A Study to Establish the Core Evaluation Items of SP Authentication using ANP(Analytic Network Process) (ANP 모형을 이용한 SP 인증 핵심 평가항목 도출 연구)

  • Software process (SP) authentication is a certification system designed in order to improve software process and ensure software reliability of the Korean small and medium enterprises (SMEs). It is composed of 3 grades, 17 evaluation items and 70 detailed evaluation items (software process). In order to certificate grade 2, 42 detailed evaluation items must be satisfied. In order to certificate grade 3, all 70 detailed evaluation items must be satisfied. SMEs are more vulnerable to manpower and material resources than the major company. Therefore, it is difficult for SMEs to invest in the long term to improve the software process. If we can provide the core evaluation items to SMEs, SMEs will be able to improve software quality more efficiently and effectively. It can be used as a short way to acquire SP authentication. The evaluation items of SP are defined as influencing each other. Therefore, the core evaluation items might be established using related techniques. Although we searched for previous researches and related informations, the associated study was insufficient. In this study, ANP (Analytic Network Process) technique was applied, and the core evaluation items of SP authentication were established. SMEs will be able to acquire SP authentication more easily using the core evaluation items. Finally, we expect that this study will be used as a basis material for creating the SP 2.0 version.

Isolation and Characterization of White Rot Fungi for Decolorization of Several Synthetic Dyes (염료의 색도 제거능력이 우수한 백색부후균 분리 및 특성연구)

  • Several white-rot fungi collected from the mountains of Korea were evaluated for their ability to decolorize azo, polymeric, and reactive dyes. Strains CJ-105, CJ-212 and CJ-315, identified as Trametes sp., Pleurotus sp. and Fomes sp., respectively, showed higher potential for decolorization of those dyes in either solid or liquid media. For Trametes sp. CJ-105, 100ppm of Remazol Brilliant blue R and 500ppm of Acid Red 264 were completely decolorized after 2 days under liquid culture. The dominating ligninolytic enzyme existing in the culture broth was laccase (E.C. 1.10.3.2). Also, Pleurotus sp. CJ-212 and Fomes sp. CJ-315 showed similar patterns in decolorization of Remazol Brilliant Blue R and Acid Red 264. The extent of decolorization of the dyes in liquid culture was found to be proportional to the activities of the ligninolytic of decolorization of the dyes in liquid culture was found to be proportional to the activities of the ligninolytic enzymes produced by each strain. In addition to that Trametes sp. CJ-105 was highly effective in degradation of polycyclic aromatic hydrocarbons and pentachlorophenol by the activity of the ligninolytic enzymes produced. In this study, we found that white-rot fungi, Trametes sp. CJ-105(KFCC 10941), Pleurotus sp. CJ-212(KFCC 10943) and Fomes sp. CJ-315(KFCC 10942), were effective in decolorizing a wide range of structurally different synthetic dyes, as well as some chemical compounds which are known to be hardly degradable.

Genome Sequences of Spinach Deltapartitivirus 1, Spinach Amalgavirus 1, and Spinach Latent Virus Identified in Spinach Transcriptome

  • Complete genome sequences of three new plant RNA viruses, Spinach deltapartitivirus 1 (SpDPV1), Spinach amalgavirus 1 (SpAV1), and Spinach latent virus (SpLV), were identified from a spinach (Spinacia oleracea) transcriptome dataset. The RNA-dependent RNA polymerases (RdRps) of SpDPV1, SpAV1, and SpLV showed 72%, 53%, and 93% amino acid sequence identities with the homologous RdRp of the most closely related virus, respectively, suggesting that SpDPV1 and SpAV1 were novel viruses. Sequence similarity and phylogenetic analyses revealed that SpDPV1 belonged to the genus Deltapartitivirus of the family Partitiviridae, SpAV1 to the genus Amalgavirus of the family Amalgaviridae, and SpLV to the genus Ilarvirus of the family Bromoviridae. Based on the demarcation criteria, SpDPV1 and SpAV1 are considered as novel species of the genera Deltapartitivirus and Amalgavirus, respectively. This is the first report of these two viruses from spinach.

Occurrence of acid producing bacteria in Meju leaves (재래식 메주중의 산생성균의 분포)

  • The distribution of acid producing bacteria and general bacteria in 23 samples of Korean traditional Meju loaves was investigated and the strains isolated from the samples identified. The acid producing bacteria occurred more in outer part than inner part and anaerobic acid producing bacteria showed higher tendency of occurrence compared with the aerobes in each part. The average number of nonhalophilic and halotolerant bacteria belonging to aerobes were counted as $24{\times}10^6$ and $33{\times}10^5$ and the average number of those belonging to anaerobes $10{\times}10^7$ and $58{\times}10^5$ cells/g, respectively. The general bacteria isolated more in outer part than inner part and its average number was $62{\times}10^7$ cells/g. In the isolates, 2 aerobic acid producing strains were identified as Micrococcus spp., 3 anaerobic acid producing strains as Streptococcus sp., Pediococcus sp. and Lactobacillus sp., and 2 strains of aerobic general bacteria as Bacillus spp.

Possibility to Develope the Multistage Culture System for Larvae Cultivation by Computer Simulation (컴퓨터 시뮬레이션을 통한 치어 사육용 다단계 배양시스템의 개발 가능성 탐색)

  • The possibility for developing multistage culture system to cultivate larvae by computer simulation with basic experiments was investigated. This culture system was composed of 3 stages. At the 1st stage, Chlorella sp. were cultivated and at 2nd stage Chlorella sp. were supplied to rotifer (Brachionus plicatilis), and rotifer were supplied to larvae at the 3rd stage. In this study, Chlorella sp. were cultivated by batch culture to search for the possibility of continuous feeding to rotifer at 2 nd stage. The maximum specific growth rate ($\mu$$_{max}$) of Chlorella sp. at the logarithmic phase was 0.56 [1/day]. Rotifer was cultivated by fed-batch culture at the feeding rate of Chlorella sp., $10^3$, $10^4$, $10^{5}$ and $10^{6}$ [cells/rotiferㆍh] to search for the relation between the feeding rate of Chlorella sp. and the growth rate of rotifer. As the results, the minimum feeding rate of Chlorella sp. was 2.8 ${\times}$ $10^4$ [cells/rotiferㆍday] in the multistage culture system, then the change of rotifer concentration at 2nd stage was simulated by computer. The required amount of rotifer for the growth of larvae was also increased as the growth of larvae. On the 9th day of the culture, the rotifer uptake rate of larvae was 250 [cells/rotiferㆍday]. Based on these basic experiments and results, It was suggested that the possibility of multistage culture system to cultivate larvae with continuous feeding of Chlorella sp. and rotifer.r.

Production of Poly-3-Hydroxyalkanhoate by Haloarcular sp. EH-1 (Haloarcular sp. EH-1으로부터 생분해성 Poly-3-Hydroxyalkanoate의 생산)

  • The extremely halophilic archaebacterium Haloarcular sp. EH-1 was isolated from solar salts. Haloarcular sp. EH-1 accumulated poly(3-hydroxyalkanoate) (PHA) as intracellular granules. PHA production in batch culture have been studied. The PHA was identified as poly(3-hydroxybutyric acid-co-3-hydroxyvaleric acid) (PHB/HV) of 3-hydroxybutyric acid and 3-hydroxyvaleric acid by the analysis of GC, IR and NMR. The melting temperature of PHB/HV was 152.46$^{\circ}C$, viscosity was 1.25 ㎗/g, and molecular weight was $1.44 X 10^5.$

Characterization of PAH (Polycyclic Aromatic Hydrocarbon)-Degrading Bacteria Isolated from Commercial Gasoline (상용 휘발유로부터 분리한 다환 방향족 탄화수소(PAH) 분해 세균의 특성)

  • BACKGROUND: Recent studies have described the importance of bacteria that can degrade polycyclic aromatic hydrocarbons (PAHs). Here we screened bacterial isolates from commercial gasoline for PAH degraders and characterized their ability to degrade PAHs, lipids and proteins as well as their enantioselective epoxide hydrolase activity, salt tolerance, and seawater survival. METHODS AND RESULTS: One hundred two bacteria isolates from commercial gasoline were screened for PAH degraders by adding selected PAHs on to the surface of agar plates by the sublimation method. A clear zone was found only around the colonies of PAH degraders, which accounted for 13 isolates. These were identified as belonging to Bacillus sp., Brevibacterium sp., Micrococcus sp., Corynebacterium sp., Arthrobacter sp., and Gordonia sp. based on 16S rRNA sequences. Six isolates belonging to Corynebacterium sp., 3 of Micrococcus sp., Arthrobacter sp. S49, and Gordonia sp. H37 were lipid degraders. Arthrobacter sp. S49 was the only isolate showing high proteolytic activity. Among the PAH-degrading bacteria, Arthrobacter sp. S49, Brevibacterium sp. S47, Corynebacterium sp. SK20, and Gordonia sp. H37 showed enantioselective epoxide hydrolase activity with biocatalytic resolution of racemic styrene oxide. Among these, highest enantioselective hydrolysis activity was seen in Gordonia sp. H37. An intrinsic resistance to kanamycin was observed in most of the isolates and Corynebacterium sp. SK20 showed resistance to additional antibiotics such as tetracycline, ampicillin, and penicillin. CONCLUSION: Of the 13 PAH-degraders isolated from commercial gasoline, Arthrobacter sp. S49 showed the highest lipid and protein degrading activity along with high active epoxide hydrolase activity, which was the highest in Gordonia sp. H37. Our results suggest that bacteria from commercial gasoline may have the potential to degrade PAHs, lipids, and proteins, and may possess enantioselective epoxide hydrolase activity, high salt tolerance, and growth potential in seawater.

Distribution of Electrochemically Active Bacteria in the Sediment (Sediment에서의 전기활성 박테리아 분포 특성)

  • Microbial fuel cells (MFC) were enriched using sediment Nakdong river, Hoidong river and protected water area in Gijang. The microbial community of sediment and enriched MFC was analyzed by FISH (fluorescent in situ hybridization) and 16S rDNA sequencing. ${\alpha}$-Proteobacteria, Acidobacter and Cyanobactia group were dominant in sediment by FISH. The coulombs of the final 10 peak of the 3 MFC (Nakdong, Hoidong, Gijang) were 0.64 C, 0.50 C, 0.61 C, respectively. When MFCs were enriched by sediment, ${\beta}$-, ${\gamma}$-Proteobacteria, Acidobacter and Firmicutes group increased 45~90%, 50~90%, 40~80% and 45~125%, respectively. In results of 16S rDNA sequencing, Roseomonas sp., Azospillium sp., Frateuria sp., Dyella sp., Enterobacter sp. and Deinocossus were isolated from Nakdong river and Azospillium sp., Delftia sp., Ralstonia sp., Klebsiella sp. and Deinococcus sp. were isolated from protected water area in Gijang and Pseudomonas sp., Klebsiella sp., Deinococcus sp., Leifsonia sp. and Bacillus sp. were isolated from Hoidong river.

Construction of Expression Vector of Bacillus sp. SSA3 Strain (Bacillus sp. SSA3 균주의 Expression Vector 개발)

  • The promoter regions from chromosomal DNA of Bacillus sp. SSA3 which is responsible for fermentation of Korean traditional soy sauce, were cloned for construction of expression vector of Bacills sp. SSA3. Recombinant plasmids were constructed by insertion of HindIIl-cleaved Bacillus sp. SSA3 chromosomal DNA fragments in front of the CAT gene of pGR71 plasmid and B-galactosidase gene of pUC18 plasmid. 6 recombinant plasmids were isolated from chloramphenicol resistant E. coli JM109 clones. All these plasmids were found to have promoter activity in Bacills sp. SSA3 and E. coli JM109. When these 6 clones of Bacills sp. SSA3 were cultivated in LB agar medium supplemented with 10% NaCI. fused CAT gene expression of 4 clones was significantly decreased in common. But the others were poorly inhibited.

Evaluation of Soil Microbial Population of Paddy Fields in Gyeongnam Province Area (경남지역의 논토양에서 미생물의 다양성 평가)

  • To use as a fundamental data for the sustainable agriculture, which is nowadays a major trend to keep the productivity and conserve the environment, 487 paddy soil samples were collected from 21 regions of the Gyeongnam Province and analyzed the chemical characteristics and microbial population of the soil. The microbial population densities were bacteria $298{\times}10^5$($4{\sim}3000{\times}10^5$ range), fungi $63{\times}10^3$($2{\sim}441{\times}10^3$ range), actinomycetes $19{\times}10^5$($0.2{\sim}1250{\times}10^5$ range), Bacillus sp. $53{\times}10^4$($4{\sim}890{\times}10^4$ range) and Pseudomonas sp. $198{\times}10^4CFU\;g^{-1}$($4{\sim}1724{\times}10^4CFU\;g^{-1}$ range), respectively. The microbial populations of the soil were in general higher in southern area than in the northern area of the Gyeongnam Province. The average ratio of bacteria/fungi population was 473. As soil clay content increased, the populations of aerobic bacteria, actinomycetes and Pseudomonas sp. were remarkably decreased. The ratio of aerobic bacteria and fungi was 1554 in sandy loam and clay loam 1144, while Bacillus sp./fungi ratio was 11 in clay loam and 10 in loam. On the topographical differences, aerobic bacteria and Bacillus sp./fungi ratio were the higher in coastal plains than any other areas. The microbial population densities from different soil types were generally lower in ill-drained paddy field than those of other paddy field. The content of $P_2O_5$, K, Ca, $NO_3-N$ and EC in soil were positively correlated to the population densities of aerobic bacteria, actinomycetes, fungi, Bacillus sp. and Pseudomonas sp.. The soil organic matter and Mg content were also positively correlated to the population densities of aerobic bacteria, actinomycetes, fungi and Bacillus sp.
