Biosurfactant as a microbial pesticide

  • Soil-borne infectious disease including Pythium aphanidermatum and Rhizoctonia solani causes severe damage to plants, such as cucumber. This soil-borne infectious disease was not controlled effectively by chemical pesticide. Since these diseases spread through the soil, chemical agents are usually ineffective. Instead, biological control, including antagonistic microbe can be used as a preferred control method. An efficient method was developed to select an antagonistic strain to be used as a biological control agent strain. In this new method, surface tension reduction potential of an isolate was included in the ‘decision factor’ in addition to the other factors, such as growth rate, and pathogen inhibition rate. Considering these 3 decision factors by a statistical method, an isolate from soil was selected and was identified as Bacillus sp. GB16. In the pot test, this strain showed the best performance among the isolated strains. The lowest disease incidence rate and fastest seed growth was observed when Bacillus sp. GB16 was used. Therefore this strain was considered as plant growth promoting rhizobacteria (PGPR). The action of surface tension reducing component was deduced as the enhancement of wetting, spreading, and residing of antagonistic strain in the rhizosphere. This result showed that new selection method was significantly effective in selecting the best antagonistic strain for biological control of soil-borne infectious plant pathogen. The antifungal substances against P. aphanidermatum and R. solani were partially purified from the culture filtrates of Bacillus sp. GB16. In this study, lipopeptide possessing antifungal activity was isolated from Bacillus sp. GB16 cultures by various purification procedures and was identified as a surfactin-like lipopeptide based on the Fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance (NMR), high performance liquid chromatography mass spectroscopy (HPLC-MS), and quadrupole time-of-flight (Q-TOF) ESI-MS/MS data. The lipopeptide, named GB16-BS, completely inhibited the growth of Pythium aphanidermatum, Rhizoctonia solani, Penicillium sp., and Botrytis cineria at concentrations of 10 and 50 mg/L, respectively. A novel method to prevent the foaming and to provide oxygen was developed. During the production of surface active agent, such as lipopeptide (surfactin), large amount of foam was produced by aeration. This resulted in the carryover of cells to the outside of the fermentor, which leads to the significant loss of cells. Instead of using cell-toxic antifoaming agents, low amount of hydrogen peroxide was added. Catalase produced by cells converted hydrogen peroxide into oxygen and water. Also addition of corn oil as an oxygen vector as well as antifoaming agent was attempted. In addition, Ca-stearate, a metal soap, was added to enhance the antifoam activity of com oil. These methods could prevent the foaming significantly and maintained high dissolved oxygen in spite of lower aeration and agitation. Using these methods, high cell density, could be achieved with increased lipopeptide productivity. In conclusion to produce an effective biological control agent for soil-borne infectious disease, following strategies were attempted i) effective screening of antagonist by including surface tension as an important decision factor ii) identification of antifungal compound produced from the isolated strain iii) novel oxygenation by $H_2O_2-catalase$ with vegetable oil for antifungal lipopeptide production.

In vitro and In vivo Activities of a Biocontrol Agent, Serratia plymuthica A2l-4, Against Phytophthora capsici

  • In vitro and in vivo activities of a biocontrol agent, Serratia plymuthica strain A2l-4, was evaluated for the control of Phytophthora blight of pepper, Strain A2l-4 inhibited mycelial growth, germination of zoosporangia and cystospores, and formation of zoospore and zoosporangia of Phytophthora capsici in vitro. In the pot experiment, incidence of Phytophthora blight of pepper in non-treated control was 100% at 14 days after inoculation, while no disease was observed in the plot treated with S. plymuthica A2l-4. In the greenhouse test, infection rate of pepper in the non-treated plots was 74.5%, while it was only 12.6% in the plots treated with A2l-4. Results indicate that S. plymuthica A2l-4 is a potential biocontrol agent for Phytophthora blight of pepper.

  • For successful microalgal biodiesel production, the strain should be selected carefully. Fast growth rate and high fatty acid contents are desired traits for algal biodiesel production. In ocean cultivation of microalgae, seawater temperature slowly changes over seasons, and rotating algal strains in accordance with their optimal temperature could improve overall productivity. Additionally, use of indigenous strain is preferred to alleviate potential impacts on the environment. In this study, five strains of Tetraselmis sp. from nearshore of Youngheung Island, Incheon, Korea, were isolated during winter and characterized for their growth patterns and fatty acid compositions in the low temperatures ($5-15^{\circ}C$). The five strains showed various characteristics in optimal growth temperature, fatty acid contents, and compositions. Compared with a strain of Tetraselmis sp., isolated from Ganghwa island in a previous study, a rapid-growing strain with 237% higher biomass productivity and an oleaginous strain with twice higher fatty acid contents at $10^{\circ}C$ were isolated. The oleaginous Tetraselmis strain showed the highest fatty acid productivity among the strains, having 438% higher productivity than the previous strain. Using the new isolates in the seasons with low seawater temperature would improve microalgal fatty acid productivity in ocean cultivation.

마그네슘합금의 초소성 특성과 응용 (Superplasticity of Magnesium Alloys and SPF Applications)

  • Magnesium alloys are of emerging interest in the automotive, aerospace and electronic industries due to their light weight, high specific strength, damping capacity, etc. However, practical applications are limited because magnesium alloys have poor formability at room temperature due to the lack of slip systems and the formation of basal texture, both of which characteristics are attributed to the hcp crystal structure. Fortunately, many magnesium alloys, even commercialized AZ or ZK series alloys, exhibit superplastic behavior and show very large tensile ductility, which means that these materials have potential application to superplastic forming (SPF) of magnesium alloy sheets. The SPF technique offers many advantages such as near net shaping, design flexibility, simple process and low die cost. Superplasticity occurs in materials having very small grain sizes of less than $10{\mu}m$ and these small grains in magnesium alloys can be achieved by thermomechanical treatment in conventional rolling or extrusion processes. Moreover, some coarse-grained magnesium alloys are reported to have superplasticity when grain refinement occurs through recrystallization during deformation in the initial stage. This report reviews the characteristics of superplastic magnesium alloys with high-strain rate and coarse grains. Finally, some examples of SPF application are suggested.

Lactobacillus plantarum KI134의 생리적 특성 및 이 균에 의한 우유 발효물의 항비만효과 (Physiological Characteristics and Anti-Obesity Effect of Milk Fermented by Lactobacillus plantarum KI134)

  • This study aimed to investigate the physiological characteristics and anti-obesity effects of milk fermented by L. plantarum KI134. The lipase, α-amylase, and α-glucosidase inhibitory activities of milk fermented by L. plantarum KI134 was 94.57±1.25%, 9.44±2.85%, and 2.74±1.24% (10 fold dilution), respectively. L. plantarum KI134 showed higher sensitivity to clindamycin and erythromycin in comparison to sixteen different antibiotics. It demonstrated the highest resistance toward ampicillin and vancomycin. The strain showed higher β-galactosidase, leucine arylamidase, valine arylamidase, acid phosphatase, β-glucosidase, and N-acetyl-β-glucosaminidase activities compared to other enzymes. It also did not produce carcinogenic enzymes, such as β-glucuronidase. The survival rate of L. plantarum KI134 in 0.3% bile was 96.90%. Moreover, the strain showed a 91.45% survival rate at a pH of 2.0. L. plantarum KI134 has resistance to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus at the rates of 70.00%, 68.18%, 59.05%, and 40.63%, respectively. L. plantarum KI134 (23.01%) showed higher adhesion ability than the positive control (16.32%) L. rhamnosus GG. These results demonstrated that milk fermented by L. plantarum KI134 demonstrated an anti-obesity effect under in vitro conditions, with confirmed potential as a probiotic.

시료 파괴 시 발생하는 SP에 관한 기초 연구 (The basic study about streaming potential generated by specimen fracture)

  • We measured potential waveform of load, displacement, micro electric signal generated by rock and mortar fracture using PXI A/D Converter. The rock type used for measurement was used granite, limestone and sandstone, and mortar specimen. we made measuring equipment of physical properties to confirm basic information of physical properties, measured physical properties of rock engineering, electric resistivity and seismic velocity. Potential waveform system was built using PXI A/D Converter and measured potential waveform of load, displacement, micro-electric signal generated using this during uniaxial compressive test by the specimen finished such test of physical properties. Using the saturated rock and mortar specimen, micro electric signal increased, and It didn't increase a signal in dried rock and mortar specimen according as load and strain rate increases. But signal also increased in saturated or dried specimen in case of sandstone. It was possible to check the close correlation relationship the signal and fracture behavior by a compressive load as the signal of fracture position was increased bigger than the other position. It was also possible to check the correlation relationship between physical properties and micro geo-electric signal.

Stress Induced Phosphate Solubilization by Aspergillus awamori bxq33110 Isolated from Waste Mushroom Bed of Agaricus bisporus

  • A fungal strain, capable of solubilizing insoluble phosphate under diverse temperature, pH and salt conditions was isolated from Waste Mushroom bed of Agaricus bisporus in South Korea. Based on 18S rRNA analysis, the strain was identified as Aspergillus awamori bxq33110. The strain showed maximum phosphate solubilization in AYG medium (525 ${\mu}g\;mL^{-1}$) followed by NBRIP medium (515 ${\mu}g\;mL^{-1}$). The strain solubilized $Ca_3(PO_4)_2$ to a greater extent and rock phosphate and $FePO_4$ to a certain extent. However $AlPO_4$ solubilizing ability of the strain was found to be very low. Glucose at the rate of 2% ($561{\mu}g\;mL^{-1}$) was found be the best carbon source for Aspergillus awamori bxq33110 to solubilize maximum amount of phosphate. However, no significant difference ($P{\leq}0.05$) in phosphorus solubilization was found between 1% and 2% glucose concentrations. $(NH_4)_2SO_4$ was the best nitrogen source for Aspergillus awamori bxq33110 followed by $NH_4Cl$ and $NH_4NO_3$. At pH 7, temperature $30^{\circ}C$ and 5% salt concentration (674 ${\mu}g\;mL^{-1}$) were found to be the optimal conditions for insoluble phosphate solubilization. However, strain Aspergillus awamori bxq33110 was shown to have the ability to solublize phosphate under different stress conditions at $30-40^{\circ}C$ temperature, pH 7-10 and 0-10% salt concentrations indicating it's potential to be used as bio-inoculants in different environmental conditions.

Thermomechanical and electrical resistance characteristics of superfine NiTi shape memory alloy wires

  • Structural health monitoring and structural vibration control are multidisciplinary and frontier research directions of civil engineering. As intelligent materials that integrate sensing and actuation capabilities, shape memory alloys (SMAs) exhibit multiple excellent characteristics, such as shape memory effect, superelasticity, corrosion resistance, fatigue resistance, and high energy density. Moreover, SMAs possess excellent resistance sensing properties and large deformation ability. Superfine NiTi SMA wires have potential applications in structural health monitoring and micro-drive system. In this study, the mechanical properties and electrical resistance sensing characteristics of superfine NiTi SMA wires were experimentally investigated. The mechanical parameters such as residual strain, hysteretic energy, secant stiffness, and equivalent damping ratio were analyzed at different training strain amplitudes and numbers of loading-unloading cycles. The results demonstrate that the detwinning process shortened with increasing training amplitude, while austenitic mechanical properties were not affected. In addition, superfine SMA wires showed good strain-resistance linear correlation, and the loading rate had little effect on their mechanical properties and electrical resistance sensing characteristics. This study aims to provide an experimental basis for the application of superfine SMA wires in engineering.

Combination of berberine and ciprofloxacin reduces multi-resistant Salmonella strain biofilm formation by depressing mRNA expressions of luxS, rpoE, and ompR

  • Bacterial biofilms have been demonstrated to be closely related to clinical infections and contribute to drug resistance. Berberine, which is the main component of Coptis chinensis, has been reported to have efficient antibacterial activity. This study aimed to investigate the potential effect of a combination of berberine with ciprofloxacin (CIP) to inhibit Salmonella biofilm formation and its effect on expressions of related genes (rpoE, luxS, and ompR). The fractional inhibitory concentration (FIC) index of the combination of berberine with CIP is 0.75 showing a synergistic antibacterial effect. The biofilm's adhesion rate and growth curve showed that the multi-resistant Salmonella strain had the potential to form a biofilm relative to that of strain CVCC528, and the antibiofilm effects were in a dose-dependent manner. Biofilm microstructures were rarely observed at $1/2{\times}MIC/FIC$ concentrations (MIC, minimal inhibition concentration), and the combination had a stronger antibiofilm effect than each of the antimicrobial agents used alone at $1/4{\times}FIC$ concentration. LuxS, rpoE, and ompR mRNA expressions were significantly repressed (p< 0.01) at $1/2{\times}MIC/FIC$ concentrations, and the berberine and CIP combination repressed mRNA expressions more strongly at the $1/4{\times}FIC$ concentration. The results indicate that the combination of berberine and CIP has a synergistic effect and is effective in inhibiting Salmonella biofilm formation via repression of luxS, rpoE, and ompR mRNA expressions.

FCC 다결정재의 집합조직 발전에 따른 이방성의 변화 (Anisotropy due to Texture Development in FCC Polycrystals)

  • The present study is concerned with the development of anisotropy and deformation texture in polycrystals. The individual grain in an aggregate is assumed to experience the viscoplastic dedformation with crystallographic slip that unsure uniquenessof the active slip systems and shearing rate onthese systems. Two different methods for updating the grain orientation are examined. Texture development for some deformation modes such as plane strain compression, uniaxial tension and simple shear are found. Changes in anisotropic flow potential due to texture development during large deformation are also given. Anisotropic behavior of polycrystals with defferent textures are examined.
