The Environmental Pollution and Geochemical Behavior of Heavy Metals in Roadside Soil and Settling Particles from Retention Pond on A-71 Motorway, France (프랑스 A-71 고속도로변 토양과 부유퇴적물의 중금속 거동 및 오염에 관한 연구)

  • Retention ponds have been dug along some of the motorways in France to minimize environmental pollution by preventing pollutants from spreading over the surrounding area. A series of studies have been conducted to determine the physicochemical characteristics of the particles settling down in such a pond to evaluate the effectiveness of the pond as a trap for heavy metals such as Pb, Zn and Cd. The highly contaminated roadside soil and the uncontaminated background soil were also studied for comparison. The settling particles exhibited heavy metal concentrations of 2 to 8 times as much as the background Sologne soil, depending on the metal species. However, the heavy metal concentrations in the roadside soil were 7 to 26 times higher than those in the· settling particles. Sequential extraction experiments illustrated that the highly contaminated roadside soil consisted mainly of the readily soluble fractions (FII, FIII and FIV) for all three heavy metals, but little W which is hardly soluble. The proportion of W considerably increased up to one third of the total in the settling particles for Pb and Zn. This result as well as the large concentration differences between the roadside soil and the settling particles indicates that most of the heavy metals were lost to the surroundings even before reaching the retention pond. Cd exhibited somewhat different behavior in that the most soluble FI, which is negligible for Pb and Zn, occupied as much as one fourth of the total in the roadside soil. In addition, FV for Cd did not increase in the settling particles.

Phytic acid does not affect the formation of colonic aberrant crypt foci in Fe-overloaded male F344 rats

  • There are accumulating evidences that high levels of dietary iron may play a role in colon carcinogenesis. Elevated iron status has been associated with oxidative stress. Phytic acid (PA) functions as an antioxidant by chelating divalent cations and prevents formation of reactive oxygen species responsible for cell injury and carcinogenesis. The protective effect of PA was investigated on formation of aberrant crypt foci (ACF) induced by azoxymethane (AOM) in iron-overloaded male F344 rats. After acclimation with AIN-93G purified diet (35 ppm Fe, normal control diet) for one week, animals were fed iron-overloaded diet (350 ppm Fe) and PA (0.5% or 2% PA in water) for 8 weeks. Animals received two (1st and 2nd week) injections of AOM (15 mg/kg b.w.) to induce colonic ACF. The colonic mucosa was examined for the total numbers of aberrant crypt (AC) and ACF after staining with methylene blue. The blood and serum were analyzed with a blood cell differential counter and an automatic serum analyzer. Iron-overloaded diet increased the concentration of iron in liver of the rats. But iron-related parameters in blood were not changed among experimental groups. The numbers of ACF per colon and AC per colon were $178.8{\pm}33.2$ and $448.4{\pm}110.2$ in the iron-overloaded F344 rats. The total AC was significantly increased, compared with normal-diet AOM control group (p < 0.05). The treatments of PA at the dose of 0.5% slightly decreased the number of ACF and AC per colon to $153.6{\pm}29.5$ and $396.3{\pm}107.5$. However, there were no significant differences in the total numbers of ACF and AC between the AOM control group and PA (0.5% or 2%)-treated groups. These results suggest that PA may not affect the formation of ACF or AC induced by AOM in ironoverloaded F344 rats.

Comparison of Volatile Compounds in Plant Parts of Angelica gigas Nakai and A. acutiloba Kitagawa (참당귀와 일당귀의 부위별 휘발성 정유성분 비교)

  • Volatile flavor compounds Angelica gigas Nakai and Angelica acutiloba Kitagawa were extracted by SDE (simultaneous steam distillation & extraction) using the mixture of n-pentane and diethylether (1:1, v/v) as an extract solvent and analyzed by GC-FID and GC-MS. The amount of essential oils of top part and root in Angelica gigas were obtained in 0.063% (v/w) and 0.389% (v/w) yields as a fresh weight base, respectively. The main compounds in top parts and the root were identified as nonane (7.51% and 24.49%, respectively), ${\alpha}-pinene$ (14.64% and 31.75%), limonene+${\beta}-phellandrene$ (14.01% and 9.66%), ${\gamma}-terpinene$ (7.85% and 1.20%), germacrene-d (5.85% and 0.22%), (E,E)-${\alpha}-farnesene$ (6.05% and 1.40%), ${\beta}-eudesmol$ (5.26% and 1.84%). Although these compounds were present in both parts. The results showed large differences in. the concentrations of them much varied. The amount of essential oils stem and leaf obtained (0.068% and 0.127% in A. gigas) and (0.153% and 0.243% in A. acutiloba) yields as a fresh weight base, respectively. More than 18 and 32 components in stem and leaf have been identified, which of main components in A. gigas were ${\alpha}-pinene$, myrcene, limonene, germacrene-d, eudesmol and butylphthalide, but germacrene-d and butylphthalide contents were also different in stem and leaf. And more than 21 and 32 components in A. acutiloba were ${\gamma}-terpinene$ and butylphthalide. Volatile compounds were very different in both species.

  • The specificity and sensitivity of oligonucleotide primers were examined for the rapid detection of Salmonella in meat samples. The oligonucleotide primers used in this study were designed with the modification of mdh and invA sequence in the chromosome of Salmonella Typhimurium. Through the subsequent analysis of the specificity and sensitivity of the primers, two types of oligonucleotide primers, SLM1 and SLT4 were selected for the detection of Salmonella genus specific and S. Typhimurium species specific, respectively. The lowest detection limit of each primer was represented as 1 cell per reaction when reacted with a prepared DNA solution. The detection efficiency of the two primers was analysed with beef and pork samples intentionally contaminated with a mixture of Salmonella culture, and three preparation methods -, namely direct reaction after extraction, enrichment after extraction, and DNA extraction after enrichment for PCR reaction, - were also compared. No differences were found in the results according to meat sources and preparation methods.

Effect of Entomopathogenic Nematodes on Egg Mass Formation by the Northern Root-knot Nematode, Meloidogyne hapia (곤충병원성 선충이 당근뿌리혹선충의 난낭 형성에 미치는 영향)

  • The entomopathogenic nematodes, Steinernema carpocapsae All strain (ScA), S.glaseri NC strain (SgN) and H. bacteriophora NC 1 strain (HbN), were evaluated for the effects on egg mass formation by the northern root-knot nematode, Meloidogyne hapla in pot experiment using tomato. In the first experiment, 2.5$\times$10$^{5}$ infective juveniles (Ijs) of entomopathogenic nematodes were inoculated to 100 g of the soil infected with ca. 450 Ijs of M. hapla/100 ㎤ in 150 $mell$ container. The number of egg mass was significantly decreased to 9.4-36.5 in ScA, to 5.7-24.7 in SgN and to 11.2-16.0 in HbN treatments compared with 62.5 in M.hapla alone. In the second experiment, ScA and S.carpocapsae Pocheon strain (ScP) and SgN and S.glaseri Dongrae strain (SgD) were treated to 350 g of the soil infected with 100, 200 M.hapla larvae/100 ㎤ in 450 $mell$ container The entomopathogenic nematodes were inoculated at the rate of 2,020 Ijs and 1.6$\times$105 Ijs in 350 g soil. The number of egg mass of M.hapla were significantly decreased in the entomopathogenic nematode treatments compared with M.hapla alone although no differences were observed among Steinernema species, strains, or infection concentrations. Treatments of entomopathogenic nematodes 3 days before M.hapla inoculation were more effective on reduction of egg mass formation than those of 3 days after M.hapla treatments. Growth of tomato was not affected by entomopathogenic nematode treatments.

Studies on the Causal Factors of Landslides on Limestone Soils in Pyeongchangkun (산사태(山沙汰) 발생요인(發生要因)에 관한 연구(硏究) -평창군(平昌郡) 석회암지대(石灰岩地帶)를 중심(中心)으로-)

  • The characteristics of landslides occurred in August 5, 1979 in pyeongchangkun were surveyed and indentified as follows. 1. Deep limestone regions. Distinct differences in soil texture between A1 and B horizon could be observed on soil profile, which is attributed to the clay illuviation. The clay illuvial horizon is supposed to be an important cause of large scale mudflows on middle slopes by the lubricant action of ground water flowing between top soil and subsoil. 2. Shallow limestone regions. Very shallow top soils (less than 50cm) laid on tilted bedrock stratification provide a proper condition of mass soil movement if the top soil is saturated and ground water flows between top soil and bedrock when concentrated heavy rainfalls shower. 3. Granite regions. Weathering granitic bedrock produces very coarse textured top soils which are very cohesionless and have many pores. Therefore, the soil has high infiltration ratio and is easy to be saturated by water and to be detached from the bedrock. The landslides abrase very severely both sides of gully with high potential energy when they flow down. The following methods for landslide prevention can be recommended. 1. The original parts of landslides on top of the gully must be treated by intensive planting of deep rooting species and check dams. 2. Clear-cutting and crop planting on steep slope (more than 25 degrees) should be controlled and prohibited. 3. Establishment of landslide prevention forest should be practised on proper site.

Quantitative analysis of the clinical signs in marine fish induced by Megalocytivirus infection (Megalocytivirus 감염 해산 어류에서 나타나는 임상증상의 정량적 변화 분석)

  • In quantitative studies of clinical signs, rock bream of adults and juveniles infected with Megalocytivirus IVS-1 isolated from rock bream (Oplegnathus fasciatus) in Korea showed average $4.49{\pm}1.13$ and $4.85{\pm}1.06$ of spleen index respectively. In challenge experiments, Megalocytivirus IVS-1 induced 100% cumulative mortality in both adult and juvenile rock bream. However we found 60% cumulative mortality in juvenile red sea bream (Pagrus major) even after 30 days of injection, which contradicted with the results of other laboratories. Interestingly, IVS-1 infected red sea bream of the same juvenile size with rock bream showed lower spleen index compared to that of rock bream. In real-time PCR, there was continuous increasing of the numbers of viral copies ($2.03{\times}10^7$ copies/mg) in the spleen of juvenile rock bream infected, which were different from those in adult rock bream showing plateau level after reaching to the peak level. Moreover, enlarged cell numbers in the infected spleen were also increased continuously in the juvenile but not in adult of rock bream, even decreased after reaching to peak level. Consequently, significant differences in clinical signs: cumulative mortality. spleen index and viral copy number were found between rock bream and red sea bream, but not between adult and juvenile rock bream. Certainly quantitative expression of clinical sign as in this study may be a way to compare the progression of megalocitiviral disease more accurately in different species or physiological conditions.

Estimation of Atmospheric Deposition Velocities and Fluxes from Weather and Ambient Pollutant Concentration Conditions : Part I. Application of multi-layer dry deposition model to measurements at north central Florida site

  • The dry deposition velocities and fluxes of air pollutants such as SO2(g), O3(g), HNO3(g), sub-micron particulates, NO3(s), and SO42-(s) were estimated according to local meteorological elements in the atmospheric boundary layer. The model used for these calculations was the multiple layer resistance model developed by Hicks et al.1). The meteorological data were recorded on an hourly basis from July, 1990 to June, 1991 at the Austin Cary forest site, near Gainesville FL. Weekly integrated samples of ambient dry deposition species were collected at the site using triple-fiter packs. For the study period, the annual average dry deposition velocities at this site were estimated as 0.87$\pm$0.07 cm/s for SO2(g), 0.65$\pm$0.11 cm/s for O3(g), 1.20$\pm$0.14cm/s for HNO3(g), 0.0045$\pm$0.0006 cm/s for sub-micron particulates, and 0.089$\pm$0.014 cm/s for NO3-(s) and SO42-(s). The trends observed in the daily mean deposition velocities were largely seasonal, indicated by larger deposition velocities for the summer season and smaller deposition velocities for the winter season. The monthly and weekly averaged values for the deposition velocities did not show large differences over the year yet did show a tendency of increased deposition velocities in the summer and decreased values in the winter. The annual mean concentrations of the air pollutants obtained by the triple filter pack every 7 days were 3.63$\pm$1.92 $\mu\textrm{g}$/m3 for SO42-, 2.00$\pm$1.22 $\mu\textrm{g}$/m-3 for SO2, 1.30$\pm$0.59 $\mu\textrm{g}$/m-3 for HNO3, and 0.704$\pm$0.419 $\mu\textrm{g}$/m3 for NO3-, respectively. The air pollutant with the largest deposition flux was SO2 followed by HNO3, SO42-(S), and NO3-(S) in order of their magnitude. The sulfur dioxide and NO3- deposition fluxes were higher in the winter than in the summer, and the nitric acid and sulfate deposition fluxes were high during the spring and summer.

Association of Diabetes Mellitus and Metabolic Syndrome with Idiopathic Pulmonary Fibrosis (폐섬유화증과 당뇨와 대사 증후군의 연관성 연구)

  • Background: Reactive oxygen species (ROS) by oxidative stress may play an important role in the pathogenesis of various chronic diseases such as diabetes mellitus, obesity, hyperlipidemia, hypertension and malignancy that are linked to metabolic syndrome. Oxidative stress has been implicated in the pathogenesis of idiopathic pulmonary fibrosis (IPF). We examined the relationship between IPF and presenting factors associated with metabolic disorders. Methods: One hundred fourteen patients who met the current consensus of IPF definition were enrolled from March 2000 to April 2006 in Gil Hospital and Samsung Medical Center in Korea. One hundred thirty-four control subjects without pulmonary diseases were selected from subjects who visited Gil hospital for routine medical examinations, including low-dose chest computed tomography from January 2002 to July 2006. Retrospectively, we analyzed the clinical characteristics, the results of blood examinations, and lung function tests from medical records of both groups. Results: IPF patients and control subjects differed in the prevalence of diabetes mellitus as assessed by univariate analysis. Multivariate analysis demonstrated that diabetes mellitus and obesity were associated with IPF. The adjusted odds ratios for diabetes mellitus were 2.733 (95% confidence interval [CI], 1.282~5.827) and 2.001 (95% [CI], 1.063~3.766) for obesity. The remaining factors tested showed no differences between the patient group and the control. Conclusion: Diabetes mellitus and obesity may be associated with IPF development.

Freezing Injury Characteristics of Evergreen Broad-Leaved Trees in Southern Urban Area, Korea (남부지역 도시녹지의 난대상록활엽수 동해피해 특성)

  • The aim of this study was to investigate the damage characteristics induced by winter freezing of evergreen broad-leaved trees distributed in urban area of warm temperate forest zone, which are gaining increased interest recently as climate change. The results of the study indicated that, as for 'stem injury', Camellia japonica Linne (59.6%) had the weakest damage, while Ternstroemia gymnanthera Sprague (83.3%) had the most severe damage. By the visual evaluation of freezing injury according to the characteristics of plantations environment, the observations of single planting (p<0.01) in planting method had weaker damage than those of group planting, the observations of under tree (p<0.001), plain (p<0.001), and organic matter plenty (p<0.05) in plantations characteristics had significant differences with relatively weaker damage than the observations of open space, slop, and scarcity, respectively. Tree height (-0.432) and crown width (-0.470) among growth factors were negatively correlated with the severity of damage, respectively. Therefore, the selection of tree species is vital for the successful creation of these urban forested area by making considerations of planting environment, and further research on evergreen broad-leaved trees is needed in this aspect.
