  • Objective: The effects of Pleurotus ostreatus on the feed utilization of broad bean stalks (BBS), rape straw (RS), paddy straw (PS), and corn stalk (CS) was examined. Methods: The four roughages were co-cultured with Pleurotus ostreatus. The chemical composition; enzyme activities of laccase, carboxymethylcellulase (CMCase) and xylanase; carbohydrate and protein fractions (based on The Cornell Net Carbohydrate and Protein System [CNCPS]) were assessed at different days after inoculation (7, 14, 21, 28 d) and un-inoculated roughages (control, 0 d). The digestibility of nutrient components and the gas production of roughage with various incubation times were monitored at 0, 2, 4, 6, 9, 12, 24, 36, 48, 60, and 72 h using an in vitro ruminal fermentation method. Results: A higher CMCase activity (0.1039 U/mL) and earlier time to peak (14 d) were detected in Pleurotus ostreatus cultured with CS (p<0.05). Significantly, the incubation length-dependent responses of cumulative gas production were observed from 24 to 72 hours post fermentation (p<0.05), and these incubation length-dependent effects on cumulative gas production of PS and CS appeared earlier (24 h) for PS and CS than those (48 h) for BBS and RS (p<0.05). The fast-degradable carbohydrate (CA) content for all four roughages significantly increased over time (p<0.05). Nonetheless, increased degradation efficiency for CA treated with Pleurotus ostreatus was detected at both 21 and 28 days of incubation (p<0.05). With the exception of PS (p<0.05), there were no significant difference among the roughages (p>0.05) in slowly-degradable carbohydrate (CB2) at different incubation times (p<0.05). Conclusion: Assessment of the alterations in chemical composition, CNCPS system fractions, and the fermentation kinetics after biological pretreatment may yield a valuable database for evaluating the biological pretreatment of Pleurotus ostreatus in ruminant feed.

  • Various enzymes produced by the Basidiomycetes, i.e. Pleurotus ostreatus, Pholiota nameko and Flammulina velutipes have been analysed. The purpose of the analyses is to produce a nutritious fertilizer from saw dust, sweet potato stems, corn stems and barley stems which are known as the farm product waste materials. The results of the analyses are as follows: 1. In order for P. ostreatus, Pho. nameko and F. velutipes to grow properly, the saw dust of broad-leaved trees, poplar and chestnut were mixed with sweet-potato stems, corn stems and barley straws at the ratio of $42{\sim}49\;:\;30$, and then to this mixture was added rice bran at the ratio of $18{\sim}35\;:\;30$ The following proved to be optimum conditions for the growth in this test: the parcentage of water content $60{\sim}70%$, $24{\sim}26^{\circ}C$ degrees of temperature. 2. We analyzed the fiber contained in the above materials after 20 days growth in various media. We found out that the fiber was decomposed up to $47{\sim}50%$ and polysaccharified. Lignin was decomposed up to $20{\sim}25%$ and about three times as much protein was contained in the product as in the original materials. 3. Our analyses revealed the possibility of producing concentrated feed from the crude fiber.

  • Seongnyu Cave have developed along the NE-SW direction in carbonate rocks of the Joseon Supergroup. The cave mostly shows horizontal passages and contains three lakes. The main passage is about 330 m long with 540 m-long branches, thus the total length of the cave is about 870 m. Through cave diving, about 85 m-long new passages were discovered. Numerous speleothems such as soda straws, stalactites, stalagmites, columns, flowstones, rimstones, cave shields, cave corals, curtains (and bacon sheets), cave pearls, cave flowers, helictites and calcite rafts can be found in the cave. Especially, some speleothems which were believed to have grown in the past were discovered in the submerged passage, and a few stalactites, stalagmites, flowstones and columns were eroded (or corroded) by the cave stream that flowed on the floor. Because these speleothems only grow in subaerial environments within limestone caves, it appears that they grew when there was no lake in the cave and became submerged as the lake level rose in the cave. The presence of these speleothems in the lakes indicate that they only grew during glacial periods when sea-level was lower. Therefore, detailed investigation of these speleothems will provide invaluable information on paleoclimatic evolution around the Korean peninsula in the future.

  • Cryopreservation of oocytes by vitrification technique may contribute a lot in the field of reproductive biotechnology. The objectives of the present study were to evaluate the effectiveness of two cryo-devices for vitrification of immature oocytes of indigenous zebu cows. Slaughter house derived immature cumulus-oocyte-complexes (COCs) of cows were vitrified using 15% dimethyl sulphoxide (DMSO) as cryoprotective agent (CPA) with 0.5 mol sucrose in TCM 199 supplemented with 20% FBS. Vitrification of COCs was completed after immediate plunging of COCs loaded cryotop or French mini straw into the liquid nitrogen ($LN_2$). Then the COCs containing cryotop or French mini straws were warmed in 0.25 mol sucrose and 20% FBS supplemented TCM 199 followed by in vitro culture in $50{\mu}l$ droplets of bicarbonate buffered TCM 199 supplemented with 10% FBS, pyruvate, FSH and oestradiol for 24 hrs at $39^{\circ}C$ with 5% CO2 in humidified air. After maturation culture, oocytes were denuded and examined under inverted microscope for presence of polar body as the indication of maturation. Denuded oocytes were also stained by whole mount technique using 1% orcein to examine the maturation by presence of MII chromosomes. The in vitro maturation rate was significantly (p<0.05) higher in oocytes vitrified and warmed using crytop ($47.1{\pm}6.9%$) than that of French mini straw ($15.9{\pm}12.5%$). Moreover, in vitro maturation rate was significantly (p<0.05) highe r in control oocytes (not vitrified) ($84.5{\pm}14.2%$) than that of vitrified oocytes. In conclusion, cryotop is better than French mini straw as cryo-device for vitrification of bovine immature oocytes.

  • As a series on the soy-bean protein and their related substances 9 samples were collected from 9 places such as straws (Rice) to obtain bacterial strains which produce protease. From these samples total of 23 strains were isolated by the use of dilution pour plate method. For all isolated strains primary screening of productivity of protease was performed and useful straines with regard to protease productivities were identified. Optimum conditions for enzyme action of protease from isolates $D_9$, $F_{20}$ strains were pH 7.5 and $40^{\circ}C$. Chung-Kook-Jang is one of the characteristic foods in Korea made from soy-bean by fermentation. The chief bacterium is Bacillus subtilis and the chief change which takes place in soy-bean during fermentation is degradation of protein. Three kinds of Chung-Kook-Jang were prepared using three different strains of Bacillus natto, $D_9\;and\;F_{20}$ from isolated. Water soluble-N, TCA soluble-N, amino-N and peptide-N were measured about the steamed soybean, Chung-Kook-Jang prepared with three strains of bacteria. Water soluble-N decreased very largely in steamed soybean, but in Chung-Kook-Jang it increased to 85% of raw soy-bean.

  • This research was investigated the relationship, in high-producing Holstein donor cows, between the number of the transferable embryos and the blood serum concentrations of Blood Urea Nitrogen (BUN), glucose and cholesterol, which affect the nutritional state of cows. CIDRs were inserted into the vaginas of twenty two heads of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_{2{\alpha}}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. The total numbers of ova collected from 3 experimental groups whose blood BUN concentrations were <10 mg/dl, 11~18 mg/dl and ${\geq}19$ mg/dl were 8.9, 12.5 and 19.0, respectively; whereas the numbers of transferable embryos were 5.8 + 1.9, 7.9 + 2.8 and 5.2 + 1.4, respectively. When glucose concentration was <60 mg/dl, the total number of collected ova was 9.9, which was smaller than when the concentration was 60~70 mg/dl or ${\geq}70$ mg/dl. When glucose concentration was 60~70 mg/dl, the number of transferable embryos was 7.1 + 2.4, which was slightly larger than the numbers 6.4 + 2.1 and 6.1 + 1.7 that were obtained when the concentrations were <60 mg/dl and ${\geq}70$ mg/dl, respectively ; however, the differences were not significant (p>0.05). When cholesterol concentrations were <150 mg/dl, 150~200 mg/dl and ${\geq}200$ mg/dl, the total numbers of collected ova were 11.2, 11.3 and 8.6, respectively. Whereas the numbers of transferable embryos were 7.1 + 2.1, 7.3 + 1.9 and 5.6 + 1.3, respectively ; however, the differences were again not significant (p>0.05). The result of this research showed no significant difference in ovum recovery rate and the number of transferable embryos according to major metabolite concentrations in high-producing Holstein donor cows. However, it is considered that the failure of maintaining proper nutritional status would cause the fall in in vivo embryo productivity.

  • This research was investigated the relationship between the number of the transferable embryos and estrus expression rate, BCS (Body Condition Score), which affect the nutritional state of the cow, in Holstein donor cows. CIDRs were inserted into the vaginas of twenty two head of Holstein cows, regardless of estrous cycle. When BCS was $$\leq_-$$2.5, the total number of collected ova was 7.3 + 1.9, which is significantly lower (p<0.05) than the numbers 15.4 + 2.8 and 15.4 + 2.1 that were obtained when BCSs were 2.75 and $$\geq_-$$3.0, respectively. Whereas the numbers of transferable embryos were 5.2 + 1.4 when BCS was $$\leq_-$$2.5, which was smaller than the numbers 6.0 + 2.1 and 8.5 + 1.8 that were obtained when BCSs were 2.75 and $$\geq_-$$3.0, respectively; however, the differences were not significant. As for estrus induction rate, the cow groups whose BCSs were 2.75 and $$\geq_-$$3.0 showed 100.0% and 95.0%, respectively. Whereas the cow group whose BCS was $$\leq_-$$2.5 showed 57.1%, and the differences were significant (p< 0.05). As for estrous expression rate, the cow groups whose BCSs were $$\leq_-$$2.5, 2.75 and $$\geq_-$$3.0 showed 100.0%, 100.0% and 85.7%, respectively; however, the differences were not significant. According to the result of this research, it is considered that the total number of collected ova and the number of transferable embryos will be affected by the nutritional state before and after in vivo embryo production and superovulation treatment, and that although the mechanism is not clear, poor stockbreeding management and nutritional level would cause the decrease of ovum recovery rate and the number of transferable embryos in high-producing cows. On the other hand, diverse researches on the superovulation treatment method that is suitable for high-producing Holstein donor cows would contribute to preventing ovarian cyclicity disorder, as well as to the early multiplication of cows with superior genes by increasing the utilization value of donor cows.

  • Fire protection countermeasure were considered on the straw thatched roof and wooden structure. For the fire resistant treatment, rice straw was soaked in the fire resistant liquid with different soaking time. After treatment, some rice straw sample was washed with water then the rice straws were tested to check the fire resistance performance. And the wood was soaked in the fire resistant liquid at an atmospheric pressure, vacuum-pressure condition and painted with brush on the surface. To analyse the fire resistant performance of rice straw, ignition delay time was measured under the radiant heat flux of cone heater. And the fire resistant performance of wood samples were tested with 45 degree fire resistant test apparatus and cone heater. Based on the cone heater test, the rice straw which most easily ignitable material shows the longer ignition delay time than not treated ones and even in the water washed straw show a longer ignition delay time than not treated ones. And fire resistant treated woods of vacuum-pressure treated sample showed the most excellent performance on fire resistance. And the fire resistant treated by brush and soaking showed a longer ignition delay time than not treated ones. From this experiment, it was found that a fire resistant treatment of rice straw and wood of the house can be protected from the fire spread.

  • In order to investigate the soil-to-plant transfer factor (TF) of $^{99}Tc$ for Korean major upland crops (soybean, radish and Chinese cabbage), pot experiments were performed in a greenhouse. Soils were collected from four upland fields (two for soybean and two for radish and Chinese cabbage) around Gyeongju radioactive-waste disposal site. Three to four weeks before sowing, dried soils were mixed with a $^{99}Tc$ solution and the mixtures were put into pots and irrigated. TF values were expressed as the ratios of the $^{99}Tc$ concentrations in plants (Bq $kg^{-1}$-dry or fresh) to those in soils (Bq $kg^{-1}$-dry). There was no great difference in the TF value between soils. The TF values for soybean seeds were extremely lower than those for the straws, indicating a very low mobility of $^{99}Tc$ to seeds. As representative TF values of $^{99}Tc$, $1.8{\times}10^{-1}$, $1.2{\times}10^1$, $3.2{\times}10^2$ and $1.3{\times}10^2$ (for dry plants), arithmetic means for two soils, were proposed for soybean seeds, radish roots, radish leaves and Chinese cabbage leaves, respectively. In the case of the vegetables, proposals for fresh plants were also made. The proposed values are not sufficiently representative so successive updates are needed.
