왕겨를 탄소원으로 사용하여 증균 배양을 실시함으로써 국내 사찰에서 제조된 된장으로부터 xylan 분해능 우수한 균주를 분리하였다. 분리균 YB-1301은 DNA gyrase subunit B 유전자(gyrB)의 염기서열에 근거하여 Bacillus safensis로 동정되었다. B. safensis YB-1301을 밀기울 또는 여러 종류의 xylan들이 첨가된 배지에서 배양하였을 때 xylanase의 생산성이 급격하게 증가되었다. 특히 birchwood xylan이 첨가된 LB 배지에서 플라스크 배양을 하였을 때 최대 340 U/ml 이상의 xylanase 생산성을 보였다. Xylan이 첨가되지 않은 LB 배지에서는 매우 소량의 xylanase가 균의 성장과 연계되어 항시적으로 생산되지만, xylan이 첨가된 배지에서는 정지기 생육단계에서 xylanase의 생산이 크게 유도되었다. 더구나 xylanase 생합성은 가수분해되지 않은 xylan 보다 xylan의 효소적 가수분해 산물에 의해 더 빠르게 유도되었다. 또한 B. safensis YB-1301의 배양상등액에 존재하는 xylanase는 55℃와 pH 6.5−7.0의 반응조건에서 최대활성을 나타냈다.
Various methods have been so far proposed to find out the directions and spectra of sound waves from the sources for provisions of noise controls. The conventional methods are generally classified into three systems such as, single microphone system, moving microphone system and multi-microphone system, which composes a resultant super directivity by giving a appropriate delay and a weighting coefficient in the output of each microphone. In case of using a single microphone there is a difficulty in providing it with desirable super directivity in the low frequency range, while in case of using multi-microphone system there has been a disadvantage that the measurement of directivity could not separately be done with the spectrum analysing. And in case of the use of a moving microphone system it needs a condition that the sound source to be detected should be stationary state and in rest. However here we introduce a method that the spectral analysing and the directivity of synthesis can be separately carried out by using a linear array of many microphones, in which each output of the microphone is multiplied by appropriate weighting coefficient and all of those products are summed after passing through adequate filters. The resultant signal is then sampled with an adequate sampling frequency and taken average for processing.
Acidithiobacillus ferrooxidans is one of the most important bacterium used in bioleaching, and can utilize $Fe^{2+}$ or sulphide as energy source. Growth curves for Acidithiobacillus ferrooxidans under phosphate starvation and normal condition have been tested, showing lag, logarithmic, stationary and aging phases as seen in other bacteria. The logarithmic phases were from 10 to 32 hours for Acidithiobacillus ferrooxidans cultivated with normal cultivating condition and from 20 to 60 hrs for Acidithiobacillus ferrooxidans cultivated phosphate starvation. Differences of protein patterns of Acidithiobacillus ferrooxidans growing in case of normal or phosphate starvation were separately investigated after cultivation at $30^{\circ}C$ by the analysis of two-dimensional gel electrophoresis (2-DE), matrix-assisted laser desorption/ionization (MALDI)-Mass spectrometry. There were total 6 protein spots identified, which were Recombination protein recA, RNA helicase, AP2 domain-containing transcription factor, NADH dehydrogenase I chain D, Hyothetical protein PF1669, and Transaldolase STY3758. From the 6 identified protein spots, 3 proteins were found to be decreased in expression at the cultivating condition of phosphate starvation, while another three upregulated.
Red blood cells (RBCs) are customarily adhered to a bio-functionalised substrate to make them stationary in interferometric phase-imaging modalities. This can make them susceptible to receive alterations in innate morphology due to their own weight. Optical tweezers (OTs) often driven by Gaussian profile of a laser beam is an alternative modality to overcome contact-induced perturbation but at the same time a steeply focused laser beam might cause photo-damage. In order to address both the photo-damage and substrate adherence induced perturbations, we were motivated to stabilize the RBC in OTs by utilizing a laser beam of ‘arbitrary intensity profile’ generated by a source having cavity imperfections per se. Thus the immobilized RBC was investigated for phase-imaging with sinusoidal interferograms generated by a compact and robust Michelson interferometer which was designed from a cubic beam splitter having one surface coated with reflective material and another adjacent coplanar surface aligned against a mirror. Reflected interferograms from bilayers membrane of a trapped RBC were recorded and analyzed. Our phase-imaging set-up is limited to work in reflection configuration only because of the availability of an upright microscope. Due to RBC’s membrane being poorly reflective for visible wavelengths, quantitative information in the signal is weak and therefore, the quality of experimental results is limited in comparison to results obtained in transmission mode by various holographic techniques reported elsewhere.
This paper describes the development and fabrication of a high temperature superconducting motor which consists of HTS rotor and air-core stator. The machine was designed for the rated power of 100hp at 1800 rpm. The HTS field windings are composed of the double-pancake coils wound with AMSC's SUS-reinforced Bi-2223 tape conductor. These were assembled on the support structure and fixed by a bandage of glass-fiber composite. The cooling system is based on the heat transfer mechanism of the thermosyphon by using GM cryocooler as cooling source. The cold head is in contact with the condenser of a Ne-filled thermosyphon. The rotor assembly was tested independently at the stationary state and combined with stator. Characteristic parameters such as reactances, inductances, and time constants were determined to obtain a consistent overview of the machine operation properties. This motor has met all design parameters by demonstrating HTS field winding, cryogenic refrigeration systems and an air-core armature winding cooled with air. The HTS field winding could be cooled down below 30K. No-load test of open-circuit characteristics(OCC) and short-circuit characteristics(SCC) and load test with resistive load bank were conducted in generator mode. Maximum operating current of field winding at 30K was 120A. From OCC and SCC test results synchronous inductance and synchronous reactance were 2.4mH, 0.49pu, respectively. Efficiency of this HTS machine was 93.3% in full load(100hp) test. This paper will present design, construction, and basic experimental test results of the 100hp HTS machine.
희박예혼합 가스터빈 연소기에 대한 3 차원 RANS 해석을 수행하였으며 PCFM(Partially Premixed Coherent Flame Model) 화염면적밀도 생성항 상수의 보정을 통하여 희박연소조건을 모사하였다. PCFM 에서 계산된 화염면적밀도에 의해 층류 예혼합 화염의 전파를 예측하고 불균일하게 분포한 기연 가스의 물성을 평형 가정에 따라 예측하였다. 복사와 대류 열전달을 모사하기 위해 냉각 조건으로서 실험과의 비교를 통해 결정된 열유속을 적용하였다. 이러한 3 차원 해석 결과를 바탕으로 파일럿 노즐과 메인 노즐에 분배되는 연료량 비에 대한 민감도 조사를 수행하였으며 CRN(Chemical Reactor Network)을 구성하여 NOx 배출량을 예측하고 측정값과 비교 분석하였다.

Human lactoferrin (hLf) is an iron-binding glycoprotein that has been considered to play many biological roles in the human, including the stimulation of the immune system, antimicrobial and anti-inflammatory effects, and regulation of iron absorption. We generated transgenic Siberian ginseng (Acanthopanax senticosus) cell cultures producing a functional hLf protein using the signal peptide sequence from the endoplasmic reticulum and driven by an oxidative stress-inducible SWPA2 promoter which is highly expressed in plant cell cultures. The production of hLf increased proportionally to cell growth and showed a maximal level (up to 3.6% of total soluble protein) at the stationary phase in suspension cultures. Full-length hLf protein was identified by immunoblot analysis in transgenic cell cultures of Siberian ginseng. Recombinant hLf (rhLf) was purified from suspension cells of Siberian ginseng by ammonium sulfate precipitation, cation-exchange and gel filtration chromatography. N-terminal sequences of rhLf were identical to native hLf (nhLf). The overall monosaccharide composition of rhLf showed the presence of plant specific xylose while sialic acid is absent. Antibacterial activity of purified rhLf was higher than that of nhLf. Taken together, we anticipate that medicinal Siberian ginseng cultured cells, as demonstrated by this study, will be a biotechnologically useful source for commercial production of functional hLf not requiring further purification.
저장사과주에서 분리(分離) 동정(同定)한 산막효모(産膜酵母) Hansenula beijerinckii FY-5의 소수성(疎水性)과 피막형성(皮膜形成)과의 관계(關係)를 조사하였다. 비(非)이온계(系) 계면활성제(界面活性劑) 첨가에 의해 균생육(菌生育)은 가능(可能)하지만 피막(皮膜)이 전혀 형성(形成)되지 않으며 소수도(疎水度)도 크게 낮아지는 사실로 미루어 볼때, 피막형성(皮膜形成)에는 효모세포(醉母細胞)를 배지표면(表面)에 보존(保持)시키는 어떤 인자(因予)가 요구되며 그 인자(因子)가 바로 효모세포표면(酵償細胞表面)외 소수성(疎水性)이 아닌가 생각된다. 산막성효모(産膜性酵母)에 있어서는 소수도(疎水度)가 클수록 피막형성(皮膜形成)이 왕성하였으나 비산막성효모(非産膜性酵母)는 대체로 소수도(疎水度)가 낮았다. 탄소원이 에타놀일때 소수도(疎水度)가 높았으며 pH의 상승에 따라 소수도(疎水度)는 감소(減少)하는 경향이었다. 배양기간별로는 균생육(菌生育)과 더불어 소수도(疎水度)도 비례적으로 증가하여 정상기(正常期)에 최대치를 보였다.
탄소원으로 palm kernel meal(PKM)과 밀기울을 함유한 배지에서 농후배양하여 작물 재배 토양으로부터 xylan과 locust bean gum(LBG)에 대한 분해활성이 있는 균을 분리하였다. 분리균 YB-1107의 16S rDNA 서열이 Cellulosimicrobium 속 균주와 유사도가 높은 균주로 판명되었다. 분리균의 mannanase는 LBG와 PKM에 의해 생산성이 증가된 반면에 xylanase는 oat spelt xylan과 밀기울에 의해 생산성이 증가되었다. Mannanase는 0.7% PKM을 첨가한 배지, xylanase는 1% 밀기울을 첨가한 배지에서 각각 최대 생산성을 보였으며 모두 정지기에서 생산이 되었다. 분리균의 배양상등액은 $55^{\circ}C$와 pH 6.5에서 mannanase의 최대활성을 보였으며, $65^{\circ}C$와 pH 5.5에서 xylanase의 최적반응 활성을 나타냈다. Mannanase에 의해 분해된 LBG와 xylanase에 의해 분해된 xylan으로부터 각각 올리고당이 관찰되었으며, 또한 이들 효소는 밀기울과 미강도 분해하여 올리고당으로 전환하는 것으로 확인되었다.
잡음의 제거는 음성을 인터페이스로 하는 기기들에 필수적이라고 할 수 있다. 실질적으로 통화 품질이나 음성 인식률은 음성 입력부의 주변에서 들어오는 원치 않는 가산성 잡음에 의해서 크게 열화된다. 본 논문에서는 기본적으로 두 개의 마이크로폰을 이용한 잡음제거 방법을 제안한다. 마이크를 여러 개 사용했을 때의 장점은 방향 정보를 이용할 수 있다는 것인데 이는 사람 목소리, 음악 소리 등의 비정상성 잡음을 제거하는 데에 유용하다. 제안된 잡음제거 알고리즘은 위너필터에 기반 한다고 볼 수 있다. 위너필터에 의한 잡음제거를 위해서는 검출하고자 하는 음성과 제거하고자 하는 잡음의 주파수 응답이 동시에 추정 가능해야 한다. 이를 위해서 주파수 영역에서 스펙트럼 분류를 시행하여 위너필터 기반의 잡음제거에 필요한 정보를 얻는다. 제안된 알고리즘을 이용한 성능은 잘 알려진 프로스트 (Frost) 알고리즘 및 적응 모드 컨트롤러를 갖는 generalized sidelobe canceller (GSC)와 비교하였다. 성능의 지표로는 객관적 음질 평가의 방법 중에서 널리 쓰이고 있는 perceptual evaluation of speech quality (PESQ) 및 음성 인식률이 사용되었다.
