Interaction of Pseudostellaria heterophylla with Quorum Sensing and Quorum Quenching Bacteria Mediated by Root Exudates in a Consecutive Monoculture System

  • Many plant-pathogenic bacteria are dependent on quorum sensing (QS) to evoke disease. In this study, the population of QS and quorum quenching (QQ) bacteria was analyzed in a consecutive monoculture system of Pseudostellaria heterophylla. The isolated QS strains were identified as Serratia marcescens with SwrIR-type QS system and exhibited a significant increase over the years of monoculture. Only one QQ strain was isolated from newly planted soil sample and was identified as Bacillus thuringiensis, which secreted lactonase to degrade QS signal molecules. Inoculation of S. marcescens to P. heterophylla root could rapidly cause wilt disease, which was alleviated by B. thuringiensis. Furthermore, the expression of lactonase encoded by the aiiA gene in S. marcescens resulted in reduction of its pathogenicity, implying that the toxic effect of S. marcescens on the seedlings was QS-regulated. Meanwhile, excess lactonase in S. marcescens led to reduction in antibacterial substances, exoenzymes, and swarming motility, which might contribute to pathogensis on the seedlings. Root exudates and root tuber extracts of P. heterophylla significantly promoted the growth of S. marcescens, whereas a slight increase of B. thuringiensis was observed in both samples. These results demonstrated that QS-regulated behaviors in S. marcescens mediated by root exudates played an important role in replanting diseases of P. heterophylla.

해양 생물에서 분리된 Phaeobacter inhibens KJ-2의 항균 활성 (Examination of Antimicrobial Activity by Phaeobacter inhibens KJ-2 Isolated from a Marine Organism)

  • 본 연구는 양식업에 막대한 경제적인 손실을 야기시키는 세균성 어류질병인 Vibrio anguillarum을 예방하고 억제하기 위한 목적을 두고, 제주 연안에 서식하는 군소 알로부터 특징적인 균주를 분리 하였다. 이를 16S rRNA 염기서열 분석, 형태학적 특성을 통해 균을 동정하여 Phaeobacter inhibens KJ-2로 명명하였다. Phaeobacter inhibens KJ-2 균주는 Vibrio sp.에 대해 강한 항균 활성을 갖고 있으며, 항균물질의 생산특성을 배양조건에 따라 검토하였으며, 그 결과 진탕 배양의 경우 $20^{\circ}C$에서 24시간 배양 후, 정치 배양의 경우 96시간 배양 후 항균 활성이 가장 높게 나타났다. 진탕 배양 및 정치 배양 모두 4시간 후부터 생육도는 낮았으나 항균 물질 생산을 시작하였다. 생육도가 거의 없는데도 불구하고 항균물질이 생산이 이루어졌는데, 이는 QS (Quorum Sensing)의 신호 물질인 AHLs과 관련이 있다고 사료되며, AHLs의 생성능은 항균 활성과 거의 일치함을 알 수 있었다. 이런 배양특성을 바탕으로 항균 활성을 검토한 결과 V. anguillarum외 다른 Vibrio sp.에 대해서도 항균활성을 나타내었다. 특히 Vibrio vulnificus, Vibrio campbellii, Vibrio mimicus에 대해 높은 저해능을 나타내었다.

  • 사회기반 시설물의 노후화에 대응해 이상 징후를 파악하고 유지보수를 위한 최적의 의사결정을 내리기 위해선 디지털 기반 SOC 시설물 유지관리 시스템의 개발이 필수적인데, 디지털 SOC 시스템은 장기간 구조물 계측을 위한 IoT 센서 시스템과 축적 데이터 처리를 위한 클라우드 컴퓨팅 기술을 요구한다. 본 연구에서는 구조물의 다물리량을 장기간 측정할 수 있는 IoT센서와 클라우드 컴퓨팅을 위한 서버 시스템을 개발하였다. 개발 IoT센서는 총 3축 가속도 및 3채널의 변형률 측정이 가능하고 24비트의 높은 해상도로 정밀한 데이터 수집을 수행한다. 또한 저전력 LTE-CAT M1 통신을 통해 데이터를 실시간으로 서버에 전송하여 별도의 중계기가 필요 없는 장점이 있다. 개발된 클라우드 서버는 센서로부터 다물리량 데이터를 수신하고 가속도, 변형률 기반 변위 융합 알고리즘을 내장하여 센서에서의 연산 없이 고성능 연산을 수행한다. 제안 방법의 검증은 2개소의 실제 교량에서 변위계와의 계측 결과 비교, 장기간 운영 테스트를 통해 이뤄졌다.

고온용 압저항센서용 크롬산화박막의 특성 (Characteristics of chromium oxide thin-films for high temperature piezoresistive sensors)

  • This paper present characteristics of chromium oxide thin-film as piezoresistive sensors, which were deposited on Si substrates by DC reactive magnetron sputtering in an argon-Oxide atmosphere for high temperature applications. The chemical composition, physical and electrical properties and thermal stability ranges of the $CrO_{x}$ sensing elements have studied. $CrO_{x}$ thin films with a linear gauge factor(GF${\fallingdotseq}$15), high electrical resistivity (${\rho}$ = $340{\mu}{\Omega}cm$) and TCR<-55 ppm/$^{\circ}C$ have been obtained. These $CrO_{x}$ thin films may allow high temperature pressure sensor miniaturization to be achieved.

대형선박의 액체 탱크용 수위 모니터링 센서 시스템 연구 (A Study on the Tank Liquid-Level Monitoring Sensor Systems for Large Scaled Vessels)

  • A fiber-optic liquid level sensor based on bending cantilever beam has been proposed. A fiber Bragg grating(FBG) embedded in the cantilever beam is used to sensing elements. The basic concept is elongation and constriction of the FBG corresponding to the liquid level variation. The best FBG position on the cantilever for obtaining the high sensitivity was 4 cm from the fixing point. When the liquid level moves up and down vertically, the Bragg wavelength is linearly shifted. But, the wavelength sensitivity of the FBG installed on the upper side of cantilever was four times better than that of the FBG equipped in the lateral side due to the difference of unit strain applied to the FBG. Intensity demodulation using the low-cost edge filter is used to interrogate the Bragg wavelength through converting the wavelength signals into the optical intensity ones. Experiment results show that the electrical output is exponentially proportional to the liquid level. But, it should be overcome for applying to the ships.

Composite Fracture Detection Capabilities of FBG Sensor and AE Sensor

  • Non-destructive testing methods of composite materials are very important for improving material reliability and safety. AE measurement is based on the detection of microscopic surface movements from stress waves in a material during the fracture process. The examination of AE is a useful tool for the sensitive detection and location of active damage in polymer and composite materials. FBG (Fiber Bragg Grating) sensors have attracted much interest owing to the important advantages of optical fiber sensing. Compared to conventional electronic sensors, fiber-optical sensors are known for their high resolution and high accuracy. Furthermore, they offer important advantages such as immunity to electromagnetic interference, and electrically passive operation. In this paper, the crack detection capability of AE (Acoustic Emission) measurement was compared with that of an FBG sensor under tensile testing and buckling test of composite materials. The AE signals of the PVDF sensor were measured and an AE signal analyzer, which had a low pass filter and a resonance filter, was designed and fabricated. Also, the wavelength variation of the FBG sensor was measured and its strain was calculated. Calculated strains were compared with those determined by finite element analysis.

재활로봇용 3축 힘/토크센서 설계 (Design of Three-Axis Force/Torque Sensor for Rehabilitation Robot)

  • In this study, we described the design of a three-axis force/torque sensor for measuring the force and torque in a lower-limb rehabilitation robot. The three-axis force/torque sensor is composed of Fx force sensor, Fz force sensor and Tz torque sensor. The sensing element for Fx force sensor and Tz torque sensor is used in a two-step parallel plate beam, and that of Fz force sensor is used in a parallel plate beam. The rated loads of Fx force sensor, Tz torque sensor and Fz force sensor are 300 N, 15 N m and 100 N, respectively. The three-axis force/torque sensor was designed using the finite element method, and manufactured using strain-gauges. The three-axis force sensor was further characterized. As a result, the interference error of the three-axis force/torque sensor was < 1.24%, the repeatability error of each sensor was < 0.03%, and the non-linearity was < 0.02%.

수직 반작용력 측정 장치 개발(II) (A Development of Device for Measurement of Vertical Ground Reaction Force(II))

  • The purpose of this study was to develop the uniaxial force plate system which is measured by the vertical force. The VGRF(vertical ground reaction force) 1.0 was composed of 2 bath digital scales, 2 indicaters, and analyzing software. This system was newly renovated to VGRF 2,0 which are 2 industrial digital scales, 2 adjustable indicators, and enforced analyzing software. Changes of the new system were as follows. First, the height of the plate was 75% lower than before. Second, sensing ability of the load cell was changed from 90 - 0.05kg to 300 - 0.1kg. Third, the speed of data processing was changed from 17 per second to 60 per second. Fourth, analyzing software was enforced to develop and calculate the data. For the test of the system, two different types(bare foot, high-heeled shoes) gait was adopted. highly skilled female walker(23yrs, height 165cm, body mass 46.8kg) participated for the experimental study. During the dynamic performance(gait analysis), the data of each load cell were very similar to the previous studies. Specifically, bare foot walking had less vertical force than high-heeled shoes. Consequently, VGRF 2.0 can sense the general dynamic movements as well as static load conditions.

복합재료내의 계면 접착 특성에 따른 지능형 구조물의 진동제어에 관한 연구 (Studies on the Vibration Controllability of Smart Structure Depending on the Interfacial Adhesion Properties of Composite Materials)

  • The success of controllability of smart structures depends on the quality of the bonding along the interface between the main structure and the attached sensing and acuating elements. Generally, the analysis procedures neglect the effect of the interfacial bond layer or assume that this bond layer behaves like viscoelastic material. Three different bond layers. two modified epoxy adhesives, and one isocyanate adhesive were prepared for their toughness and moduli. Bond layer of the chosen adhesive provides an almost perfect bonding condition between the composite structure and the PZT while bended significantly like arrow-shape. The perfect bonding condition is tested by considering various material properties of the bond layers. and based on this perfect bonding condition, the effects of the interfacial bond layer on the dynamic behavior and controllability of the test structure is experimentally studied. Once the perfect bonding condition is achieved. dynamic effects of the bond layer itself on the dynamic characteristics of the main structure is negligible. but the contribution of the attached PZT elements on the stiffness of the multi-layered structure becomes significant when the thickness of the bond layer increased.

근전의수의 제어시스템에 관한 연구 (A Study on the Control System of Myoelectric Hand Prosthesis)

  • This paper presents a myoelectric hand prosthesis(MHP) with two degree of freedom(2-DOF), which consists of a mechanical hand, a surface myoelectric sensor(SMES) for measuring myoelectric signal, a control system and a charging battery. The actuation for the 2-DOF hand functions such as grasping and wrist rotation was performed by two DC-motors, and controlled by myoelectric signal measured from the residual forearm muscle. The grip force of the MHP was automatically changed by a mechanical automatic speed reducer mounted on the hand. The skin interface of SMES was composed of the electrodes using the SUS440 metal in order to endure a wet condition due to the sweat. The sensor was embedded with a amplifier and a filter circuit for rejecting the offset voltage caused by power line noises. The control system was composed of the grip force sensor, the slip sensor, and the two controllers. The two controllers were made of a RISC-type microprocessor, and its software was executed on a real-time kernel. The control system used Force Sensing Resistors, FSR, as slip pick-ups at the fingertip of a thumb and the grip force information was obtained from a strain-gauge on the lever of the MHP. The experimental results were showed that the proposed control system is feasible for the MHP.
