Comparative Analysis of Terpenoids in in vitro Culture Media of Metabolically Engineered Transgenic and Wild Type Spearmint (Mentha spicata L.) (대사 제어된 스피아민트와 야생 스피아민트 기내배양배지의 터페노이드 성분 비교 분석)

  • IPP isomerase (Iso) and Limonene synthase (Limo) are important enzymes in terpenoids biosynthesis pathway. The wild type and each metabolically engineered (Iso and Limo) transgenic spearmint (Mentha spicata Linne) plants were compared for their growth patterns and the contents of essential oil in in vitro culture media. The profile of terpenoid metabolites was obtained from the essential oil of the metabolically engineered transgenic spearmint, which was extracted using a modified SDE method, by GC-MS analysis. The growth of wild spearmint was more profuse in B5 culture medium than in other media. Significant differences in leaf and root growth patterns were observed between metabolically engineered transgenic and wild type spearmint plants. The leaves of the transgenic spearmint plants were slightly elongated but were dramatically narrower than those of wild type spearmints. The content of essential oil of transgenic spearmint was different slightly depending on the target terpenoid genes. The content of essential oils in Limo transgenic plants was higher than that of Iso, except for transgenic plant in B5 medium. The transgenic spearmint produced more terpenoids than the wild type. Iso spearmint extracts showed eleven terpenoids and a phenylpropane, while Limo spearmint extracts contained nine terpenoids. However, extracts from the wild type showed the presence of only four terpenoids.

Insecticidal Activity of Spearmint Oil against Trialeurodes vaporariorum and Bemisia tabaci Adults (온실가루이와 담배가루이에 대한 Spearmint Oil의 살충활성)

  • These studies were carried out to investigate the fumigation and contact toxicities of spearmint oil (Mentha spicata) against adults of greenhouse whitefly, Trialeurodes vaporariorum and sweet-potato whitefly, Bemisia tabaci. And we carried out the constituent analysis of spearmint oil using gas chromatograph (GC) and gas chromatograph mass spectrometry (GC/MS). Spearmint oil showed $99.1\%,\;91.7\%,\;41.1\%$ fumigation toxicity against T. vaporariorum adults at $10{\mu}L/954mL,\;5{\mu}L/954mL,\;1{\mu}L/954mL$ air concentration, respectively. In case of B. tabaci adults, spearmint oil showed $100\%,\;100\%,\;61.3\%$ fumigation toxicity, respectively. However, spearmint oil showed < $30\%$ contact toxicity against adults of T. vaporariorum and B. tabaci. Through the constituent analysis using GC and GC/MS, we confirmed main constituents of spearmint oil were limonene ($16.1\%$), ${\gamma}$-terpinene($13.8\%$), ${\rho}$-cymene($5.8\%$), 3-octanol($6.9\%$), carvone($40.9\%$). Carvone, major constituent of spearmint oil, also showed $100\%$ fumigation toxicity at $10{\mu}L/954mL$ air concentration.

Fumigant Toxicity of Pennyroyal and Spearmint oils against Western Flower Thrips, Frankliniella occidentalis (꽃노랑총채벌레에 대한 Pennyroyal oil과 Spearmint oil의 훈증효과)

  • Fumigant toxicity of four plant essential oils (muguet flower, patchouli, pennyroyal, spearmint) were tested against the adults of western flower thrips, Frankliniella occidentalis. Among them, pennyroyal and spearmint oils showed 100% mortality at $1{\mu}{\ell}/45m{\ell}$ air dose. Through the constituent analysis using GC and GC-MS, we confirmed that main constituents of pennyroyal oil were pulegone(100.0%), and spearmint oil were $\beta-myrcene(3.5%)$, limonene(12.1%), carvone(85.4%). Among them, carvone and pulegone showed 100% fumigation at $1{\mu}{\ell}/45m{\ell}$ air dose, respectively. It can concluded that two oils are potential control agents against F. occidentalis.

  • The chemical composition of essential oils obtained from aerial parts in spearmint, apple mint and chocolate mint, was investigated by gas chromatography/mass spectrometry analyses. (-)-Carvone (33.0%) was quantitatively major compound in spearmint, followed by R-(+)-limonene (11.7%) and ${\beta}$-phellandrene (9.7%); (-)-carvone (37.4%) and germacrene D (11.9%) in apple mint; and (-)-menthol (34.3%), p-menthone (18.4%) and menthofuran (9.8%) in chocolate mint. Hierarchical cluster analysis and principle components analysis showed the clear difference in chemical composition of the three mint oils.

A Research on Satisfaction of Toothpaste Added Natural Herbal Extracts (천연허브추출물을 첨가한 치약의 만족도 조사)

  • Purpose: The study aims to evaluate satisfaction of toothpaste adding natural materials extracted from Zanthoxylum schinifolium and various herbs (Pine, Lemongrass, Spearmint). Methods: We performed the first survey to collect basic data for the development of natural herbal toothpaste. Participants were the general public in Jeonju and Iksan of Jeollabuk-do, and a total of 273 questionnaires were analyzed for the study. By adding anthoxylum schinifolium and herbal extracts (Pine, Lemongrass, Spearmint) to the toothpaste, four kinds of natural herbal toothpaste prototypes were manufactured. The second survey was conducted to assess the satisfaction one week after using the natural herbal toothpaste prototypes. Results: Satisfaction with current toothpaste was 3.52. The most important criterion of toothpaste was the prevention of tooth caries. 55.3% participants obtained toothpaste information through advertising. The survey indicated the most worrisome ingredient of toothpaste was surfactant, showing 55.3%. The overall satisfaction of natural herbal toothpaste was 3.99; among the four natural herbal toothpaste, toothpaste 2 (Zanthoxylum schinifolium, Lemongrass) showed the highest satisfaction as 3.99; and toothpaste 3 (Zanthoxylum schinifolium, Spearmint) showed the lowest satisfaction as 3.56. Conclusion: Four kinds of natural herbal toothpaste showed higher satisfaction as 3.99. In particular, the toothpaste adding Zanthoxylum schinifolium and Lemongrass extracts.

Corrosion Inhibition of Steel for Water Pipe Line by Adding a Non-Toxic Spearmint Extracts

  • Purpose: To investigate the corrosion inhibition effect of the natural spearmint oil extracted from Mentha Spicata plants on 304 stainless steel in different concentrations of hydrochloric acid. Method: The anti-corrosive effect has been investigated in 0.5m, 1m and 2m HCl using weight loss test and electrochemical polarization method as a function of inhibitor concentration and immersion time in strong chloride environment. The surface morphology was analysed by scanning electron microscopy (SEM). Results: The corrosion rate of steel decreased and inhibition efficiency increased with the increase in inhibitor concentration. Microscopic evaluation revealed significant corrosion in the specimens immersed in uninhibited conditions. Potentiodynamic polarization test results showed an increase in corrosion potential (Ecorr) and decrease in corrosion current (icorr) value with increasing concentration of inhibitor. Conclusions: Immersion of steel in higher concentration of inhibitor resulted in greater surface coverage value and hence lesser number of surface corrosion sites/pores were formed; thus lowering the corrosion rate.

Enhancing Production of Terpenoids in Metabolically Engineered Transgenic Spearmint (Mentha spicata L.) by Salt and Fungal Elicitors

  • Forest tree species usually takes for long periods to be harvested and cultivated but spearmints are a good model system for woody plant because of reducing and shortening cultivation time. Spearmints are good model plants (Mentha species) for research about terpenoids production and industrial essential oil manufacture. Isopentenyl pyrophosphate isomerase (Iso) and limonene synthase (Limo) are the key enzymes of terpenoid biosynthesis pathway. Transgenic and wild spearmints (Mentha spicata, MS) were cultured in vitro and assessed for the essential oil contents. The content of essential oil of transgenic spearmint also was enhanced slightly depending on the target terpenoid genes. In an attempt to increase productivity of terpenoids further, salt and fungal elicitation strategy was adopted on transgenic Mentha spicata. The salt (800 mM NaCl) as abiotic and two fungi (Botrytis cinerea and Glomerella cingulata) as biotic were used for elicitors. In the absence of salt stress four terpenoids were detected from the spearmint extracts, all of them being monoterpenes. On the other hand, the transgenic (MSIso) extracts contained eleven terpenoids (10 monoterpenes and 1 phenylpropene) while transgenic (MSLimo) extracts contained seven monoterpenes. After 3 days of fungal infection, the resistance indices further increased to 4.38, 3.89 and 2.04 for wild type, MSIso and MSLimo, respectively. The salt and fungal elicitators proved beneficial towards modifying both the terpenoids profile and improvement in the composition of essential oil. These results have important applications for the large-scale production of essential oils and forest biotechnology with respect to spearmint.

Repellency of Herb Plants and Essential Oils Against the Green Peach Aphid, Myzus persicae (허브류 식물과 정유의 복숭아혹진딧물에 대한 기피 효과)

  • Repellencies of herb plants and essential oils against alate Myzus persicae were investigated in a insect cage, a screen house, and a tobacco field. Numbers of nymphs deposited by alate or apterous Myzur persicae were lower on rosemary(Rosmarinus officinalis), silver thyme (Thymus vulgaris), and lemon balm(Melissa of fcianazis). Rosemary and silver thyme show little repellency against alate Myzus persicae on a nearby tobacco plant Rosemary oil, peppermint (Mentha piperita) oil, and ginger oil showed repellency against alate Myzus persicae, but spearmint (Mentha spicata) oil and lavender oil showed little repellency in a screen house. Rosemary oil, ginger oil, peppermint oil, lavender oil, and spearmint oil showed repellency against alate Myzus persicae in a tobacco field, but there was no significant difference among oils.

Effect of Jasmonic Acid and NaCl on the Growth of Spearmint(Mentha spicata L.) (Jasmonic Acid 및 NaCl 처리가 스피아민트의 생육에 미치는 영향)

  • This study investigated the effects of NaCl and jasmonic acid (JA) on the growth and physiological responses of spearmint (Mentha spicata L.). Spearmint was hydroponically grown for 3 weeks in modified Hoagland solution containing 0 (untreated control), JA ($20{\mu}M$ JA pretreatment), NaCl (50 mM NaCl treatment) and JA + NaCl ($20{\mu}M$ JA pretreatment + 50 mM NaCl treatment). Growth characteristics, chlorophyll, vitamin C, proline contents, DPPH scavenging activity and inorganic ion contents were evaluated. As a results, there were significant decreases in the plant height, leaf length, leaf width, and fresh weight of plants, treated with NaCl compared with control. On the other hand, the dry matters of shoot and root treated with JA + NaCl combination were better than control or NaCl treatment. Chlorophyll a and b contents in JA treatment was the highest. Vitamin C, antioxidant activity, and proline content in shoot were increased in NaCl treatment which showed low level of growth rate. The K/Na ratio, which is known to indirectly reflect the balance of ion uptake, was higher in a single treatment of JA than the control group, while lower in salt treatment (NaCl and JA + NaCl) because of high $Na^+$ absorption. In conclusion, these results showed that moderate stress treatment such as low level salt treatment and plant growth regulator jasmonic acid (JA) application would be potential strategies to improve the quality of spearmint by inducing the accumulation of secondary metabolites containing high antioxidant activity and essential oil.

Development of Ice Cream with Improved Microbiological Safety and Acceptable Organoleptic Quality Using Irradiation (감마선 이용 미생물학적 및 관능적 품질이 우수한 아이스크림의 개발)

  • To develop the manufacturing method of ice cream with microbiologically safe and proper sensory quality using irradiation for sensitive consumer, 3 different flavors, which were resistant to their flavors against irradiation, were selected and used for ice cream manufacturing to reduce the irradiation-induced off-flavor problem. The general composition was not different among treatments. Total aerobic bacteria were detected as 2.38, 1.23, 1.38, and 1.15 log CFU/g level in ice cream with control(no flavor added), spearmint, mint, and citrus flavor, respectively. No viable cells were observed by irradiation at 1 kGy except for the control. Sensory evaluation indicated that the irradiated ice cream with spearmint flavor at 1 kGy and citrus flavor at 3 kGy had higher overall acceptability. Therefore, a low dose irradiation (less than 3 kGy) with mint or citrus flavors may enhance the safety of ice cream with proper sensory quality for sensitive consumer.
