Enhancement of Alcohol Metabolism by Sprouted Peanut Extract in SD Rats

  • Excessive ethanol intake is known to induce a number of physiological symptoms, including headache, dizziness and vertigo. In this study, we investigated the attenuation effect of sprouted peanut extract (SPE) on ethanol-induced hangover in male Sprague-Dawley rats. The animals were divided into five groups: the control group, which was administered ethanol only; the ethanol plus SPE experimental groups, which were administered ethanol and 100, 200, or 400 mg SPE/kg b.w.; and the positive control group, which was administered ethanol plus DAWN808$^{(R)}$, a commercial product. SPE-suspended water was delivered to rats via gavage 15 h and 30 min before the administration of ethanol. Blood was collected from the tail 0, 1, 3, and 5 h after ethanol administration. The results showed that serum ethanol concentrations were significantly lower in SPE treated groups than in the control group. Furthermore, hepatic alcohol and acetaldehyde dehydrogenase activities were enhanced by SPE in a dose dependent manner. These results suggest that SPE could be useful in attenuating hangover after alcohol consumption.

Study on the Hepatoprotective Effect and Cytochrome P450 Regulation of Scutellaria Radix (황금의 간보호활성 및 Cytochrome P450 발현조절에 관한 연구)

  • In this study, the liver protective effect of the hot water extracts of Scutellaria radix (SR) was investigated. The SR exhibited a hepatoprotective activity against $CCl_4$-induced liver damage in Sprague-Dawley (SD) rats and Chang cell. And the SR also showed significant decrease of malodialdehyde (MDA) and increase of glutathion (GSH), catalase activity in rat liver homogenate. The expression of cytochrome P450 2E1 (CYP2E1), measured by RT-PCR and western blot, was significantly decreased in the SR treated SD rats and Chang cell. But $CCl_4$ and SR has no significant effect on 1A1 and 3A1 isoform of cytochrome P450. Based on these findings, it is suggested that hepatoprotective effects of SR possibly related to antioxidative effects and downregulation of CYP2E1 expression.

Acute toxicity study of 77 herbal formulas according to GLP guidelines in rats (흰쥐에서 GLP 기준에 의한 한약처방 77종의 단회 투여 급성 독성 연구)

  • Objectives: This study aimed to investigate the acute oral toxicity of 77 herbal formulas and performed in male and female Sprague-Dawley (SD) rats as per the guidelines mentioned in Ministry of Food and Drug Safety. Methods: Each sex of SD rat were administered a single dose (2000 or 5000 mg/kg) of 77 herbal formulas via oral gavage; the control group received vehicle only. After administration, the mortality, clinical signs, gross findings, and body weight were followed up for 15 days. Results: After administration of 77 herbal formulas, mortality, clinical signs, body weight changes, and gross findings related to the test substances were not observed in both male and female groups. Conclusion: Our results demonstrate the single-dose oral administration of 77 herbal formulas produced no mortality indicating the approximate lethal dose is greater than 2000 or 5000 mg/kg body weight.

Activity of Superoxide dismutase(SOD) by fermented soybean (발효 대두 식품의 Superoxide dismutase(SOD) 활성)

  • This study was performed to evaluate the inhibition effects of fermented soybean on lipid perosidation and antioxidative relative enzyme activity. in vivo. Fermented soybean was induced the high SOD activity, while significantly inhibited on the peroxide value of linoleic acid and lipid perxidation from rat microsome induced by Fe$^{2+}$ ascorbate system, Sprague-Dawley(SD) male rats were fed basic diet, and experimental diets group added 200 or 500 mg/kg fermented soybean for 2 weeks. The effect of fermented soybean is also significantly increased catalase and glutathione peroxidase activities, while significantly inhibited the lipid peroxidation of rat liver microsome in a dose dependent manner. Therefore, these results suggest that fermented soybean has antioxidative activity which is related enzyme to prevention of oxidative stress.s.

Transdermal permeation and Anti-inflammatory Effects of Piroxicam Gel by Phonophoresis (음프록시캄 겔을 이용한 음파영동이 경피투과와 염증에 미치는 효과)

  • This study was performed that how phonophoresis using ultrasound for piroxicam affects transdermal permeation and anti-inflammative effects. Transdermal permeation study conducted by using hairless mouse had two categories: control group and ultrasound group. Transdermal permeation was observed according to duty cycle and intensity. Anti-inflammatory effects were determined using in Sprague-Dawley rat. The subjects were divided into three groups of six SD rat each 24 hour, 48 hour, 72 hour. The results of this study were as follows: 1. Transdermal permeation of piroxicam was measured according to ultrasound duty cycle. This research demonstrates that ultrasound group retains more transdermal permeation than control group, and that pulsed ultrasound group holds a little more transdermal permeation than continuous ultrasound group. 2. The transdermal permeation of piroxicam is closely related with ultrasound intensity. Effect of each group of transdermal permeation was significant rises in proportion to ultrasound intensity. 3. By observing inflammation of the tissue caused by trauma, phonophoresis group showed more significant of anti-inflammatory effect. The conclusion of phonophoresis was found to improve significantly the transdermal permeation and the anti-inflammatory effect.

Optimal Level for the Protection of Carbon Tetrachloride-induced Sprague-Dawley Rat Liver Damage by Mycelial Cultures of Lentinus edodes (Sprague-Dawley 쥐의 사염화탄소 유도 간 손상에 대한 표고버섯균사체의 효과)

  • The protective effects of a powder mixed with solid-cultured and liquid-cultured Lentinus edodes mycelia (2 : 1, w/w) (designate LED) with different doses of carbon tetrachloride ($CCl_4$) on induced hepatotoxicity in male Sprague-Dawley (SD) rats was investigated. The rats were divided into seven groups (6 rats/group) and the following substances were administered orally to each group: Vehicle (0.2 ml distilled water), Control (0.2 ml distilled water), LED (LED 100, 200, 300 and 400 mg/kg BW in 0.2 ml distilled water), and Silymarin (200 mg/Kg BW in 0.2 ml distilled water). After two weeks of daily administration, all groups except for the Vehiclegroup were subjected to abdominal injection with $CCl_4$ ($CCl_4$ : corn oil, 1 : 1 v/v; 0.5 ml/kg BW). One day later, blood and liver samples were collected to analyze biomarkers. All LED treatments elevated hepatic superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH peroxidase) activities, and reduced thiobarbituric reactive substances (TBARS), tumor necrosis factor-$\alpha$ (TNF-$\alpha$), interleukin-$1{\beta}$ (IL-$1{\beta}$) and interleukin-6 (IL-6), resulting in the reduction of glutamate-oxalate transaminase (GOT), glutamate-pyruvate transaminase (GPT) and lactic acid dehydrogenase (LDH) activities in plasma. These results indicate that LED effectively protected SD rat hepatotoxicity induced by $CCl_4$ through its antioxidative activity and reduction of some cytokines. The highest efficacy was found in LED 200 mg/kg BW, showing potential as a useful material for protection from hepatotoxicity in humans.

Structure of hemolymph node in rats (랫드의 혈림프절 구조)

  • It has been known that the hemolymph node is one of the lymphoid organs found normally in ruminants and some rodents, and this organ shares morphological and functional characteristics of lymph node and spleen. To clarify the rigorous definition of morphological structures and functions of the hemolymph node in Sprague-Dawley(SD) rats, we examined these organs of SD rats gross anatomically and light microscopically. The hemolymph nodes were normally found in the abdominal cavity and in the neck of SD rats. This organ was surrounded by a thin connective tissue capsule and there was a hilus. The parenchyma comprised a cortex of lymphatic nodules and diffuse lymphatic tissues, and a medulla of diffuse lymphatic tissues arranged in cords. Afferent and efferent lymph vessels were observed but there was no extensive subcapsular and medullary sinuses. These sinuses were filled with erythrocytes. The stroma of hemolymph nodes was composed of reticular cells and fibers, and many lymphocytes, granulocytes, erythrocytes, plasma cells, macrophages and megakaryocytes were supported by the reticular network. The above findings suggest that the hemolymph nodes of SD rats may take part in blood formation, blood filtration and immune reaction.

Acute Oral Toxicity of Alismatis Rhizoma in SD Rats (택사(Alismatis Rhizoma) 추출물의 급성 경구투여 독성 연구)

Methods : male and female rats were administered orally with Alismatis Rhizoma water extract of 1,000 mg/kg (low dosage group), 2,000 mg/kg(middle dosage group) and 4,000 mg/kg(high dosage group). We daily observed number of deaths, clinical signs and gross findings for 7 days. After 7 days, we measured body and organs weight. Also we analyzed hematological changes. Results : No dead SD rats and no clinical signs were found during the experiment period. Also other specific changes were not found between control and treated groups in hematology and serum biochemistry. In addition no significant changes of gross body and individual organs weight. Conclusions : These results suggest that water soluble extract of Alismatis Rhizoma has not acute oral toxicity and oral $LD_{50}$ value was over 4,000 mg/kg in SD rats.

Methods : Male and female rats were administered orally with Dictamnus dasycarpus Turcz water extract of 1,000 mg/kg(low dosage group), 2,000 mg/kg(middle dosage group) and 4,000 mg/kg(high dosage group). We daily observed number of deaths, clinical signs and gross findings for 7 days. After 7 days, we measured body and organs weight. Also we analyzed hematological changes. Results : No dead SD rats and no clinical signs were found during the experiment period. Also other specific changes were not found between control and treated groups in hematology. But we found out subtle changes in body weight and individual organ weight of the female group. In addition specific changes were observed in serum biochemical value of female group. Conclusions : These results suggest that water soluble extract of Dictamnus dasycarpus Turcz has not acute oral toxicity and oral $LD_{50}$ value was over 4,000 mg/kg in SD rats. Also Dictamnus dasycarpus Turcz is expected to be sensitive with respect to the female.

Methods : Male and female rats were administered orally with Alismatis Rhizoma water extract of 500 mg/kg(low dosage group), 1,000 mg/kg(middle dosage group) and 2,000 mg/kg(high dosage group). We daily observed number of deaths, clinical signs and gross findings for 14 days(twice a day). After 14 days, we measured body and organs weight. Also we analyzed hematological changes. Results : No dead SD rats and no clinical signs were found during the experiment period. Also other specific changes were not found between control and treated groups in hematology and serum biochemistry. In addition no significant changes of gross body and individual organs weight. Conclusions : These results suggest that water soluble extract of Alismatis Rhizoma has not repeated dose oral toxicity and oral LD50 value was over 2,000 mg/kg in SD rats. As a result, we can determine Alismatis Rhizoma is a relatively safe substance.
