Effect of Sawdust Culture on Oak Mushroom, Lentinula edodes (Berk.) Pegler by Inoculation of the Liquid Spawn (액체종균(液體種菌) 접종(接種)에 의한 표고톱밥재배(栽培) 효과(效果))

  • For cultivation on sawdust-bed of oak-mushroom until present time, inoculation of spawn on sawdust bed has been performed by sawdust spawn. But, liquid spawn may have advantages for rapid mass production of spawn, and now, sawdust-cultivation by liquid spawn inoculation should be applied instead of sawdust spawn. Therefore, investigations were performed to evaluate the effect of sawdust-cultivation by liquid spawn inoculation. The results were as follows: 1. When 11 kinds of liquid media were applied, the oak-mushroom culture medium was the most excellent in growth. Most suitable temperature at PDA was $25^{\circ}C$, and $22.5\sim27.5^{\circ}C$ in range were optimal for liquid culture. In liquid culture, amount of mycelial growth increases rapidly up to 40 days of cultivation. Incubation at fermentor brought yield of 106mg dry mycelia per 40ml media after 17 days. 2. In 1l-spawn bottle, growth of mycelium by inoculation of 20ml-liquid spawns were faster than 6g-sawdust spawn in spread of mycelia. On 2kg-bag culture, inoculations of 10ml-, 20ml- and 30ml-liquid spawns were all slower than 20g-sawdust spawn in mycelial spread. So, amount increasement in ampunt of liquid spawn should be discussed. Yields of mushrooms until third sproutings of 2kg-bag culture were 580g in 30ml-liquid spawn inoculation, but 510g, 486g and 470g from 20g-sawdust spawn, 20ml-liquid spawn and 10ml-liquid spawn, respectively. Thus, 30ml-liquid spawn inoculation was highest in yield.

Effect of Tree Species, Inoculation Amount and Inoculation Methods on Mycelium Growth and Sclerotia Formatino of Poria cocos Wolf (樹種, 接種量 및 接種方法이 茯笭 菌絲生長과 結笭에 미치는 영향)

  • When medicinal mushroom, Poria cocos, is cultured , inoculation method of spawn is cross slice inoculation of which the both sides of pine tree were peeled and spawn of P.cocos was inoculated. However, this method required lots of inoculation amount. This study was carried out to improve the culturing method of P. cocos. A good growth of P.cocos was observed in MCM(mushroom complete medium), showing proper mycelia growth and density. In inoculation amount, conventional method(cross slice inoculation) requires 20 bottles of spawn. In contrast, short log method required 8 bottles of spawn and drilling inoculation method 2~3 bottles, which could save by 60% and 85-90% respectively. In the selectrion of tree species, pine and larch had better condition for spawn culture and sclerotia formation condition.In terms of yield , pine was 33.7kg/3.3$m^2$. In the yield of pine, conventional method was 23.4kg/3.3$m^2$, drilling inoculation 29.4kg/3.3$m^2$, short log inoculation 31.7kg/3.3$m^2$, therefore drilling inoculation could increase by 25% and short log inoculation 35%, In addition, management cost was also saved.

Effects of Multipore Spawn Inoculation on Productivity of Lentinus edodes (Berk) Sing (다공 접종재배가 표고 생산성에 미치는 영향)

  • The effects of multipore spawn inoculation of Lentinus edodes on the mycelial growth stage and on the quantitative and qualitative yields of fruit-bodies for 5 years were investigated at natural environmental conditions. The spawn inoculation quantity did not affect the mycelial rooting at a range of 3.0kg-6.0kg per $m^3$ of bed-logs while the increase of spawn quantity by multipore inoculation increased the mycelial growth and the inner erosion of bed-logs, resulting in the increased yields of total fruit-bodies of L. edodes per $m^3$ of bed-logs for 5 years but resulting in the shortened major production period of fruit-bodies to the the first three years. Major production of fruit-bodies occurred during May-June and August-September, which was not affected by the spawn inoculation quantity. No significant differences in dry weight of a fruit-body were observed as increasing the spawn quantity by multipore inoculation; however the heaviest dry weight of a fruit-body was 1.63g at the spawn inoculation of 4.0kg per $m^3$/TEX> of bed-logs.

Comparison of Cultivation, Mushroom Yield, and Fruiting Body Characteristics of Lentinula edodes Strains according to the Inoculation Method

  • The cultivation in sawdust media, mushroom productivity, and fruiting body characteristics of Lentinula edodes strains NIFoS 2778 and NIFoS 3363 were compared according to the inoculation conditions. The cultivation period was 5% shorter when liquid spawn was used. Fruiting bodies were induced after 113 days of incubation on media inoculated with liquid spawn, and the cultivation period was 119 days on media inoculated with solid spawn. Mushroom productivity of NIFoS 2778 was the highest (661.4 g) when 36 mL of liquid spawn was used. For NIFoS 3363, mushroom production was higher under liquid inoculation conditions when the same amount of liquid and solid spawns were used. The mushroom characteristics of the two strains were not significantly different, except for gill width and stipe diameter.

Studies on Improvement of Artificial Cultivation and Antioxidative Activity of Poria cocos (복령의 인공 재배법 개선과 항산화활성에 관한 연구)

  • This study was carried out to improve an artificial culture techniques and antioxidative activity of the crude extract isolated from sclerotia of Poria cocos(Fr.) Wolf. In the test of different spawns and inoculation method, the sclerotia formation, number of sclerotia and production yield were excellent in the both sides inoculation method of log spawn, whereas the both side inoculation method of sawdust spawn was poor in sclerotia formation and yield. The optimal spawn and inoculation method for the quality and productivity of P. cocos was in the order of log spawn (both sides inoculation > log spawn(cutting section inoculation) > sclerotia (both sides inoculation) > sawdust spawn (both sides inoculation). The physiological activity substance, crude extract content of P. cocos NIAST 13007 was about 83%. As the concentration of crude extracts increased, the relative viscosity tended to be increased. However, as the concentration of sodium chloride increased, the relative viscosity did not affected. In antioxidative activities, electron donating ability (EDA) of P. cocos was about 10% of butylated hydroxytoluene (BHT). Thiobarbituric acid (TBA) value was similar to that of the vitamin C, however the peroxide value (POV) was lower than those of BHT and vitamin C.

Production of Pleurotus ostreatus and Flammulina velutipes Using Liquid Spawn Inoculation System (느타리 팽나무버섯 재배에서 액체종균 배양 및 접종시스템 적용방법의 구명)

  • This research aimed at developing the efficient method and device applicable to the inoculation of mushroom using (spawn was intended to utilize) liquid spawn for the stable production of mushroom. For the mass production of liquid culture, optimal inoculum volume and cultural period were $5{\sim}6%$ and 4 days for Flammulina velutipes and 4% and 5 days for Pleurotus ostreatus. Fruit body weight in 850 ml polythene bottle was highest at 6% or 15 ml liquid inoculum for P. ostreatus and $4{\sim}6%$ or $10{\sim}15ml$ for F. velutipes. Weight of fruit body by the application of liquid spawn inoculation system increased up to 33.7% for P. ostreatus and 32.8% for F. velutipes, respetively, compared to conventional spawn making system. The system of liquid spawn inoculation was successfully operated without malfunction in opening or closing the lid, and it took 26 min to inoculate 1200 bottles.

Condition of the most suitable inoculation and manufacture of spawn of ear mushroom (목이재배를 위한 배지선발 및 최적 접종조건)

  • Studies were made to optimize the media composition and cultural condition for mycelial growth of ear mushroom(Auricularia auricula). Sawdust spawn of media composition for optimal growth were found to be quercus sawdust combination of 20% rice bran and poplar sawdust combination of 10% wheat bran were the best of the optimal combination. The optimal condition of the most suitable inoculation amount showed that quercus spawn and poplar spawn were 15g, 25g, respectively.

The Analysis of Management and the Method of Cultivation of Lentinus edodes I. for Full-Development of Mycelium in Bed Logs (표고재배(栽培)의 관리분석(管理分析)과 종균활착(種菌活着)을 위한 골목관리(管理)에 관(關)한 연구(硏究))

  • This study was carried out to offer the successive method of cultivation and increase the productivity of mushroom yield with good quality through the elevation of rate of spawn development for Lentinus edodes. Studied about the analysis of current management of actural cultivation, a base of these, researched and presented for the upward method of productivity through an experiment of the high rate of spawn development and cultivation, putting first cultural environment. The results obtained were as follows ; 1. As the result of the analysis of current management in actural cultivation, many cultivators had a tendency to neglect managements of cultivation. These were reason for the deficiency of labour, funds and the lack of knowledge of cultivation, etc. 2. Water contents in bed logs according to the date of inoculation was shown as the decreasing order of 28.63%(3/12), 25.20%(3/25) and 23.19%(4/10). For the purpose of the maintenance of the water contents, the full-development of mycelium in bed logs and the dispersion of labour, the date of inoculation should be started in the early March. 3. The difference of the rate of spawn development among species was not shown, 100%(Mori 465). 98.98%(Mori 3046) on the spawn in high temperature and 98.97%(Mori 290) on the spawn in low temperature. The relative rate of spawn development was 97.70%(Mori 465), 82.45%(Mori 3046) on the spawn in high temperature and 88.87%(Mori 290) on the spawn in low temperature, it showed the difference. The spawn should be selected carefully in the future, as the spawn of cultivater's preference showed the difference for the development of mycelium. 4. The rate of spawn development following the date of inoculation was 100.0%(3/12), 98.98%(3/25) and 96.79%(4/10) on the spawn in high temperature and 99.09%(3/12), 98.97%(3/25) and 97.89% (4/10) in low temperature, it showed little difference. And the relative rate of spawn development was 97.70%(3/12), 82.45%(3/25) and 81.42%(4/10) on the spawn in high temperature and 93.27%(3/12), 89.67%(3/25) and 88.87%(4/10) that in low temperature, As the result of the relative rate, the time of inoculation of spawn should begin in the early March. 5. The height of stock logs on temporary placing should be less than 60cm at most on the surface, because of the low rate of water contents.

Response of Barley Genotypes to Fusarium Head Blight under Natural Infection and Artificial Inoculation Conditions

  • Forty-eight spring barley genotypes were evaluated for deoxynivalenol (DON) concentration under natural infection across 5 years at Harrington, Prince Edward Island. These genotypes were also evaluated for Fusarium head blight (FHB) severity and DON concentration under field nurseries with artificial inoculation of Fusarium graminearum by the grain spawn method across 2 years at Ottawa, Ontario, and one year at Hangzhou, China. Additionally, these genotypes were also evaluated for FHB severity under greenhouse conditions with artificial inoculation of F. graminearum by conidial suspension spray method across 3 years at Ottawa, Ontario. The objective of the study was to investigate if reactions of barley genotypes to artificial FHB inoculation correlate with reactions to natural FHB infection. DON concentration under natural infection was positively correlated with DON concentration (r = 0.47, P < 0.01) and FHB incidence (r = 0.56, P < 0.01) in the artificially inoculated nursery with grain spawn method. Therefore, the grain spawn method can be used to effectively screen for low DON. FHB severity, generated from greenhouse spray, however, was not correlated with DON concentration (r = 0.12, P > 0.05) under natural infection and it was not correlated with DON concentration (r = -0.23, P > 0.05) and FHB incidence (r = 0.19, P > 0.05) in the artificially inoculated nursery with grain spawn method. FHB severity, DON concentration, and yield were affected by year, genotype, and the genotype × year interaction. The effectiveness of greenhouse spray inoculation for indirect selection for low DON concentration requires further studies. Nine of the 48 genotypes were found to contain low DON under natural infection. Island barley had low DON and also had high yield.

Comparison of Productivity according to Different Quantity of Sawdust and Liquid Spawn for Sawdust Cultivation of Shiitake (표고톱밥재배시 톱밥종균과 액체종균의 접종량 차이에 따른 생산성 비교)

  • Studies were made to investigate the difference of the shiitake productivity according to the use of sawdust and liquid spawn on middle-temperature type strain. Treatments inoculated with sawdust spawn have shown that yields were dispersed according to flushing periods and fruit-bodies occurred sporadically on medium. However, treatments inoculated with liquid spawn have shown that yield was relatively concentrated at 1st flushing period and fruit-bodies were densely occurred on medium. At total yield and the number of fresh fruit-bodies until seventh flushing, treatments inoculated with liquid spawn were more than those inoculated with sawdust spawn. Also, biological efficiency (B.E.) of inoculation with 20 ml liquid spawn which produced 30% of the medium weight was highest as 69%, but biological efficiency of inoculation with 10 g sawdust spawn was lowest as 42%. And, although rate of fruit-bodies over 10 g in total yield was high on treatments inoculated with sawdust spawn, amounts of fruit-bodies over 10 g were not different among treatments.
