Alteromonas sp. SH-1균 유래의 α-agarase의 특성조사 (Characterization of α-agarase from Alteromonas sp. SH-1)

  • A novel agar-degrading marine bacterium, SH-1 strain, was isolated from seashore of Namhae at Gyeongnam province, Korea. The SH-1 strain exhibited 98% similarity with Alteromonas species based on 16S rDNA sequencing and named as Alteromonas sp. SH-1. Alteromonas sp. SH-1 showed agarase activity of 348.3 U/L (1.67 U/mg protein). The molecular masses of the enzymes were predicted as about 85 kDa and 110 kDa by SDS-PAGE and zymogram. The enzymatic activity was optimal at $30^{\circ}C$ and the relative agarase activity was decreased as temperature increase from $30^{\circ}C$ and thus about 90% and 70% activities were shown at $40^{\circ}C$ and $50^{\circ}C$, respectively. The optimum pH was 6.0 for agarase activity in 20 mM Tris-HCl buffer and activities were less than 70% and 85% activity at pH 5.0 and pH 7.0, respectively, compared with that at pH 6. Agarase activity has remained over 90% at $20^{\circ}C$ after 1.5 hour exposure at this temperature. However, its activity was less than 60% at $30^{\circ}C$ after 0.5 h exposure at this temperature. The enzymes produced agarooligosaccharides such as agaropentaose and agarotriose from agarose, indicating that the agarases are ${\alpha}$-agarases. Thus, Alteromonas sp. SH-1 and its agarases would be useful for the industrial production of agarooligosaccharides which are known as having anticancer and antioxidation activities.

국내 식품으로부터 분리한 Listeria Species의 RAPD 분석 (Randomly Amplified Polymorphic DNA Analysis of Listeria Species Isolated from Foods in Korea)

  • Randomly Amplified Polymorphic DNA (RAPD) 기술을 이용하여 국내 식품으 로부터 분리한 Listeria sp. 분리균주에 대한 DNA polymorphism을 분석하고 유연환계를 비 교하며, 유용 marker를 개발할 목적으로 10가지 10-mer primer를 이용하여 PCR을 수행한 결과 5개의 primer(OPA-01, OP-26-01, OP-26-02, OPB-01, OP-26-10)가 선별되었고, 76개 의 DNA 단편이 증폭되었다. 이 중 OPA-01과 OP-26-10 promer에 의한 약 1.5 kb와 0.7 kb의 증폭 band는 모든 Listeria 분리균에서 관찰할수 있었으나, 이 증폭된 DNA 단편은 Listeria sp.에만 특이적인 것은 아니었다. NTSYS 프로그램을 이용해서 Listeria sp. 분리구 간의 유전적 유연관계를 알아본 결과 7개의 cluster로 나누어졌고 유사도는 대체로 0.54~ 0.93사이였으며, 특히, No.3과 No.20은 93%로 가장 높은 유사도를 나타내었고, No.7과 No.24 또는 No.7과 No.45는 54%로 가장 낮은 유사도를 나타내었다. 이러한 결과는 RAPS 기술을 이용하여 쉽게 Listeria sp.를 subspecies로 분류할 수 있음을 시사하였다.

Granulosicoccaceae fam. nov., to Include Granulosicoccus antarcticus gen. nov., sp. nov., a Non-phototrophic, Obligately Aerobic Chemoheterotroph in the Order Chromatiales, Isolated from Antarctic Seawater

  • A Gram-negative, motile by tuft flagella, obligately aerobic chemoorganoheterotrophic, sphere-form bacterium, designated $IMCC3135^T$, was isolated from the Antarctic surface seawater of King George Island, West Antarctica. The strain was mesophilic, neutrophilic, and requiring NaCl for growth, but neither halophilic nor halotolerant. The 16S rRNA gene sequence analysis indicated that the strain was most closely related to genera of the order Chromatiales in the class Gammaproteobacteria. The most closely related genera showed less than 90% 16S rRNA gene sequence similarity and included Thioalkalispira (89.9%), Thioalkalivibrio (88.0%-89.5%), Ectothiorhodospira (87.9%-89.3%), Chromatium (88.3%-88.9%), and Lamprocystis (87.7%-88.9%), which represent three different families of the order Chromatiales. Phylogenetic analyses showed that this Antarctic strain represented a distinct phylogenetic lineage in the order Chromatiales and could not be assigned to any of the defined families in the order. Phenotypic characteristics, including primarily non-phototrophic, non-alkaliphilic, non-halophilic, and obligately aerobic chemoheterotrophic properties, differentiated the strain from other related genera. The very low sequence similarities (<90%) and distant relationships between the strain and members of the order suggested that the strain merited classification as a novel genus within a novel family in the order Chromatiales. On the basis of these taxonomic traits, a novel genus and species is proposed, Granulosicoccus antarcticus gen. nov., sp. nov., in a new family Granulosicoccaceae fam. nov. Strain $IMCC3135^T\;(=KCCM42676^T=NBRC\;102684^T)$ is the type strain of Granulosicoccus antarcticus.

순천만에 자생하는 염생식물 근권에서 유래한 해양세균의 계통학적 분석 및 다양성 (Phylogenetic Analysis and Diversity of Marine Bacteria Isolated from Rhizosphere Soils of Halophyte in Suncheon Bay)

  • 순천만의 염생식물 근권에서 정주하는 세균의 분리를 위해 순천만에서 우점하는 자생식물인 칠면초 군락 3개 지점을 선발하여 샘플링 하였다. 시료는 marine broth, tryptic soy broth 한천배지를 통해 분리되었으며, 형태학적인 구분을 통해 순수분리되었다. 분리주의 16S rDNA를 분석하여 총 92 균주가 동정되었다. 이들의 유연관계 확인을 위한 계통수 작성 결과, 각각 firmicutes (56.5%), gamma-proteobacteria (29.3%), alpha-proteobacteria (5.4%), actinobacteria (5.4%), bacteroidetes (3.3%)에 속하였다. Shannon’s Diversity index (H')를 산출하였을 때 각각 1.675, 1.924, 2.04로, 채취 지점별로 종 다양성의 차이를 보였다.

우리나라에 분포하는 고추와 토마토 풋마름병균(Ralstonia solanacearum) 계통들의 유전적 다양성 (Genetic Diversity of Ralstonia solanacearum Strains Isolated from Pepper and Tomato Plants in Korea)

  • 풋마름병징을 나타내는 고추와 토마토 식물체로부터 35개의 풋마름병균 계통들을 분리하여 생리.생화학 실험, 병원성 실험, 유전학적 실험을 통해 유전적 다양성을 연구하였다. 생리 생화학 실험과 종특이적 polymerase chain reaction(PCR)을 실시한 결과 풋마름병균 계통들은 모두 Ralstonia solanacerum biovar 4로 동정되었다. 분리균은 고추와 토마토 유묘를 이용해 병원성이 확인되었다. Repetitive sequence-based PCR(rep-PCR) 결과를 토대로 계통도 분석을 한 결과 고추와 토마토 분리균은 6개의 group으로 나뉘었으며, 유전적 다양성은 높게 나타났다. 풋마름병균 계통들의 그룹간에는 지역별, 기주별 특이성은 관찰되지 않았다.

충북지방의 뿌리혹병 감염 포도나무 뿌리에서 분리한 Agrobacterium속 균의 특성 (Characterization of Agrobacterium spp. Isolated from Roots of the Crown Gall-infected Grapevine in Chungbuk)

  • 최근 혹병이 심하게 발생한 충북지방의 포도나무의 뿌리로부터 선택배지를 이용하여 Agrobacterium속 세균을 분리하고 동정하였다. 분리균의 지방산 분석을 통한 MIDI에 의한 동정, 16S rDNA 염기서열, 생화학적 특성, 종 특이적 primer을 이용한 PCR 결과로 13개 분리균은 모두 A. tumefaciens로 동정 되었으며, 포도나무 혹병균인 A. vitis는 분리되지 않았다. 모든 분리균은 토마토와 포도나무의 줄기와 뿌리에 병원성을 나타내지 않았다. Rep-PCR 결과 분리균은 A. tumefaciens와 일부 유사성이 있지만 전체적으로 병원균인 A. tumefaciens와 A. vitis와는 유사성이 낮았다. 이상의 결과는 충북 일부 지방의 MBA와 거봉에서 발생한 혹병을 일으키는 병원균은 토양이나 뿌리로부터 전반되지 않고 묘목을 통해서 전반되었을 가능성을 시사하고 있다.

Phylogenetic Relationships of 3 Korean Neodiplostomum Species (Digenea: Neodiplostomidae) Based on Partial CO1 Gene

  • The phylogenetic relationships of the 3 Neodiplostomum spp. (Digenea: Neodiplostomidae) occurring in Korea (N. seoulense, N. leei, and N. boryongense) were analyzed using the partial mitochondrial cytochrome c oxidase subunit 1 (CO1) gene. The adult flukes were recovered from Sprague-Dawley rats (N. seoulense) and newborn chicks (N. leei and N. boryongense) experimentally infected with the neodiplostomula from the grass snake, Rhabdophis tigrinus tigrinus. The genomic DNA was amplified using specific primers, and the sequence of CO1 was obtained. According to the results, the pairwise similarity was 96.1% between N. boryongense and N. seoulense, but was 95.0% between N. boryongense and N. leei and 94.2% between N. leei and N. seoulense. The results demonstrated a closer phylogenetic relationship between N. seoulense and N. boryongense. This high relationship of N. seoulense and N. boryongense may be related to their similar morphologic features including the limited distribution of vitellaria and the presence of a genital cone. N. leei is distinct on the other hand with an extensive distribution of vitellaria and the absence of a genital cone.

Chlamydophila psittaci에 감염된 앵무새의 임상병리학적 특징과 원인균의 ompA 유전자 비교분석 (Clinico-pathological Features of Chlamydophila psittaci Infection in Parrots and Genetic Characterization of the Isolates)

  • Avian chlamydiosis is caused by Chlamydophila psittaci and considered as one of an important zoonotic disease throughout the world. Among more than 400 avian species including poultry and pet birds susceptible to the disease, psittacine birds were known to be mostly susceptible hosts. In Korea, no outbreak of the disease and genetic analysis of the agent in poultry and pet birds have been reported. With histopathological findings and genetic identification of a causative agent, avian chlamydiosis was identified in parrots submitted from the same pet bird farm in 2006 and 2009 for the diagnosis. Based on genetic sequences and phylogenetic analysis of ompA gene, the two isolates of Chlamydophila psittaci showed 100% of genetic similarity and belonged to genotype A, suggesting that the same agent might be continuously circulated in the farm. This result indicates that serological survey of the disease in pet bird farms and impact of the disease on significance in public health may be further studied.

DGGE를 이용한 대청호 수화 발생시기의 세균군집 분석 (Analysis of Microbial Communities During Cyanobacterial Bloom in Daechung Reservoir by DGGE)

  • 대청호에서 수화 발생시기인 2003년 7월에서 10월까지 분자생태학적 방법의 하나인 DGGE (denaturing gradient gel electrophoresis)를 이용하여 시간에 따른 세균군집구조의 변화를 연구하였다. 조사기간 동안 출현한 식물플랑크톤을 형태학적으로 분류한 결과 cyanobacteria, 규조류 및 녹조류가 발견되었고, 이 중 Microcystis, Chroococcus, Oscillatoria, Phormidium 속이 크게 우점하였다. 16S rDNA의 DGGE pronto 분석에 의하여 Microcystis flos-aquae와 Oscillatoria spp.가 우점하는 것으로 확인되었으며, Aphanizomenon flos-aquae는 8월 중순에 우점하는 것으로 확인되었다. DGGE profile을 토대로 cluster analysis를 적용하여 다양한 미생물 군집의 유사성을 비교한 결과, 9월 2일의 미생물 군집이 다른 시기의 시료와 확연히 다른 그룹으로 구분되었다. 결과적으로 분자 생물학적 방법은 형태적 분석방법과 유사한 결과를 보였으며, 일부 세균의 분포 및 변화, 미생물 군집의 유사성 등에 대한 추가적 정보를 제공하였다.

Comparative Analysis of Nucleotide Sequence and Codon Usage of Arylphorin Gene Cloned from Four Silk-Producing Insects and Their Molicular Phylogenetics

  • To determine phylogenetic relatedness of four silk-producing silkmoths (B. mori, B. mandarina, A. yamamai and A. pernyi), internal coding region of arylphorin which is a storage protein in hemolymph protein of insects were amplified by polymerase chain reaction and then sequenced and compared each other. The nucleotide composition was biased toward adenine and thymine(59% A+T) and a strong bias for use of C in the third position of codons was found for Phe and Tyr. Together TTC(Phe) and TAC(Tyr) account for about 16.8% (10 for TTC and 8 for TAC) of all codon usage. The nucleotide similarity of arylphorin gene from B. mori showed 99%, 98% and 97% homology with those of B. mandarina, A. yamamai and A. pernyi, respectively. Also, the nucleotide sequence of arylphorin gene from B. mandarina showed 98% and 97% homology with those of A. yamamai and A.pernyi, respectively. Between A. yamamai and A. pernyi, the sequence homology was 97%. The deduced amino acid sequences in B. mori, B. mandarina and A. yamamai showed almost 99% homology. Although the aryphorin gene provided insufficient variability among the four insect species, A UPGMA tree is generated that supported the monophyly of silk-producing insects, with M. sexta placed basal to it. It is suggest that silk-producing insects have a close relationship and a homogeneous genetic background from comparison with those of other insects.
