  • The roots of Saposhnikovia divaricata Schischk. (Umbelliferae) have been known to possess analgesic, anti-inflammatory, anti-parasitic and anti-bacterial activities, and used for curing headaches, fever and arthralgia. In this study, we aimed to isolate active constituents to provide phytochemical data for the quality control of this plant. Nine coumarins, eight chromones, three sterols and a coumarolignan were isolated from EtOAc-soluble fraction of the roots of S. divaricata through repetive column chromatography method using silica gel, ODS gel, Sephadex-LH 20, MPLC and HPLC. By analyses of spectroscopic data and comparison of their data with those of published values, the compounds were identified as 3'-O-angeloylhamaudol (1), ${\beta}$-sitosterol (2), marmesin (3), phellopterin (4), anomalin (5), imperatorin (6), xanthotoxin (7), deltoin (8), bergapten (9), stigmasterol (10), ledebouriellol (11), hamaudol (12), 8'-epicleomiscosin A (13), xanthoarnol (14), cimifugin (15), 5-O-methylvisamminol (16), daucosterol (17), 4'-O-${\beta}$-D-glucosyl-5-O-methylvisamminol (18), nodakenin (19), sec-O-glucosylhamaudol (20), prim-O-glucosylcimifugin (21). Among them, 8'-epicleomiscosin (13) was firstly reported from Umbelliferae family and xanthoarnol (14) and nodakenin (19) were isolated from this plant for the first time.

From EtOAc fraction, three sterols were isolated on repeated silica gel and ODS column chromatographies. The chemical structures were determined as ${\beta}-sitosterol$ (1), daucosterol (2) and ergosterol peroxide (3) by NMR, MS and IR, which is the first to be isolated from Cymbidium goeringii REICHB. fil.

  • The sprouts of Brassica oleracea var. gongylodes were extracted with 100% MeOH and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2O$, successively. From the EtOAc fraction, three sterols were isolated through the repeated silica gel and ODS column chromatographies. On the basis of physico-chemical and spectroscopic data including NMR, MS, and IR, the chemical structures of the sterols were determined as ${\beta}$-sitosterol (1), brassicasterol (2), and 7-ketobrassicasterol (3). Compound 1 is usually observed in plant. Compounds 2 is observed in Brassica sp., and compounds 3 have very rarely occurred in natural source including plant.

Variation of Properties of Lipid Components in Rice Endosperm Affected on Palatability (식미에 영향을 미치는 쌀 배유조성 지질성분특성 변이)

  • In order to understand the difference in rice eating quality, two rice varieties having different eating qualities were examined from the correlation between palatability score (Toyo value) and components of lipid in rice endosperm. We also analyzed the relationship between lipid properties and physicochemical characteristics of milled rice. Major fatty acids of milled rice starch-lipids, known to exist as inclusion complexes with amylose in starch granules, were palmitic (38.25% in Gopum and 39.75% in Palgong) and linoleic aicds (33.13% in Gopum and 31.93% in Palgong). In addition, the unsaturated fatty acid contents of rice endosperm showed a significant relationship with palatability. Plant sterols in two rice samples were identified, Gopumbyeo of good eating quality had high contents of squlaene and cycloartenol and similar contents of campesterol, stigmasterol and sitosterol. There were significant differences in pasting properties of rice flours and starches between the two cultivars. These results suggested that rice lipids impact on pasting properties after cooking, subsequently relating to palatability.

Hypocholesterolemic Effect of Tangerine Pulp, Sea Tangle or Prickly Pear Cactus on Lipid Level, Intestinal Cholesterol Absorption, Platelet Aggregation and Liver Tissue in Hypercholesterolemic Rats (감귤박, 다시마, 손바닥 선이장 분말을 함유한 식이의 급여가 고콜레스테롤혈증 흰쥐의 체내 지질수준과 장내 콜레스테롤 흡수, 혈소판 응집성 및 간 조직에 미치는 영향)

  • This study done to investigate the hypocholesterolemic effect of ow dietary fiber in a high cholesterol diet in aspect of cholesterol absorption and excretion using Sprague Dawley rats. After feeding diets containing 0.5% cholesterol and 5% tangerine pulp, sea tangle or prickly pear cactus for four weeks, we measured the levels of plasma and liver cholesterol and triglyceride, fecal neutral sterols and fecal radioactivity after ingestion 14(sup)C-cholesterol. We also examined platelet aggregation and histological change in liver tissues in association of hypercholesterolemia. The liver to body weight ratio was significantly(p<0.01) lower in rats fed prickly pear cactus than in other groups. The levels of plasma total cholesterol, LDL-cholesterol and triglyceride were decreased significantly(p<0.01) in the prickly pear cactus group compared with the control, while there was no difference in the liver levels of total cholesterol and triglyceride among groups. Fecal corprostanol and cholesterol were significantly(p<0.01) higher groups of control and prickly pear cactus compared to the other two groups. Radioisotope excretion after ingestion of 14(sup)C-cholesterol was higher in the control group than in tangerine pulp group or sea tangle group, with the highest in prickly pear group. Radioisotope excretion was the highest during the 2nd and 3rd days in all groups. Hematocrit and platelet aggregation were decreased in all fiber groups compared to the control, but not statistically different. Microscopic examination showed that cholesterol diet cause a fat accumulation in the liver and prickly pear cactus decreased the fat accumulation. Result indicates that prickly pear cactus has hypocholesterolemic effect by decreasing absorption and increasing excretion of cholesterol, thereby protective effect on fatty liver. Control group fed diet containing high cholesterol and low fiber seems to have a self control system in cholesterol absorption and excretion preventing hypercholesterolemia.   • The Korean origin sunflower (Helianthus Annus Linn.) seed of netural lipid were analysed by thinchrography, High performance liquid chromatography, preparative Thinlayer and Gas liquid chromatography. 1) The seed oil triglyceride components were conveniently separated based on their degree of unsaturation by employing the chromatography on silica gel sintered rod impregnated with 12.5% silver nitrate. Sunflower seed oil was composed of triglyceride, especially trilinolein 57. 74% triolein 25.28%, tripalmitin 7 55% ana tristearin 9.43% by a thinctrography. 2) The fatty acid compositions of seed oil have been determined by a high performance liquid chromatographic analysis using a ALC/GPC 244 type from Waters Association (Japan) with ${\mu}$ Bondapak FFAA column. It contained stearic acid 8.59%, oleic acid 27. 19%, palmitic acid 7.50% and linoleic acid 56.72% respectively. 3) The composition of sterols were determined by a preparative Thinlayer and Gas liquid chromatographic analysis. It was noted that sitosterol was the major sterol in the Korean sunflower seed. The results showed that contents of sterols were cholesterol trace, campesterol $13_.^{22\sim}13.9%$, stigmasterol $13.8{\sim}14.1%$, If, sitosterol $58.4{\sim}60.7%$, ${\vartriangle}^7$-stigmastenol $10.2{\sim}10.5%$ and ${\vartriangle}^{7,24(25)}$-stigmastenol $3.6{\sim}3.8%$ by method of planimetry and triangulation.

Changes in Lipid Components During Germination of MungBean (녹두발아(綠豆發芽)중 지방질성분(脂肪質成分)의 변화(變化))

  • This investigation was conducted to observe changes of lipid components (phospholipids, free sterols, free fatty acids, triglycerides and estrified sterols) and fatty composition in diethyl ether extract and 86% MeOH extract from cotyledon and seedling axis of mung bean which were germinated in the dark at 25-$27^{\circ}C$ for 7 days. The total lipid contents in cotyledonn and seedling axis gradually decreased during germination. The triglyceride content in total lipid from cotyledon decreased and free fatty acid content increased, but triglyceride content from seedling axis decreased slightly and free fatty acid content decreased. Free sterol, esterified sterol and phospholipid content in cotyledon increased continuously, but their content in seedling axis dcreased slightly. The major fatty acids in mung bean were palmitic acid, linoleic acid and linolenic acid.

  • The flowers of Chionanthus retusus Lindl. & Paxton were extracted with 80% aqueous MeOH at room temperature. The concentrated extract was partitioned as EtOAc, n-BuOH, and $H_2O$ fractions. From the EtOAc fraction, two triterpenoids and two sterols were isolated using the repeated silica gel ($SiO_2$) and octadecyl $SiO_2$ (ODS) column chromatographies. According to the results of physico-chemical and spectroscopic data including nuclear magnetic resonance, mass spectrometry, and infrared. The chemical structures of the compounds were respectively determined as ursolic acid (1), corosolic acid (2), ${\beta}$-sitosterol (3), and daucosterol (4). All the compounds were isolated for the first time from the flowers of Chionanthus retusus Lindl. & Paxton.
