Genotypic Variation in Leaf Water Status of Soybean

  • Plant water status during growth is directly and indirectly associated with seed yield. The objective of the present study was to determine the genotypic differences in leaf water characteristics at an early growth stage of soybean [Glycine max (L.) Merrill] plants through the pressure-bomb technique. Measurements of water potential as well as relative water content (RWC) were made at the third leaf from the fully-expanded top leaf of eight different soybean genotypes grown for 31 to 35 days after field emergence. On the basis of the modified exponential model, pressure-volume (PV) curves were fitted well ($R^2$=0.92** to 0.99** for the curvi-linear region and R=0.67** to 0.96** for the linear region), indicating that a segmented model using PROC NLIN of SAS could be used effectively to estimate the leaf water characteristics. The regression analysis for the pressure-volume (PV) curve revealed genotypic variation in the solute potential at saturation (Ψ$_{s,sat}$ :-10.7 to -14.8 bar), solute potential at incipient plasmolysis (Ψ$_{s,ip}$ : -14.3 to -18.3 bar), RWC at incipient plasmolysis (RW $C_{ip}$ : 83.3 to 91.7%), high integrated turgor pressure from saturation to plasmolysis ( $_1$$^{b}$ : 0.39 to 0.81), and maximum volumetric modulus of elasticity ($\varepsilon$$_{max}$ : 150 to 445 bar).).

콩 품종 교호재배가 광합성능력 및 수량에 미치는 영향 (Effects of Alternative Planting of Cultivars on Photosynthetic Ability and Yield in Soybean)

  • 초관높이와 도복저항성 차이가 있는 콩 두 품종의 교호재배가 광합성능력과 수량에 미치는 영향을 분석한 결과는 다음과 같았다. 1. 초관의 높이는 황금콩 1열+태광콩 1열에서 가장 높았으며, 도복은 황금콩 1열+태광콩 1열 및 황금콩 1열+태광콩 2열에서 경감 효과가 가장 컸다. 2. 교호재배에서 정엽 및 제 9본엽의 광합성능력은 태광콩열이 단일재배에 비하여 높았으며, 황금콩 정엽의 광합성능력은 낮았다. 3.수량은 황금콩 1열+태광콩 1열이 황금콩 단일재배에 비하여 $17\%$, 태광콩 단일재배에 비하여는 $8\%$증수되었다.

Genetics and Breeding for Modified Fatty Acid Profile in Soybean Seed Oil

  • Soybean [Glycine max(L.) Merr.] oil is versatile and used in many products. Modifying the fatty acid profile would make soy oil more functional in food and other products. The ideal oil with the most end uses would have saturates(palmitic + stearic acids) reduced from 15 to < 7%, oleic acid increased from 23 to > 55%, and linolenic acid reduced from 8 to < 3%. Reduced palmitic acid(16:0) is conditioned by three or more recessive alleles at the Fap locus. QTLs for reduced palmitic acid have mapped to linkage groups(LGs) A1, A2, B2, H, J, and L. Genes at the Fad locus control oleic acid content(18:1). Six QTLs($R^2$=4-25%) for increased 18:1 in N00-3350(50 to 60% 18:1) explained four to 25% of the phenotypic variation. M23, a Japanese mutant line with 40 to 50% 18:1 is controlled by a single recessive gene, ol. A candidate gene for FAD2-1A can be used in marker-assisted breeding for high 18:1 from M23. Low linolenic acid(18:3) is desirable in soy oil to reduce hydrogenation and trans-fat accumulation. Three independent recessive genes affecting omega-3 fatty acid desaturase enzyme activity are responsible for the lower 18:3 content in soybeans. Linolenic acid can be reduced from 8 to about 4, 2, and 1% from copies of one, two, or three genes, respectively. Using a candidate gene approach perfect markers for three microsomal omega-3 desaturase genes have been characterized and can readily be used in for marker assisted selection in breeding for low 18:3.

재래종대두의 결실일수군별과 제형질과의 관계 (Relationship between Fruiting Period and Agronomic Characters in Korean Local Soybean Collections)

  • The present study was conducted to evaluate agronomic characters of the soybean germplasm which was collected from the sites covering whole areas of southern Korean peninsula. A total of 2,748 Korean native soybeans was grouped for fruiting period and in this group frequency distribution of the important agronomic characters and relationships among the characters were determined. The results obtained were summarized as follows; The soybean germplasm was comprised of SeoulㆍKyonggi-do 7.5%, Kangwon-do 13.3%, Chungchongbuk-6.6%. Chungchongnam-do 12.3%, Chollabuk-do 10.7%, Chollanam-do 12.6%, Kyongsangbuk-do 20.8%, Kyongsangnam-do 15.3% and Cheju-do 0.6% in number of collections. Frequency distribution of the germplasm for fruiting period was 0.14% in Group I (below 55 days). 1.67% in Group II (56-60 days), 10.58% in (Group III(61-65 days), 23.18% in Group IV(66-70 days), 25.91% in Group V(71-75 days) 19.79% in Group Ⅵ(76-80 days), 15.1% in Group Ⅶ(81-85 days) and 3.6% in Group Ⅷ(over 85 days). The lines over 70 days in fruiting period were as much as 64.4% of germplasm. The fruiting period varied largely from 45 days to 91 days. Fruiting period was positively correlated to seed weight, days to maturity, days to flowering, plant height and number of branches per plant, respectively. While. it was negatively correlated to number of pods, lodging and virus infectivity, repectively, and in particular negative correlation was shown significantly between yield and fruiting period. Average yield was very high as much a 1.389kg/ha in fruiting period Group I, and decreased with increment of fruiting period, 1.400kg/ha in Group II. 1.384kg/ha in Group III. 1,299kg/ha in Group IV, 1,197kg/ha in Group V, 1,117kg/ha in Group Ⅵ, 967kg/ha in Group Ⅶ and 832kg/ha in Group Ⅷ.

대두 갈색무늬병에 의한 수량감소의 평가 (Assessment of the Soybean Yield Reduction due to Infection of Septoria Brown Spot, Septoria glycines Hemmi)

  • 2년간에 걸쳐 조사한 대두의 수량감소는 갈색무늬병을 접종한 경우 농약 살포구에 비해 $15.7-16.4\%$ 감소 차였으며 자연감염의 경우에는 $8.2-9.2\%$의 감소를 나타냈다. 갈색무늬병의 감염은 초장, 주당절수(株當節數), 주당분지수(分枝數)에는 큰 영향이 없었으나 주당협수(狹數)와 종실중(種實重)에는 현저한 감소 현상을 보였다. 대두의 수량감소율을 갈색무늬병의 이병율과 정(正)의 상관을 보였으며 그 상관도는 Van der Plank's infection rate (0.156) 보다는 병 진전국선면적(AUBC) 과의 상관도가 높았다$(r=0.388^*)$. 병진전곡선 면적으로 표시된 갈색무늬병의 감염율과 대두수량 감소율과의 관계는 y=4.38+0.5X $(r=0.686^{**},\;df=25)$으로 나타났다.

한국산 장류콩 종자 렉틴의 생화학적 특성 (Biochemical Properties of Seed Lectin from Korean Soybean Cultivars Developed for Soy Source)

  • 한국산 장류콩 종자로부터 최종적으로 Sephadex G-100상에서 분리한 lectin의 분자량, 적혈구 응집력, 온도, 열 안정성, 및 pH의 생화학적 특성을 조사하였다. 분리된 lectin을 SDS-PAGE한 결과, 분자량이 32 kDa와 22 kDa인 2개의 band가 나타났으며, Native PAGE에 의해서 108 kDa의 tetramer임을 알 수 있었다. Lectin은 trypsin을 처리한 사람의 A, B, AB, O 혈액과 쥐 혈액에서 혈구응집 반응이 일어나지 않았으나 토끼에서는 응집 반응이 일어났으며, trypsin을 처리하지 않은 모든 혈액에서는 응집반응이 일어나지 않았다. 이 lectin은 최적온도가 20-$50^{\circ}C$이며, 10-$60^{\circ}C$ 에서 열 안정성을 보였다. 최적 pH는 7.2로서 6.2 이하와 8.0 이상에서는 활성이 상설되었다.

Association Analysis of Charcoal Rot Disease Resistance in Soybean

  • In this research, the relationships among the 31 microsatellite markers with charcoal rot disease resistance related indices in 130 different soybean cultivars and lines were evaluated using association analysis based on the general linear model (GLM) and the mixed linear model (MLM) by the Structure and Tassel software. The results of microsatellite markers showed that the genetic structure of the studied population has three subpopulations (K=3) which the results of bar plat also confirmed it. In association analysis based on GLM and MLM models, 31 and 35 loci showed significant relationships with the evaluated traits, respectively, and confirmed considerable variation of the studied traits. The identified markers related to some of the studied traits were the same which can probably be due to pleiotropic effects or tight linkage among the genomic regions controlling these traits. Some of these relationships were including, the relationship between Sat_252 marker with amount of charcoal rot disease, Satt359, Satt190 and Sat_169 markers with number of microsclerota in stem, amount of charcoal rot disease and severity of charcoal rot disease, Sat_416 marker with number of microsclerota in stem and amount of charcoal rot disease and the Satt460 marker with number of microsclerota in stem and severity of charcoal rot disease. The results of this research and the linked microsatellite markers with the charcoal rot disease-related characteristics can be used to identify the suitable parents and to improve the soybean population in future breeding programs.

Variations of Yield Components and Anthocyanin Contents in Soritae and Yakkong Black Soybean Landraces Collected from Different Areas

  • In this study, we cultivated 115 Soritae and 86 Yakkong black soybean landraces collected from ten different locations in Korea. Then, the variations of three yield components (one-hundred seeds weight (HSW), number of seeds per pod (SPP), and yield per plant (YPP)) and three anthocyanins (cyanidin-3-O-glucoside (C-3-O-G), delphinidin-3-O-glucoside (D-3-O-G) and petunidin-3-O-glucoside (Pt-3-O-G)) were investigated according to landrace type and collection area. Both yield components and anthocyanin contents significantly varied between the soybeans demonstrating genetic differences. Soritae landraces had the highest average HSW and TAC, whereas Yakkong landraces displayed the highest average SPP irrespective of collection area. Relatively, Yakkong landraces from Gyeongsangnam-do (1697.29 mg/100 g) and Soritae landraces from Gyeonggi-do (2340.94 mg/100 g) had the highest average TAC. Principal component analysis clearly separated Soritae and Yakkong landraces. Moreover, TAC and C-3-O-G showed positive and significant associations in both Soritae (r = 0.972) and Yakkong (r = 0.885) landraces, while yield components showed negative or weak correlations with each other. Overall, ten landraces were identified as important resources owing to their high yield (>150 g/plant) and high level of TAC (>2300 mg/100g). This study could lay foundations to molecular level investigations and reinforce the use of Yakkong and Soritae landraces during cultivar development.

Assessment of environmental impact of vitamin A-enhanced soybeans and hybrid soybeans

  • An understanding of safety problems pursuant to environmental release of GM (Genetically Modified) crops is considered important. Among the recognized safety problems, the possibilities of weediness and ecosystem invasion are constantly being validated. We herein compared the growth characteristics and germination rate of soybeans formed by hybridization with vitamin A-enhanced soybeans carrying an introduced gene that increases β-carotene content. We also examined overwintering, survival, and weed competitiveness to evaluate hybrid ecological impact on long-term unmanaged cultivatable land. These studies revealed that the hybrid soybeans exhibited intermediate growth characteristics and germination rate compared with the vitamin A-enhanced soybeans and wild soybeans, or exhibited traits similar to those of the maternal strain. Overwintering experiments were conducted by planting seeds at depths of 0, 5, 10, and 20 cm and recovering them after three or five months. After five months, all seeds at depths more than 5 cm lost viability. Among seeds recovered after three months, only wild soybeans retained viability at depths of more than 5 cm. Survival and weed competitiveness were assessed by sowing each type of seed and performing no irrigation, or pest or weed control. Quantitative assessment of numbers of individual soybean plants that appeared in the experimental plot revealed that all plants germinated after sowing, but only wild type plants survived overwintering. These studies suggest that both GM soybeans and hybrid soybeans cannot survive in uncultivated land even if they are released into the environment, which indicates less possibility of ecosystem invasion and weediness.

장류용 내병 내재해 기계수확 적응 콩 신품종 '남풍' (A New Soy-paste Soybean Cultivar, 'Nampung' with Disease Resistance, Good Combining Adaptability and High Yielding)

  • '남풍'은 기계수확이 가능한 다수성 콩 신품종 육성을 목표로 소분지 내재해 다수성 '수원190호'를 모본으로 하고 대립 다수성 '보광콩'을 부본으로 1997년도에 인공교배하고 세대단축 기간을 거쳐 선발 육성한 기계수확 적성이 우수하고 내병내재해 다수성 품종으로 품종보호권 등록번호는 2660이며 주요특성을 요약하면 다음과 같다.1. 유한신육형으로 화색은 백색이고 엽형은 피침형이며 모용색은 갈색이다. 종자모양은 구형이고 종피색은 황색이며 배꼽색은 연한갈색이다. 2. 성숙기는 10월 2일로서 '태광콩'과 같으나 개화기가 빨라 등숙기간이 5일이 길다. 경장은 58 cm로서 '태광콩'과 비슷하고 도복에 강하며, 100립중은 19.9 g으로 '태광콩'에 비하여 2.6 g 정도 가벼운 중립종이다. 3. 불마름병에 강하고 콩 바이러스 접종시 모자이크나 괴저반응을 보이지 않았으며, 내습성과 내한발성, 뿌리썩음병 및 종실 이병립율은 '태광콩'과 비슷하였다. 착협 위치가 평균 19.6 cm로서 높고 분지수가 적으며 내탈립성이 강하여 기계수확에 적합한 품종이다. 4. 조단백질 및 조지방 함량은 '태광콩'과 비슷하였으나, 불포화지방산 비율과 총 아이소플라본 함량은 '태광콩'에 비해 다소 높았다. 메주 및 청국장 수율이 '태광콩'보다 다소 높았고 발효정도가 '태광콩'보다 양호하였다. 5. 수량성은 생산력검정시험 결과 '태광콩'에 비하여 23%증수되었으며, 3개년간 실시한 지역적응시험에서 중남부 이모작 6개소 평균 ha당 2.97톤으로서 표준품종인 '태광콩'보다 21% 증수하였다.
