Factors Affecting Plant Regeneration in the Culture of Different Explants of Stevia(Stebia rebaudiana Bertoni) (Stevia 절편체 종류의 배양에 따른 식물체 재분화)

  • This study was conducted to determine the optimal concentrations of plant hormones (2,4-D, picloram, dicamba, NAA, kinetin) and the suitable explants among seeds, hypocotyl, and cotyledons on calls formation and plant regeneration of stevia(Stevia rebaudiana Bertoni). The frequency of cellus formation was higher in the young leaf-explants then the older ones, and in the seeds then the hypocotyls and cotyledons on MS medium with 1mg/L 2,4-D. After transfer of seed-derived stevia callus producing embryogenic callus on plant-regeneration medium, the frequency of plant regeneration from callus was 23.8% in MS medium with 1mg/L NAA and 3mg/L kinetin.

  • The consumption of sugar in our diet increased so great that much U. S. dollars are spended to import sugar source from foreign contries. To replace a part of it we adopted mother plant and seeds of the well-known Stevia (Stevia rebaudiana Bertoni M.) originated from south America, from the Agricultural Experimental Station of Hokkaido, Japan, in 1974. In order to determine the growth condition of Stevia in our climate and soil condition, the results of survey and determination were as follows: 1. Germination rate of Stevia was averaged 33.6% at 35 th day after seedling and the maximum germination period was between 5 and 20 days after seedling. 2. The seedling rate of Stevia was about 8%. 3. Establishment of roots of Stevia propagated by cutting method was above 90% but only S-1 strain showed lower rate., 75%. 4. S-2 strain showed the best growth rate that was about 160cm by plant height and it was decreased by the following order : S-4, S-3, S-1, S-6 and S-5 strains. 5. S-4 strain resulted in 45 branches that was the highest number among strains tested and S-2 strain was the next. 6. The content of sweetening compound of Stevia, stevioside, was 5.3% in S-1, S-2, S-5 and S-4 strains but S-3 and $S^*$-6 strains were relatively low content, 4.9%. From the above results we obtained Stevia could be propagated by cutting method and the growth conditions including plant height, number of branches and content of stevioside were relatively better in S-2 and S-4 strains that were differentiated by the shape of leaf.

Effects of Gamma-ray and Chemical Mutagens on the Germination and Seedling Growth in Stevia rebaudiana Bert. (감마선 및 화학적 돌연변이원 처리가 스테비아 (Stevia rebaudiana Bert.)의 종자 발아 및 초기 생장에 미치는 영향)

  • This study was carried out to develop the improved useful mutants for yield or composition of stevia plants using the gamma ray or chemical mutagens treatments. The seeds of stevia 'Suwon No. 11' were irradiated up to 400 Gy of gamma ray. Chemical mutagens were treated on the seeds of the 'Suwon No. 11' using 0.07% colchicine, 10 mM sodium azide, or 10 mM NMU for various durations. The germination rate, and shoot and root growth of seedling were estimated at 30 days after gamma ray irradiation or chemical mutagen treatment, and the plant height, the number of branches, and leaf length and width were examined at 3 months after mutagenesis treatments. In the case of gamma ray treatments, the germination rate and early-stage growth were decreased as the increase of radiation dose, and the 50% lethal dose was found to be 200 Gy. the plant height was decreased as the increase of radiation dose, while the number of branches per plant and leaf length were increased. Leaf shape was modified to the relatively longer one compared to the control, which was identified more apparently at the treatments of higher than 150 Gy. In the treatment of chemical mutagens, the rate of germination and survival were decreased as the increase of incubation time. The 50% lethal dose for germination rate were identified as the conditions of the 15 hours incubation in 0.07% colchicine, the 4 hrs in 10 mM sodium azide, and the 2 hrs in 10 mM NMU, in the three chemical mutagens treatments. Chemical mutagens had no influence on shoot growth, while root growth was increased, especially as the incubation time was extended. The highest root growth occurred in the NMU treatment at 6 hrs incubation time. The plant height was decreased as the increase of incubation time in the chemical mutagens treatments. Among the chemical mutagens, NMU was the most effective to induce the mutants with long-shaped or the least lobed leaves.

  • In Korea. the cultivating standard of stevia is not established yet. Because stevia is across-fertilizing plant. planting a cutting is its available propagation method. This study was made to know the effective propagation growth and adaptability of stevia in Jeju-do, according to the wintering effect and the planting density.

Antioxidant and Antimelanogenic Effects of Stevia rebaudiana Flower Extract

  • Stevia rebaudiana (Asteraceae), a perennial plant, has been used as a low-calorie sweetener and is being developed as a therapeutic agent for diabetes, hypertension, myocardial diseases, and microbial infections. Despite the common use of its leaves and stem, the bioavailability of the components present in S. rebaudiana flowers, when used as ingredients of cosmetics, has not been well investigated. Herein, we investigated the antioxidative and antimelanogenic effects of an aqueous extract of S. rebaudiana flowers (Stevia-F). Total flavonoid and phenolic content in Stevia-F were determined to be $8.64{\pm}0.23mg$ of quercetin equivalents/100 g and $631.5{\pm}2.01mg$ of gallic acid equivalents/100 g, respectively. The $IC_{50}$ values of Stevia-F for reducing power, and 2,2-diphenyl-1-picryl-hydrazyl-hydrate radical, hydrogen peroxide, and nitric oxide scavenging activities were 5541.96, 131.39, 466.34, and $10.44{\mu}g/mL$, respectively. Stevia-F showed inhibitory effects on the tyrosinase ($IC_{50}=134.74{\mu}g/mL$) and ${\alpha}$-glucosidase ($IC_{50}=114.81{\mu}g/mL$) activities. No significant cytotoxicity of Stevia-F was observed in B16F10 cells, treated with up to $100{\mu}g/mL$ of the extract for 24 and 48 h (p > 0.05). Stevia-F ($1-100{\mu}g/mL$) suppressed ${\alpha}$-melanocyte stimulating hormone-induced melanin production in B16F10 cells (p < 0.05) and also inhibited the cellular tyrosinase activity (p < 0.05). Overall, our results show that Stevia-F possesses potential for inhibiting tyrosinase and ${\alpha}$-glucosidase activities and has significant antioxidant capacity. The antimelanogenic potential of Stevia-F should extend the usage of S. rebaudiana flowers in the development of skin-whitening products.

Effect of Stevia rebaudiana on the Bioactive Compounds from Agarwood Leaf (Aquilaria spp.) by Lactic Fermentation and Spray Drying

  • Agarwood (Aquilaria spp) has high economic value. However, essential oil production from agarwood is a time-consuming process. Additionally, agarwood leaves have not been utilized even though they contain various bioactive ingredients. In this study, agarwood leaves were fermented using Lactobacillus plantarum ATCC 8014 with or without Stevia (4, 8, and 12%; v/v). The fermented fluid was mixed with maltodextrin (15%; w/v) and subjected to spray drying (inlet temperature, 120℃; outlet temperature, 65-70℃). The contents of polyphenols, polysaccharides, saponins, and flavonoids and the viability of L. plantarum were determined. Fermentation enhanced the levels of bioactive compounds. The contents of polyphenol (69.19 ± 4.05 mg GAE/g of sample), polysaccharide (20.75 ± 0.98 mg GE/g of sample), saponin (305.23 ± 4.21 mg OAE/g of sample), and flavonoid (7.86 ± 0.72 mg QE/g of sample), and the viability of L. plantarum (8.72 ± 0.17 log CFU/ml) were markedly upregulated in the samples containing Stevia (12%; v/v). This indicated that the supplementation of Stevia during fermentation decreases the fermentation time (9 h), upregulates bioactive compound production in agarwood leaves, enhances microencapsulation during spray drying, and increases the viability of L. plantarum under simulated gastric digestion conditions.

Selection of Self-Fertilization Stevia and Development of Multiple Propagation Method using Hypocotyl Explants (자가수정 스테비아의 선발과 배축절편체를 이용한 대량증식 방법 개발)

  • This study was carried out to assess the potential of hypocotyl explants for multiple-propagation by intensive sweetener variety with high yielding in stevia of self-fertilization. Agronomical character of self-fertilization shown a height range 72.0~120.7 cm, branch's 4.9~56.6 in stevia. Selection of high content of Rebaudioside A (RA) from segregating population in mixing variety have a variety, 'DA1' (55.2%) from HPLC analysis. On a developing of multiple-propagation in stevia, hypocotyl explants used to select for reasonable medium under the influence of each plant growth regulators concentrations. In a Mix treatment of plant growth regulators, as Murashige and Skoog medium with 1.5 mg/L BA and 0.5 mg/L Kinetin showed the most favorable. We got the multiple-propagation of 30 shoots from one inoculated hypocotyl explants as the result of these bases. Rooting induced superbly on MS medium with 0.1 mg/L IBA with inoculated shoots. All of the rooting plants were adapted to a normal seedling. This method of multi-propagation is used effectively in Rebaudioside A productivity elevation.

The Quality Characteristics of Stevia (Stevia rebaudiana Bert) Leaf Tea according to Different Manufacturing Processes (스테비아 잎차의 제조 방법에 따른 품질 특성)

  • In order to develop tea by using the leaves of stevia, which is a herbal plant, and to solve the disadvantages of stevia dried leaf tea, we have manufactured the steamed tea, stir-fried tea and fermented tea by changing the manufacturing processes. As a result of the sensory tests, the steamed tea, stir-fried tea and fermented tea received higher evaluations than the dried leaf tea. In terms of efficiency, it is desired that the total number of steaming and stir-frying is only once, but the fermentation is found to be the most desirable for 2 days. There are no trends for changes in the general ingredients, mineral and free amino acid contents of stevia leaf teas by different manufacturing processes. As a result for the measurement of antioxidant activities, the steamed tea and dried leaf tea did not show significant differences, but the stir-fried tea and the fermented tea show significantly low antioxidant activities as compared to the steamed tea. The contents of stevioside in both the stir-fried tea and the fermented tea were less than that in the dried leaf tea, but in the steamed tea, there was no significant difference in the content of stevioside. Base on the present observations, this study supports high potentials of steaming process in order to produce new stevia leaf tea.

Identification of Effective Microorganisms Isolated from Fermented Stevia Extract and Their Antimicrobial Activity (스테비아 추출물 발효액에서 분리된 유효 미생물들의 동정 및 항미생물 활성)

  • Stevia rebaudiana Bertoni is a sweet herb of the Asteraceae family originally derived from South America. Twenty three bacterial strains and ten yeast strains were isolated from fermented Stevia extract and identified by general taxonomic methods and molecular genetic method. Isolated strains from fermented Stevia extract include ten species of bacteria which belong to five genus and one species of yeast. Based on 16S and 18S rDNA sequence analysis, phylogenetic trees were constructed. Antimicrobial activity of the isolated strains was examined against various bacteria and plant-pathogenic fungi. Among them, Lactobacillus paracasei SB13 showed strong antibacterial activity towards a wide range of bacteria. These results may be useful to develop environmentally friendly microbial agent for soil improvement.
