A review of the genus $Adoxophyes$ (Lepidoptera Tortricidae) in Korea, with description of $A.$ $paraorana$ sp. nov.

  • We reviewed the genus $Adoxophyes$ (Lepidoptera: Tortricidae) from Korea and recognized three species of the genus. Among them, $A.$ $paraorana$ sp. nov., formerly misidentified as $A.$ $orana$ in Korea, is described as new to science. $Adoxophyes$ $orana$, a notorious pest known in most Eurasian countries for a long time, turns out to be only recently introduced or found in Korea. Photographs of the adults and genitalia of the species are provided. Specific distinction was supported by the COI barcode study.

  • 형태학적특징으로 분류된 6개의 그룹(GI, GII, GIII, GIV, GV, GVI)에 속하는 수종의 Phytophthora species 간의 RAPD fingerprinting 양상을 살펴보았다. Phytophthora의 일부 균주는 배양시 균사생장 형태가 매우 유사하며, 분리를 위한 여러 가지 특징이 중첩되기 때문에 정확한 동정이 난해하다. 본 실험결과 감자에서 분리한 균주 뿐만 아니라 토마토에서 분리한 P. infestans에서 RAED 분석을 통해서 동정이 가능함을 알 수 있었다. 증폭된 절편들은 $0.3{\sim}3.2\;kb$의 범위에 속했다. 전체 145개 밴드 중에서 109개의 polymorphic band를 얻었다. 7개의 P. infestans 간에는 최저 0.92에서 최고 0.99의 높은 유사성을 보였으며 감자를 기주로한 isolate 3 과는 $0.93{\sim}0.95$를 보였고, P. capsici 간에는 $0.77{\sim}0.86$의 유연성을 보였으며 약 80%의 수준에서 효과적으로 grouping 되었다. 특히 OPA-20를 사용하여 genus Phytophthora 내에 공통으로 존재하는 600 bp의 common band와 P. infestans에만 형성되는 680 bp의 specific band를 얻어 marker로서의 활용이 가능함을 알 수 있었다. 형태학적으로는 GVI 속하는 P. cinnamomi와 P. cryptogea간의 RAPD fingerprinting pattern에서 상이한 band 패턴을 나타냈다. 특히 OPA-04, OPA-17, OPA-18, OPA-19, OPB-12에서는 major band의 molecular weight와 생성밴드의 수에서 뚜렷한 차이를 보였다. 그리고 두 균주간의 유사성은 0.67로 나타났다. GV에 속하는 P. megasperma와 P. sojae의 경우, 유사성 정도가 0.65으로 나타났으며 이는 다른 균주와의 유사정도 보다 가장 높은 수준을 보였다. 또한 이들 균주는 OPA-08, OPA-17, OPA-19를 사용했을 때 단일 major band에서 큰 차이를 보였다.

Composition and Diversity of Tree Species in Kamalachari Natural Forest of Chittagong South Forest Division, Bangladesh

  • Information on plant diversity and community structure are required to chalk out necessary actions for conservation management. The present study assessed the composition and diversity of tree species in Kamalachari Natural Forest of Chittagong South Forest Division, Bangladesh, during April 2010 to November 2011. A total of 107 tree species belonging to 72 genera and 37 families were recorded, where Moraceae family was represented by maximum (11) species. Density, Basal area and volume of tree species were $418{\pm}20.09stem/ha$, $21.10{\pm}2.62m^2/ha$ and $417.4{\pm}79.8m^3/ha$ respectively. Diameter and height class distribution of tree species revealed an almost reverse J-shaped curve. Both the number of species and percentage of tree individuals were maximum in the lower DBH and height ranges. Anthropogenic disturbances like illegal tree cutting, over extraction, settlement inside forest area etc. were noticed during the study, which are supposed to cause gradual decrease of both tree species and individuals in the higher DBH and height classes. However, Artocarpus chama was found dominant showing maximum IVI followed by Schima wallichii, Aporosa wallichii, and Lithocarpus acuminata. The quantitative structure of the tree species of Kamalachari natural forest is comparable to other tree species rich tropical natural forests. The findings of the study may help in monitoring future plant population changes of the identified species and adopting species specific conservation programs in Kamalachari natural forest.

Nested PCR 기법을 이용한 인삼 뿌리썩음병원균의 특이적 검출 (Specific Detection of Root Rot Pathogen, Cylindrocarpon destructans, Using Nested PCR from Ginseng Seedlings)

  • Cylindrocarpon destructans는 인삼 및 수목에 뿌리썩음병을 일으키는 토양 전염병 식물병원균이다. 신속 정확한 검출 가능성을 알아보기 위하여 종 특이적인 primer와 nested PCR 기법을 활용하여 인삼 유묘로부터 뿌리썩음 병균 C. destructans로 2차 PCR증폭을 실시한 결과 병원성이 확인된 C.destructans에서만 400bp의 종특이적 증폭산물을 얻을 수 있었다. 종 특이성 primer 와 nested PCR 기법을 이용한 인삼뿌리썩음병균 DNA에 대한 반응 민감도는 최저 약 1fg으로 나타나 단 몇 개의 포자만 존재해도 검출이 가능하였다. 또한, nested PCR 기법은 실제 이병토양에 심었을 경우에도 C.destructans 에 감염된 인삼 유묘로부터만 정확하게 병원균을 검출해 내었다. 종특이적 primer 와 nested PCR 기법을 이용한 본 연구 결과는 실제 재배농가에서 인삼 경작시 뿌리썩음병 진단에 매우 유용하게 활용될 수 있을 것으로 판단된다.

베타글루칸과 잔탄검 계열 바이오폴리머 신소재의 국내 하천 식물종에 대한 생육 촉진 영향 (β-Glucan- and Xanthan gum-based Biopolymer Stimulated the Growth of Dominant Plant Species in the Korean Riverbanks)

  • 신설 제방의 사면 안정화를 위해 사용하는 토목 자재는 완공 이후 유입되는 식생의 종류 및 생장에 큰 영향을 미친다. 최근 미생물 유래 베타글루칸과 잔탄검 계열 바이오폴리머가 식물의 생육을 촉진시킬 수 있는 새로운 친환경 제방 사면 안정화 물질로 주목 받고 있다. 본 연구는 바이오폴리머 신소재의 생태성 평가를 목적으로, 특히 생태계 내에서 다수를 차지하는 우점 식물종들이 생태계의 특성을 크게 좌우하는 점에 주목하여, 바이오폴리머 신소재가 국내 하천에서 흔히 발견되는 자생 초본 식물 8종과 녹화용 식물 2종의 발아 및 생육에 미치는 영향을 시험하였다. 시험 식물종들의 종자 발아율은 바이오폴리머가 혼합된 토양의 영향을 받지 않았다. 반면 식물 생육은 바이오폴리머가 혼합된 토양에서 증가하였는데, 특히 돌피의 경우 약 2배정도의 총 생물량이 증가하였다. 식물 뿌리의 생육이 증가하는 경향을 보였고, 잎의 건중량 대비 엽면적이 증가하였다. 뿌리의 생육 증진과 잎의 건중량 대비 엽면적 증가가 환경 스트레스에 적응된 식물종들의 대표적 기능 형질인 점을 감안하면, 제방 사면에 처리된 바이오폴리머 신소재는 국내 하천 우점종들의 생육 및 환경 스트레스 저항성을 높일 것으로 예측된다.

Microbe-Based Plant Defense with a Novel Conprimycin Producing Streptomyces Species

  • Crops lack genetic resistance to most necrotrophic soil-borne pathogens and parasitic nematodes that are ubiquitous in agroecosystems worldwide. To overcome this disadvantage, plants recruit and nurture specific group of antagonistic microorganisms from the soil microbiome to defend their roots against pathogens and other pests. The best example of this microbe-based defense of roots is observed in disease-suppressive soils in which the suppressiveness is induced by continuously growing crops that are susceptible to a pathogen. Suppressive soils occur globally yet the microbial basis of most is still poorly described. Fusarium wilt, caused by Fusarium oxysporum f. sp. fragariae is a major disease of strawberry and is naturally suppressed in Korean fields that have undergone continuous strawberry monoculture. Here we show that members of the genus Streptomyces are the specific bacterial components of the microbiome responsible for the suppressiveness that controls Fusarium wilt of strawberry. Furthermore, genome sequencing revealed that Streptomyces griseus, which produces a novel thiopetide antibiotic, is the principal species involved in the suppressiveness. Finally, chemical-genetic studies demonstrated that S. griseus antagonizes F. oxysporum by interfering with fungal cell wall synthesis. An attack by F. oxysporum initiates a defensive "cry for help" by strawberry root and the mustering of microbial defenses led by Streptomyces. These results provide a model for future studies to elucidate the basis of microbially-based defense systems and soil suppressiveness from the field to the molecular level.

  • Rice stripe virus causes severe damage to rice in Korea, Japan and China. RSV is a type member of the tenuivirus group and transmitted by the small brown planthopper, Laodeiphax striatellus, in a persistant manner. Until now, occurrence of RSV is limited in of southern part of Korea. But recently occurrence of RSV is increasing and spreading in central part of Korea including Chungcheong and Kyonggi province. So we analyzed recent occurrence trend of RSV which is increased and cloned and sequenced coat protein gene for isolating of RSV strain. Infected rice of each species(Ilpumbyeo, Sindongjinbyeo, Keumobyeo-2, Dongjinbyeo, Jongnambyeo, Samcheonbyeo, etc.)is collected, we extracted total RNA from infected leaves and detected RSV viral RNA by reverse transcription(RT)-PCR using specific primer of coat protein gene. The result of RT-PCR, we observed specific band. We already cloned cDNA from the band, is analyzing sequence variety and homology of each species.

Five Species of Syrphidae (Insecta: Diptera) Newly Recorded in Korea

  • In Korean Peninsula, a total of 175 syrphid species are currently known. As a result of ongoing systematic study of the family Syrphidae, we have discovered the following five species for the first time in the Korean Peninsula: Mallota rossica Portschinsky, 1877, M. shatalkini Mutin, 1999, Sphiximorpha rachmaninovi (Violovitsh, 1981), Volucella bivitta Huo et al., 2007, and V. inanoides Hervé-Bazin, 1923. Among these taxa, Sphiximorpha Rondani, 1850, is the genus recorded for the first time in Korea. In total, three subfamilies, 16 tribes, 70 genera, and 180 species are now officially recognized for the Korean syrphid fauna. In order to facilitate their identification, we here provide specific diagnoses and color photographs for the species listed.

Characterization and RT-PCR Detection of dsRNA Mycoviruses from the Oyster Mushroom, Pleurotus ostreatus

  • The partial nucleotide sequences of the genomic dsRNA mycovirus infecting Pleurotus ostreatus isolates ASI2223 and Suhan were determined and compared with those of mycoviruses belonging to partitiviruses and totiviruses. Partial nucleotide sequences of the purified dsRNA from ASI2223 and Suhan showed RNA-dependent RNA polymerase sequences that are closely related to those of partitiviruses, including Fusarium poae virus 1, Fusarium solani virus, Rhizoctoniasolani virus, Discula destructiva virus 2, and Oyster mushroom isometric virus 2. Specific primers were designed for RT-PCR detection of dsRNA viruses from the P. ostreatus isolate ASI2223 and Suhan. Two virus specific primer sets were found to specifically detect each virus among six sets of designed oligonucleotide primers. Collectively, these results suggest that dsRNA mycoviruses from P. ostreatus isolates ASI2223 and Suhan belong to the family Partitiviridae, although, they are not the same virus species. Our results also suggest that these virus-specific primer sets can be employed for the specific detection of each viral sequence in infected tissues.
