Correlation of virus replication and spleen index in rock bream iridovirus infected rock bream Oplegnathus fasciatus

  • Rock bream iridovirus (RBIV) is a member of the Megalocytivirus genus that causes severe mortality to rock bream (Oplegnathus fasciatus) with characteristic clinical signs of spleen enlargement. In this study, we assessed spleen size and RBIV copy number patterns in RBIV-infected rock bream to determine lethal and safe levels of virus copy number/spleen index that may define disease progress. We found that rock bream infected with RBIV ($1.1{\times}10^7virus\;copy\;number/100{\mu}l$) and held at 29, 26, 23 or $20^{\circ}C$ exhibited significantly higher levels of spleen size compared to $17^{\circ}C$. In dead condition (100% mortality at $20{\sim}29^{\circ}C$), the spleen index ($spleen\;weight/fish\;weight{\times}100$) and virus copy number were 3.00~5.38 and $10^6{\sim}10^8/{\mu}l$, respectively. Conversely, in survived condition (0% mortality at $17^{\circ}C$), spleen index and virus copy number was as low as not-infected control ($0.34{\sim}1.22/10^0{\sim}10^1/{\mu}l$, respectively). These findings suggest that spleen index can be an indicator of disease severity of RBIV disease.

복부 CT 영상에서 비장의 웨이브 형태를 이용한 비장 비대의 자동 진단 (Computer-Aided Diagnosis of Splenic Enlargement Using Wave Pattern of Spleen in Abdominal CT Images)

  • 일반적으로 간경변에 동반된 비장은 비대한 것으로 알려져 있다. 본 연구는 간경변을 보유한 환자의 복부 CT 영상을 이용하여 비장의 웨이브 패턴(wave patten)을 관찰하였는데 정상간을 보유한 사람의 복부 CT 영상과 차이가 있음을 발견하였다. 간경변을 가지고 있는 환자의 복부 CT 영상에서 비장은 왼쪽 측면에 깊은 웨이브 부분이 존재하였다. 정상간의 경우에도 왼쪽 측면에 웨이브가 존재하기도 하나 깊이가 깊지 않음을 알 수 있다. 그러므로, 간경변을 가진 비장의 웨이브 부분들의 면적의 합이 정상간을 가진 비장의 웨이브 면적보다 크다는 것을 알 수 있다. 나아가, 간경변을 보유한 영상에서 비장의 웨이브 부분을 추출하여 원형성(circularity)을 살펴보았을 때 정상간에 수반된 비장의 웨이브 부분보다 더 원형에 가깝다는 것을 알 수 있었다. 본 논문은 위와 같이 관찰된 원리를 바탕으로 복부 CT 영상에서 비장의 웨이브 패턴을 이용하여 비장 비대를 효과적으로 진단하는 새로운 방법을 제시한다. 이는 단순히 비장의 크기만을 이용하지 않고 비장의 형태 변화로써 국소적 비장 비대를 자동 판정해 낼 수 있음을 말해주는 것이다.

참돔이리도바이러스 감염 돌돔에서 임상적 변화와 viral genome copy number 간의 상관관계 (Correlation between clinical changes and viral genome copy number in rock bream infected with red sea bream iridovirus)

  • In this study, the correlation between clinical changes and RSIV genome copy number was investigated to determine the quantitative criteria for the characteristics of RSIV infection. The rock bream (Oplegnathus fasciatus) was intraperitoneally injected with three different doses (1.0×101, 1.0×103 and 1.0×105 viral genome copies/fish) as low, medium, and high doses, respectively. The clinical signs (spleen enlargement, death) observation and real-time PCR were conducted at 5, 10 and 14 days post-injection. During the experiment, spleen index as a quantitative indicator for spleen enlargement was continuously increased in the medium- (up to 2.26) and high-dose (up to 4.99) challenge groups, respectively. Notably, when the spleen index was over 1.5, 2.0, 2.5 and 3.0, a positive correlation was revealed with average viral genome copy numbers of 2.51, 3.37, 4.97 and 5.43×107 viral genome copies/mg, respectively. Moreover, the threshold of spleen index over 1.5 was 1.0×106 viral genome copies/mg, while the thresholds of spleen index over 2.0 and dead was 2.51×107 viral genome copies/mg and the thresholds of spleen index over 2.5 and 3 was 3.98×107 viral genome copies/mg. These findings suggest the possibility of quantitatively analyzing the characteristics and development process of RSIV infection.

랫드를 이용한 Acetanilide의 반복투여 및 생식/발생독성 병행시험 (Repeated Dose and Reproductive/Developmental Toxicities of Acetanilide in Rats)

  • The study was conducted to assess the repeated dose and reproduction and developmental toxicities of acetanilide, an intermediate for drug production, as a part of OECD Screening Information Data Set (SIDS) program. The test agent was administered by gavage at dose levels of 0, 22, 67, 200 and 600 mg/kg to Sprague-Dawley rats (12/group/sex) during pre-mating and mating period for males(up to 30 days) and females and pregnancy and early lactation period for females (up to 39-50 days). At 22 mg/kg, decreases in HGB, HCT (males) and MCHC (females), hyperplasia of spleen red pulp, hyperplasia of femur bone marrow (both sexes) were observed. At 67 mg/kg, salivation (males), reduced food consumption (both sexes), decreases in RBC, HGB, HCT and MCHC (males), increases in MCV (males) and spleen weight (males), hyperplasia of spleen red pulp and femur bone marrow (both sexes) were observed. At 200 mg/kg, decreases in locomotor activity and salivation (both sexes), reduced food consumption (both sexes), decreases in RBC, HGB, HCT and increases in MCV, MCH, BUN, T-BIL (males), enlargement of spleen (both sexes), increased weight of spleen (males), hyperplasia of spleen red pulp and femur bone marrow and extramedullary hematopoiesis of liver (both sexes), atrophy of thymus and corpus luteum hyperplasia of ovary (females) were observed. At 600 mg/kg, decreases in locomotor activity, cyanosis (both sexes), reddish tear, and salivation (males), mortality (4 out of 12 females), decreased body weight (females), reduced food consumption (both sexes), decreases in RBC, HGB, HCT and MCHC and increases in WBC, MCV, MCH, reticulocyte, neutrophil, lymphocyte, monocyte, AST, ALT, BUN, T-BIL, ALB, Ca and A/G ratio (males), enlargement of spleen, increased weights of spleen (both sexes), liver (males), kidney and ovary, decreased weights of thymus (females), hyperplasia of spleen red pulp, hyperplasia of femur bone marrow and extramedullary hematopoiesis of liver (both sexes), and atrophy of thymus and corpus luteum hyperplasia of ovary (females) were observed. Regarding the reproduction and development toxicities, there were no treatment-related changes in precoital time, mating index, fertility index and pregnancy index at all doses tested. At 22 and 67 mg/kg, there were no adverse effects on all the parameters observed. At 200 mg/ kg, decreased body weight of pups (day 4 p.p.) were observed. At 600 mg/kg, decreased body weight of pups (day 0 and 4 p.p.) and viability index (day 4 p.p.), increased incidence of newborns dead or with abnormal clinical signs were observed. The results suggest that the NOAELs for general toxicity are < 22 mg/kg, LOAELs are 22 mg/kg and the NOAELs for reproductive toxicity are 67 mg/kg.

초음파검사에 의한 20대 정상 성인의 비장 크기 (A Spleen Size Measured by Ultrasound in 20's Normal Adults)

  • 비장종대를 일으킬 수 있는 질환에는 감염성질환, 간경화, 기타 간질환, 비정상혈구에 의한 혈류질환, 림프계질환, 면역성질환 등이 있다. 따라서, 정상적인 비장의 크기를 아는 것은 비장의 정상 여부와 비장종대의 진단에 도움을 주며, 추적검사에도 유용하게 이용될 수 있다. 초음파검사에서 최대길이 12cm 미만, 두께 5cm 미만이면 정상적인 크기라고 여겨지나 성별, 인종에 따라 다를 수 있다. 저자들은 우리나라 20대 학생들을 대상으로, 초음파 검사로 비장크기를 측정하여, 비장 크기의 정상범위를 제시하고, 신장과 체중에 따른 비종종대 기준의 상한치 조정이 필요한지 알아보고자 한다. 측정결과 남자 비장의 길이는 10.95±1.07 cm(8.9~13.9), 폭은 4.48±0.61 cm 이었으며, 여자 비장의 길이는 9.20±1.30 cm(6.8~12.0), 폭은 3.55±0.44 cm으로 남녀 간에 평균에 유의한 차이가 있었다(p<0.001). 신장의 크기가 증가할수록 비장의 길이는 증가하였으며(r=.57), 체중이 증가함에 따라서 비장의 길이가 증가하는 뚜렷한 양적 선형관계가 있었다(r=.63). 정상 성인에서 비장의 길이가 12cm를 초과한 남자는 5명, 여자는 2명 이었으며, 13cm를 초과한 경우는 남성 2명이었다.

Dose Dependence of the Severity of Radiation-Induced Thymic Lymphoma in Mice

  • The dose dependence of the severity of radiation-induced thymic lymphoma in C57BL/6J mice was studied. Mice were exposed to fractionated irradiation at the total doses of 4.0, 6.0 and 8.0 Gy (four irradiations at 8-day intervals) starting from 33 days after birth. Pathological and histological changes of each mouse were observed after periodical sacrifice at day 75, 100, 125, 150, 175, 200, 250, 300 after the first irradiation. The severity of cancers were classified into 4 stages by clinical signs with respect to the enlargement of the thymus, spleen, liver, the progression of the cancer in the thymus, and the metastasis to the spleen, liver, lung and the lymphatic nodes. Among the 490 mice observed, 146 mice had thymic lymphoma. A clear dose-effect relationship was observed as well as the dose-response relationship. Also, periodical observation showed that thymic lymphoma was first induced in mice sacrificed at day 100 (130days old), and metastasize in the order of spleen, lung, liver and then the lymphatic nodes. The results suggest that radiation may be involved not only as a tumor initiator but also as a tumor promoter, and a tumor progression-enhancing agent.

Molecular and Histopathological Evidence of Mycobacteriosis in Paradise Fish Macropodus opercularis Imported into Korea

  • We report on mycobacteriosis in an imported tropical ornamental fish Macropodus opercularis commonly known as the paradise fish. Mass mortality occurred in paradise fish imported to Korea from Southeast Asia in 2008. The affected fish did not show any outward clinical signs, but enlargement of the spleen, kidneys, and liver was observed on dissection. Histopathological examination revealed numerous granulomas in the spleen, and acid-fast bacilli were observed in the centers of the granulomas. About 65% of spleen DNA samples were PCR positive using mycobacteria-specific primers targeting the 16S rRNA and hsp65 genes. The nucleotide identities of the 16S rRNA and hsp65 genes with those of Mycobacterium marinum were 99.5% and 99.4%, respectively. Although the bacterium was not cultured, the molecular diagnosis and histopathological findings were consistent with mycobacteriosis in paradise fish.

  • 1. The obesity is the aggregation of symptoms accompanied with the metabolic disorder and the increase of fatty layer accumulation by the enlargement or increase of fatty cells, which is the condition that has 20% overweight than the standard. 2. The final metabolic of fat depends on the defensive energy. The shape of fatty layer is changed by the point where is the defensive energy is replenish or not. So Goin, Bein and Yuckin is made a discriminated and there is different between Besudaeso and Herlgidaso. 3. In case of liver overacting spleen by seven emotions, we can use reinforcing the spleen and don't use depressing energy. 4. Damp, weakness of Qi(氣), weakness of spleen came from seven emotion, and social stress can occur obesity. Obesity is reason of C.V.A, dizziness, dementia, schizophrenia, etc. 5. The obesity which can occurs various adult diseases came from eating disorder and various stresses at the standpoint of western medical. This is cured by diet, kinesiatrics, operation, pharmacotherapy, etc.

Expressional Analysis of STAT2 Gene in Rock Bream, Oplegnathus faciatus, Under LPS or Poly I:C Stimulation and Megalocityvirus Infection

  • Rock bream iridovirus (RBIV) is a megalocytivirus widely infected in various fish species in Korea, causing symptoms of acute inflammation and enlargement of spleen. In our previous study, RBIV induced the initial upregulation but later down-regulation of proinflammatory cytokines and IFN1 gene expression. Signal transducers and activators of transcriptions (STAT) are transcription factors involved in the regulation of immune genes including IFNs. This study was conducted to analyse the expression of STAT2. The expressional study of STAT2 gene was performed in head kidney and spleen upon RBIV infection and immune stimulants like LPS or poly I:C in vitro. Consequently, STAT2 gene expression pattern was different in head kidney and spleen as it was significantly up-regulated by LPS from 4 h to 8 h but down-regulated at 24 h while up-regulated by poly I:C at 8 h in head kidney while, in spleen, STAT2 gene expression was down regulated by LPS but significantly up-regulated by poly I:C. Upon RBIV stimulation, STAT2 gene expression was significantly down-regulated by high dose RBIV at 4 h but up-regulated at 8 h and 24 h in head kidney. In spleen cells, it was up-regulated by medium dose RBIV at 4 h and by high dose RBIV at 4 h and 8 h but down regulated later then. In vivo, STAT2 gene expression was not significantly affected by RBIV infection while significant up-regulated by vaccination at day 7 post-vaccination, indicating STAT2 gene can be involved in adaptive immune response in rock bream.

마우스에서의 고선량(高線量) 방사선(放射線)에 의한 만성장해(慢性障害) (Late Biological Effect of High-dose Radiation in the Mice)

  • Radiation-induced life shortening, carcinogenesis and other pathological changes were investigated in NIH(GP) mice after $^{60}Co-{\gamma}$ ray irradiation(100~700rads). The results were summarized as follows: 1. There were little difference in body weights, hematological examination and other clinical findings between normal and irradiated groups. 2. Mean survival time of the mice after irradiation were decreased dose-dependently. 3. Main gross findings of the mice irradiated were appeared as enlargement of spleen, thymus and lymph nodes, tumorous nodules of lung and cyst of ovary. Especially, enlargement of thymus was promineort in high dose group. 4. Microscopically, there were various findings including myeloid leukemia, thymic lymphoma, lung adenoma, adenosquamous cell carcinoma of pancreas, pneumonia and other pathological changes. Especially thymic lymphoma was highly frequent in the 700 rads group.
