Effects of Cold Pretreatment and Medium Composition on Anther Culture Initiation in Strawberry

  • Callus culture initiation of strawberry (Fragaria${\times}$ananassa Duch.) was investigated at different Murashige and Skoog (MS) medium strengths, types and concentrations of plant growth regulators, and incorporating a cold pretreatment period to determine the optimal nutritional and environmental conditions. No high quality callus was induced on MS media without auxin regardless of medium strength. When 6-benzylaminopurine (BA) was combined with indole acetic acid (IAA), naphthalene acetic acid (NAA), and 2,4-dichlorophenoxyacetic acid (2,4-D), high quality callus were highly induced compared to medium supplemented with auxin alone. When $0.5mg{\cdot}L^{-1}$ BA was combined with IAA, NAA, and 2,4-D, high quality callus induction was more effective than the medium supplemented with the other BA concentrations. The best combination of auxin and cytokinin for high quality callus induction was $1.0mg{\cdot}L^{-1}$ NAA and $0.5mg{\cdot}L^{-1}$ BA. Although the differences in callus induction were not significant, high quality callus induction at half strength MS medium was more effective than at full strength medium. When $30g{\cdot}L^{-1}$ sucrose was added to the half strength MS medium, the rate of high quality callus induction increased. The optimum cold pretreatment temperature and period for high quality callus induction were $4^{\circ}C$ and 72 h, respectively. Regeneration rate of high quality callus increased in MS medium supplemented with thidiazuron.

Sugar content and expression of sugar metabolism-related gene in strawberry fruits from various cultivars

  • Strawberry (Fragaria ${\times}$ ananassa) is a globally-cultivated and popular fruit crop, prized for its flavor and nutritional value. Sweetness, a key determinant of fruit quality, depends on the sugar composition and concentration. We selected eight strawberry cultivars based on the fruit soluble solids content to represent high and low sugar content groups. The average soluble solid content was $13.6^{\circ}Brix$ (Okmae, Geumsil, Aram, and Maehyang) and $2.9^{\circ}Brix$ (Missionary, Camino Real, Portola, and Gilgyung53), for the high and low sugar content groups, respectively. Sucrose was the main sugar in the cultivars with high sugar content, whereas fructose was the main component in the low sugar content cultivars. Fruit starch concentration ranged from $3.247{\pm}0.056$ to $3.850{\pm}0.055g/100g$, with a 12% higher concentration in the high sugar content cultivars. Additionally, we identified 41 sugar metabolism-related genes in Fragaria ${\times}$ ananassa and analyzed the relationship between their transcripts and the sugar accumulation in fruit. FaGPT1, FaTMT1, FaHXK1, FaPHS1, FaINVA-3, and FacxINV2-1 were highly expressed in the high sugar content cultivars, while FapGlcT, FaTMT2-1, FaPHS2-1, FaSUSY1-1, and FaSUSY1-2 were highly expressed in the low sugar content cultivars. In general, a greater number of genes encoding sugar transporters or involved in sugar synthesis were highly expressed in the high sugar content cultivars. Contrarily, genes involved in sugar degradation were preferentially transcribed in the low sugar content cultivars. Although gene expression was not perfectly proportional to sugar content or concentration, our analysis of the genes involved in sugar metabolism and accumulation in strawberries provides a framework for further studies and for the subsequent engineering of sugar metabolism to enhance fruit quality.

pH 조절제를 이용한 농축 딸기 퓌레 열처리 시 품질저하 억제 (Prevention of Quality Deterioration in Concentrated Strawberry Puree by Employing pH Regulators during Heat Treatment)

  • 본 연구는 농축 딸기 퓌레를 제조 시 구연산(CA) 또는 산성 메타인산나트륨(ASM)을 산미료로 이용하여 열처리 과정에서 농축 딸기 퓌레의 폴리페놀과 안토시아닌과 같은 기능성 성분 농도와 항산화 활성의 저하를 최소화시키는 데 있다. 열처리한 농축 딸기 퓌레의 색가는 1% CA+1% ASM 처리군이 13.027로 가장 높았고, 1% CA 처리군이 9.539, 대조군이 6.905의 순으로 나타났다. 총안토시아닌 함량에서도 1% CA+1% ASM 처리군이 3.049 mg/100 g, 1% CA 처리군, 대조군 순으로 각각 1.140 mg/100 g, 0.757 mg/100 g으로 나타났다. 항산화 활성으로써 DPPH 라디칼 소거능의 경우 대조군은 58.148%, 1% CA 처리군은 72.638%, 1% CA+1% ASM 처리군은 83.763%로 나타났다. 모든 분석치에서 대조군과 1% CA+1% ASM 처리군 간에는 5% 내에서 유의성 차이가 있었다. 기호도 검사 결과 전체적인 기호도에서는 1% CA 처리군이 가장 높았고 1% CA+1% ASM 처리군, 대조군 순이었으나 세 처리군 간에 5% 내에서 유의적 차이를 보이지 않았다. 본 결과를 통해 ASM 처리가 열처리 과정에서 기능성 성분과 생리활성의 저하를 억제해 주는 역할을 하는 것으로 나타났으며 새로운 산미료로서 활용할 수 있을 것으로 기대한다.

Effects of LED Light Quality of Urban Agricultural Plant Factories on the Growth of Daughter Plants of 'Seolhyang' Strawberry

  • This study was conducted to examine the influence of Light-Emitting Diode (LED) light quality in urban agricultural plant factories on the growth and development of Seolhyang strawberry daughter plants in order to improve the efficiency of daughter plant growth and urban agriculture. LED light quality by demonstrated that above-ground growth and development were greatest for daughter plant 2. Daughter plant 1 showed the next highest growth and development, followed by daughter plant 3. Among the different qualities of LED light, the stem was thickest and growth rate of leaves was highest for R + B III (LED quality: red 660 nm + blue 450 nm/photosynthetic photon flux density (PPFD): $241-243{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and lowest for R (red $660nm/115-117{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). Plant height, leaf width, petiole length, and the leaf growth rate were highest for W (white fluorescent lamp/$241-243{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and lowest for R + B I (red 660nm+blue 450nm/$80-82{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$). For above-ground growth and development, as the plants surpassed the seedling age, mixed light (red + blue), rather than monochromatic light (red or blue), and higher PPFD values tended to increase development. Regarding the quality of the LED light, daughter plant 2 showed the highest chlorophyll content, followed by daughter plant 1, and daughter plant 3 showed the least chlorophyll content. When the wavelength was monochromatic, chlorophyll content increased, compared to that when PPFD values were increased. Mixed light vitality was highest in daughter plant 2, followed by 1, and 3, showed increased photosynthesis when PPFD values were high with mixed light, in contrast to the results observed for chlorophyll content.

과일류의 염소 소독 방법에 따른 이화학적 및 관능적 품질 특성 변화 (Changes in Physicochemical and Sensory Properties of Fruits as Affected by Chlorine Sterilization)

  • The purpose of this study was to investigate the changes in physicochemical and sensory properties of raw fruits during washing and chlorine treatments. Strawberry and banana were pre-prepared at different concentration of chlorinated water(0 ppm, 50 ppm and 100 ppm), immersion time(3 min and 5 min), and number of post-rinsing(1 time, 2 times and 3 times). The physicochemical properties such as pH, sugar contents, residual chlorine contents, color values and hardness of the fruits were analyzed, and the sensory quality were evaluated throughout the sterilization treatment process. After washing strawberry with 100 ppm chlorinated water and 3 times of post-rinsing, pH and residual chlorine contents were showed a little difference, while sugar contents, hardness, and color values(L, a and b) were reduced. In case of banana, pH, sugar contents and residual chlorine contents were not affected, and hardness and L color value were reduced. However, a and b color values of banana were gradually increased as the development of brown discoloration. Sensory properties of the samples were affected by the chlorine sterilization treatment. In overall acceptance, strawberry and banana treated with 100 ppm chlorinated water showed the lowest scores among treatments. Therefore it could be suggested that the application of 50 ppm chlorinated water for $3{\sim}5$ minutes with over 3 times of post-rinsing was the effective pre-preparation method without affecting the quality of the fruits.

수확 후 고농도 CO2 단기처리가 '매향' 딸기의 저장에 미치는 영향 (Effects of short-term treatment of high pressure CO2 on the changes in fruit quality during the storage of 'Maehyang' strawberries)

  • This experiment was conducted to examine the effects of a short-term treatment of high pressure $CO_2$ on shelf-life of strawberry fruit. A short-term treatment (12 hr) of 100% $CO_2$ resulted in the increase of fruit firmness up to 71.9% compared to that at harvest. The firmness of $CO_2$ treated fruit remained a significantly higher than that of control (air) up to 15 days. The alteration of pectic polymers was observed by $CO_2$ treatment such as an increase of EDTA soluble pectins and decrease of water soluble ones. The $CO_2$ treatment resulted in the increase of total amount of wall bound calcium. Pectate lyase activity, an important agent of strawberry fruit softening, was also significantly reduced by $CO_2$ treatment. Contents of soluble solids and acids of $CO_2$ treated fruit were higher than those of control fruit. Short-term treatment of high pressure $CO_2$ affected shelf-life through firmness increase whereas the visual quality and decay incidence of strawberry fruit were not affected.

수출딸기 선별장에서 잿빛곰팡이병원균 밀도조사와 저장온도에 따른 잿빛곰팡이병 발생 연구 (Studies on Botrytis cinerea Density in Packing Shed and Gray Mold Incidence Following Storage-Temperature in Exported Strawberry)

  • 국내 딸기는 현재 홍콩, 싱가포르 및 동남아시아 지역으로 수출량이 증대되고 있는 가운데 유통과정에서 환경조건에 따라 품질에 손상이 크게 발생하고 있다. 주로 수출딸기에 품질손상을 일으키는 잿빛곰팡이병은 다른 국가로 이동하는 동안 혹은 수입국에서 소비자에게 판매되기까지의 기간 동안 발생하고 있다. 본 연구는 수출딸기 선별장 공기중 부유중인 잿빛곰팡이병원균의 밀도와 저장중 발병률에 관한 상관관계를 규명하기 위하여 수행되었다. 공기중 병원균의 밀도가 증가함에 따라 저온 저장중 병 발생율이 최대 16%로 나타났다. 상온저장의 경우 상대적으로 공기중 병원균의 밀도가 낮게 조사된 12월 말부터 발병율 100%가 관찰 되었다. 본 연구결과, 선별장의 잿빛곰팡이 병원균 밀도와 저장중 병발생의 상관관계가 구명되었다.

칼슘처리와 MA포장이 딸기의 품질에 미치는 영향 (Effects on Quality of Strawberry Fruit by Dipping of Calcium Solution and MA Packaging)

  • 딸기를 $20^{\circ}C$$45^{\circ}C$$1\%\;CaCl_2$ 용액에 각각 15분간 침지시킨 후, PE film (0.011mm)으로 포장한 군과 포장하지 않은 군으로 나누어 $5^{\circ}C$에 저장하였다. 그리고 $CaCl_2$, 용액에 침지시키지 않고 PE film(0.011mm)으로 포장한 군과 포장하지 않은 군으로 나누어 $5^{\circ}C$에 저장하였다. $45^{\circ}C\;CaCl_2$ 용액에 침지시킨 후 포장한 처리구에서 부패율은 낮고 경도가 높게 나타났다. 또한 비타민 C함량이 높았고 외관이 가장 좋은 것으로 나타났으며 다른 처리군에 비해 pH가 낮고 산도가 높게 나타났다. 그러나 당도는 처리군 간에 유의성이 없었다. 따라서 수확 후 칼슘 처리와 포장의 병행은 딸기의 저장에 효과적인 것으로 사료된다.

수확 후 CO2 처리 시기 및 농도에 따른 '설향' 딸기 저장 중 품질변화 (Effect of different days of postharvest treatment and CO2 concentrations on the quality of 'Seolhyang' strawberry during storage)

  • '설향' 딸기 수확 후 $CO_2$ 처리에 의한 선도유지 효과 및 적정 처리조건을 구명하고자 연구를 수행하였다. 착색도 70% 수준의 '설향' 딸기를 수확 후 1, 3일 후에 $CO_2$ 농도 0, 5, 15 및 30%에서 3시간 처리한 다음 $4^{\circ}C$에서 각각 12일, 10일 저장(수확 후 13일) 하였다. 수확 후 1일에 $CO_2$ 처리한 '설향' 딸기는 15% 및 30% $CO_2$ 처리에서 과실의 경도를 증가시켜, 저장 10일까지 저장 전 보다 경도가 높게 유지되었고, $CO_2$ 농도가 높아질수록 물러짐 발생이 적게 나타났으며, 부패과율도 낮게 나타났다. 특히 30% $CO_2$ 처리한 딸기의 색은 적색도(Hunter a값)가 가장 낮았고, 외관에 대한 관능점수는 가장 높게 나타났다. 그러나 수확 후 3일에 $CO_2$ 처리한 딸기는 $CO_2$ 농도에 관계없이 딸기 저장 중 품질에 영향을 미치지 않았고, 물러짐 발생이 수확 후 1일 $CO_2$ 처리보다 높았으며, 이취 및 부패과율이 높아 품질이 나쁜 것으로 나타났다. 따라서 '설향' 딸기를 수확 후 1일 이내에 15% 및 30% $CO_2$ 처리하면 선도유지에 효과적인 것으로 나타났으며, 특히 30% $CO_2$ 농도 3시간 처리는 수확 후 10일 까지 '설향' 딸기의 품질을 유지할 수 있었다.
