Comparative Histomorphology on Low and High Rubber Yielding Guayule Varieties of India

  • A preliminary comparative histodimensional studies, such as wood to bark ratio, total area of parenchyma cells and bast fibers, height and diameter of vascular rays, pith diameter and phenological parameters, height of the plant, perimeter of the stem, dry weight of the stem and leaves have been investigated for the low and high rubber bearing Guayule plants growing in same and different climatic regions of India. A plausible correlation is found between the percentage of rubber content and anatomical characters in the investigated varieties.

  • Morphological and anatomical evaluation of grafted P. merkusii have been undertaken to obtain the information about compatible and incompatible symptoms of 18 years old grafts based on morphological observation and microscopic analysis. Samples of compatible and incompatible grafts were obtained from previous research conducted by the Silviculture Departement Team in 1994. Result showed that compatible grafts have normal stem form and secondary growth (diameter growth), but some abnormality symptoms like undulated pattern of annual growth rings, phloem thickening and abnormality resin ducts in inner and middle parts of the union area occurred. Incompatible ones showed abnormality of the stem form, cortex-bark necrosis and swelling in the union area. Microscopic observation showed abnormality of all parts of the union, undulated pattern of annual growth rings, phloem thickening, abnormal resin ducts, low numbers and discontinuity of vascular elements in the union area.

  • Two medicinal plant of Sundarbans mangrove forest has been tested for the evaluation of growth inhibitory and antibacterial activity. The methanol extract of Xylocarpus granatum stem bark showed potent wheat rootlet ($IC_{50}=0.01{\mu}g/ml$) and shoot ($IC_{50}=0.23{\mu}g/ml$) growth inhibitory activity in a concentration related manner. The growth inhibitory activity was markedly decreased in residual methanol extract. The methanol extract showed antibacterial activity (MIC > 3 mg/ml) against Bacillus subtilis, Staphylococcus aureous and Proteus vulgaris. The residual methanol extract did not show any antibacterial activity. The results suggest the bioactive principle(s) of Xylocarpus granatum may be relatively non polar compound(s). The methanol extract and residual methanol extract of Sarcolobus globosus stem showed poor wheat rootlet and shoot growth inhibitory activity and no antibacterial activity.

Allometric equations, stem density and biomass expansion factors for Cryptomeria japonica in Mount Halla, Jeju Island, Korea

  • This study was conducted to develop allometric equations and to determine the stem density and biomass expansion factor (BEF) for the estimation of the aboveground and belowground biomass of Cryptomeria japonica in Jeju Island, Korea. A total of 18 trees were harvested from the 40-year-old C. japonica stands in Hannam experimental forest, Jeju Island. The mean biomass of the C. japonica was $50.4Mg\;ha^{-1}$ in stem wood, $23.1Mg\;ha^{-1}$ in root, $9.6Mg\;ha^{-1}$ in branch, $4.6Mg\;ha^{-1}$ in needle and $4.3Mg\;ha^{-1}$ in stem bark. The diameter at breast height (DBH) was selected as independent variable for the development of allometric equations. To evaluate the performance of these equations, coefficient of determination ($R^2$) and root mean square error (RMSE) were used and results of the evaluation showed that $R^2$ ranged from 71% (root biomass equation) to 96% (aboveground biomass equation) and the RMSE ranged from 0.10 (aboveground biomass equation) to 0.33 (root biomass equation). The mean stem density of C. japonica was $0.37g\;cm^{-3}$ and the mean aboveground BEF was $1.28g\;g^{-1}$. Furthermore, the ratio of the root biomass to aboveground biomass was 0.32.

Inhibitory Effect of Rosmarinic acid Extrcted from Euonymus Alatus on Cyclooxygenase-2

  • Objectives and methods : Previous mechanistic studies suggest the cyclooxygenase-2 (COX-2) inhibitors represent the good candidates against tumor progression. MeOH extract of the stem barks of Euonymus alatus induced the strong inhibition of COX-2. A phenolic compound responsible for the anti- COX-2 known to involve in tumor adhesion and invasion has been studied through the methanol extracts. The compound, rosmarinic acid (ROS-A) was an ester of caffeic acid and 3,4-dihydroxyphenyllactic acid. ROS-A showed a strong inhibitory effect of COX-2 activity in a concentration-dependent manner. Then we have measured the IL-1${\beta}$, IL-6 and TNF-${\alpha}$ production related the immune regulation, induction of inflammatory related genes. Results and Conclusions :Hep3B cells produce proinflammatory cytokines of IL-1${\beta}$, IL-6 and TNF-${\alpha}$ while ROS A inhibited the cytokines production. Since IL-1${\beta}$, IL-6 and TNF-${\alpha}$ need the transcription factors such as nuclear factor- ${\kappa}$B (NF-${\kappa}$B) and activated protein-1 (AP-1), we measured the transcription factors. ROS-A inhibited the activation of p65, p50, c-Rel subunits of NF-${\kappa}$B and AP-1 transcription factors. These findings indicate that ROS A from the stem bark of E. alatus inhibits proliferation in metastatic cancer cells. It was suggested that stem barks of E. alatus could be suitable for anti-cancer drugs.

Honokiol Suppresses Renal Cancer Cells' Metastasis via Dual-Blocking Epithelial-Mesenchymal Transition and Cancer Stem Cell Properties through Modulating miR-141/ZEB2 Signaling

  • Renal cell carcinoma (RCC) is associated with a high frequency of metastasis and only few therapies substantially prolong survival. Honokiol, isolated from Magnolia spp. bark, has been shown to exhibit pleiotropic anticancer effects in many cancer types. However, whether honokiol could suppress RCC metastasis has not been fully elucidated. In the present study, we found that honokiol suppressed renal cancer cells' metastasis via dual-blocking epithelial-mesenchymal transition (EMT) and cancer stem cell (CSC) properties. In addition, honokiol inhibited tumor growth in vivo. It was found that honokiol could upregulate miR-141, which targeted ZEB2 and modulated ZEB2 expression. Honokiol reversed EMT and suppressed CSC properties partly through the miR-141/ZEB2 axis. Our study suggested that honokiol may be a suitable therapeutic strategy for RCC treatment.

Anti-inflammatory and PPAR Subtypes Transactivational Activities of Phenolics and Lignans from the Stem Bark of Kalopanax pictus

  • A new compound, kalopanaxin F (3), and 11 known compounds (1, 2, 4-12), were isolated from the stem bark of Kalopanax pictus. Their structures were elucidated on the basis of chemical and spectroscopic methods. Five of the compounds (2, 3, 5, 6, and 12) significantly inhibited $TNF{\alpha}$-induced NF-${\kappa}B$ transcriptional activity in HepG2 cells in a dose-dependent manner, with $IC_{50}$ values ranging from 6.2 to 9.1 ${\mu}M$. Furthermore, the transcriptional inhibitory function of these compounds was confirmed based on decreases in COX-2 and iNOS gene expression in HepG2 cells. Compounds 3-7, 9, and 12 significantly activated the transcriptional activity of PPARs dose-dependently, with $EC_{50}$ values ranging from 4.1-$12.7{\mu}M$. Compounds 4 and 5 exhibited $PPAR{\alpha}$, $PPAR{\gamma}$, and $PPAR{\beta}({\delta})$ transactivational activities in a dose-dependent manner, with $EC_{50}$ values of 16.0 and 17.0, 8.7 and 16.5, 26.2 and 26.3 ${\mu}M$, respectively.

The Protective and Inhibitory Effect of Antioxidants Found in Broussonetia kazinoki Siebold against Oxidative DNA Damage

  • Oxidative DNA damage negatively affects humans and the research is currently ongoing to find ways to reduce oxidative stress. Oxidative stress has been identified as a key factor in triggering various diseases. Thus, its alleviation is important for human health. Broussonetia kazinoki (B. kazinoki) has been used in traditional Korean medicine as a dermatological therapy to treat burns, pruritus, and acne. B. kazinoki is generally segregated into peeled root (PR), root bark (RB), peeled stem (PS), and stem bark (SB). To assess these components for their antioxidant activity and protection against DNA damage, their ethyl acetate fractions were examined by 1,1-diphenyl-2-picryl hydrazyl (DPPH) and 2,2'-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging assay. As a result of confirming the expression of factors involved in attenuating DNA damage, the protective effect of SB on oxidative stress suppressed the expression of p-p53 and γ-H2AX. Additionally, the levels of p53 and H2AX mRNA were significantly downregulated. In conclusion, these results indicated that the SB component of B. kazinoki had the potential to be used as an effective natural antioxidant compared to the other parts of the plant.

아까시재목버섯이 옻나무 껍질의 urushiol 제거에 미치는 영향 (Effect of Perenniporia fraxinea on Eliminating Urushiol from Rhus verniciflua Stokes Stem Bark)

  • 본 연구에서는 urushiol 제거에 적합한 장수버섯 종균용 원료에 대하여 평가하였다. 세 가지 곡물조, 현미, 통밀에서의 장수버섯 균사체 증식속도는 각각 4.92±0.05, 2.20±0.03, 1.93±0.03 mm/day 이었다. 장수버섯이 배양된 곡물종균의 laccase의 활성은 각각 0.86±0.02, 0.04±0.01, 0.01±0.00 U/mL 이었다. 증식속도와 효소활성 측면에서 장수버섯 종균용 곡물로는 조를 사용하는 것이 적합하였다. 조에 배양된 장수버섯 종균을 옻나무에 배양하였더니 3일차에 urushiol이 최대 86.6% 감소하였고 7일차에 98.5%까지 감소하였다. Urushiol 제거를 위한 적정 배양기간은 3일이었으며 이때 flavonoid 잔존량은 68%, phenolic 성분의 잔존량은 42% 이었다. 액체종균을 사용하여 배양한 것과 비교했을 때 urushiol을 제거하기 위한 증식기간이 10일에서 3일로 단축되었다.
