Genetic Divergence and Speciation of Eurasian and American Yellow Perch Based on the Nucleotide Sequence of Cytochrome b Gene

  • Eurasian yellow perch (Perca fluviatilis) and American yellow perch (Perca flavescens) are known to be endemic species in Eurasia and North America, respectively. The presence of endemic species on each continent suggests their independent evolutionary history. However, because of the morphological similarity, distribution pattern, and only recent fossil record, their divergence time and speciation of the two Perca species has long been controversial. Here, from the comparison of the entire nucleotide sequences of cytochrome b gene, large genetic divergence between the two Perca species is observed although they are morphologically similar each other. Among 1,140 base pairs, interspecific nucleotide differences are found at 130 sites $(11.4\%)$. The differences varies with codon position, showing 22 sites in the first, 5 sites in the second, and 103 sites in the third codon position. Considering the types of nucleotide changes, transitional differences are much more than transversional differences and its ratio turned out to be 5.19. The estimated divergence time of the two Perca species indicates that they were separated each other approximately in the late Miocene period, which implies the long history of speciation. With comparison of the inferred amino acid sequences, strong structural and functional constraints which seem to be maintained by the highly conservative amino acid residues or protein regions, as found in other taxonomic groups of organisms, are also recognized in the cytochrome b of the fishes examined.

Chemical Speciation of Heavy Metals in Soils of Jeju Island, Korea (제주도 토양 중 중금속의 화학적 형태)

  • For 26 soil series distributed more than 1% among 63 soil series in Jeju Island, natural uncultivated soil samples were collected. For these soils, the chemical speciation of eight heavy metals (Cd, Cr, Cu, Mn, Ni, Pb, V, and Zn) was examined. Further, the Plant Bioavailability (PB) and Mobility Factor (MF) of these heavy metals were evaluated using Tessier's 5-step sequential extraction method (exchangeable, carbonate, reducible (bound to Fe/Mn oxides), oxidizable (bound to organic matter), and residual fraction). The main form present was residual fraction for Cd and Zn; residual and oxidizable fractions for Cr, Cu, Ni, and Pb; reducible fraction for Mn; and carbonate fraction for V. The average plant availability and average mobility factor were found to be V (57.37%) > Zn (12.49%) > Cd (11.76%) > Cu (11.19%) > Pb (9.37%) > Cr (9.09%) > Mn (3.13%) > Ni (2.63%), and Mn (61.04%) > V (59.94%) > Zn (31.54%) > Cd (17.65%) > Cr (15.66%) > Ni (13.89%) > Pb (13.80%) > Cu (13.53%), respectively.

  • Horizontal starch gel electrophoresis for 29 populations (n=543) of the wrinkled frog, Rana rugosa, from Korea and Japan was peformed to assess the degree of genic variation and genetic diversity, and to understand the biogeographic pattern of distribution and speciation. A sum of 22 presumptive loci was screened from 17 enzymes and general proteins. Four loci, Aco, Est-3, Me-2, and Pgm, demonstrated high levels of polymorphism. The degree of average genetic variation of R. rugosa was P=22.7% (9.1-40.9%), Ho=0.086 (0.048-0.165) and He=0.090 (0.042-0.168). In the south-eastern region of the Korean peninsula (Chongsong, Yongchon, Ulsan, Kyongju, Pohang, yongdok and Ulchin), a few unique alleles in the Mpi locus were detected and their biogeographic implications were considered. The degree of genetic differentiation among the Korean populations was moderate (S=0.900), whereas the degree of genetic diversity between Korean and Japanese populations was notably high (S=0.687, D=0.293). This result corresponds with the data obtained by the mitochondrial cytochrome b gene sequence (Lee et al., 1999) suggesting that the Korean and Japanese R. rugosa might have evolved a specific level of genetic differentiation since their geographic isolation.

Speciation of Some Heavy Metals in Surface and Core Sediments of Kyeonggi Bay, West Coast of Korea

  • Chemical speciation of five heavy metals (Cr, Cu, Ni, Pb, Zn) has been analyzed from 37 surface and 2 core sediments of Kyeonggi Bay, using the modified sequential extraction method based on Tessier et at. (1979). The results show that heavy metals in the Kyeonggi Bay surface sediments are associated dominantly with the crystal lattice fraction. But in the polluted sediments of the Incheon North Harbor, the importance of the labile fractions increased while that of the lattice fraction decreased. In particular, the adsorbed and the easily reducible fractions showed a noticeable increase. In the core samples emerged a speciation pattern which differed significantly from that of the surface sediments. A sharp increase in the percentage of the reducible and organic/sulfide fractions and a decrease in the lattice fraction were observed. Throughout the vertical column, however, the metal contents in the lattice fraction showed stability while those of the labile fractions showed an upward increase. The strong association of heavy metals with the organic/sulfide fraction could be attributed in part to the sulfate reduction prevailing in the polluted harbor sediments.

Speciation Analysis of Arsenic Species in Surface Water (수중의 비소 종 분리 분석)

  • In this study, a technique of speciation and determination of the trace inorganic arsenic(As(III) and As(V)) in water sample using HPLC-DRC-ICP-MS has been developed. Isocratic mobile phase of 10 mM ammonium nitrate and 10 mM ammonium phosphate monobasic was used and methanol(5 v/v%) was used as flushing solvent. Selection of the best flow rate of reaction gas, O$_2$, and optimization of the parameters such as pH and flow rate of mobile phase, and injection volume of sample for the separation and detection of arsenic species were carried out. The oxygen flow rate of 0.5 mL/min, pH of 9.4 and flow rate of 1.5 mL/min of mobile phase, and injection volume of sample of 100 $\mu$L were found to be the best parameters for the speciation and determination of arsenic species. The analytical features of the method were detection limit 0.10 and 0.08 $\mu$g/L, precision(RSD) 4.3% and 3.6%, and recovery 95.2% and 96.4% for As(III) and As(V), respectively. Analysis time was 4 minutes per sample. Linear calibration graphs with r$^2$ = 0.998 were obtained for both As(III) and As(V). Speciation analysis of arsenic species in the raw water samples collected from the tributary streams to Han River and main stream of Paldnag were performed by the proposed method. The concentrations of As(III) ranged from 0.10 to 0.22 $\mu$g/L and As(V) concentrations ranged from 0.44 to 1.19 $\mu$g/L, and 93.5% of total arsenic was found to be As(V).

Study on the Chemical Speciation of Hydrolysis Compounds of U(VI) by Using Time-Resolved Laser-Induced Fluorescence Spectroscopy (시간분해 레이저 유도 형광 분광학을 이용한 우라늄(VI) 가수분해 화학종 규명 연구)

  • Study on the chemical speciation of uranium(VI) species, ${UO_2}^{2+}$, $UO_2(OH)^+$, ${(UO_2)}_2{(OH)_2}^{2+}$, ${(UO_2)}_3{(OH)_5}^+$, has been peformed by using time-resolved laser-induced fluorescence spectroscopy. Speciation sensitivity which depends on the excitation wavelength was investigated. We obtained the speciation sensitivity in the order of $10^{-9}$ M concentration of U(VI) compounds at the excitation wavelength of 266 nm. The fluorescence spectrum and lifetime of ${UO_2}^{2+}$ were carefully measured at pH 1 and ion strength of 0.1 M. The spectrum showed the four characteristic peaks located around 488, 509, 533, 559nm and the fluorescence lifetime of $1.92{\pm}0.17{\mu}s$. The wavelength shifts of fluorescence peaks and the change of lifetimes for uranium hydrolysis compounds were compared with those of ${UO_2}^{2+}$. We report on the characteristic features, the shifts of peaks to the longer wavelength direction and the prolonged lifetimes, in the fluorescence of the U(VI) hydrolysis compounds.

  • Low and high resolution inductively coupled plasma mass spectrometry (ICP-MS) coupled with ion chromatography (IC) has been investigated for speciation of Cr(III) and Cr(VI). In particular, the interference of ArC^+formed by the carbon in a sample on the simultaneous determination of Cr(III) and Cr(VI) has been studied. In chemical speciation, this study shows that quadrupole type ICP-MS with low resolution has a limitation of simultaneous determination fo chromium species if the sample contains the carbon elements. The interference problems can be solved by high resolution ICP-MS.

  • 한국 고유종인 새코미꾸리(Cobitis rotundicaudata)의 집단간 유전적 차이에 따른 종 분화 여부를 밝히고자 4개집단을 대상으로mitochondrial DNA(mtDNA)의 RFLP분석을 실시 하였다. C. rotundicaudata mtDNA를 10개의 6-base cutting 제한요소로 처리한 다음 그 절 편 양상을 비교, 분석한 결과 4개 집단 공히 mtDNA의 전체 genome 크기는 약 16.5$\pm$ 0.5Kbp였으며 공통절편수(F)에서 한강 2개 집단(가평, 진부)과 동해안 마읍천 짐단간의 F값 은 0.911로 매우 가까웠으나 낙동강의 산청집단은 타 3개 집단과 F=0.375로 차이가 있었다. 또한 염기치환율(p)에 있어서도 한강 2개 집단 및 마읍천 집단간은 평균 p=0.005로 매우 유 사하였으나, 산청 집단은 타 집단들과 염기치환율에 있어 p=0.059로 종수준의 뚜렷한 차이 를 나타내어서 이들은 각각 별종으로 사료된다.
