A Comparative Study on the Lipid Components of Barley and Malt -I. Composition of Neutral Lipids- (보리와 맥아(麥芽)의 지방질(脂肪質) 성분(成分)에 관한 비교(比較) 연구 -제(第)1보(報) : 중성지질(中性脂質)의 조성(組成)-)

  • Neutral lipids of the total lipid extracted from 4 representative varieties of barley grown in Korea and their corresponding malt were studied. Total lipids of barley and malt were solvent extracted with chloroform:methanol:water (1.0 : 1.0 : 0.9, v/v), The total lipids were fractionated into neutral and polar lipids by silicic acid column chromatography, and neutral lipids fraction was separated by thin layer chromatography and quantitated by TLC scanner. The fatty acid compositions was determined by gas liquid chromatography. The average content of total lipid in the 4 barleys and their malts were 3.3 and 2.9%, and average of neutral lipids content in the barley and malt lipids were 73.8 and 68.5%, respectively. Among the neutral lipids, triglycerides and free fatty acids were the major components, and triglycerides content decreased and free fatty acids content increased during malting. Sterol esters, free sterols, 1,3-and 1,2-diglycerides were the minor components of the neutral lipids, and contents of those components showed increasing tendency during malting. The major fatty acid composition of the total lipids were linoleic, palmitic and oleic acids, and in general, the malts had lower amounts of unsautrated fatty acids and high amounts of saturated fatty acids. Fatty acid composition of neutral lipids was of almost the same pattern as that of the total lipids.

Effect of Different Application of P, K, and Ca on Lipid Content and Composition of Perilla (인산(燐酸), 가리(加里), 석회(石灰) 시용(施用)이 들깨의 유지함량(油脂含量)과 그 조성(組成)에 미치는 영향(影響))

  • This study was carried out to investigate the effects of P, K and Ca on Perilla lipid content and composition. The results were summarized as follows; Seed yield of Perilla was more increased with P application than K and Ca. K application resulted in increased lipid content of Perilla seed, while P and Ca had no effect on lipid content. Triglyceride content in Perilla lipid was increased with application of P, particulary up to 92.1% with P+K+Ca application. Main fatty acids in Perilla lipid were palmitic, oleic, linoleic, and linolenic acid that is the most predominating fatty acid. P and Ca applicatin without K decreased the proportion of linolenic acid, and therefore degree of unsaturation when P and Ca were respectively applicated was 1.80 and 1.84, but when together with K they were respectively increased to 2.06 and 2.11. 4-Desmethyl sterols which is a predominating fraction in unsaponifiables of seed lipid were identified as campesterol, stigmasterol, and sitosterol. Of the 4-desmethylsterols, sitosterol showed the highest proportion at all pots with different kinds of fertilizers.

Isolation of Growth Inhibition Substance on Food borne Microorganisms from Hypericum ascyron L. and Application to Food Preservation (물레나물(Hypericum ascyron L.)의 식중독 미생물 증식 억제 물질의 분리 및 식품적용)

  • The ethanol extract and n-hexane fraction from Hypericum ascyron L. showed strong growth inhibition at 25 ppm on 5 strains of Listeria monocytogenes for 72 hr at $32^{\circ}C$. The purified substance, H2-5-2 fraction, was isolated by silica gel column and preparative thin layer chromatography from n-hexane fraction of Hypericum ascyron L. The H2-5-2 fraction showed a strong bacteriostatic activity on 5 strains of L. monocytogenes at 10 ppm in tryptic soy broth, and the viable cell was reduced 1 log cycle compared to initial cell number. The n-hexane fraction of Hypericum ascyron L. showed strong growth inhibition at 25 ppm on Bacillus cereus and Staphylococcus aureus, and at 50 ppm on Vibrio parahaemolyticus for 72 hr. The purified antimicrobial substance, the H2-5-2 fraction, was assumed as high unsaturated sterol by $^1H-NMR$ and $^{13}C-NMR$. On application test using minced Alaska pollack and ground beef, the n-hexane fraction of Hypericum ascyron L. at the level of 250 ppm was applied at $32^{\circ}C$ and $5^{\circ}C$. At $32^{\circ}C$ storage condition, the antimicrobial substances did not reduced L. monocytogenes ATCC 19113, meanwhile at $5^{\circ}C$ storage condition, L. monocytogenes ATCC 19113 was reduced in viable number.

  • The sterol compositions of the Pelecypoda, M. sultataria, S. sachalinensis, and the Gastropoda, H. discus hannai Inc, T. cornutus were investigated. The results areas follows: 1. The contents of the unsaponifiables and sterols of the Pelecypoda, M. sulcataria, S. sathalinensis, and the Gastropoda, H. discus hannai Ino, T. cornutus, are $12.0\%,\;11.8\%,\;and\;16.2\%,\;15.3\%$, respectively. 2. The complex sterols from the Pelecypoda and Gastropoda are well separated on Silica Gel HF 254 TLC impregnated with $15\%$ silver nitrate. 3. The prominent sterols of the Pelecypoda, M. sulcateria and S. sachalinensis, are 22-trans-24-norcholesta-5, $22-dien-3\beta-ol$ $3.0\%\;3.9\%$ 22-dehydrocholesterol $6.7\%,\;10.2\%$, cholesterol $39.0\%,\;48.6\%$ brassicasterol $14.1\%,\;13.8\%$ 24-methylenecholesterol $19.4\%,\;11.5\%$, stigmasterol $2.4\%,\;0\%$ $\beta-sitosterol\;10.5\%,\;11.9\%$, and fucosterol $4.3\%,\;0\%$. 4. Abalone, H. discus hannai Ino, and T. cornutus contain cholesterol $98.0\%,=97.5\% as main component with small amounts of 22-dehydrocholesterol, brassicasterol and desmosterol. In H. discus hannai rno, 24-methylenecholesterot and fucosterol are also found.

Studies on the Lipid and Fatty Acid Compositions of Ark-Shell, Anadara broughtonii (피조개 지질 및 지방산 조성에 관한 연구)

  • The present study was designed to analyze the lipid and fatty acid compositions of ark shell, Anadara broughtonii. The crude lipid was extracted by Bligh and Dyer's method, and then fractionated by TLC and quantitatively analyzed by TLC scanner. Lipid extracted from ark shell was fractionated into neutral and polar lipid by column chromatography with silicic acid. The fatty acid composition of lipid fractions were determined by gas liquid chromatography. Total lipid content of ark shell was 0.83% base on wet weight. The content of unsaponifiable matter was 20.19%, and iodine value was 156.13. The main components of total lipids were triglyceride, diglyceride, hydrocarbon, and sterol ester. The fatty acid composition of total lipid chiefly consisted of $C_{17 : 0}$, $C_{16 : 0}$, $C_{18 : 1}$ and $CT_{16 : 1}$. The main fatty acids of neutral lipid were $C_{16 : 0}$, $C_{18 : 1}$, $C_{22 : 1}$, $C_{18 : 0}$ and $C_{16 : 1}$. The major fatty acids of polar lipid were $C_{16 : 0}$, $C_{18 : 2}$, $C_{20 : 5}$ and $C_{22 : 6}$. In total lipid fractionation, saturated acid contents were high in all (SA>MA> PA), in neutral lipid fractionation, menoenoic acid contents were high in all (MA > SA> PA), and in polar lipid fractionation, saturated acid con-tents were high in all (SA> PA> MA).

Changes in lipid and Fatty Acid Composition in Korean Native Meju during Fermentation (한국 재래식 메주 발효과정에 있어서 지질 및 지방산 조성의 변화)

  • Changes in lipid and fatty acid composition in Korean native Meju were investigated at one week interval over 6 weeks of fermentation. For the systematically, salicic acid column chromatography and gas liquid chromatography were used. Following results were obtained. 1. The lipid fraction obtained from the soared soybean and the cooked soybean were mainly composed of $72.47%{\sim}92.35%$ of neutral lipid, phospholipid and glycolipid were 4.64% and 4.88%, respectively. During fermentation period, lipid content decreased to 80.59%, but glycolipid and phospholipid contents increased. 2. The triglyceride contents of nonpolar lipids prepared from the cooked soybean and the soaked sobean was 89.66% and 87.83% respectively. Free fatty acid, diglyceride and sterol contents increased during fermentation, whereas triglyceride content decreased. 3. Lipids extracted from the soaked soybean and the cooked soybean were composed of 54.58% linoleic acid, $22{\sim}20%$ oleic acid, $10{\sim}12%$ palmitic acid and $3{\sim}5%$ stearic acid. 4. During the Korean native Meju fermentation in palmitic acid decreased from the second week and stearic acid through $3rd{\sim}4th$ week. Oleic acid and linoleic acid content decreased gradually, but linolenic acid content increased. 5. During the fermentation, myristic acid content of glycolipid fraction increased. Lipase activity reached to the maximum at the 3rd week.

A Study on the Lipid Components of Silkworn Pupae (잠용의 지질성분(脂質成分)에 관(關)한 연구(硏究))

  • To study the composition of silkworm pupae oil, lipid of silkworm pupae was separated into two fractions, ether extractable and 85% methanol extractable, and the lipid components of each fraction were analyzed by using silicic acid column, thin layer chromatography and gas chromatography. Silkworm pupae contains 35.4% crude fat (dry-basis) of which consists 34.4% diethyl ether-extract and 0.9% of 85% methanolextract. The diethyl ether-extract contained 96.1% of neutral lipid, 2.9% of glycolipid and 1.0% of phospholipid while methanol-extract was consisted of 47.4% of neutral lipid, 14.6% of glycolipid and 38.1% of phospholipid. The major components of phospholipid were phosphatidyl glycol(41.0%), and phosphatidyl choline(28.2%) and phosphatidyl ethanolamine(21.2%) in the diethyl ether-extract and phosphatidyl glycol(48.4%), phosphatidyl inositol(22.8%) and phosphatidyl choline(17.9%) in the methanol-extract. The major fatty acids of the total lipid were oleic acid(33.5%), linolenic acid(31.0%) and palmitic acid(23.1%).

A Network Pharmacology-based Study to Explore the Potential Mechanism of Artemisia capillaris Thunb. for Psoriasis Vulgaris (네트워크 약리학을 활용한 심상성 건선에 대한 인진호(茵蔯蒿)의 잠재적 작용 기전 탐색 연구)

  • Objectives : The purpose of this study is to investigate the potential mechanism of Artemisia capillaris Thunb. for psoriasis vulgaris. Methods : We conducted the network pharmacological analysis. It contains the process that search the compounds of the herb, the target proteins of the compounds, related genes of psoariasis vulgaris and the pathway/terms of the common protein lists between psoriasis vulgaris and Artemisia capillaris Thunb.. Results : 13 compounds and 30 protein targets of Artemisia Capillaris Herba were searched. And 997 psoriasis-related genes were searched. The common proteins were 11, and the core genes were 3; AKT1, CASP3, MAPK8. The related pathway/terms of 11 proteins were analyzed. ω-hydroxylase P450 pathway(60%), nitric oxide(NO) biosynthetic process(20%) were resulted. Also, 19 proteins of Artemisia Capillaris Herba were analyzed, and sterol homeostasis(78.95%), sterol biosynthetic process(15.79%), Type 2 diabetes mellitus(5.26%) were resulted. Conclusion : The Artemisia Capillaris Herba can potentially act through the ω-hydroxylase P450 pathway and nitric oxide(NO) biosynthetic process for psoriasis. Also, the metabolism of sterol biosynthesis and homeostasis can be involved in a roundabout way for psoriasis.

Primary study of sterols composition of Rhodiola sachalinensis by using GC/MS (GC/MS를 이용한 고산 홍경천의 스테롤 구성에 대한 초기연구)

  • The steroid compounds in Rhodiola sachalinensis were determined with adsorption column chromatographic purification and GC/MS. Sterols were extracted by sonication and Soxhlet with ethanol and dichloromethane, respectively. The extract was partitioned with chloroform and water using liquid-liquid extraction, and purified with a silica column after the sterols had been converted to the corresponding silyl derivatives with BSTFA. Eighteen free sterols, including $\beta$-sitosterol, stigmasterol and cycloartenol, and nine sterol conjugates were found from Rhodiola sachalinensis by GC/MS. Among them, cholest-5-ene-3-ol, cholesterol, stigmasterol, $\beta$-sitosterol were confirmed and quantified with sterol standards. Most sterols were presented in the chloroform part, with $C_{29}$ being the most abundant group in this sterol group. $\beta$-sitosterol was the most abundant compound with a relative content of 45.94% followed by ergost-7-ene-3-ol (11.33%), 4,14-dimethyl-ergosta-8,24(28)-diene-3-ol (7.07%), stigmasterol (6.09%), cycloartenol (5.43%) and 4-methyl-cholest-5-ene-3-ol (5.39%).

  • Continuing to previous report, seven compounds were isolated from the aerial parts of Ajuga multiflora. The structures of them were established as Di-2-ethylhexyl phthalate (1), ursolic acid (2), sterol glucoside (3), 20-hydroxyecdysone (4), makisterone A (5). cyasterone (6) and apigenin 7-glucuronide (7), respectively.
