Simultaneous Determination of Baicalein, Baicalin, Wogonin, and Wogonoside in Rat Plasma by LC-MS/MS for Studying the Pharmacokinetics of the Standardized Extract of Scutellariae Radix

  • A new composition of standardized Scutellariae Radix extract (HPO12) was developed for treatment of Alzheimer's disease. For the preclinical pharmacokinetic study of HPO12, a rapid, sensitive, and selective LCMS/MS method was developed and validated for the simultaneous determination of 4 bioactive compounds, baicalein, baicalin, wogonin, and wogonoside. After extraction with ethylacetate, chromatographic analysis was performed on a Thermo $C_{18}$ column ($150mm{\times}2.1mm$, $3{\mu}m$) with a mobile phase consisting of 0.1% formic acid (A) and 0.1% formic acid in 95% acetonitrile (B) by using gradient elution at a flow rate of $250{\mu}L/min$. Analytes introduced to a mass spectrometer were monitored by multiple reaction monitoring (MRM) in positive ion mode. Using $25{\mu}L$ of plasma sample, the method was validated over the following concentration ranges: 25-5000 ng/mL for baicalein, 20-40000 ng/mL for baicalin, 1-1000 ng/mL for wogonin, and 5-10000 ng/mL for wogonoside. The intra- and inter-day precision and accuracy of the quality control samples at the 4 concentrations showed $\leq$ 13.7% relative standard deviation (RSD) and 86.6-105.5% accuracy. The method was successfully applied to determine the concentrations of baicalein, baicalin, wogonin, and wogonoside in rat plasma after intraperitoneal and oral administrations of HPO12.

  • In this paper, a very new web-based software for renewable energy system (RES) design and economic evaluation was introduced. This solution would provide the precise RES estimation service including not only photovoltaic (PV), wind turbine (WT) and fuel cell (FC) individually but also energy storage system (ESS) as combined forms with PV or WT. The three reasons why we ought to develop it are: First, the standardized tool suitable to the domestic environment for estimating power generation from RES facilities and economic evaluation is required. Secondly, the standardized tool is needed to spread domestic RES supply policy and to promote the new industry in the micro-grid field. The last, the reliability of economic evaluation should be enhanced more for new facilities. To achieve those aims, the weather database of one hundred locations have established and the RES facility database has also constructed. For the energy management, mathematical models for PV, WT, ESS and FC were developed. As a final phase, the analytical process to evaluate economics has performed with field data verification.

A Systematic Study on the Effect of Social StoryTM Intervention for Individual with Autism Spectrum Disorder (자폐스펙트럼장애 아동을 위한 상황이야기 중재의 효과 연구에 대한 체계적 고찰)

  • Purpose : The purpose of this study was to systematically review the studies on the effects of social story intervention on the individuals with autism spectrum disorders(ASD) to provide the basis for evidence-based practice. Methods : In order to find out the studies on the effect of social story intervention on individuals with ASD, studies published from 2011 to December 2020 were searched on Google Academic Search. The keywords used were "autism spectrum disorder AND social story". A total of 16,900 studies were searched, and from these, 12 studies were selected based on the application of the selection and exclusion criteria. The included studies comprised of 10 single subject design studies and 2 randomized controlled trials design studies. The included studies were analyzed in accordance to population, intervention, outcome measures and results. Results : As a result of analysing the subjects of the included studies, we found out that pre-school children and adolescents were the most common groups of individuals diagnozsed with ASD. In addition to ASD, the diagnosis of the subjects included intellectual disability, Asperger, and Prader-willi. We found out that there were more interventions that were mixed with other interventions, than just the pure social story interventions. In particular, video modeling was found to be the most frequent intervention. This was followed by photo-based social stories. For outcome measure, autism social skills profile was found to be the most often used standardized assessment. For a non-standardized assessment, social desired behavior was found to be the most frequently evaluated behavior, followed by personal problem behavior. Of all the dependent variables, the dependent variable reported as "effective" was the most, followed by "some effective". Conclusion : This study is organized to help the individuals with ASD, families, researchers, and therapists understand the effects of social story intervention on the individuals with ASD in an easy fashion. Further, therapists can use this study as the basic data for evidence-based practice.

The standardized Korean Red Ginseng extract and its ingredient ginsenoside Rg3 inhibit manifestation of breast cancer stem cell-like properties through modulation of self-renewal signaling

  • Background: The ginsenoside Rg3, one of active components of red ginseng, has chemopreventive and anticancer potential. Cancer stem cells retain self-renewal properties which account for cancer recurrence and resistance to anticancer therapy. In our present study, we investigated whether the standardized Korean Red Ginseng extract (RGE) and Rg3 could modulate the manifestation of breast cancer stem cell-like features through regulation of self-renewal activity. Methods: The effects of RGE and Rg3 on the proportion of $CD44^{high}/CD24^{low}$ cells, as representative characteristics of stem-like breast cancer cells, were determined by flow cytometry. The mammosphere formation assay was performed to assess self-renewal capacities of breast cancer cells. Aldehyde dehydrogenase activity of MCF-7 mammospheres was measured by the ALDEFLUOR assay. The expression levels of Sox-2, Bmi-1, and P-Akt and the nuclear localization of hypoxia inducible $factor-1{\alpha}$ in MCF-7 mammospheres were verified by immunoblot analysis. Results: Both RGE and Rg3 decreased the viability of breast cancer cells and significantly reduced the populations of $CD44^{high}/CD24^{low}$ in MDA-MB-231 cells. RGE and Rg3 treatment attenuated the expression of Sox-2 and Bmi-1 by inhibiting the nuclear localization of hypoxia inducible $factor-1{\alpha}$ in MCF-7 mammospheres. Suppression of the manifestation of breast cancer stem cell-like properties by Rg3 was mediated through the blockade of Akt-mediated self-renewal signaling. Conclusion: This study suggests that Rg3 has a therapeutic potential targeting breast cancer stem cells.

Parkinson's Disease as Risk Factor in Osteoporosis and Osteoporotic Vertebral Fracture : Prevalence Study Using National Inpatient Sample Database in Korea

  • Objective : To determine the prevalence of osteoporosis (OP) and osteoporotic vertebral fracture (OVF) in people with Parkinson's disease (PD) in Korea and its association with socioeconomic status. Methods : Using Health Insurance Review and Assessment Service-National Inpatient Sample (HIRA-NIS) data from 2009 to 2013, we estimated the annual prevalence of PD, OP, and OVF and investigated its association with socioeconomic status using data from National Health Insurance (NHI) beneficiaries and Medical Aid (MA) recipients. This study was supported by research funding from Korean Society for Bone and Mineral Research 2015. There were no study-specific biases related to conflicts of interest. Results : The number of PD patients in the HIRA-NIS increased each year from 2009 to 2013. Among patients with PD, the standardized prevalence rates of OP and OVF increased from 2009 to 2013; from 23.2 to 27.8 and from 2.8 to 4.2, respectively. Among patients with PD with OP, the prevalence of OVF were 12.2% and 15.1% in 2009 and 2013, respectively. The standardized prevalence rates of PD with OP and PD with OVF were significantly higher in MA recipients than in NHI beneficiaries. Conclusion : The prevalence of PD both with OP and with OVF increased and the prevalence was higher in MA recipients than in NHI beneficiaries. These findings may suggest that age over 65 years, female and low income may be a significant factor related to PD occurring with OP and OVF.

  • In the era of the 4th industrial revolution symbolized by the Internet of Things, big data and artificial intelligence, various embedded devices are increasing exponentially. These devices have communication functions despite their low specifications, so the possibility of personal information leakage is increasing, and security threats are also increasing. Embedded devices can have security issues at most levels, from hardware to services over the network. In addition, it is difficult to apply general security techniques because it has characteristics of resource constraints such as low specifications and low power, and the related technology has not been standardized. In this study, we present vulnerabilities and possible problems and considerations in applying SDN to embedded devices in consideration of structural characteristics and real-world discovered cases. This study presents vulnerabilities and possible problems and considerations when applying SDN to embedded devices. From a hardware perspective, we consider the problems of Wi-Fi chips and Bluetooth, the problems of open flow implementation, SDN controllers, and examples of structural properties. SDN separates the data plane and the control plane, and provides a standardized interface between the two, enabling efficient communication control. It can respond to the security limitations of existing network technologies that are difficult to respond to rapid changes.

Meta-analysis of the Effect of Proprioceptive Neuromuscular Facilitation Training on Gait Ability in Patients with Storke

  • The purpose of this meta-analysis was to determine the efficacy of proprioceptive neuromuscular facilitation training on Gait. We included all randomized controlled trials assessing the efficacy of proprioceptive neuromuscular facilitation training on gait ability control in patients after stroke. This study was conducted according to the PRISMA guideline. CINAHL, PubMed and RISS were searched for studies published up to December 2020, and all randomized controlled trails assessing PNF intervention were included. This analysis included only randomized controlled trials. A total of 12 studies were selected from 1475 records obtained from the databases. The meta-analysis was performed using the R software. The overall intervention effect was moderate (standardized mean difference (SMD): 0.67, 95% confidence interval (CI): 0.50-0.84). Additionally, Gait rite (SMD: 0.69), 10m walking test (SMD: 0.59) had medium effect sizes. These findings indicate that PNF is an effective intervention for improving gait ability in patients stroke.

Standardized ileal digestibility of amino acids of protein sources associated with exogenous enzymes for broilers

  • Objective: Two experiments were conducted to evaluate the effect of enzyme complex (EC) on the standardized ileal digestibility (SID) of amino acids (AA) in corn gluten meal (60%) (CGM), soy protein concentrate (SPC), dried bovine plasma (DBP), and poultry offal meal (POM). Experiments I and II were conducted with broilers in the pre-starter (1 to 7 days of age) and starter (1 to 21 days of age) phases, respectively. Methods: The treatments consisted of a protein-free diet (PFD) containing feedstuffs either supplemented with EC (xylanase, amylase, and protease) or not. In Experiment I, a total of 360 one-day-old male Cobb-500 broiler chicks were randomly housed in 45 pens, resulting in five replicates with eight birds each, totalizing eight treatments and one PFD group. In Experiment II a total of 270 one-day-old male Cobb-500 broiler chicks were randomly housed in 45 pens, resulting in five replicates with six birds each, totalizing eight treatments and one PFD group. The PFD groups were used to assess the endogenous AA losses. The birds were slaughtered to collect the ileal content. Results: In the pre-starter phase, the SID of arginine, branched chain-aminoacids, glycine, serine, aspartate, and glutamic acid increased with EC addition. The EC improved the SID of arginine and glutamic acid of CGM; the SID of valine and cystine of SPC; the SID of leucine, glycine, and aspartate of POM and the SID of isoleucine of DBP. In the starter phase, the SID of isoleucine, phenylalanine and glycine increased in EC-supplemented diets. The EC improved the SID of isoleucine of DBP; the SID of phenylalanine of CGM and POM. The SID of AA of SPC was not influenced by the EC. Conclusion: The addition of an EC to broiler pre-starter and starter diets is efficient in increasing the SID of AA on SPC, POM, and DBP.

A Study on How to Build an Optimal Learning Model for Artificial Intelligence-based Object Recognition (인공지능 기반 객체 인식을 위한 최적 학습모델 구축 방안에 관한 연구)

  • The Fourth Industrial Revolution is bringing about great changes in many industrial fields, and among them, active research is being conducted on convergence technology using artificial intelligence. Among them, the demand is increasing day by day in the field of object recognition using artificial intelligence and digital transformation using recognition results. In this paper, we proposed an optimal learning model construction method to accurately recognize letters, symbols, and lines in images and save the recognition results as files in a standardized format so that they can be used in simulations. In order to recognize letters, symbols, and lines in images, the characteristics of each recognition target were analyzed and the optimal recognition technique was selected. Next, a method to build an optimal learning model was proposed to improve the recognition rate for each recognition target. The recognition results were confirmed by setting different order and weights for character, symbol, and line recognition, and a plan for recognition post-processing was also prepared. The final recognition results were saved in a standardized format that can be used for various processing such as simulation. The excellent performance of building the optimal learning model proposed in this paper was confirmed through experiments.

A Study on the Data Collection and Analysis System for Learning Experiences in Learner-Centered Customized Education (학습자 중심의 맞춤형 교육을 위한 학습 경험 데이터 수집 및 분석 체계 연구)

  • This study investigates the comprehensive system for collecting intelligent learning activity data tailored to learner-centered personalized education. We compared and analyzed the characteristics of xAPI, Caliper analytics, and cmi5, which are learning activity data collection standards, and established a system that allows not only standardized data but also non-standardized learning activity data to be stored as big data for artificial intelligence learning analysis. As a result, the system was structured into five stages: defining data types, standardizing learning data using xAPI, storing big data, conducting learning analysis (statistical and AI-based), and providing learner-tailored services. The aim was to establish a foundation for analyzing learning data using artificial intelligence technology. In future research, we will divide the entire system into three stages, implement and execute it, and correct and supplement any shortcomings in the design.
