Retinoic acid loaded with chitosan nanoparticles improves spermatogenesis in scrotal hyperthermia in mice

  • Objective: High temperatures can trigger cellular oxidative stress and disrupt spermatogenesis, potentially leading to male infertility. We investigated the effects of retinoic acid (RA), chitosan nanoparticles (CHNPs), and retinoic acid loaded with chitosan nanoparticles (RACHNPs) on spermatogenesis in mice induced by scrotal hyperthermia (Hyp). Methods: Thirty mice (weighing 25 to 30 g) were divided into five experimental groups of six mice each. The groups were as follows: control, Hyp induced by a water bath (43 ℃C for 30 minutes/day for 5 weeks), Hyp+RA (2 mg/kg/day), Hyp+CHNPs (2 mg/kg/72 hours), and Hyp+RACHNPs (4 mg/kg/72 hours). The mice were treated for 35 days. After the experimental treatments, the animals were euthanized. Sperm samples were collected for analysis of sperm parameters, and blood serum was isolated for testosterone measurement. Testis samples were also collected for histopathology assessment, reactive oxygen species (ROS) evaluation, and RNA extraction, which was done to compare the expression levels of the bax, bcl2, p53, Fas, and FasL genes among groups. Additionally, immunohistochemical staining was performed. Results: Treatment with RACHNPs significantly increased stereological parameters such as testicular volume, seminiferous tubule length, and testicular cell count. Additionally, it increased testosterone concentration and improved sperm parameters. We observed significant decreases in ROS production and caspase-3 immunostaining in the RACHNP group. Moreover, the expression levels of bax, p53, Fas, and FasL significantly decreased in the groups treated with RACHNPs and RA. Conclusion: RACHNPs can be considered a potent antioxidative and antiapoptotic agent for therapeutic strategies in reproductive and regenerative medicine.

액상 정액의 보존 기간이 정자 기능 및 정자염색질 구조 분석에 미치는 영향과 인공수정 분만율과의 상관관계 (Effect of Storage Times on Sperm Function, Sperm Chromatin Structure Assay (SCSA) and Correlations Between Fertility and SCSA in Boars)

  • 본 연구는 인공수정 센터에서 이용되고 있는 종모돈의 인공수정 분만율의 차이를 기준으로 하여 정액의 보존 일에 따른 정자의 생존율, CASA, HOST, IVP 및 SCSA를 조사하여 이들 정자의 분석법들과 인공수정 분만율과의 상관관계를 조사하여 종모돈의 수정 능력을 예측할 수 있는 정액 평가법을 개발하고자 실시하였다. 종모돈의 액상정액을 대상으로 CASA, HOST, IVP 및 SCSA 기법을 적용한 정액의 평가 결과는 다음과 같았다. 정액의 보존 일별 HOST 결과는 보존 일이 경과함에 따라 유의적으로 감소하였고(P<0.05), IVP 결과 난자 당 침투 정자 수는 보존 3일째에 유의적으로 감소하였다(P<0.05). 보존 0일과 6일에 HOST는 분만율이 80% 이상인 종모돈 군이 70% 미만인 종모돈 군 보다 유의적으로 높았고, 3일에서는 80% 이상인 종모돈 군이 다른 종모돈 군에 비하여 높았다(P<0.05). IVP는 보존 0일에 분만율이 80% 이상인 종모돈 군이 70% 미만인 종모돈 군보다 난자당 침투 정자 수가 유의적으로 많았다(P<0.05). %Red는 보존 0일과 3일에는 분만율이 80% 이상과 70% 미만인 종모돈 군 간에 유의적인 차이를 보였으나 보존 6일에서는 모든 종모돈 군간에 유의적인 차이가 있었다(P<0.05). %Red(r=0.79, P<0.01 ; r=0.86, P<0.01 ; r=0.88, P<0.01)는 정액의 보존 일이 증가함에 따라 분만율과의 부의 상관 계수가 증가하였다. 이상의 결과를 종합해 보면 SCSA는 정액의 수정 능력을 평가하는데 있어서 매우 유용한 방법이며, 정액의 보존 일은 여러 가지 정자 기능과 SCSA 결과에 크게 영향을 주는 것으로 나타났다. 정액의 수정 능력을 평가함에 있어서 정액의 보존 일령을 고려하고 정자 기능 검사와 SCSA를 병행하여 실시할 경우 종모돈의 수정 능력 평가 시 정확성을 더욱 높일 수 있는 유용한 방법이 될 수 있을 것으로 사료된다.

미세술기를 이용한 정계정맥류절제술이 정자 핵 내 DNA Integrity에 미치는 효과 (Effect of Microsurgical Varicocelectomy on Human Sperm Nucleus DNA Integrity)

  • 목 적: 인간 정자 핵 내의 DNA integrity는 배아의 발달 및 임신 유지에 중요한 역할을 하여 DNA integrity가 손상된 경우 불임과 유산의 원인이 된다고 하며, 정계정맥류는 DNA 손상을 일으키는 대표적인 원인 중 하나이다. 본 연구에서는 미세술기를 이용한 정계정맥류절제술로 정계정맥류를 교정을 하였을 때 정자 핵 내 DNA integrity가 어떠한 영향을 받는지에 대하여 알아보았다. 연구방법: 2006년 4월부터 2007년 4월까지 불임을 주소로 미세술기를 이용한 정계정맥류절제술을 받았던 18명의 환자에서 수술 전 후에 정액검사의 다른 지표들과 함께 정자 핵 내 DNA integrity가 어떻게 변화하였는지 조사하였다. 정자 핵 내 DNA integrity를 측정하는 방법으로 comet assay를 시행하였고, comet assay를 통한 DNA 손상 정도는 DNA fragmentation index (DFI)로 나타내었다. 결 과: 수술 후 4개월에 모든 환자에서 재발의 소견은 보이지 않았으며, DNA 손상 정도를 나타내는 평균 DFI는 수술 전에 19.3%, 수술 후에 13.7%로 유의한 변화를 보였다. 수술 전 DFI가 10 이상으로 비정상인 14명의 환자들 중 12명 (85%)에서 개선 소견을 보였으나, 수술 전 DFI가 10 미만인 정상 환자 4명에서는 1명 (25%)만이 개선 소견을 보였다. 수술 후 정자의 밀도, 운동성, 생존성에서 호전 양상을 보였으나 유의한 차이는 없었다. 결 론: 미세술기를 이용한 정계정맥류절제술을 통한 수술적 교정은 정액검사상의 다른 지표의 개선 뿐 아니라, 정자 핵 내 DNA 손상을 감소시킬 수 있다. 이상에서 정계정맥류의 수술적 교정으로 정자 핵 내 DNA integrity의 개선을 기대할 수 있으며, 이는 보다 양호한 정자를 많이 얻을 수 있어 자연임신이나 보조 생식술의 성공 가능성을 높일 수 있다는 점을 제시한다.

감마선에 조사된 생쥐에 있어서 방사선방어효과 평가를 위한 생물학적 파라메타 (Biological Parameters for Assessing Radioprotective Effects in ${\gamma}-irradiated$ Mice)

  • This study deals with the biological changes in mice after ${\gamma}-irradiated$. Four weeks old BALA/c mice were irradiated with 6.5Gy of ${\gamma}-ray$ on the fifth day after oral administration of radioprotectants such as ascorbic acid, tocopherol and cysteine. Control group was irradiated with 6.5Gy without pre-administration of radioprotectors. Blood cells and sperm cells were counted and body, testis and spleen were weighed 3 days after irradiation. And also liver antioxidant activity and range of spleen immune cells were measured. Differences in most biological parameters were not clearly distinguished between experimental groups. However, the relative spleen weight, the relative testis weight and the population size of spleen immune cells such as T helper cells, B cells and macrophages measured by means of FACS showed significant difference between irradiated and radioprotectant administered group. It is concluded that the relative spleen weight, the relative testis weight and the population size of spleen immune cells are easy and useful parameters for assessing the effect of radioprotective substances and for quantifying biological damage of radiation, as well.

쇄양(鎖陽)의 농도별(濃度別) 투약(投藥)이 수컷 생쥐 생식능력(生殖能力)에 미치는 영향(影響) (Effects of Cynomorii Herba Extract Solution on Reproductive Capacities in Mice)

  • Purpose : These studies were undertaken to evaluate the effects of the Cynomorii Herba (CH) on the spermatogenic abilities such as the concentration, motility and morphological normality of sperm from the testis, and the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. Materials and Methods : We used the 2-month-old mice and administered 0.2ml extract solution of CH in the 0.1mg/ml, 1mg/ml, 10mg/ml and 100mg/ml once a day for 60days. The control group was administered the distilled water in the same way. After the administration of extract solution, we examined the number of total, motile and normal sperm from the cauda epididymis, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. We observed the histological changes of isolated testis and compared to the testicular tissue especially seminiferous tubules between control and CH groups by histochemical method. Results : The concentration of total sperm and the motility of spermatozoa were significantly increased in the 1mg/ml, 10mg/ml and 100mg/ml CH groups, especially in 10mg/ml group, compared to the control group. The significant differences were observed in the normality of spermatozoa of the CH groups compared to the control group. In the histolocal analysis of the testicular tissues, the enlargement of testicular lobe diameter and apparent vasculogenesis between testicular lobes were observed in the CH groups compared to the control group. Also, the activity of hyaluronidase was significantly increased in the CH groups compared to the control group. In the antioxidant activity analysis, the activities of testicular peroxidase and testicular catalase were significantly increased in the CH groups compared to the control group, respectively. Conclusion : This study shows that CH has the beneficial effect on the concentration, morphology and motility of sperm, the activities of sperm hyaluronidase, testicular peroxidase and testicular catalase. We can suggest that CH extract solution be useful for the treatment of male sexual dysfunctions and infertility.

정자의 여러가지 형태학적 특징에 따른 정자 침투능의 평가 (Evaluation of Sperm Penetration Ability according to Various Morphological Characteristics of Spermatozoa)

  • Morphological estimation of human spermatozoa is complicated by the fact that there is great natural variation in shape. This natural variation in shapes makes it difficult to say which forms are associated with infertility and which are normal variations. Possibly post coital test or in vitro cervical mucus penetration tests will help to clarify this question by showing which sperm are capable of penetration. The purpose of this investigation was performed to assess distribution of various morphological abnormalities according to the ability of sperm to penetrate cervical mucus. The sperm-mucus penetration using hen's egg white as substituting mucus for human cervical mucus was done in 45 fertile men with normal semen analysis and 122 infertile men with abnormal seminal parameters more than one. The female partners of 122 infertile couples showed normal results in the female fundamental test for fertility. Conventional semen analysis was evaluated according to the WHO standard normal(l980). The detailed classification of the abnormal sperm was made according to David et al(l975). The vitality of the sperm samples determined by eosin yellow-nigrosin stainig according to the method of Eliasson(l977). Results were as follw; 1. The patients had significantly lower total sperm count, motility (%), normal morphology (%), viability and total functional sperm fractions(TFSF) than fertile donors. 2. The mean value of sperm penetration distance of the patients(28.69${\pm}$11.02mm) showed significantly lower than fertile donors(37.33${\pm}$5.49mm). And 43/45 fertile donors(95.5%) as well as 57/122 patients(46.7%) had over 30mm in sperm penetration distance respectively. While 2/45 fertile donors(4.5 %) and 65/122 patient(53.3%) had under 30mm in sperm penetration distance respectively. 3. The morphological abnormalities in fertile donors were significantly lower 23.04${\pm}$5.83% (head = 12.89${\pm}$4.98, neck=6.11${\pm}$3.83%, and tail=3.43${\pm}$2.65%), compared to 36.03${\pm}$14. 40% in patients(head = 15.98 8.60%, neck 11.20${\pm}$6.56% and tail=8.70${\pm}$6.55%). Also, 3 types of sperm abnormalities including head, neck and tail were significantly lower in patient than fertile donors, respectively. Both the patients and fertile donors showed higher distribution of sperm with abnormal head than abnormal neck and tail. 4. The mean morphological abnormalities(SP>30mm) of the patients(30.68 11.64%; head = 15.95${\pm}$9.35%, neck=8.14${\pm}$4.21 %, tail=6.56${\pm}$5.64%) were significantly lower compared to patients(40.72${\pm}$15.01 %; head=16.02${\pm}$7.69%, neck 13.89${\pm}$7.82%, tail=1O.58${\pm}$6.75%) under 30mm in sperm penetration distance. Also, both groups over 30mm and under 30mm in sperm penetration showed distance higher distribution of sperm with abnormal head than abnormal neck and tail. The morphological abnormalities of head did not show significant difference but abnormal neck and tail were significant difference between the over 30mm and under 30mm group in sperm penetration distance.

Evaluation of sperm protamine deficiency and apoptosis in infertile men with idiopathic teratozoospermia

  • Objective: Sperm morphology plays an important role in infertility, especially in cases of defects in the heads of spermatozoa. Tapered-head or elongated-head spermatozoa are examples of morphological abnormalities. The aim of this study was to compare the semen parameters, levels of protamine deficiency, and frequency of apoptosis between patients with normozoospermia and those with teratozoospermia with tapered-head spermatozoa. Methods: Fifty-two semen samples (27 patients with tapered-head sperm and 25 fertile men) were collected and semen analysis was performed according to the World Health Organization criteria for each sample. Protamine deficiency and the percentage of apoptotic spermatozoa were evaluated using chromomycin A3 (CMA3) staining and terminal deoxynucleotidyl transferase dUTP nick-end labelling (TUNEL) assays, respectively. Results: Sperm concentration, motility, and normal morphology in the tapered-head spermatozoa (cases) were significantly lower than in the normozoospermic samples (controls). CMA3-reactive spermatozoa (CMA3+) in the case group were more common than in the controls. Apoptotic spermatozoa (TUNEL-positive) were significantly more common in the cases than in the controls. Conclusion: This analysis showed that tapered-head spermatozoa contained abnormal chromatin packaging and exhibited a high rate of apoptosis, which can be considered to be an important reason for the impaired fertility potential in teratozoospermic patients with tapered-head spermatozoa.

3-MCPD의 생식ㆍ발생독성에 관한 연구 (Study on the Reproductive and Developmental Toxicity of 3-MCPD)

  • 3-Monochloro-1,2-propanediol(3-MCPD) is a toxic compound, often present in different foods containing acid hydrolyzed(AH) protein, like seasonings and savory food products. The purpose of the present study was to investigate the effects of 3-MCPD on male fertility, sperm and testosterone secretion. In vivo male fertility test was performed for observing the adverse effects of 3-MCPD on the function of male reproductive system and pregnancy outcome. 0.01, 0.05, 0.25, 1 and 5 mg/kg b.w. of 3-MCPD was given daily by gavage to groups of 15 adult male SD rats for 4 weeks. At the end of pre-treatment period, males were mated overnight with normal females. Following morning, males demonstrating successful induction of pregnancy were sacrificed on that day to assess sperm parameters and histopathology of reproductive organs. The resulting pregnant females were sacrificed on day 20 of gestation to evaluate pregnancy outcome. As a result, four-week paternal administration with 3-MCPD resulted in adverse effects on male fertility and pregnancy outcome without remarkable histopathological changes in testes and epididymides; sperm motility, copulation index and fertility index were markedly decreased in the treated group and numbers of live fetuses showed steep dose-response curves. Also, spermatogenesis was investigated in this experiment. However, no effect was observed on production of sperm in testes treated with 3-MCPD for 4 weeks. Hormone assay was performed for observing the effects of 3-MCPD on testosterone and luteinizing hormone (LH) in blood and testes of male SD rats and cultured primary Leydig cell. In result, significant changes of related hormones did not observed by treatment of 3-MCPD. These results indicated that paternal treatment with 3-MCPD induced spermatotoxic effect, which caused an antifertility on male.

Washing solution and centrifugation affect kinematics of cryopreserved boar semen

  • Cryopreservation is a widely-used efficient means of long-term sperm preservation. However, unlike other types of semen, cryopreserved boar semen has reduced fertility and the efforts continue to optimize post-thawing sperm recovery. In this study, we evaluated the effects of various washing solutions (Hulsen solution, lab-made DPBS and commercial DPBS) on post-thawing porcine sperm kinematics (CASA system), viability (SYBR-14/PI) and acrosome integrity (PSA/FITC). We also examined the effect of washing-centrifugation on frozen-thawed semen kinematics. The results indicate that type of washing solution and post-thawing centrifugation alters parameters linked to sperm quality (total motility, progressive motility, viability and acrosome integrity). Significantly higher (p < 0.05) motility and progressive motility were obtained when cryopreserved semen was processed with Hulsen solution. The post-thaw percentage of live and intact acrosomal sperm was significantly higher in group 1 (Hulsen solution) as compared to other groups. Following thawing-centrifugation, the results showed significantly higher motility and progressive motility in group 1 than other groups. However, the latter two DPBS groups did not differ statistically. Taken together, Frozen-thawed spermatozoa motility, acrosome integrity and viability can be affected by the type of washing solution used. Moreover, centrifugation of frozen-thawed semen has an unfavorable effect on total motility and progressive motility.

Green tea extract addition into a Tris-based egg yolk extender improves Bali bull sperm quality

  • Objective: The conservation of Bali bulls, the Indonesian native breed of cattle, is crucial for cattle breeding in Indonesia. To guarantee the spread of Bali bulls through artificial insemination the quality of the frozen semen must be high. To this end, adding an extender material to semen that increases spermatozoa's survival during cryopreservation is important. Green tea extract (GTE) can be used as cryoprotectant because its high antioxidant activity can help avoid reactive oxygen species formation. Methods: Semen of five Bali bulls from the National Artificial Insemination Center at Singosari, Indonesia was collected routinely twice a week. First, fresh semen inspection was performed to determine the feasibility of using Bali bulls as animal samples. The extender used in this study was Tris-based egg yolk. The samples were divided into four treatments: T0, no GTE added to the extender; T1, 0.05 mg GTE plus 100 mL extender; T2, 0.10 mg GTE plus 100 mL extender; and T3, 0.15 mg GTE plus 100 mL extender. The semen freezing process was conducted according to standard procedures and sperm quality parameters, i.e., sperm motility, viability, abnormalities, and membrane integrity observed pre-freezing and post-thawing. Results: There were significant differences in total motility, progressive motility, viability, and integrity membrane of Bali bull sperm at both pre-freezing and post-thawing after adding GTE into the extender. In contrast, there were no differences in abnormalities among treatments. Conclusion: Adding GTE at a 0.15 mg into 100 mL Tris-based egg yolk extender can improve the quality of cryopreserved Bali bull sperm.
