Annual reproductive cycle of the file fish, Stephanolepis cirrhifer (Temminck et Schlegel), was histologically investigated. Samples were collected monthly in the coastal waters of Chungmun, south of Cheju Island, Korea from July 1997 to June 1999. In males and females of S. cirrhifer, the values of GSI (gonadosomatic index) were similar to the changes of water temperature and day length. GSI values in males and females were reached the maximum in June. Reproductive cycle could be divided into the following successive stages: in females, growing stage (February to May), mature stage (May to June), spawning stage (June to August), degenerative and resting stage (September to December), and in males, growing stage (February to April), mature stage (May to June), spent stage (June to August), degenerative and resting stage (September to January), respectively. Throughout histological observation of the ovary in the spawning period, the species belong to an asynchronous and multiple spawner.
Genetic diversities, population genetic structures and demographic histories of the thread-sail filefish Stephanolepis cirrhifer were investigated by nucleotide sequencing of 336 base pairs of the mitochondrial DNA (mtDNA) control region in 111 individuals collected from six populations in Korean coastal waters. A total of 70 haplotypes were defined by 58 variable nucleotide sites. The neighbor-joining tree of the 70 haplotypes was shallow and did not provide evidence of geographical associations. Expansion of S. cirrhifer populations began approximate 51,000 to 102,000 years before present, correlating with the period of sea level rise since the late Pleistocene glacial maximum. High levels of haplotype diversities ($0.974{\pm}0.029$ to $1.000{\pm}0.076$) and nucleotide diversities (0.014 to 0.019), and low levels of genetic differentiation among populations inferred from pairwise population FST values (-0.007 to 0.107), support an expansion of the S. cirrhifer population. Hierarchical analysis of molecular variance (AMOVA) revealed weak but significant genetic structures among three groups ($F_{CT}$ = 0.028, p<0.05), and no genetic variation within groups (0.53%; $F_{SC}$ = 0.005, p = 0.23). These results may help establish appropriate fishery management strategies for stocks of S. cirrhifer and related species.
An epidemic was occurred in Stephanolepis cirrhifer during acclimation in laboratory tanks (water temperature was about $17^{\circ}C$). Diseased fish showed an unique external sign, large cloudy skin surface. We investigated the cause of the disease, and isolated one pure cultured bacterium, that was identified as Vibrio anguillarum by biochemical analysis and sequence analysis of the l6S rRNA and recA genes. During the outbreak, about 30% of S. cirrhifer showed the large cloudy skin surface, and approximately 10% of the stocked fish died. Fish were effectively cured by treatment with oxolinic acid.
The maturation and spawning of filefish, Stephanolepis cirrhifer were investigated using samples randomly collected in the East Sea of Korea from August 2009 to July 2010. The total length at 50% group maturity of filefish was estimated to be 11.7 cm. Based on monthly changes in maturity stages and gonadosomatic index (GSI), the spawning period was estimated to be from June to September, and the peak spawning from August to September. The fecundity (F) ranged from 693,981 eggs to 7,383,429 eggs at 23.6 cm between minimum and maximum total length (TL) 13.3 cm and 23.6 cm respectively. The relationship between TL and F was expressed as F=$12.19TL^{4.208}$ ($R^2$=0.727). The sex ratio of male to female was 1 to 0.9.
The genetic diversity and population genetic structure of thread-sail filefish, Stephanolepis cirrhifer (Temminck & Schlegel), were examined with a nucleotide sequence analysis of a 495bp fragment of the 5'-end of the cytochrome b gene in 113 fish collected from five populations from the south and east coasts of the Korean Peninsula. Seventeen variable nucleotide sites and 16 haplotypes were defined. The observed haplotypes had a shallow haplotype genealogy and no geographical association. Most of the populations had high haplotype diversity and low nucleotide diversity, and significant negative values for Fu's $F_S$, suggesting rapid, recent population growth from an ancestral population and sudden population expansion. The estimated pairwise fixation indices ($F_{ST}$) indicate that substantial gene flow occurs among these populations. Thread-sail filefish in the South Sea of Korea and East Sea Korean populations forms a single panmictic population. Thus, thread-sail filefish in these areas should be treated as one management unit.

The feeding sounds of three fishes, Lateolabrax japonicus, stephanolepis cirrhifer and Fugu vermicularis were recorded in the tank, and the frequency and the sound pressure level were analyzed. The recorded sounds were emitted again into the tank and the response of fishes were observed. The results obtained are summarized as follows: 1) The frequency and the sound pressure level of the feeding sound of Lateolabrax japonicus, Stephanolepis cirrhifer, and Fugu vermicularis are 80~350 Hz and 250~500 Hz, and 101~103 dB, 106~115 dB and 102~112dB. 2) It was most effective to increase the sound pressure level as much as dB that make the fishes respond positive reaction to the feeding sound. 3) Lateolabrax japonicus and stephanolepis cirrhifer showed positive response and Fugu vermicularis showed little to the emitted feeding sound. 4) The fishes showed positive response to the sound until 5 minutes and then showed negative response, as the sound emitting succeed 10 minutes.
A total of 43 species, 493 individuals, and 89,367.1 g of fishes were collected by a gill net in the coastal water off Goeje Island. Samples were collected by bimonthly from February to October, 1996. The dominant species were Stephanolepis cirrhifer, Ditrema temmincki, Hexagrammos otakii, Limanda yokohamae, Sebastes schlegeli, Paralichthys olivaceus, which accounted for 61.7% of the total numbers and 66.3% of biomass of fish collected. Fishes collected in the study area were mainly consisted of coastal species, but a few fishes were migratory species including Stephanolepis cirrhifer, Pleuroniohthys cornutus, Paralichthys oliuaceus, Thamnaconus modestus, Pagrus major, Engraulis japonicus. The number of species and abundance showed a peak in October, and low in February. Monthly species diversity indices ranged between 1.99 and 2.81, and high in June.

The relationship between width of line target and distance at the limit of discrimination was examined by means of the behavioral method, for filefish Stephanolepis cirrhifer from 11 to 15cm body length. Target distance was distance from beginning of partition board to target plate, and was varied from 50cm to 200cm. The target plate was made of white acrylic resin with a vertical black line in the center. The width of line target was varied from 0.2mm to 8.0mm. Fish were trained to respond to a line target and the width of line target reduced until the minimum width required to elicit a response was established. Rate of success was expressed as the percentage of target choices in 90 trials. The line acuity of filefish was found to be 0.58 at a target distance of 50cm. The rate of success decreased slowly as line target width decreased from 8.0mm to 1.5mm, and decreased suddenly for target widths less than about 1.5mm. The width of the line target D(mm) at the limit of discrimination was shown to be an exponential function of the target distance L(cm) as follows : D=exp(9.947$\times$$10^-3$.L+0.146)

To determine the predation rate of threadsail filefish, we conducted feeding experiment of threadsail filefish, Stephanolepis cirrhifer on different life stages (medusae, ephyrae, and polyps) of Aurelia aurita s.l. in the laboratory. All size groups of threadsail filefish actively preyed on moon jellyfish Aurelia aurita s.l. and predation rate was proportional to the wet weight of threadsail filefish. Average wet weight 0.5, 1, 2.2 g of threadsail filefish preyed on average $0.77{\pm}0.51g\;fish^{-1}h^{-1}$ of medusae. Average wet weight 150 and 344 g of threadsail filefish preyed on $9.26{\pm}1.61$ and $15.89{\pm}3.67g\;fish^{-1}h^{-1}$ of medusae respectively. In ephyrae and polyps, average wet weight 3.7 g of juvenile of threadsail filefish preyed on 100 ephyrae within 10 minutes and average wet weight 2.2 g of juvenile of threadsail filefish preyed on $5.73{\pm}0.59$ polyps $g^{-1}$ of fish $h^{-1}$. Whereas adults of threadsail filefish did not prey on polyps. In feeding preference experiments to elucidate whether threadsail filefish prey on moon jellyfish in the sea, adults of threadsail filefish preyed on mostly oysters (ca. 60%) and polychaetes (ca. 80%). Medusae were consumed only 2% of total. However, juvenile of threadsail filefish preyed on ephyrae and polyps as well as mysid shrimps. Besides, polyps were consistently consumed during the experiment. Although juvenile of threadsail filefish consumed all kinds of prey, they ceased feeding at temperatures below $11^{\circ}C$, which suggests that possibility of predation on ephyrae may be very low in the sea because ephyrae are released at low temperatures below $11^{\circ}C$ from March to April. Whereas polyps are inhabitable in all seasons in Korean coastal waters, juvenile of threadsail filefish may prey on polyps in the sea. These results suggest that removing moon jellyfish at polyp stage by using juvenile of threadsail filefish is the most effective way for extermination of moon jellyfish.
This study investigated the antibacterial effects of BioZone atmospheric plasma (AP) against Bacillus cereus (F4810/72) and Staphylococcus aureus (ATCC 6538) as the major foodborne bacteria on the surface of dried filefish (Stephanolepis cirrhifer) fillets. The fillets were experimentally contaminated with 7-8 log CFU/mL of B. cereus or S. aureus using a spot inoculation method. Bacterial counts were measured by standard plate method on tryptic soy agar, and were significantly reduced with the increase in the treatment time (1, 3, 5 or 20 min) of AP on the fillets (p < 0.05). The reductions of the pathogens by AP treatment ranged from 0.9 to 2.93 logCFU/g for B. cereus and from 1.04 to 2.55 logCFU/g for S. aureus. A reduction of >1-logCFU/g for B. cereus and S. aureus was observed on the fillets treated with AP for >3 min. The differences in color on the Hunter scale (L=light vs. dark, a=red vs. green, b=yellow vs. blue) of the fillets were not significantly different between the nontreated (control) and AP-treated fillets (p>0.05). This study suggested that 3 min of AP could be effective in reducing >90% of the bacteria without causing any concomitant changes in the color of the fillets.
