Relationship between Entomopathogenic Nematode and Entomopathogenic Fungus, Beauveria brongniartii

  • 곤충병원성 선충의 이용 효율을 높이기 위하여 곤충병원성 선충 상호간 또는 선충과 곤충병원성 곰팡이와의 상호관계를 알아보았다. Steinernema glaseri는 기주의 무게에 따라 성충수에서는 차이가 없었으나 침입태 유충수에서는 차이가 있었다. 선충의 접종 농도에 따른 침입 발육된 성충수는 기주당 20마리 이하의 접종에서는 차이를 보였으나 40마리 이상 1,000마리 접종구에서는 차이가 없었고, 증식된 유충수도 기주당 1,000마리 접종구에서 현저히 적은 것을 제외하고는 차이가 없었다. 동일 기주에서의 선충 상호간 관계에서는 steinernematid 선충에 의한 기주 치사율이 heterorhabditid 선충에 의한 것보다 높았다. 즉, 서로 다른 종의 곤충병원성 선충을 동일 기주에 접종하였을 때, S. carpocapsae 포천에 의한 기주 치사율은 $76.2\pm$4.8%였고 Heterorhabditis bacteriophora 함양에 의한 치사율은 $23.8\pm$4.8%였다. 또한 S. carpocapsae 포천에 의한 기주치사율이 각각 $90.5\pm$4.8%와 $80.9\pm$4.8%, H. bacteriophora NC 1에 의한 것은 9.5$\pm$4.8%였다. S. glaseri NC와 H. bacteriophora 함양 및 H. bacteriophora NC 1을 동시 접종하였을 때는 S. glaseri NC에 의한 것이 각각 $61.9\pm$9.65%와 $80.9\pm$4.8%, H. bacteriophora 함양에 의한 것이 $38.1\pm$9.5%, H. bacteriophora NC 1에 의한 것이 $19.1\pm$4.8%였다. 그러나 두 선충의 동시 감염은 관찰되지 않았다. 그리고 S. carpocapsae All과 곤충병원성 곰팡이인 Beauveria brongniartii를 동시 또는 곰팡이를 먼저 처리했을 때는 곰팡이 12시간 전 처리부터 선충과 곰팡이의 동시 감염이 관찰되었고, H. bacteriophora NC 1는 곰팡이 6시간 전 처리부터 동시 감염이 관찰되었다. 선충에 의한 감염과, 곰팡이에 의한 감염, 선충과 곰팡이 동시 감염은 곰팡이 48시간 전 처리부터 관찰되었다. 그러나 유충 증식수는 선충 단독 감염보다 동시 감염충에서 현저히 떨어졌다.

The Detection and Multiplicity of Entomopathogenic Nematodes Using Silkworms (Bombyx mori) (누에를 이용한 곤충병원성 선충의 검색 및 배양)

  • Entomopathogenic nematodes were isolated through the investigation in soils collected from cultivated and non-cultivated fields using silkworms (Bombyx mori) and Galleria mellonella trap. The detectable rate of entomopathogenic nematodes of silkworms trap was higher than the G. mellonella trap. This study indicates the detection of entomopathogenic nematodes from soils that silkworms are sensitive superior to the G. mellonella to entomopathogenic nematodes. The steinernema, rhabditidae, and diplogatroidae strains successfully cultured on the silkworms host as well as on artificial media. Reproductivity in the living silkworm larva and pupa was 1.5 to 3.5${\times}$ 10$\^$5/ nematodes per host However, G. mellonella could be multiplied less than 5${\times}$l0$\^$5/ nematodes. The dried pupa of the silkworm following mositurize was cultured 0.5 to 2${\times}$10$\^$5/) nematodes per host. The culture methods of the steinernema, rhabditidae, and diplogatroidae strains, using silkworm powder, extracted chicken intestine, and food waste fertilizer could be applicative, but rate of reproduction was low.

Immunosuppressive Activity of an Entomopathogenic Bacteria, Xenorhabdus ehlersii KSY, and Its Application to Enhance Insecticidal Activity of Bacillus thuringiensis (곤충병원세균(Xenorhabdus ehlersii KSY)의 곤충면역 억제 능력과 이를 이용한 Bacillus thuringiensis 의 살충력 증가 효과)

  • An entomopathogenic bacterium, Xenorhabdus ehlersii KSY, is symbiotic to a nematode, Steinernema longicaudum, and exhibits high entomopathogenic virulence against lepidopteran insects. This study showed that the bacterial pathogenicity is induced by its inhibitory activity against eicosanoid biosynthesis of target insects, resulting in immunosuppression. To be applied for insect pest control, the bacteria should be infected to insect hemocoel. To deliver X. ehlersii to inset hemocoel, Bacillus thuringiensis (Bt) was mixed with the bacteria to breakdown the physical barrier (= midgut epithelium) from midgut lumen to hemocoel. The bacterial mixture significantly enhanced insecticidal activity of Bt only against larvae of Plutella xylostella and Maruca vitrata. For formulation, X. ehlersii cells were freeze-dried and mixed with sporulated Bt cells. The formulated bacterial mixture was applied to semi-field cultivating cabbage crop infested by P. xylostella. The bacterial mixture treatment showed over 95% control efficacy, while Bt alone gave 80% control efficacy. These results suggest that X. ehlersii can be applied to develop a novel insect control agent.

Pathogenicity bioassay of entomopathogenic nematode, Steinernema carpocapsae GSN1 strain (Nematoda: Steinernematidae) on the shiitake fungus moth, Morophagoides moriutii (Lepidoptera: Tineidae) (표고버섯좀나방에 대한 곤충병원성 선충의 병원성 검정)

  • Pathogenicity of Steinernema carpocapsae GSN1 strain was evaluated against Morophagoides moriutii larvae. In Petridish tests, insect mortality by the nematode was dose dependent, which increased with dose from 5 to 160 infective juveniles(IJs)/larva. Pathogenicity against fourth-instar larvae was higher than the rate of corresponding second- and third-instar larvae, showing 100% insect mortality with the dose of 40 IJs/$4^{th}$ instar larva and 80 IJs/$2^{nd}$ or $3^{rd}$ instar larvae. Lethal concentration values at 50% ($LC_{50}$) of S. carpocapsae GSN1 strain were 4.2 IJs/$2^{nd}$ instar larva; 8.5 IJs/$3^{rd}$ instar larva; and 2.3 IJs/$4^{th}$ instar larva, respectively. The number of nematodes established in M. moriutii larvae after infection increased in the increment of dose and insect developmental stage. The highest number of nematodes was harvested from fourth instar larvae of M. moriutii at a dose of 160 IJs per larva, showing 22.5 nematodes per insect larva. Nematode reproductive capacity was related to insect developmental stage, showing 6,335 IJs/$2^{nd}$ instar larva, 21,660 IJs/$3^{rd}$ instar larvae, and 88,700 IJs/$4^{th}$ instar larvae.

Insecticidal Toxin from Xenorhabdus nematopilus, Sysbiotic Bacterium Associated with Entomopathogenic Nematode Sreinernema glaseri

  • Entomopathogenic nematodes are being used for insect control. We purified a toxin secreted by the insect-pathogenic bacterium, Xenorhadbus nematophilus, which lives in the gut of entomopathogenic nematodes. Culture broth of X. nematophilus was separated by centrifugation and concentrated by ultration. The concentrated culture broth was applied to a DEAE Sephadex A-50 column, and proteins were eluted stepwise with increasing concentrations of KCI. Fractions column. The molecty weight of purified toxin was39 kDa on SDS-PAGE, and Fourier tranformed infrared (FTIR) spectroscopy indicated that this toxin could be a new protein exhiting the charactristics of C=O stretching peak near 1650cm-1.

Pathogenicity of Entomopathogenic Nematodes to Some Agro-Forest Insect Pests (농림해충에 대한 곤충병원성 선충의 병원성)

  • Mortality of some agro-forest insect pests, Agrotis segetum, Blitopertha orientalis, Agerastica coerulea, Glyphodes perspectalis and Acantholyda parki caused by entomopathogenic nematodes was investigated in laboratory and in pot. Steinernema carpocapsae All and Pocheon strain were effective against 2nd of 3rd instar larvae of A. segetum showing 100% mortalities at the concentration of 10Ijs/larva but not effective against 4th inster larvae. Mortality of B. orientalis was 100% after 26 days in H. bacteriopora NC 1, H. bacteriophora Hamyang and S. glageri NC strain treatment. S. monticulum also showed 100% mortalities against A. coerulea andd G. perspectalis at the concentration of 80 and 40 Ijs/larva. However, H. bacteriophora Cheju and S. glaseri Cheju strain were not effective against A. parki, i.e., showing 23.3 and 20.0% mortalities, respectively at the concentration of 160Ijs/larva. S. glaseri Hanrim strain was more effective than H. bacteriophora Cheju strain against Pryeria sinica.

Biological Control of Japanese Pine Sawyer, Monochamus alternatus (Coleoptera: Cerambycidae) using Korean Entomopathogenic Nematode Isolates (한국산 곤충병원성선충을 이용한 솔수염하늘소(Monochamus alternatus)의 생물적 방제)

  • Japanese pine sawyer (JPS), Monochamus alternatus is a vector insect of pine wood nematode, Bersaphlenchus xylophilus in Korea and Japanese pine forest. This study was to evaluate the possibility of biological control of JPS with Korean entomopathogenic nematodes (EPNs). Korean EPNs (Heterorhabditis sp. Gyeongsan, Steinernema carpocapsae GSN1, S. glaseri Dongrae and S. longicaudum Nonsan strain) were infested bate insect, Galleria mellonella in log of dead pine tree. Result showed that, S. carpocapsae GSN1 strain has the highest pathogenicity. Heterorhabditis sp. Gyeongsan and S. carpocapsae GSN1 had infested bate insect, which located in 7.5 cm depth of pine log. EPN has pathogenicity against larva and adult of JPS. Spray application of EPNs against adult of JPS, Heterorhabditis sp. Gyeongsan strain showed higher pathogenicity than S. carpocapsae GSN1 strain. EPN infested larva of JPS was detected on bark spray treatment with S. carpocapsae GSN1 strain in dead pine log that naturally infested larva of JPS however, no EPN infested JPS was detected on soaking of pine log with suspension of S. carpocapsae GSN1 strain. Though EPN had found with low efficacy against JPS but still possible to apply to control the boring insect pest as they can control log dwelling insect.

Evaluation of Entomopathogenic Nematodes against Armyworm, Pseudaletia separata on Tall Fescue, Festuca arundinacea (톨페스큐에서 곤충병원성선충의 멸강나방에 대한 효과 검정)

  • The armyworm, Pseudaletia separata was occurred suddenly in a golf club in Namhae, Gyeongnam province in 2013. Thus, pathogenicity of seven species of entomopathogenic nematodes (Steinernema carpocapsae GSN1 strain, S. glaseri Dongrae strain, S. longicaudum Nonsan strain, S. monticolum Jiri strain and S. siamkayai, Heterorhabditis sp. Gyeongsan strain, and H. bacteriophora Hamyang strain) was evaluated against armyworm on tall fescue in pot and golf course to control this pest environmentally friendly. The pathogenicity against P. separata larvae was significantly different depending on nematode species. The corrected mortality of fifth instar of P. separata was 100% in the treatment with S. carpocapsae GSN1 in 7 days in tall fescue pot. However, there was not different in the mortality of fifth instar of P. separata (80 to 100%) at the rate of 385 to 6,160 infective juveniles (Ijs) ($=2.5{\times}10^2$ to $4{\times}10^5Ijs/m^2$) of S. carpocapsae GSN1 strain in each pot. Corrected mortality of P. separata was 65 and 60% at the rate of $10^5Ijs/m^2$ of S. carpocapsae GSN1 and S. longicaudum Nonsan strain, respectively in the tall fescue of golf course.
