Effect of Cordyceps militaris on Testosterone Production in Sprague-Dawley Rats

  • Some of men have been suffered from the insufficient secretion of testosterone causing by physical factors, social and psychological factors. Testosterone is an essential steroid hormone controlling male reproductive function. Alternative medicines in plants, fungi, and insects have been studied to enhance sexuality. $Cordyceps$ species including $Cordyceps$ $sinensis$ (CS) and $C.$ $militaris$(CM) has been used as for the enhancement of sexual functionfor hundreds of years in Far East Asian. In the present study, we determined the effect of fruiting bodies of $C.$ $militaris$ which cultured on bee drone medium (CMD) and brown rice medium (CMB) on testosterone concentration in Sprague-Dawley rats. Eighteen rats per group were housed to regular diet or diet supplemented with CMB and CDD, respectively for 4 weeks. Serum was collected from 6 rats per group. Results showed that changes of the body weight, food and water intake of the rats were not observed in this study. However, both CMB and CDD increased the serum testosterone concentration in rats. Furthermore, CMD significantly stimulated testosterone production (p <0.05) compared to the control. Hence, it suggests that $C.$ $militaris$ fruiting bodymight be developed as a complementary medicine to improve sexual hormones.

Intravenous Single-dose Toxicity of Mountain Ginseng Pharmacopuncture in Sprague-Dawley Rats

  • Objectives: Mountain ginseng pharmacopuncture (MGP) is an extract distilled from either mountain cultivated ginseng or mountain wild ginseng. This is the first intravenous injection of pharmacopuncture in Korea. The word intravenous does not discriminate between arteries, veins, and capillaries in Oriental Medicine, but only the vein is used for MGP. The aim of this study is to evaluate the intravenous injection toxicity of MGP through a single-dose test in Sprague-Dawley (SD) rats. Methods: Male and female 6-week-old SD rats were injected intravenously with MGP (high dosage of 20 mL/kg or low dosage of 10 mL/kg). Normal saline was injected into the rats in the control group by using the same method. After the rats has treated, we conducted clinical observations, body-weight measurements and histological observations. Results: In this study, no mortalities were observed in any of the experimental groups. Also, no significant changes by the intravenous injection of MGP were observed in the body weights, or the histological observations in any of the experimental groups compared to the control group. The lethal dose for intravenous injection of MGP was found to be over 20 mL/kg in SD rats. Conclusion: Considering that the dosage of MGP generally used each time in clinical practice is about 0.3 mL/kg, we concluded with confidence that MGP is safe pharmacopuncture.

Single-dose Intravenous Injection Toxicity of Water-soluble Danggui Pharmacopuncture (WDP) in Sprague-Dawley Rats

  • Objective:This study is to evaluate both the single-dose intravenous injection toxicity and the approximate lethal dose of Water-soluble Danggui Pharmacopuncture (WDP) in Sprague-Dawley (SD) rats. Methods: Toxicity experiments were conducted at Good Laboratory Practice (GLP) laboratory in Biotoxtech Co., according to the regulations of GLP. WDP injection of dose 0.125, 0.25, and 0.5 mL/animal were experimental groups and normal saline injection group was control group. WDP and normal saline were injected once to 6- week old 5 male and 5 female SD rats at the tail veins at approximately 2 mL/min. During 14 days after the injection, general symptoms were observed and weight were measured. After the observation period, hematological and blood biochemical examination, macroscopic autopsy, topical resistance test at the injection area were performed. Results: RThe WDP 0.5 mL/animal injection group in 4 cases of male rats and all cases of female rats showed hematuria 30 minutes after the administration. However, after 1 hour, no more abnormal general symptoms were observed. The WDP did not affect weight, hematological and blood biochemical examination, macroscopic autopsy, and topical resistance test at the injection area. Conclusion: WDP single dose intravenous injection results showed that WDP have no toxic effects and a lethal dose of WDP should be over 0.5 mL/animal in male and female rats under the study condition. So WDP may be safe.

수영과 저출력 레이저 치료가 류마티스 관절염 흰쥐의 혈청 인터루킨-6 농도에 미치는 영향 (The Effects of Swimming and Low Power Laser on the Concentration of Interleukin-6 for Rheumatoid Arthritic Rats)

  • This study was designed to determine the effects of swimming and low power laser on rheumatoid arthritis in Sprague-Dawley rats. Rheumatoid arthritis was induced in 36 rats among 48 Sprague-Dawley rats by the subcutaneous injection of .05 $m{\ell}$ Freund's Complete Adjuvant into the right hind paw and .05 $m{\ell}$ Freund's Complete Adjuvant into the right hind knee joint capsule. A second injection was performed by the same method using .1 $m{\ell}$ Freund's Complete Adjuvant per a rat. Arthritic rats were divided into 8 groups: each 1 week and 2 weeks of arthritic swimming, arthritic laser, arthritic case control and normal group. In this study, several experimental tests were performed to determine the concentration of Interleukin-6, the space of the knee joint and the thickness of the hind paw. The concentration of Interleukin-6 and hind paw thickness decreased in the swimming group and laser group as compared to the control group. The space of the knee joint increased significantly after the swimming exercise. Swimming and low power laser therapy positively affect rheumatoid arthritis in rats affect by decreasing the concentration of Interleukin-6 and hind paw thickness, and increasing the space of the knee joint.

재조합 인간상피세포 성장인자(rhEGF, DWP401)가 랫드의 수태능, 태자와 신생자 발달 및 모체기능에 미치는 영향 (Fertility and Reproductive & Developmental Toxicity Study on Recombinant Human Epidermal Growth Factor (rhEGF, DWP401) in Rats)

  • This study was conducted to investigate for its effects on reproductive and developmental toxicity of recombinant human epidermal growth factor (rhEGF) in Sprague-Dawley rats. Male rats were administered rhEGF at doses of 1, 10, 100, and 1000$\mu$g/kg/day, respective1y, by subcutaneous injection from 63 days before and throughout to mating period until the day before sacrifice. Female rats were administered rhEGF at the same doses from 14 days before mating to day 20 of gestation or to day 21 of lactation. We examined the male and female fertility indices and maternal toxicity of F0 parental animals. Also, we examined the external, visceral, or skeletal malformation of fetuses, growth and development, behavior, and/or reproductive performance of F1 animals. At the highest dose (1,000 $\mu$g/kg), the mean body weights of F0 animals were significantly increased in males and females at 3 or 2 weeks after treatment, respective1y. No clinical signs and food intakes were observed at any time during the experimental period by rhEGF treatment. In autopsy examination, the relative and absolute liver weights significantly increased in both sexes of 1,000 $\mu$g/kg. At the highest dose (1,000 $\mu$g/kg), there was a statistically significant increase of pregnancy period and the number of dead fetuses. Moreover, significant increase of mean fetal body weight and decrease of number of live fetuses, which related to the difficult dilivery were observed in highest dose group. In Fl examination, no adverse effects on external, visceral, and skeletal malformation, physical and functional development, behavior or reproductive ability of Fl animals were observed in any group. Also, there was no significant difference between control and treated groups in copulation or fertility indices of Fl animals. These results indicate that rhEGF had no adverse effect on fertility and reproductive ability of Sprague-Dawley rats.

참마 분획물이 당뇨 유발 흰쥐의 혈당 및 에너지대사물농도에 미치는 영향 (Effects of Fractions of Dioscorea Japonica Thumb on Blood Glucose Level and Energy Metabolism in Streptozotocin Induced Diabetic Rats)

  • Dioscorea japonica Thunb its effects has been used in f31k remedies f9r various purposes including treatment of diabetes, on hypoglycemic actiity and energy metabolism were investigated. The plant was extracted with methanol(MeOH) and fractionated into four layers hexane, chloroform(CHCI$_3$), butanol(BuOH), and $H_2O$. Diabetes mellitus was induced in male Sprague-Dawley rats by the injection of streptozotocin(STZ) into tail vein at a dose of 45mg/kg body weight(BW). Sprague-Dawley male rats weighing 160-200g were divided into five groups a diabetic control and four experimental groups such as hexane group, CHCl$_3$ group, BuOH group, and $H_2O$ group. The rats of all groups were fed on a AIN-76 diet and the four experimental groups were orally administered each fraction(500mg/kg BW) for 12 days. The diabetic control group was orally administered 5% carboxymethyl cellulose. The body weights were monitored and the concentrations of blood glucose were determined. The levels of glycogen and protein in liver were also measured. The plasma levels of cholesterol, triglyceride (TG), and fee fatty acid(FFA) were also analysed. The body weight gain was higher in the $H_2O$ group than in the control group. Heart weight was significantly reduced by administrations of Dioscorea japonica Thunb. The extents of blood glucose decrement in BuOH and $H_2O$ group were greater than that found in the control group. The muscle protein levels showed significantly higher amounts in all experimental groups. Glycogen levels were higher in the BuOH group than in the control group. The levels of TG were decreased in all experimental groups and the levels of plasma FFA were lower in the BuOH group. The plasma cholesterol levels were not influenced by these four fractions in diabetic rats. These results suggest that the orally administered H2O fraction of Dioscorea japonica Thunb exhibited hypoglycemic effects in STZ induced diabetic rats. (Korean J Nutrition 31(7) : 1093-1099, 1998)

The effect of seamustard on blood lipid profiles and glucose level of rats fed diet with different energy composition

  • Recently, Korean people are consuming seaweeds almost 3.5 times more now than three decades ago. It is well known that seaweeds contain lots of soluble dietary fiber in addition to micronutrients such as ${\beta}$-carotene, iodine and some bioactive components. Seaweeds are considered to be effective for preventing chronic diseases including obesity, diabetes mellitus, atherosclerosis, cancer or constipation. This study was conducted to investigate the effect of seamustard intake on body weight gain, blood glucose level and lipid profiles in rats fed diets with different energy nutrient composition. Male Sprague-Dawley rats (average initial weight 103.7g) were divided into groups for two experiments as follows; Control, M2.5 & M5 groups (Exp. I) and M5, M10, HCM5, HCM10, HFM5 & HFM10 groups (Exp. II). The rats were fed diet and water ad libitum for 4 weeks. In general, there was no significant difference in blood glucose and triglyceride concentration among groups. In Exp. I, serum LDL-cholesterol level of rats fed diet with 5% seamustard powder (M5) was significantly lower than that of control group, while HDL-cholesterol level, TC/LDL ratio and weight of adrenal gland were higher. In Exp. II, food intake, body weight gain and EER of high fat diet with 10% seamustard group (HFM10) were the lowest among groups. Except gastrocnemius muscle, all organ weights of HFM10 group were the lowest. Fecal cholesterol excretion and serum LDL-cholesterol concentration of HFM10 group were the highest, while serum HDL-cholesterol level was the lowest among groups. Interestingly, HDL-cholesterol concentration was the highest in HCM5 group among groups. From these results, it was suggested that seamustard intake might be more effective for body weight control, but not for improving blood lipid profiles in high fat diet than in high carbohydrate diet.

톳(Hizikia fusiformis)의 Sprague-Dawley계 흰쥐에서 항고지혈증 및 항동맥경화증 효능 (Antihyperlipidemia and Antiarteriosclerosis of Hizikia fusiformis in Sprague-Dawley Rats)

  • The effects of dietary supplementation with Hizikia fusiformis on anti-hyperlipidemic activities were investigated in an animal study in which normal rats were fed Hizikia fusiformis, Hizikia fusiformis ethanol extract and Hizikia fusiformis ethanol extract residue. Serum total lipid and triglyceride levels were significantly (P<0.05) reduced in rats fed Hizikia fusiformis ethanol extract residue at a dose of 200 mg/kg body weight when compared to hyperlipidemic control rats. There were also significant decreases in serum total cholesterol and LDL-cholesterol levels in the rats fed Hizikia fusiformis ethanol extract residue at 200 mg/kg body weight. In addition, the atheroscrelosis index and superoxide dismutase in blood lipids were significantly (P<0.05) lowered in rats fed Hizikia fusiformis ethanol extract residue at 200 mg/kg body weight, as compared to control rats. In conclusion, Hizikia fusiformis and Hizikia fusiformis ethanol extract residue had beneficial antihyperlipidemic and antiarteriosclerosis effects.
