Propagation by Leafy Stem Cuttings Containing Xylem of Populus alba × P. glandulosa Clone Bongwha1

  • The study was conducted to establish a method for the proliferation of hybrid poplar (P. alba × P. glandulosa) clone Bongwha1, an excellent biomass species. It was found that to collect the cuttings of Bonghwa1, it was necessary to use the main stem rather than the axillary branch. Stem growth by green-wood cuttings showed a tendency to decrease as the length of the collected cuttings increased, but the survival rate was low. Therefore, modified leafy stem cutting was attempted to increase the survival rate of the cuttings. In the modified leafy stem cutting method, 4 leaves were included in the cuttings, and especially, cuttings were performed using cuttings containing 2-4 cm xylem parts. Leafy stem cutting increased root growth and the number of stems, as well as the survival rate of hybrid poplar clone Bongwha1 compared to green-wood cuttings. The root growth of the leafy stem cutting poplar was better as there was more xylem part. Using two-year-old nursery stocks, the leafy stem cutting was used to produce about 66 cuttings. This study is expected to contribute to the mass propagation of high-quality nursery stocks.

단기순환림 생산을 위한 삽목 이식기 개발 (Development of Stem-cutting Transplanter for Short-term Rotation Coppice)

  • Since SRC (Short-term Rotation Coppice) such as poplar and willow can be harvested in three years, they are known to be a potential forest biomass as fuel for a power plant. The production system including transplanting and harvesting is, however, necessary to be mechanized because such a biomass should be handled in a massive volumetric size. A pull-type stem-cutting transplanter was developed in the research as the first step to realize the production of SRC. A needle-like transplanting device pushes a stem-cutting into the prepared soil bed by a pneumatic cylinder, and another device firms soil around a stem-cutting transplanted. Since this is an intermittent operation, it was necessary to develop a zero horizontal velocity mechanism which enabled only the transplanting needle part to continue a zero horizontal movement relative to the ground during the transplanting operation even when the tractor kept moving forward. The 2-row transplanter can transplant stem-cuttings at the rate of 6.5 seconds per row without missing a single attempt. The planting depth and distance were well maintained and controlled. Their CVs were between 2.1~3.4% and 0.87~1.7% for the depth and the distance, respectively. Although, the transplanted stem-cuttings tended to lean outward from the back-view and forward from the side view, they were planted within the range of $3^{\circ}$ from the upright position.

  • This paper presents a novel stem-cutting device using a twisted string actuation system combined with the cinch bag-typed gripper proposed in previous research. The suggested cutting device was developed to cut the stem of a tomato using two motors. The relationship between contact time and motor angle was mathematically induced, and the contacting time was verified through the experiments. The contact time has decreased as the offset of each pair of strings at the disk increases. The contact time and its deviation were reduced by increasing the radius of the twisted string bundle, and the motor torque to exert an equivalent cutting force was surged at the same time. The proposed cutting mechanism with 16 strands of twisted string bundle and 40 mm of offset can cut the given tomato stems and stalks, exerting up to 132.4 N of cutting force in 4.6 to 6.5seconds.

Mass Propagation of Plug Seedling using Stem Cutting and Their Tuber Yield in Potato

  • For the mass production of plug seedlings in cultivar ‘Dejima’ potato (Solanum tuberosum L.) the optimal apical cutting diameter for rooting and rapid multiplication of stem cuttings in hydroponics were determined. In addition, the best planting date was predicted to increase tuber yield of plug seedlings at fall cropping in Cheju-Do, Korea. Days to initial rooting decreased as the cutting diameter was reduced. Plant height, leaf number, root length and root weight per plant were favorable as the cutting diameter was small. The ideal cutting diameter was 1-2 mm in this experiment. In the hydroponic cultures, the Japanese standard (JS) nutrient solution was the most effective for multiplication of stem cuttings. It was able to propagate more than 20 times a month from a single mother plant. Viability of plants, which were derived from plug seedlings using stem cuttings, was excellent when transplanted to the field. The number of tubers and tuber yield in both of the plug seedlings and seed potato planting plots were high when planted on 25 August. The number and yield were reduced when planted on 15 August, 5 September and 15 September. The degree of decrease of tuber yield in the plug seedling planting plot however, was lower than that of seed potatoes when the planting date was late. In the case of small tubers (under 30 g), the number of tubers and tuber yield were evidently increased in the seed potato tuber planting plot; the yield of large tuber (over 80g) in the plug seedling planting plot was higher than that of the seed potato. The total tuber yield per plant in the plug seedling planting plot was less than that of the seed potato; therefore, in order to increase tuber yield it was necessary to increase field plant density.

단근, 줄기 절단과 식재 밀도에 따른 음나무(Kalopanax septemlobus) 묘목의 활착 및 생장 특성 (Effects of Root Pruning, Stem Cutting and Planting Density on Survival and Growth Characteristics in Kalopanax septemlobus Seedlings)

  • Kalopanax septemlobus (Thunb. ex Murray) Koidz. is natively distributed in Korea. The importance of this species has been increased not only for high quality timber but for medicinal and edible uses. However, increasing market demand of K. septemlobus with illegal cutting and overexploitation has resulted in its rapid depletion and destruction of natural habitat. This study was conducted to understand the survival rate and growth characteristics of planted K. septemlobus seedlings with treatment of root pruning, stem cutting and planting density. The survival rate and growth of height and root-collar diameter for one- and two-year old seedlings with different planting densities were investigated in the clear-cut area of a Pinus densiflora stand for five years. One-year-old seedlings were treated with or without root pruning and planted with three density levels (5,000 trees $ha^{-1}$, 10,000 trees $ha^{-1}$, and 40,000 trees $ha^{-1}$). Two-year-old seedlings were treated with and without stem cutting and planted with the density of 5,000 trees $ha^{-1}$. The survival rate of one-year-old seedlings with root pruning treatment in the density of 10,000 trees $ha^{-1}$ was 92%, while that without root pruning in the density of 40,000 trees $ha^{-1}$ was 67% after five years. The height of one-year-old seedlings has been significantly affected only by planting density in the $5^{th}$ year. The survival rate of the two-year-old seedlings with stem cutting was 75.5% and greater than control (67.3%) in the $5^{th}$ year but no difference in height was shown between the two treatments from three years after plantation.

감자 'Solara' 경삽묘의 육묘기간에 따른 묘소질 및 수경재배에서의 수량 특성 (Effect of Seedling Quality on the Seedling Raising Period of Stem Cutting and Yield Characteristics of 'Solara' Potatoes in Aeroponics Cultivation)

  • 이 시험은 '솔라라' 품종의 종자생산을 위해 분무경 수경재배에 적합한 묘를 구명하고자 2015년 3월 16일 배양순화묘 등 8처리의 묘를 분무경 베드에 정식하여 정식 후 70, 78일에 각각 생육과 수량특성을 조사하였다. 경장은 경삽 15일묘와 배양순화묘가 작은 편이었고 경삽묘의 육묘일수가 늘어남에 따라 길어지는 경향이었다. 생체중도 경삽 40일묘가 주당 122 g으로 가장 무거웠고 무발근묘와 배양순화묘는 가장 가벼웠다. 1차 복지는 경삽 35일묘에서 주당 12.9개로 가장 많았고 2차 복지는 경삽 35일묘, 배양순화묘, 경삽 30일묘에서 많이 발생하였다. 10주당 총괴경수는 배양순화묘가 145개로 많았으나 3~50 g(상품) 괴경수는 경삽 35일묘가 108개로 가장 많았고, 괴경 무게는 경삽 35일묘가 1,947 g으로 가장 무거웠고 다음으로 경삽 30일묘였다. '솔라라' 품종 수경재배 시에는 경삽한 후 30일~35일(경장 15 cm, 마디 12절) 묘로 정식하는 것이 수량성을 높일 수 있었다.

여주 삽목 시 삽수절단 위치와 온도조건이 발근에 미치는 영향 (The Effect of Cutting Positions and Temperature on the Rooting of Bitter Gourd)

  • 본 실험은 종자 가격이 비싼 여주를 삽목하여 증식하고자 할 때 적절한 줄기 절단위치와 삽목상내 온도를 구명하기 위하여 실험하였다. 시험품종 'NS454'(NS)와 '드레곤'(DR) 품종을 대상으로 삽수 절단 위치를 3번째 잎의 마디를 절단한 처리(I) 와 3번째와 4번째 잎의 중간 부위를 절단한 처리(II)를 하였다. 삽목상내 온도는 18, 23, 28, $35^{\circ}C$ 처리를 하였다. 삽목상내 명기는 16시간유지하였으며, 광합성유효광량자속은 약 $150-200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 정도였으며, 상대습도는 85% 이상 유지시켰다. 그 결과 삽목 후 10일에 생존율은 두 품종 모두 $18^{\circ}C$$23^{\circ}C$ 처리구에서는 삽수의 절단 위치와 상관없이 100%의 생존율을 보였지만, $18^{\circ}C$ 처리구의 경우, 뿌리 발근이 되지 않아 많은 개체가 고사하게 되었다. 뿌리 발근율에 있어서는 $28^{\circ}C$ 처리구가 DR과 NS 품종 모두 삽수의 절단위치와 상관없이 100% 새뿌리가 발생하였고, $23^{\circ}C$ 처리구에서는 DR 품종의 경우 마디를 절단한 처리는 90%, 마디 중간지점을 절단한 처리는 40%이었으며 NS 품종의 경우도 마디를 절단한 처리는 50%, 중간지점을 절단한 처리는 40% 로 마디를 절단한 처리구에서 두 품종 모두 발근율이 높았다. 온도 $35^{\circ}C$ 처리구에서는 발근율이 현저하게 낮아졌으며 NS 품종의 경우는 절단위치에 상관없이 발근이 되지 않았다. 뿌리의 개체당 발근수는 DR 품종의 경우, $28^{\circ}C$ 처리구는 마디 중간지점을 절단한 처리구에서 5.3개로 처리구중에서 가장 많았고 마디를 절단한 처리구는 2.7개이었다. 따라서 여주의 삽목시 절단위치는 마디부위를 절단하거나 마디 중간지점을 절단해도 되지만 마디 중간지점을 절단하는 것이 생존율을 높일 수 있을 것으로 사료되고, 또한 삽목상내 기온을 $28^{\circ}C$ 정도를 유지해 주는 것이 바람직할 것으로 판단된다.

  • 본 실험은 Dendrobium nobile 계통의 교잡종인 'Hamana Lake Dream' ${\times}$ 'No. 55' 유묘의 기내삽목 시 증식 및 생육에 알맞은 배지와 배양조건을 구명하고자 실시하였다. Hyponex배지보다 MS배지에서 생장이 잘되었는데 MS배지에 활성탄을 0.1과 1.0g/L 수준에서 첨가했을 때 줄기수와 엽장, 근수, 생체중이 양호 하였으나 2.0g/L이상에서는 감소하였으나 통계적 유의성은 없었다. MS배지에 agar, sucrose, peptone, gelite 를 농도별로 첨가한 실험에서는 agar 5g/L 첨가 시 줄기 길이와 엽폭, 생체중의 값이 높았고, 절간장과 근수는 sucrose 40g/L을 첨가했을 때 효과적이었다. 또한 줄기수와 엽수는 peptone 1g/L에서 효과적이었으나 gelite는 모든 농도에서 대조구에 비해 저조하였다. 기내삽목 시 배양온도는 $28^{\circ}C$가 알맞았고 동일한 온도조건에서는 MS배지가 Hyponex배지보다 더 효과적이었다. 또한 sucrose 농도별 실험에서는 $28^{\circ}C$에서 40 g/L를 첨가하는 것이 좋았다.

돌나물의 가을 노지삽목에서 삽수부위 및 삽식방법에 따른 생육특성 (Growth Characteristics as Influenced by Cutting Site and Planting Method in Autumn Field Cutting of Sedum sarmentosum)

  • 돌나물의 노지 가을삽식을 위하여, 줄기의 삽수 부위(상부, 중부, 하부) 및 삽식 방법(점파, 조파 산파)에 따른 월동전후 생육특성을 조사하였다. 삽수부위에 따른 삽목후 40일째의 월동전 생육은 상부 삽수에서 초장, 근장, 줄기수, 지경수, 지상부 생체중 및 건물중 등에서 유의하게 높았으며, 월동후 개화전(May 10th) 생육은 개체당 줄기수, $m^2$당 지상부 생체중 및 건물중에서 하부 삽수보다 유의한 증가를 보였다. 삽식 방법에 따른 삽목후 40일째의 월동전 생육은 점파($10{\times}10cm$)가 조파나 산파보다 양호하였고, $m^2$당 지상부 생체중 및 건물중도 점파>산파>조파 순으로 높았다. 월동후 생육(May 10th)은 개체당 줄기수, $m^2$당 생체중 및 건물중에서 점파가 조파보다 유의하게 증가하였다. 돌나물의 가을 노지 삽식은 줄기 상부의 삽수를 이용하는 것이 월동 전후 생육 및 수량성에 유리하였으며, 삽식방법은 점파에서의 수량성이 가장 양호하였으나, 생력화를 위해서는 월동후 생체중에서 점파와 유의한 차이를 보이지 않은 혼파방법이 바람직할 것으로 기대되었다.

Effects of Culture Type and Inoculation Quantity in Bioreactor on Production of Potato Plantlets

  • Potato (Solamum tuberosum 'Dejima') plantlets were investigated on culture type and initial quantity of inoculation in bioreactor and survival rate by hydroponics for mass production. rode stems (1 to 1.5cm in length) of potato plantlets multiplied in vitro were grown for 3 weeks in liquid Murashige and Skoog (MS) medium with sucrose $30 g\; L^{-1}$. When plantlets (80-node inoculation) were raised in 10L balloon type bubble (BB) bioreactor, the healthiest growth of plantlets was obtained from explants cultured in ebb & flow culture with medium supplied periodically 12 times per day. The suitable inoculation quantity of 20L BB bioreactor was 120 pieces of stem segments (mean 2.2g fresh weight) in ebb & flow culture. Number of nodal shoot was eight on the average. In controlled culture room, survival rate of plantlets at 7 days after stem cutting was above $70\%$ when they were acclimatized by hydroponics grown in deep flow and solid medium culture. The highest survival rate of the stem cutting plantlets was in nutrient solution adjusted to EC $1.4dS{\cdot}m^{-1}$. Stem cutting plantlets through one culture could be obtained $670\~900$, when plantlets were grown in ebb & flow culture during 3 weeks using a 20L bioreactor with initial 120 pieces of nodal segments. 11 is possible In do mass production of seedlings cultured in bioreactor and hydroponics.
