Effect of graded levels of soapnut (Sapindus mukorossi) shell powder on reproductive performance in broiler breeders

  • Objective: This study investigated the effects of soapnut (Sapindus mukorossi) shell powder (SSP) on serum hormone level, egg quality, semen characteristics and reproductive performance of broiler breeders fed with a maize-soybean meal based diet. Methods: Ninety six female and twenty four male CARIBRO-VISHAL broiler breeders, 38-week old, were individually caged and randomly allocated to four treatment groups (24 female breeders/treatment and 6 male breeders/treatment): an un-supplemented control (T1) and three groups with 0.0176% SSP (group T2), 0.026% SSP (group T3) and 0.0528% SSP (group T4), to have supplementary saponin at 0, 50, 75, and 150 ppm, respectively, for 42 days. Results: The results indicated that serum (p<0.001) and seminal plasma (p<0.05) testosterone level, semen volume (p<0.001), mass motility (p<0.001), and live spermatozoa count (p<0.001) was increased in groups T3 and T4 compared to T2 and control groups. Compared with control group, total sperm count was increased (p<0.001) and dead spermatozoa count was decreased (p<0.001) in SSP supplemented groups. Supplementation of SSP did not affected the quality of egg lay. Compared with control group, fertility (p<0.01) and hatchability (total eggs set and fertile eggs set) (p<0.001) were significantly improved in SSP supplemented groups with the highest improvement in T3 treatment group. Embryonic death was decreased (p<0.001) in SSP supplemented groups compared to control; lowest embryonic death was recorded in T3 treatment group. Conclusion: Thus, it was concluded that dietary supplementation of 0.026% SSP (saponin equivalent 75 ppm) improved the reproductive performance of broiler breeders.

Dual trigger in normally-responding assisted reproductive technology patients increases the number of top-quality embryos

  • Objective: The feasibility of a gonadotropin-releasing hormone agonist (GnRHa) trigger in normal responders is still a matter of debate. The aim of this study was to compare the number of mature oocytes, the number of good-quality embryos, and the live birth rate in normal responders triggered by GnRHa alone, GnRHa and human chorionic gonadotropin (hCG; a dual trigger), and hCG alone. Methods: A retrospective cohort study was conducted at the infertility clinic of a university hospital. Data from 200 normal responders who underwent controlled ovarian hyperstimulation and intracytoplasmic sperm injection with a GnRH antagonist protocol between January 2016 and January 2017 were reviewed. The first study group consisted of patients with cycles triggered by GnRHa alone. The second study group consisted of patients with cycles triggered by both GnRHa and low-dose hCG (a dual trigger). The control group consisted of patients with cycles triggered by hCG alone. Results: The groups were comparable in terms of demographics and cycle characteristics. The numbers of total oocytes retrieved and metaphase II oocytes were similar between the groups. The total numbers of top-quality embryos were 3.2±2.9 in the GnRHa group, 4.4±3.2 in the dual-trigger group, and 2.9±2.1 in the hCG group (p=0.014). The live birth rates were 21.4%, 30.5%, and 28.2% in those groups, respectively (p=0.126). Conclusion: In normal responders, a dual-trigger approach appears superior to an hCG trigger alone with regard to the number of top-quality embryos produced. However, no clinical benefit was apparent in terms of live birth rates.

임신중인 생쥐에 Bisphenol A 투여 후 태어난 차산자의 생식독성과 혈액성분에 관한 연구 (Studies on the Reproductive Toxicant and Blood Metabolite in Pups Born After Bisphenol A Administration in Pregnant Mice)

  • Bisphenol A (BPA) is a monomer used in the manufacture of a multitude of chemical products, including epoxy resins and polycarbonate. The objective of this study was to evaluate the effects of BPA administration on reproductive characteristics and blood hematological and chemical values in offspring of pregnant dams treated with BPA. BPA was administrated to pregnant mice by intraperitoneally injection with 0, 0.05, 0.5 and 5.0 mg/kg B.W. for 5 times at 3 days interval on gestation days 1-16. There were no treatment-related effects of BPA on reproductive organ weight in male offsprings at 45 days-of-age, but body weight was the lowest in 5.0mg BPA group when compared to other groups (P<0.05). No differences in semen characteristics (sperm concentration, viability, motility and abnormality) were observed between the control and BPA treatment groups. The WBC, HB, HT, MCV, MCH, MCHC, albumin, BUN and total protein of blood hematological and chemical values in male offsprings were not difference for any treatment groups, but RBC value in BPA groups was significantly increased comparing to the control group (P<0.05). The PLT value was slightly higher in 5.0mg BPA groups than in any other group, but not significantly difference among the experimental groups. In female offsprings, the effects of BPA didn't affect to the body and ovary weight, but the uterus weight in 5.0mg BPA group was slightly heavier than that of control group (P>0.05). No statically significant difference in blood hematological values in female offsprings were observed between the control group and BPA groups, but the concentration of albumin and BUN were significantly higher in 0.5mg BPA group when compared to control and other BPA treatment groups (P<0.05). The histological evaluation of testis and ovary in growing offspring at 45 days-of-age was not difference between the control group and BPA groups, but endometriosis of the uterus in female offspring was dramatically increased in 0.5 and 5.0mg BPA groups. These founding suggest that low concentration of BPA might not have a important role on reproductive ability or blood metabolite in offspring of pregnant dams treated with BPA.

  • Objective: This study was to compare the characteristics between parthenogenetic mES (P-mES) cells and in vitro fertilization mES cells. Materials and Methods: Mouse oocytes were recovered from superovulated 4 wks hybrid F1 (C57BL/6xCBA/N) female mice. For parthenogenetic activation, oocytes were treated with 7% ethanol for 5 min and $5{\mu}g$/ml cytochalasin-B for 4 h. For IVF, oocytes were inseminated with epididymal sperm of hybrid F1 male mice ($1{times}10^6/ml$). IVF and parthenogenetic embryos were cultured in M16 medium for 4 days. Cell number count of blastocysts in those two groups was taken by differential labelling using propidium iodide (red) and bisbenzimide (blue). To establish ES cells, b1astocysts in IVF and parthenogenetic groups were treated by immunosurgery and recovered inner cell mass (ICM) cells were cultured in LIF added ES culture medium. To identify ES cells, the surface markers alkaline phosphatase, SSEA-1, 3,4 and Oct4 staining were examined in rep1ated ICM colonies. Chromosome numbers in P-mES and mES were checked. Also, in vitro differentiation potential of P-mES and mES was examined. Results: Although the cleavage rate (${\geq}$2-cell) was not different between IVF (76.3%) and parthenogenetic group (67.0%), in vitro development rate was significantly low in parthenogenetic group (24.0%) than IVF group (68.4%) (p<0.05). Cell number count of ICM and total cell in parthenogenetic b1astocysts ($9.6{\pm}3.1,\;35.1{\pm}5.2$) were signficantly lower than those of IVF blastocysts ($19.5{\pm}4.7,\;63.2{\pm}13.0$) (p<0.05). Through the serial treatment procedure such as immunosurgery, plating of ICM and colony formation, two ICM colonies in IVF group (mES, 10.0%) and three ICM colonies (P-mES, 42.9%) in parthenogenetic group were able to culture for extended duration (25 and 20 passages, respectively). Using surface markers, alkaline phosphatase, SSEA-l and Oct4 in P-mES and mES colony were positively stained. The number of chromosome was normal in ES colony from two groups. Also, in vitro neural and cardiac cell differentiation derived from mES or P-mES cells was confirmed. Conclusion: This study suggested that P-mES cells can be successfully established and that those cell lines have similar characteristics to mES cells.

  • 본 연구는 개 정액 채취방법에 따른 정액성상과 동결전 pentoxifylline의 첨가가 미치는 영향을 조사하였고, CASA를 이용하여 정자의 운동성 측정 및 생존율, 첨체 온전성, 저삼투성 팽창화를 조사하였다. 그리고 자연발정과 발정유기된 암캐에서 동결정액 주입의 임신율을 조사하였다. 1.품종별 정액성상을 조사한 결과 Beagle, English cockers spanie, Collie 및 Grey hound의 정액량과 정자 수에서 큰 차이가 없었으나, Shihtzu에서 정액량과 정자수가 유의적은 아니나 다소 적었다. 2.동결희석액에 pentoxifylline 0, 1 및 3mM 첨가에서 융해 후 정자활력은 각각 46.4, 56.4 및 47.2%였으며 1mM 처리구가 다른 처리구보다 유의적으로 높았다. 3.동결희석액에 pentoxifylline 첨가는 융해 후 정자의 생존성, 첨체막 온전성 및 원형질막 온전성(HOST) 모두 유리한 결과를 나타냈으며, 1mM의 첨가에서 정자의 생존성과 기능성이 향상되었다.4.자연발정에서 발정개시 후 11일, 13일 및 15일째 초음파기로 측정한 난포 크기는 각각 6.5mm, 11.5mm 및 8.0mm이였다. 배란은 13-15일 사이에 일어났다. 5.발정유기한 개체에서 수정후 임신 20일째부터 5일 간격으로 3회 측정한 태낭의 크기는 13.7mm, 28.5mm 및 40.5mm 이었다. 6.자연발정과 발정을 유기한 개체에서 pen- toxifylline 1mM이 첨가된 정액을 동결융해하여 인공수정 후 임신율은 71.4%와 75%로 두 방법간에 차이가 없었으나, 자연발정군에서 평균산자수는 6.6두였고, 발정유군의 평균산자수는 2.6두였다.

돼지 액상정액의 보존액, 보존온도 및 기간이 정액성상과 번식성적에 미치는 영향 (Effect of Extender, Preservation Temperature and Period of liquid Boar Semen on Semen Characteristics and Reproductive Performance)

  • 돼지 인공수정센터에서 사육중인 인공수정용 종모돈을 이용하여 1995년부터 2000년까지 보존액 종류, 보존온도 및 보존기간에 따른 정액성상 변화와 번식성적을 조사하여 돼지 인공수정시 번식성적 향상과 실용화에 기여코자 본 연구를 실시하였다. 1. 돼지 액상정액의 보존액 종류에 따른 보존기간별 활력은 Androhep 보존액은 3일째부터, BTS 및 Modena는 5일째부터 현저하게 감소하는 경향을 보였고 (P<0.05), pH변화는 6.24~7.06 범위에서 변이가 심하게 관찰되었으며 Androhep 보존액이 산도가 낮은 경향을 보였으나 전반적으로 규칙 적 인 경향을 나타내었다. 보존액 종류별 보존기간에 따른 분만율은 BTS, Modena 및 Androhep 보존액 모두 5일 보존까지 차이가 없었다. 보존액별 분만율은 Androhep 보존액이 BTS 보존액과는 차이가 없었으나 1일 보존과 5일 보존에서 Modena보존액 보다 우수하였다 (P<0.05). 산자수는 보존 기간별 보존액간에 차이는 없었으나 Androhep 보존액은 3일 보존부터 산자수가 유의적으로 감소하였다 (P<0.05). 2. 보존온도에 따른 액상정액의 운동성과 정상첨체 비율에서 17$^{\circ}C$ 보존정액의 운동성은 3일째부터, 정상 첨체율은 4일째부터 감소하였으나 5$^{\circ}C$ 보존정액은 4일 보존까지 정액성상의 변화가 크지 않은 것으로 나타났다. 17$^{\circ}C$에 보존한 액상정액이 5$^{\circ}C$에 보존한 것보다 분만율은 현저히 높았으나 산자수는 비슷한 경향을 보였고, 분만율은 보존 2일째부터 산자수는 3일째부터 감소하였다. 5$^{\circ}C$ 보존정액은 4일까지 분만율에 큰 변화가 없었으나 산자수는 다소 감소하는 경향을 보였다.

천연기념물 어름치 Hemibarbus mylodon (Pisces: Cyprinidae)의 난 발생 및 초기생활사 (Egg Development and Early Life History of the Natural Monument Species Hemibarbus mylodon (Pisces: Cyprinidae) in Korea)

  • 천연기념물 어름치 Hemibarbus mylodon의 복원학적, 발생학적 특징을 밝히기 위하여 난 발생 및 초기생활사 연구를 실시하였다. 자연에서 친어를 채집한 후 실험실로 옮겨 사육하면서 약 15개월 후에 암 수의 인공 성숙에 성공하였다. 성숙한 암컷과 수컷은 Ovaprim (0.5 mL/kg)을 주사하여 산란을 유도하였고 건식법으로 인공수정시켰다. 산란된 성숙란(n=30)은 $2.21{\pm}0.06mm$이었으며, 접착성을 띤 불투명한 회색이었다. 수정란은 수온 $20{\pm}1^{\circ}C$에서 수정 후 78시간(50%)만에 부화하였으며, 부화 직후의 자어는 전장 $6.6{\pm}0.75mm$ (n=10)이었다. 부화 후 14일에 전장 $13.5{\pm}0.23mm$ (n=10)로 난황흡수가 완료되어 후기자어로 이행하였다. 부화 후 21일에는 전장 $14.8{\pm}0.45mm$ (n=10)로 모든 지느러미 기조가 정수가 되어 치어기로 이행하였다. 100일 후에 전장 $33.0{\pm}4.25mm$ (n=10)로 1쌍의 입수염이 나타나고 체측의 반문과 체형이 비교적 성어와 유사하였다. 본 연구에서 얻어진 사육 기술 및 초기생활사 특징은 어름치의 보전생물학적 측면에 도움이 될 것으로 사료되었다.

형질전환 돼지의 정자와 일반돼지의 정자성상에 대한 비교평가 (Comparative Evaluation on Sperm Parameter of Transgenic Pigs with General Pigs)

  • Pig has been known to be one of the most feasible animals as a bioreactor to produce pharmaceuticals in milk and as a mediator in xenotransplantation research. Previously, we generated transgenic pigs for both purposes, which were expressing Factor 8, vWF, hTPA, and hEPO in milk, along with expression of MCP at GalT gene locus ($GalT^{-MCP/-MCP}$) as well as expressing MCP at GalT gene loci with CD73 expression ($GalT^{-MCP/+}/CD73$). In this study, we performed comparative analyses of sperm parameters between wild type male (WT) pig and those transgenic males to examine the effects of transgenes integrated into the pigs on motility, morphology, viability, and acrosome integrity of the spermatozoa. Our results showed that the rates of actively motile spermatozoa of WT, Factor 8, vWF, hTPA, hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs were 85.0%, 83.3%, 82.5%, 83.3%, 82.5%, 77.5%, and 78.7%, respectively. Whereas, the rates of morphologically normal spermatozoa of WT, Factor 8, vWF, hTPA, hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs were 90.0%, 80.0%, 80.0%, 83.3%, 85.0%, 91.8%, and 80.8%, respectively. In addition, the viability in spermatozoa of WT, Factor 8, vWF, hTPA, hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs were 93.9%, 82.4%, 89.9%, 83.9%, 87.4%, 92.8%, and 83.6%, respectively. The rates of spermatozoa with normal acrosome integrity in WT, Factor 8, vWF, hTPA, hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs were 98.1%, 98.6%, 98.6%, 98.7%, 98.1%, 99.5%, and 95.1%, respectively. There were no significant differences in motility, morphology, viability, and acrosome integrity of the spermatozoa among WT, Factor 8, vWF, hTPA, and hEPO, $GalT^{-MCP/+}/CD73$, and $GalT^{-MCP/-MCP}$ pigs. These mean that neither random integration nor targeted integration of the transgene into chromosome of pig effect on characteristics of spermatozoa. Ultimately, the transgenic male pigs subjected in this study could apply to propagate their progenies for production of human therapeutic proteins and advancing the xenotransplantation research.

코끼리조개, Panopea japonica (A. Adams)의 수정란 발생과 유생 성장 (Development and growth in fertilized eggs and larvae of the Japanese geoduck, Panopea japonica reared in the laboratory)

  • 코끼리조개의 대량 인공종묘생산을 추진하기 위한 기초연구로서 수정란, 유생 및 치패의 발달을 조사하였다. 생식체는 잘 성숙한 어미에서 추출하여 희석 정자 용액으로 수정하였으며, 발생유생은 수온 $19{\pm}1^{\circ}C$로 유지하였다. 수정란의 난경은 평균 $81.6{\mu}m$ 이었으며, 수정 후 14시간 만에 담륜자 유생이 되어 부상하고, 27시간 만에 D상 유생 (각장 $116{\mu}m$)으로 발달하고, 20일 후에는 각장 $311.2{\mu}m$로 성장하여 바닥에 안착하였다. $18.5-21.5^{\circ}C$에서 정상적으로 부화 및 유생사육이 가능하였으며, 채묘기질은 모래가 우수하였으나 캐시미론도 양호하였다. 부유유생이 착저한 후 108일간 사육한 결과 각장 13 mm, 전중량 0.4 g으로 성장하였고 일간성장률은 각장 $117.5{\mu}m$ 이었으며, 자연해수로 연중 실내육성이 가능하였다.

자궁강내인공수정시 황체기 보강으로서 경구 미분화 프로게스테론과 디드로게스테론의 비교 (Comparison of Oral Micronized Progesterone and Dydrogesterone as a Luteal Support in Intrauterine Insemination Cycle)

  • 목 적: 과배란유도 후 자궁강내 인공수정 시술시 황체기 보강으로서 경구 미분화 프로게스테론 투여법과 경구 디드로게스테론 투여법의 임상적 효용성을 비교하고자 하였다. 연구방법: 2007년 1월부터 2009년 8월까지 시행된 과배란유도 후 자궁강내 인공수정 시술 183주기를 후향적으로 분석하였다. 과배란유도는 성선자극호르몬 단독 또는 클로미펜과 성선자극호르몬의 병합요법을 사용하였다. 136주기에서는 황체기 보강으로서 경구 미분화 프로게스테론을 하루 300 mg으로 투여하였고 47주기에서는 디드로게스테론을 일일 20 mg으로 투여하였다. 결 과: 여성의 연령, 불임 인자, 성숙난포수 ($\geq$16 mm), 총운동성정자수, triggering 날의 자궁내막 두께는 두 군간 유의한 차이가 없었다. 자궁내 태낭이 확인되는 임상적 임신율은 미분화 프로게스테론 투여군에서 21.3%, 디드로게 스테론 투여군에서 19.1%로 차이가 없었다 (p=0.92). 유산율은 미분화 프로게스테론 투여군에서 다소 높은 경향을 보였으나 통계학적인 차이는 없었다 (34.5% vs. 11.1%, p=0.36). 결 론: 황체기 보강으로서 경구 디드로게스테론 투여법은 경구 미분화 프로게스테론 투여에 비하여 비슷한 임신율과 유산율을 보였다. 그러나 상대적으로 디드로게스테론 투여군의 수가 적어 좀더 많은 환자를 대상으로 한 전향적 연구가 필요하다.
