A Study on Mineral and Alginic acid Contents by Different Parts of Sea Mustards(Undaria pinnatifida) (미역의 부위별 무기성분 및 알긴산 함량에 관한 연구)

  • This study was conducted to examine the proximate composition, alginic acid and mineral contents and the types of calcium and magnesium in the parts(frond, stipe, sporophyll) of sea mustards. Carbohydrates and the crude fat was the highest in sporophyll. The contents of alginic acid were $25.9{\sim}32.2%$. Total alginic acid(32.2%) and insoluble alginic acid in water(27.7%) was e highest in sporophyll. Calcium and magnesium was the highest in frond. Phosphorous, potassium and sulfur was the highest in sporophyll. Iron, zinc and mangane was the highest in frond. Ca/P ratio in frond and stipe was about $1.7{\sim}1.8:1$ levels. Calcium and magnesium soluble in sodium chloride was the highest in sea mustards. Calcium and magnesium soluble in water was the highest in frond. Calcium and magnesium soluble in hydrochloric acid was the highest in sporophyll.

Fatty Acid Compositions of Nutural Lipids and Polar Lipids in the Parts of Miyeok (Undaria pinnatifida) (미역의 부위별 중성 및 극성 지방질의 지방산 조성)

  • Fatty acid composition of frond, spororphyll and stipe of Miyeok (Undaria pinnatifida) were extensively analyzed to evaluate a nutrient significance of n-3 polyenes, n-6 polyenes, ${\gamma}$-linolenic acid (18:3, n-6) and $\alpha$-linolenic acid (18:3, n-3). Polyenes of $\alpha$-linolenic acid (22:6, n-3), stearidonic acid (18:4, n-3), eicosapentaenoic acid (20:5, n-3) and docosahexaenoic acid (22:6, n-3) were as high as 29.69%, but ${\gamma}$-linolenic acid (18:3, n-6) was as low as 0.90% when compared to total polyenes of 44.18% in frond. The n-3 polyenes of neutural lipids, glycolipids and phospholipids were 22.40%, 35.82% and 3.53% in frond, 5.26%, 9.51% and 2.37% in stipe and 8.01%, 4.49% and 2.14% in sporophyll, respectively. These results suggest that total polyenes and n-3 polyenes in frond of Miyeok may be used as resource of functional food.

Characteristics of Ganoderma lucidum during short log cultivation with soil contact barrier treatment (영지버섯(Ganoderma lucidum) 단목재배시 토양접촉차단재 처리별 자실체 특성)

  • Arthrographis cuboidea is a wood rot fungus found in soil. It causes disease in repeated cultivation areas of Ganoderma lucidum, which damage the mushroom logs and causes considerable loss of income to mushroom farmers. This study aimed to investigate the physicochemical properties of soil and characteristics of the growth environment such as the temperature of the Ganoderma lucidum cultivated land. The time required for primordia initiation was 12-13 days. In the chromaticity survey, the degree of redness (a) was 19.5 for vinyl, 20.2 for control, and 19.5 for nonwoven. The yellowness index (b) was 12.7 for vinyl, 13.3 for control, and 12.4 for nonwoven. In the study of growth characteristics, in case of no treatment (control), the length and thickness of the pileus were measured as 145.7 mm and 23.6 mm and those of the stipe were 73.9 mm and 11.5 mm, respectively, and the weight was 130 g. Treatment with non-woven fabric resulted in 157 mm and 93.3 mm long and 22.3 mm and 11.9 mm thick pileus and stipe, respectively, and the weight was 164 g. This result indicates that treatment with non-woven fabric is better than no treatment with respect to Ganoderma lucidum growth.

Physicochemical characteristics and antioxidant activities of Lentinula edodes cultivated with liquid spawn according to harvest cycle (액체종균에 의한 표고의 수확 주기에 따른 이화학적 특성 및 항산화 활성)

  • The physicochemical characteristics and antioxidant activities of Lentinula edodes cultivated with liquid spawn were compared according to the harvest cycle (1-3 cycles). There was no significant difference in moisture content. The crude lipid and crude protein contents tended to decrease according to harvest cycle. The content of β-glucan was the highest in mushrooms after one cycle. The β-glucan content in cycles 2 and 3 was 68.5% and 62.3% that of the content after cycle 1, respectively. Mineral content in the pileus and stipe was similar. The mineral content was highest in the pileus after two cycles. Composition and contents of free amino acids tended to decrease according to the harvest cycle. Contents of essential amino acids were higher in mushroom after one cycle. Total phenol content was highest in the pileus (31.96 mg/100 g) and the stipe (21.10 mg/100 g) after one cycle and tended to decrease with subsequent cycles. The flavonoid content was 11.13 mg/100 g after one cycle and significantly decreased according to the harvest cycle. Antioxidant activities were also highest after one cycle and significantly decreased according to the harvest cycle. When compared to L. edodes cultured for one cycle, L. edodes cultivated with liquid spawn for three cycles was considered to have similar marketability in terms of quality characteristics.

  • The species of Grateloupia filicina, Grateloupia divaricata in East Sea were investigated taxonmically in order to clarify taxonomic position. The ecological character, external morphology, anatomy of vegetative structure. Blade length are $15{\sim}40\;cm$, erect from discoidal holdfast of $3{\sim}10\;mm$ in diameter. Stipe $1{\sim}2.5\;cm$ long, narrowly cylindrical below, compressed above Grateloupia filicina. Main axis are long and compressed, $3{\sim}7\;mm$ broad in broadest part. Colors are scarlet to light red. Blade length are $10{\sim}25\;cm$, erect from discoidal holdfast of $3{\sim}8\;cm$ in diameter Grateloupia divaricata. Stipe are single and simple $2{\sim}5\;mm$ broad. Thallus composed of cortex and medulla in section ; cortex composed of $9{\sim}10$ layers of anticlinally arranged cortical cell, divided into outer, middle and inner parts. Partial fragments of nuclear 18S rDNAs from the two species of Grateloupia (Rhodophyta) were amplified using the PCR reaction and sequenced to compare their similarity. The partial sequences showed 98.9% similarity each other. Grateloupia filicina has 371 bp sizes and Grateloupia divaricata has 372 bp size. The G+C contents of Grateloupia filicina is 54.3% and Grateloupia divaricata is 53.64%.

Characteristics and breeding of a new cultivar Pleurotus eryngii var. ferulae, 'Beesan No.2' (아위느타리 신품종 '비산2호'의 육성 및 자실체 특성)

  • To develop a new cultivar of King oyster mushroom white variety (Pleurotus ferulae), GW10-68 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2798-24 derived from ASI 2798 and dikaryotic strain ASI 2803. The GW10-95(ASI 2803 ${\times}$ ASI 2798-24) was shown the best cultural characteristics, selected to be a new cultivar and designed as 'Beesan No.2'. The 'Beesan No.2' was formed incompatibility line distinctly in the confrontation growth of parental strains ASI 2803, ASI 2798 and Beesan No.1. Analysis of the genetic characteristics of the new cultivar 'Beesan No.2' showed a different DNA profile as that of the control strains, ASI 2803, ASI 2798 and 'Beesan No.1', when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were $25^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about 123.3 g. And also the stipe was long. This new cultivar 'Beesan No.2' of Pleurotus ferulae was characterized white-like variety of King oyster mushroom in the color of stipe, that was long and low yield compared to that of other cultuvar 'Beesan No.1'. We therefore expect that this new strain will increase of export and substitute cultivar of King oyster mushroom.

This new cultivar 'Beesan No.1' of Pleurotus ferulae was characterized by fast fruitbody formation, the stipe was thick and long and high quality yield compared to that of other cultuvars. We therefore expect that this new strain will increase of farmer's income by construction of stabilized production system.

Characteristics of a new variety 『Gonji10ho』 in Neolentinus lepideus (잣버섯 신품종 『곤지10호』 특성)

  • 'Gonji10ho', a new variety of Neolentinus lepideus, was bred by mating two monokaryotic strains isolated from 'GMNL66037' and 'GMNL66014' in Mushroom Research Institute, Gyonggi-Do ARES in 2012. The optimum temperature for the mycelial growth of 'Gonji10ho' was $29^{\circ}C$ on PDA medium and that for the primordia formation and the growth of fruiting body of 'Gonji10ho' was $20{\pm}1^{\circ}C$. In bottle cultivation of 'Gonji10ho', the period of spawn running was around 35 days at $22{\pm}1^{\circ}C$ and the period from scratching of inoculum to harvesting was 27 days. 'Gonji10ho' had thicker pileus and stipe than control variety('Solhyang'). The number of available stipe was fewer than that of control variety. During the cultivation of 'Conji10ho', the occurrence rate of Trichoderma spp. was lower than that in control variety. In bottle cultivation, the yield of fruiting bodies of 'Gonji10ho', which was 38% higher than that of control variety, was 69.6 g/900 ml bottle, and in bag cultivation, that of 'Gonji10ho', which was 10% higher than that of control variety, was 114.2 g/1 kg bag. In physical characteristics, the strongness and brittleness of the pileus of 'Gonji10ho' were higher than those of control variety. When considered in the period available for sale, the shelf life of 'Gonji10ho', which was 5 days longer than that of control variety, was 34 days.

  • To develop a new cultivar of king oyster mushroom(Pleurotus eryngii), G09-21 as parental strain was selected by the method of Di-mon crossing between monokaryotic strain ASI 2824-21 derived from ASI 2824(Keunneutari No. 2) and dikaryotic strain ASI 2887(Aeryni 3). The Pe21-53(G09-21-10 x ASI 2844-9) was shown the best cultural characteristics, selected to be a new cultivar and designated as 'Seolsong'. The 'Seolsong' was distinctly formed incompatibility line in the confrontation growth of parental strains Keunneutari No. 2, Aeryni 3 and ASI 2844. Analysis of the genetic characteristics of the new cultivar 'Seolsong' showed a different DNA profile as that of the control strains, Keunneutari No. 2, Aeryni 3 and ASI 2844, when RAPD(Random Amplified Polymorphic DNA) primer URP4 was used. The optimum temperature and pH arrange for mycelial growth were $25{\sim}30^{\circ}C$ and pH 5~8, respectively. This new cultivar 'Seolsong' of fruiting body production per bottle was about $131.4{{\pm}}43.1$ g which is about 102% quantity compared to that of other cultivar Keunneutari No. 2. And also the stipe is thick and long, but the number of available stipe is few. Particularly, it was tolerant of high moisture above 90% during the growth period after primodia formation. We therefore expect that this new strain will save of labor and cost of cultivation by without culling work.

Production of Flammulina velutipes by Using Culture Medium Containing Herb Medicine Refuse and Analysis of Characteristics of Its Fruiting Bodies (폐 한방슬러지 첨가배지를 이용한 팽이버섯의 생산 및 자실체 특성 검토)

  • This study aims to reduce the production cost of mushrooms and therefore boost the income of mushroom producers. The addition of a 10% concentration of herb medicine refuse was adequate for promoting mycelium growth in Flammulina velutipes. A moisture content of 60% in the culture medium was adequate for promoting hyphal growth. The optimum temperature and humidity were $16^{\circ}C$ and 70%-85%, respectively. The average yield of Flammulina velutipes fruting body per 1,100 ml bottle was 275-282 g, which is similar to that of the control. The outbreak of diseases such as mildew and bacterial blotch in mushroom was never observed in the bottle treated with herb medicine refuse. Fewer deformed and second-class quality mushrooms were observed than in the control group. Deviation reduced, and mushroom growth was observed to be significantly uniform. The winter mushroom had a white fruiting body with a hemispherical shape. The diameter of the mushroom cap was 9.2-9.3 mm, stipe length was 12.7-12.8 cm, and thickness was 3.3-3.4 mm; these were almost the same as those in the control group. The partial hardness of winter mushrooms in both the experimental and the control groups showed that the stipe ($54.6-57.3g/cm^2$) is a little harder than the mushroom cap ($46.8-47.6g/cm^2$). The calories and nutrients per 100 g of mushrooms in the control and samples were similar to each other.
