centigrade. As above results, pulsed Nd:YAG Laser has well-absorbing affinity to black dye. And antibacterial effect of pulsed Nd:YAG Laser against Streptococcus mutans is seemed to be due to increase of temperature.

S. mutans에 대한 대나무 숯의 항균력에 관한 연구 (Antibacterial effect of bamboo charcoal on Streptococcus mutans)

  • Objectives : The purpose of the present study was to investigate the effect of bamboo charcoal on Streptococcus mutans which is one of the most important causative agents of dental caries. Methods : S. mutans was incubated with or without bamboo charcoal and then changes were observed in its cell viability and antibacterial effect. Oral epithelial cells viabillity(human gingival fibroblast, HGF) was performed using MTT assay. Antibacterial effect was analyzed using a dilution plating method and agar diffusion method. Results : Oral epithelial cells, human gingival fibroblast (HGF) showed a tendency to increase in bamboo charcoal treatment solution concentrations(0.5, 1, 2, 3, 5, 10%). The bamboo charcoal had an antibacterial effect on S. mutans. Antibacterial effect of bamboo charcoal for the bacterium was 58%. Charcoal concentration of 2% and 5% in the inhibition zone showed a minimal growth, but the concentration of 10% bamboo charcoal in inhibition zone revealed a conspicuous antibacterial activity. Conclusions : Overall results suggested that the bamboo charcoal proved to be bactericidal effect on S. mutans.

Evaluation of antimicrobial effects of commercial mouthwashes utilized in South Korea

  • Streptococcus mutans is frequently associated with dental caries. Bacterial fermentation of food debris generates an acidic environment on the tooth surface, ultimately resulting in tooth deterioration. Therefore, various mouthwashes have been used to reduce and prevent Streptococcus mutans. The aim of this study was to evaluate the antimicrobial activities of 4 commercial mouthwashes and those of 10% and 20% ethanol solutions (formula A, B, C, D, E and F) against Streptococcus mutans using biofilm and planktonic methods. The range of reduction in the viable cell count of Streptococcus mutans as estimated by the biofilm and planktonic methods was 0.05-5.51 log ($P{\leq}0.01$) and 1.23-7.51 log ($P{\leq}0.001$) compared with the negative control, respectively, indicating that the planktonic method had a stronger antibacterial effect against S. mutans. Among the tested formulations, formula A (Garglin $regular^{(R)}$ mouthwash) was the most effective against Streptococcus mutans ($P{\leq}0.001$).
