The Experimental Study on Anti-bacterial Potency of Variou Herb-medicine used for eye disease on Staphylococcus epidermidis Keratitis (다종의 안질환 사용 처방이 각막염 유발균인 Staphylococcus epidermidis에 미치는 영향)

  • Objective : This experimental study was performed to investigate the effect of Herbe-medicine used for eye disease(Sean-tang, Jinpi-san, Tangpo-san, Copitdis Rhizoma) on Staphylococcus epidermidis keratitis. Methods : After administering various herbal eye drops on Staphylococcus epidermidis, I measured MIC and the size of inhibition zone. MIC was measured by dropping from 20 to $50{\mu}{\ell}$ according to density. Anti-bacterial potency was measured by the size of inhibition zone with change of volume under 2 days culture condition. Also continuous anti-bacterial potency of herbal eye drops was measured by the size of inhibition zone according to 2 days and 6 days culture each under the $50{\mu}{\ell}$ condition. Results : 1. MIC on Staphylococcus epidermidis in Sean-tang, Jinpi-san was 1%, $50{\mu}{\ell}$ 2. MIC on Staphylococcus epidermidis in Tangpo-san, Coptidis rhizoma was 10%, $50{\mu}{\ell}$ 3. MIC on Staphylococcus epidermidis in Cravit was 0.1%, $20{\mu}{\ell}$. 4. Under the 2 days culture condition, the size of inhibition zone on Staphylococcus epidermidis by volume for Sean-tang was 10.0mm in $50{\mu}{\ell}$, Jinpi-san was 16.0mm in $50{\mu}{\ell}$, Tangpo-san was 17.5mm in $50{\mu}{\ell}$, Coptidis rhizoma was 31.0mm in $50{\mu}{\ell}$ and Cravit was 34mm in $50{\mu}{\ell}$, showed the highest anti-bacterial potency. 5. Under the $50{\mu}{\ell}$ condition, the size of inhibition zone on Staphylococcus epidermidis by 2 and 6 culture days for Sean-tang was 47.5mm in 6days, Jinpi-san was 36.0mm in 6days, Tangpo-san was 45.0mm in 6 days and Cravit was 48.0mm in 6 days, which showed the highest anti-bacterial potency. 6. Under the $50{\mu}{\ell}$ culture condition, the size of inhibition zone on Staphylococcus epidermidis by 2 and 6 culture days for Coptidis rhizoma was 31.0mm in 2 days and 6 days, which showed the same anti-bacteria1 potency. Conclusions : The present author think that Herbe-medicine used for eye disease can be used to cure Staphylococcus epidermidis keratitis and if further study is performed, the use of the Herbe-medicine used for eye disease will be valuable and beneficial in the clinical medicines.

The Experimental study of safety and efficacy in using Soyeom Pharmacopuncture solution as eyedrops (점안용 소염약침액의 안전성 및 유효성 평가)

  • Objective : This experimental study was performed to investigate the safety of Soyeom Pharmacopunture solution manufactured by extraction of alcohol and water. To identify the use of it as eyedrops, the eye irritation test of rabbits and antibacterial test of Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans was performed. Methods : 1. The eye irritation test of this material was performed according to the Regulation of Korea Food & Drug Administration(2005. 10. 21, KFDA 2005-60). After Soyeom pharmacopuncture solution was administered in the left eye of the rabbits, eye irritation of the cornea, iris and conjunctiva was observed at 1, 2, 3, 4 & 7day. 2. After administering Soyeom Pharmacopuncture solution on bacterial species (Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) which cause Keratitis, MIC(Minimum Inhibition Concentration) and the size of inhibition zone were measured. Anti-bacterial potency was also measured using the size of inhibition zone. Results : 1. After Soyeom pharmacopuncture solution was administered in the left eye of the rabbits, it was found that none of nine rabbits have abnormal signs and weight changes. 2. After Soyeom pharmacopuncture solution was medicated in the left eye of the rabbits, no eye irritation of the cornea, iris and conjunctiva was observed at 1, 2, 3, 4 & 7day. 3. There was no response to MIC on bacterial species (Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans) after Soyeom pharmacopuncture solution was medicated. Conclusions : The present study suggests that Soyeom pharmacopuncture solution is a non-toxic and non-irritant medicine, which does not cause eye irritation in rabbits, but dosen't have anti-bacterial effects on bacterial species which cause Keratitis. These study result recommends that more research on other herbal medicines of eye drop for Keratitis are required.

  • Objectives : This experimental study was performed to investigate the effect of Fel Ursi & Bovis Calculus pharmacopuncture solution(FUBCPS) manufactured by using alcohol/water extraction method for identify ing the use of it as eyedrops. Methods: FUBCPS was manufactured by using alcohol/water extraction method. Measure the size of inhibition zone and MIC(Minimum Inhibition Concentration) after administering FUBCPS on bacterial species. Staphylococcus aureus. Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum and Candida albicans, which cause keratitis. Administering cravit(Levonoxacin medicine) on bacterial species also performed to compare the anti-bacterial potency of this material, measured by using the size of inhibition zone Results : After administering FUBCPS on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Candida albicans). there was no response to MIC and there was no anti -bacterial potency also. Conclusions : This study suggests that FUBCPS dose not have anti-bacterial effects on bacterial species which cause Keratitis. These study result recommends that we need to research more on herbal medicines of eye drop which have anti-bacterial effects on keratitis

  • Objective : This experimental study was performed to investigate the effect of Soyum Herbal-acupuncture, Hwangryunhaedoktang & Coptidis rhizoma on Staphylococcus epidermidis that induce keratitis. Methods : MIC was measured by dropping to $50{\mu}l$ according to density Soyum Herbal-acupuncture, Hwangryunhaedoktang & Coptidis rhizoma(100%, 10%, 1%, 0.1%). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Results : 1. MIC on Staphylococcus epidermidis in Soyum Herbal-acupuncture dippear all sample. 2. MIC on Staphylococcus epidermidis in Hwangryunhaedoktang disappear all sample. 3. MIC on Staphylococcus epidermidis in Coptidis rhizoma was 100%, $50{\mu}l$ and anti-bacterial potency increase compare with quantity. Conclusions : The present author think that Soyum Herbal-acupuncture must be extracted a new method for anti-bacterial potency and Coptidis rhizoma was made Herbal-acupuncture for eye drops.

황련의 증량에 따른 전탕액이 Staphylococcus epidermidis에 미치는 항균효과에 대한 실험적 연구

  • Objective : This experimental study was performed to investigate the effect of Coptidis rhizoma extract compared with quantity on Staphylococcus epidermidis that induce keratitis. Methods : MIC was measured by dropping to 50${\mu}l$ according to density Coptidis rhizoma extract(100%, 10%, 1%, 0.1%) compared with quantity(40g, 80g, 160g). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Results : 1. MIC on Staphylococcus epidermidis in Coptidis rhizoma extract was showed anti-bacterial potency compared with quantity and density in 100% and 10% of all samples(40g, 80g, 160g). 2. MIC on Staphylococcus epidermidis in Coptidis rhizoma extract(40g, 80g, 160g) was showed anti-bacterial potency compared with quantity all samples($20{\mu}l$, $30{\mu}l$, $40{\mu}l$, $50{\mu}l$) in 100% density, but anti-bacterial potency of 80g, $50{\mu}l$ Coptidis rhizoma extract decreased compared with 40g, $50{\mu}l$. 3. MIC on Staphylococcus epidermidis in 40g Coptidis rhizoma extract was showed in $50{\mu}l$, $40{\mu}l$ of 10% density, 80g Coptidis rhizoma extract was showed in $50{\mu}l$ of 10% density, 160g Coptidis rhizoma extract was showed in $50{\mu}l$, $40{\mu}l$, $30{\mu}l$ of 10% density. Conclusions : Coptidis rhizoma extract was showed anti-bacterial potency compare with quantity and density. In herbal drug, anti-bacterial potency compare with quantity and density must be studied.

Testing of the Safety and the Effectiveness of Using Samjeong Pharmacopuncture Solution as Eye drops

  • Objective: This experimental study was designed to investigate the safety and the effectiveness of Samjeong pharmacopuncture solution (SPS) manufactured by using a the lowtemperature extract on process. Methods: To identify the safety and the effectiveness of using SPS as eye drops, we performed applied eye irritation tests on rabbits and antibacterial tests for Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans. The eye irritation test was performed according to the toxicity testing regulation of the Korea Food & Drug Administration (2009. 8. 24, KFDA 2009-116). After SPS had been applied on the left eye of the rabbits, eye irritation in the cornea, iris and conjunctiva was observed on the 1st, 2nd, 3rd, 4th & 7th day. After SPS had been dropped on bacterial species that cause keratitis, the minimum inhibition concentration and the size of the inhibition zone were measured. The anti-bacterial potency was also measured by taking the size of inhibition zone. Results: After SPS had been administered on the left eye of the rabbits, none of nine rabbits were found to show abnormal signs or weight changes. After SPS had been administered on the left eye of the rabbits, no eye irritation in the cornea, iris and conjunctiva was observed on the 1st, 2nd, 3rd, 4th & 7th day. No specific response was detected in MIC for bacterial species Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, and Candida albicans after SPS had been applied. Conclusions: This study suggests that SPS is a non-toxic and non-irritant medicine that does not cause any of eye irritation in rabbits, but it has no antibacterial effects on bacterial species that are well known to cause keratitis. These results suggest that more research is required on extracts from herbal medicines for treating keratitis.

The Bacterial Contamination in Glasses for Vision Correction (시력 교정용 안경의 세균 오염)

  • Purpose: Recently, bacterial contamination of equipment and accessories required for vision correction has become a main causal factor in ophthalmic diseases. Thus, We investigated on both the actual condition of bacterial contamination from glasses of vision correction. Methods: Investigation of microorganisms was carried out with a group of 145 glasses wearers, composed of 36 elementary school students, 37 middle school students, 38 high school students, 10 college students, and 32 aged men. Results: Seventeen species of bacteria are detected from glasses of vision correction: B. cereus, B. licheniformis, Bacillus sp., CNS, Enterococcus sp., Escherichia coli, Proteus sp., Pseudomonas sp., Serretia sp., Streptococcus sp., Staphylococcus epidermidis, Staphylococcus aureus, Streptococcus hemolyticus,, Acinetobacter sp., Enterobacter cloacae, GNR, and Pseudomonas aeruginosa. Among 17 species of bacteria, there are some potential causative agents for keratitis, corneal ulcer, Acute dacryocystitis, Orbital cellulitis, Periphlebitis retinae, Marginal blepharitis, and Acute conjunctivitis. Enterobacter cloacae, Pseudomonas aeruginosa and Staphylococcus epidermidis cause keratitis. Pseudomonas sp., and Staphylococcus aureus cause corneal ulcer. Staphylococcus aureus causes acute dacryocystitis, orbital cellulitis, periphlebitis retinae, marginal belpharitis. Streptococcus hemolyticus causes acute conjunctivitis. Conclusions: In summation, it is verified that hazardous, opportunistic and infectious microorganisms exist in glasses for vision correction. Ophthalmic diseases are predicted. Therefore, supplementary research on the development of a cleaning solution to cleanse the infection and of an effective method to remove microorganisms is required.

The Experimental Study on Antibiosis of Decoctions Made by Cassiae Semen, Celosiae Semen and Buddlejae Flos (결명자(決明子), 청상자(靑葙子), 밀몽화(密蒙花) 전탕액(前湯液)의 항균성에 대한 실험적 연구)

  • Objectives : This experimental study was performed to investigate antibiosis of decoctions made by Cassiae Semen(CaS), Celosiae Semen(CeS) and Buddlejae Flos(BF). Methods : Decoctions made by CaS. CeS and BF were prepared. After administering decoctions made by CaS. CeS and BF on bacterial species(Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Aspergillus niger, Fusarium oxysporum, Condida albicans) which cause Keratitis, the size of inhibition zone and MIC(Minimum Inhibition Concentration) were measured Results : The inhibition zone on bacterial species didn't appear, after administering decoctions made by CaS, CeS and BF. Conclusions : This experimental study is showed that decoctions made by CaS, CeS and BF don't have antibiosis on bacterial species which cause Keratitis.

Antimicrobial Activity of Hwangryunhaedok-tang Extract against Keratitis (황련해독탕 추출액의 각막염 원인균에 대한 항균효과)

  • Objectives: The goal of this project was to measure the antibacterial effectiveness of Hwangryunhaedok-tang (barberry root decoction for detoxification) and its constituents. All of the active ingredients including the final product were prepared using high-pressure sterilization for use as medicinal eye drops. The varying forms of Hwangryunhaedok-tang (barberry root decoction for detoxification) were used to treat Staphylococcus epidermidis and Enterococcus faecalis which are found in keratitis. Methods: The antibacterial effect was measured by observing the presence or absence of antimicrobial activity when treated with varying concentrations of Hwangryunhaedok-tang extract. The tests were performed using a dosage of $70{\mu}l$ dosages of 100%, 50%, 10% and 1% the extracted solution by the minimum growth inhibitory concentration measurement. Antimicrobial activity was measured by examining the correlation between dosage strength and bacterial activity from $70{\mu}l$ to $10{\mu}l$ at the same concentration. Results: 1. Hwangryunhaedok-tang (barberry root decoction for detoxification), Phellodendri Cortex, and gardenia didn't show any antimicrobial effects against S. aureus, S. epidermidis, or E. faecalis. 2. Barberry root showed antimicrobial effects against S. aureus and S. epidermidis depending on the levels of concentration but didn't show any antimicrobial effects against E. faecalis. 3. Skullcap showed antimicrobial effects against S. aureus and S. epidermidis when a dosage of 100% extract $70{\mu}l$ was used. However, did not show any antimicrobial effects at all against E. faecalis. Conclusions: Hwangryunhaedok-tang (barberry root decoction for detoxification) and its constituents such as barberry root, phellodendri cortex, skullcap, and gardenia, can be used as an alternative to antibiotic medicinal eye drops to treat keratitis. However, further research on effective uses of and efficient extraction methods are needed.
