Effects of Foliar-sprayed Diniconazole on Contents of Endogenous Gibberellic Acids and Abscisic Acid in Lilium davuricum (Diniconazole 엽면살포가 날개하늘나리의 내생 GA 및 ABA 함량에 미치는 영향)

  • Plant growth retardants reduce the plant height by inhibiting stem elongation in Lilium davuricum. To investigate the plant hormones related to stem elongation, we sprayed 50 $mg{\cdot}L^{-1}$ diniconazole to young plants of L. davuricum and quantified the contents of endogenous gibberellic acids (GA) and abscisic acid (ABA). In GA biosynthesis, L. davuricum had not only the early C-13 hydroxylation ($GA_{19}{\rightarrow}GA_{20}{\rightarrow}GA_1$) pathway resulting in $GA_1$ as the active form but also the non C-13 hydroxylation (NCH, $GA_{12}{\rightarrow}GA_{24}{\rightarrow}GA_9{\rightarrow}GA_4$) with $GA_4$ as the active form. However, the main pathway was NCH because $GA_4$ concentration of 55 $ng{\cdot}g^{-1}$ dry wt was much higher than $GA_1$ content of 0.23 $ng{\cdot}g^{-1}$ dry wt in control plant. Diniconazole inhibited GA biosynthesis through NCH pathway from its early stage. $GA_{12}$ content decreased by diniconazole up to 6% level of that of control and this effect continued to $GA_4$. Diniconazole reduced $GA_{12}$ content by 12.7 $ng{\cdot}g^{-1}$ dry wt, whereas that of control plant was 213.8 $ng{\cdot}g^{-1}$ dry wt. ABA content decreased up to one third of control by diniconazole application. From the contents of endogenous $GA_4$, $GA_1$, and ABA in this study, we could conclude that diniconazole reduces the plant height by inhibiting $GA_4$ biosynthesis in L. davuricum.

Developmental Stage and Temperature Influence Elongation Response of Petiole to Low Irradiance in Cyclamen persicum (저광도에 대한 시클라멘 엽병의 발육 단계 및 온도 조건별 신장 반응)

  • Reduced irradiance promotes shoot elongation depending on developmental stage and environmental factors and decreases plant quality in $Cyclamen$ $persicum$ Mill. To determine the petiole elongation responses to low irradiance, 'Metis Scarlet Red' cyclamen at different developmental stages [juvenile (5-6 unfolded leaves), transitional (1-3 visible flower buds), or mature (1-3 elongating peduncles)] was grown in growth modules at 60 (low light, LL) or 240 (high light, HL) ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD within the growth chambers at different temperatures [16/12 (low temperature, LT), 22/18 (medium temperature, MT), or 28/$24^{\circ}C$ (high temperature, HT) (day/night)]. In Experiment I, juvenile plants were either kept in an LL or HL module during the entire treatment of 4 weeks or were transferred to the other module at 1, 2, or 3 weeks after treatment in an MT chamber. In Experiment II, juvenile, transitional, or mature plants were moved to the HL module at 0, 3, 6, 9, or 12 days after being placed in the LL module at the MT chamber and grown for 21 days. In Experiment III, transitional plants were moved to the HL module at 0, 3, 6, 9, or 12 days after being placed in the LL module at the LT, MT, or HT chambers. As the exposure duration to LL increased from 0 to 4 weeks or from 0 to 12 days, petiole length and plant height increased at all temperatures and developmental stages. In Experiment I, the exposure to LL during the latter period, rather than the early period, increased elongation rate. In Experiment II, petiole elongation in transitional plants was more sensitive to LL than juvenile or mature plants during the early period of the treatment for 12 days. In Experiment III, petiole length increased with increasing temperature and exposure duration to LL. Petiole elongation rate at HT increased rapidly from the beginning of LL exposure as compared to LT. Increase of $6^{\circ}C$ in temperature had the similar effect to LL exposure for 3 days in petiole elongation. To conclude, transitional cyclamen under higher temperatures responds more immediately to low irradiance and elongates its petioles.

Growth and Flowering of Cut Spray Chrysanthemum 'Charming Eye' and 'Pink Pride' by Daminozide (스프레이 절화국화 '챠밍아이'와 '핑크프라이드'의 daminozide에 의한 생육 및 개화)

  • This study was conducted to improve commercial quality of cut spray chrysanthemums 'Charming Eye' and 'Pink Pride' bred in Korea by foliar application of daminozide, suppressing excessive elongation of peduncle caused by high temperature in greenhouse. Applications were made at two floral-bud-developmental stages and concentrations used were 0, 500, 1000, and $2000mg{\cdot}L^{-1}$. As for 'Charming Eye', cut flower length, peduncle diameter, stem diameter, flower bud diameter, and the number of flower buds did not show any significant difference among all treatments including control. However, the suppressing effect of peduncle elongation, widening angle of flower cluster arrangement on apical part, and increasing parallel flower buds in stage I showed better than those in stage II regardless of daminozide concentration. As for 'Pink Pride', cut flower length, peduncle diameter, and stem diameter did not show any significant difference among all treatments including control but angle of flower cluster on apical part increased compared to control as daminozide was sprayed at stage I and II except $2,000mg{\cdot}L^{-1}$ daminozide-sprayed at stage II. The number of flower buds and flower bud diameter showed the greatest increment through $1,000mg{\cdot}L^{-1}$ daminozide-sprayed at stage I and did the least values as sprayed with $2,000mg{\cdot}L^{-1}$ daminozide at stage II. Daminozide also gradually reduced peduncle length in a concentration-dependant manner but elongation of peduncle foliar-sprayed at stage I showed the more suppressing effect than that at stage II. Increasing the number of parallel flower buds showed the best results when sprayed with $2,000mg{\cdot}L^{-1}$ at stage I. In conclusion, we recommended that foliar spraying with $500-1,000mg{\cdot}L^{-1}$ daminozide at stage I and foliar spraying with $1,000-2,000mg{\cdot}L^{-1}$ daminozide at stage I improved cut flower quality of 'Channing Eye' and 'Pink Pride', respectively.

  • A visualization study on the effect of forcing amplitude in tone-excited jet diffusion flames has been conducted. Visualization techniques are employed using optical schemes. which are a light scattering photography. Flame stability curve is attained according to Reynolds number and forcing amplitude at a fuel tube resonant frequency. Flame behavior is globally grouped into two from attached flame to blown-out flame according to forcing amplitude: one sticks the tradition flame behavior which has been observed in general jet diffusion flames and the other shows a variety of flame modes such as the flame of a feeble forcing amplitude where traditionally well-organized vortex motion evolves, a fat flame. an elongated flame. and an in-burning flame. Particular attention is focused on an elongation flame. which is associated with a turnabout phenomenon of vortex motion and on a reversal of the direction of vortex roll-up. It is found that the flame length with forcing amplitude is the direct outcome of the evolution process of the formed inner flow structure. Especially the negative part of the acoustic cycle under the influence of a strong negative pressure gradient causes the shapes of the fuel stem and fuel branch part and even the direction of vortex roll-up to dramatically change.

Virus Free Stock Production by In vitro Stem Cutting of Shoot Tip Cultures of Grapes (포도 경정배양에서 얻은 유묘의 기내삽목에 의한 무병묘 생산)

  • The experiment was conducted to know the effect of plant growth regulators on axillary bud elongation from in vitro stem cutting and the possibility of virus-free stock production. Axillary buds were well elongated in 3/4 strength MS medium supplemented with 0.1 or 0.5 mg/1 BA and 0.05 mg/1 NAA. Transferred plantlets could be established well in vermiculite and peat moss mixture (3:1, v/v) compare to other mixtures. In virus indexing, all the varieties of mother plants were infected by GLRV Ⅲ. Infected percentages of the three varieties were ranged from 30% to 75%. But negative response was revealed against the other species of virus, GLRV Ⅰ, GFLV and ArMV. Plantlet of 'Schuyler' and 'Muscat of Alexandria', which were cultured in vitro, showed positive response against GLRV Ⅲ and infected percentage of the former was 37.5% but the latter, 12.5%. On the other hand, that of 'Campbell Early' negativiely responded against all the species of virus indexed.

  • The objective of this paper is to compare and summarize the procedure and effectiveness of some bioassay systems and to point out ways to obtain reliable results from each bioassay. Detailed C:escriptions were given for those widely-adapted bioassay methods, such as mungbean rooting (auxin), Avena first internode straight growth (auxin), dwarf rice growth (gibberellin), dwarf pea epicotyl elongation (gibberellin), radish cotyledon expansion test (cytokinin), and tobacco stem pith callus growth (cytokinin), and the effects of various plant growth regulators including some recently introduced growth retardants (Paclobutrazol, Uniconazol, etc.) were also summarized.

In Vitro Shoot Tip Culture of Haemaria discolor (Haemaria discolor 경정의 기내배양)

  • The experiments were conducted to obtain culture conditions on optimal cultural conditions for propagation of a topical orchid, Haemaria discolor, through shoot tip cultrue in vitro. Survival percentage of shoot tip explant, in initial culture media was higher in H$_3$P$_4$ medium than in Murashige and Skoog's medium. Explants, cultured in H$_3$P$_4$ medium containing 1.0 mg/L kinetin, showed more shoot elongation when compared with those on other media. When the distal part of stem (pseudobulb) was longitudinally sectioned into half and cultured in H3P4 medium containing 0.1 mg/L 2ip, 0.1, or 1.0 mg/L kinetin, shoots from axillary bud were much more elongated under light condition.

Comparison of Agronomic Traits According to Plant Types before Wintering in Malting Barley Lines (맥주맥의 월동전 초형에 따른 주요 형질의 비교)

  • So as to compare the differences of major agronomic traits according to fall plant types, two cultivars and eight lines which were derived from the same pedigree, were planted and analyzed. The heading and mature times, and culum length of erect plant types were earlier or longer, but the spike number per ㎡ and yield were smaller than the those of prostrate types. Also, 1000 grain weith, assortment rate and light penetration rate in erect types were greater than in prostrate types. The heading time was positively correlated with mature time and yield, but negatively correlated with culum length, 1000 grain weight, assortment rate and light penetration rate. Yield was negatively correlated with light penetration rate. If the lines with prostrate types in fall season, fast regrowth after wintering and rapid elongation of stem were bred and selected, it will be of great benefit in the area requiring sever winter hardiness.

Effect of Phytohormones on Multiple Shoot Bud Induction in cv. NARI-6 of Safflower (Carthamus tinctorius L.)

  • In the present study, in vitro multiple shoot induction was achieved from cotyledonary node and stem nodal explants of cv. NARI-6 of safflower (Carthamus tinctorius L.). Among various growth regulators tested, MS salts and B5 vitamins supplemented with BA (6-Benzy-laminopurine) $17.76\;{\mu}M$ and KN (Kinetin) $6.96\;{\mu}M$ phytohormonal combination was found to be the most effective in initiating numerous shoot buds after 30 days of culture than BA ($4.44-44.39\;{\mu}M$) or KN ($2.32-46.40\;{\mu}M$) alone in the medium. In addition, 0.8% (w/v) agar (Hi-media) and 3.0% sucrose (w/v) was the optimum level for the formation of adventitious shoots. Further results showed the maximum shoot elongation occurred on MS medium with BA ($8.88\;{\mu}M$) and $GA_3$($11.56\;{\mu}M$) combinations. Efficient rooting occurred on quarter strength MS medium with NAA $10.74\;{\mu}M$. The regenerated plantlets were acclimatized and successfully transferred to the field.

Morphogenetic Responses according to Media and Growth Regulators in vitro Culture of Eight Populus spp. (배지(培地) 및 생장(生長)호르몬에 따른 기내배양(器內培養)된 포푸라속(屬) 8개(個) 수종(樹種)의 기관형성반응(器官形成反應))

  • To examine the morphogenetic response, stem segments of 8 Populus spp. and 3 different explants of P. nigra var. italica were cultured on MS (Murashige and Skoog 1962) and WPM (Woody Plant Medium) medium containing various phytohormones. The results obtained were as follows: 1. Shoot regeneration and development from stem segment of 8 Populus spp. showed a quite difference according to the section and the species. All of the species of Leuce and Tacamahaca section did not form adventitious buds, while most of explants showed axillary or dormant bud elongation after 4 weeks. But P. nigra var. italica of Aigeiros section showed a successful adventitious bud formation (mean 5.4 buds per explant). 2. Leaf, petiole, and internode segment of P. nigra var. italica showed a quite differences according to media and ex plants upon the morphogenetic response. Adventitious bud formation from leaf was more abundant and readily initiated on the abaxial side than on the adaxial side. Mean number of 103 adventitious buds per explant was obtained from abaxial side of leaf segment cultured on WPM medium containing $0.2mg/{\ell}$ BAP for 5 weeks. 3. 2,4-D (2,4-dichlorophenoxy acetic acid) supplemented to media appeared to be negative upon the adventitious bud formation of P. nigra var. italica, while it promoted callus formation from all explants. Especially, NAA (${\alpha}$-naphtalene acetic acid) or NAA combination with BAP (6-benzylaminopurine) promoted root regeneration from the all explant of P. nigra var. italica in this study.
